引导高校网络青年自组织走向规范的对策

Abstract :

高校网络青年自组织的生成是内在动因和外部因素综合作用的结果,具有组织的自发性、成员关系的非强制性,沟通方式的特殊性,以及价值内核的公共性等特征,除了发挥正向效能外,还兼具诱发青年网瘾的多米诺骨牌效应,消解规范约束导致青年道德行为失范,使得高校组织与网络自组织两个目标发生冲突等负向影响。文章分析高校网络青年自组织的结构规制和文化规制,提出发挥共青团积极引导作用,引导其走向规范。

Abstract :

Nostoc flagelliforme is a terrestrial cyanobacterium species whose metabolism follows an obvious diurnal pattern. Diurnal changes at physiological and proteomic levels of N. flagelliforme were obtained. In the morning (7:00 H), net photosynthesis, dark respiration, as well as the activities of total Rubisco, nitrogenase, glutamine synthetase, SOD and CAT were comparatively high. All these physiological activities significantly decreased in the afternoon (13:00 H), and then slowly increased in the evening (19:00 H). Thirty-one differentially expressed proteins with a variety of important functions were reproducibly detected and identified over a diurnal cycle. These proteins were categorized according to their predicted functions into secretion and regulation (15.79 %), antioxidative processes (21.05 %), nitrogen metabolism (10.53 %), carbohydrate and energy metabolism (10.53 %), as well as cell division (2.63 %). The remaining proteins had unclassified/unknown functions (21.05 %) or were unidentified (18.42 %). The results suggested a metabolic shift from active (7:00 H) to quiescent (13:00 H) and then to active (19:00 H) over the diurnal cycle. The differential expression level of ferritin, Mn-CAT, SOD and Fe-SOD may serve as molecular markers for the diurnal metabolism in N. flagelliforme.

Abstract :

Kandelia candel is being established as a model xylophyte for ecoadaptation due to its salt tolerance. To adapt to high salinity, the photosynthesis apparatus must function efficiently under these conditions. Proteomic analysis of chloroplasts isolated from plants under different degrees of salt stress was performed to quantify the changes of individual proteins and to gain a global view of the total chloroplast protein dynamics. Among the 1030 proteins quantified (unique peptide >= 1), 76 showed a more than 1.5-fold change in abundance, of which 36 are involved in the light-dependent reactions and 12 in the Calvin cycle. The dynamic change of these proteins indicates that light-dependent reactions are maintained by up-regulating the levels of component proteins at both moderate and high salinity, and the Calvin cycle remained functional at moderate salinity but showed a decline at high salinity. In addition to proteins related to photosynthesis, some known abiotic-stress proteins and plastoglobuli were up-regulated in salt-stressed plants. Plastoglobuli might contribute to maintaining membrane integrity and fluidity. In conclusion, this extensive proteomic investigation on intact chloroplasts of the salt-tolerant xylophyte under salt stress provides some important novel information on adaptative mechanisms involving photosynthesis in responses to salt stress in K. candel.

Abstract :

Nostoc flagelliforme must undergo a dehydration/rehydration cycle during its growth stages; the mechanisms underlying this constraint are examined. The novel insights into N. flagelliforme's response to desiccation and rehydration at ultrastructural, physiological and proteomic levels were offered. The structure of colonies and cells remained unchanged in response to dehydration and rehydration treatments except that the sheath appeared shrunken, and both the quantity and volume of vacuoles were decreased when dehydrated compared with rehydration. A significant increase in photosynthesis, respiration, total Rubisco activity, superoxide anion level, SOD, CAT, POD, nitrogenase and glutamine synthetase (GS) activities in response to rehydration was noted, whereas H2O2, ammonium, proline and glutamate contents all registered a decrease. 32 differentially expressed proteins between dehydrated and rehydrated colonies were categorized according to their predicted functions into secretion, signaling, transcription and translation, antioxidative processes, nitrogen metabolism, energy metabolism, lipid metabolism and chaperonin. The dehydration is a quiescent state in which metabolism is down-regulated, upon rehydration, a metabolic shift occurs from quiescent to active. The specific metabolic and regulated mechanisms to accommodate the dehydration/rehydration cycle in N. flagelliforme is reported here. (C) 2012 Elsevier B.V. All rights reserved.

Abstract :

Two-dimensional gel electrophoresis (2-DE) was used to analyze the proteins related to floral reversion in Dimocarpus longan Lour. Proteins were extracted from buds undergoing the normal process of flowering and from those undergoing floral reversion in three developing stages in D. longan. Differentially expressed proteins were identified from the gels after 2-DE analysis, which were confirmed using matrix-assisted laser desorption/ionization-time of flying-mass spectroscopy and protein database search. A total of 39 proteins, including 18 up-regulated and 21 down-regulated proteins, were classified into different categories, such as energy and substance metabolism, protein translation, secondary metabolism, phytohormone, cytoskeleton structure, regulation, and stress tolerance. Among these, the largest functional class was associated with primary metabolism. Down-regulated proteins were involved in photosynthesis, transcription, and translation, whereas up-regulated proteins were involved in respiration. Decreased flavonoid synthesis and up-regulated GA20ox might be involved in the floral reversion process. Up-regulated 14-3-3 proteins played a role in the regulation of floral reversion in D. longan by responding to abiotic stress. Observations via transmission electron microscopy revealed the ultrastructure changes in shedding buds undergoing floral reversion. Overall, the results provided insights into the molecular basis for the floral reversion mechanism in D. longan. (C) 2011 Elsevier B.V. All rights reserved.

Abstract :

To establish a suitable and effective protocol of protein extraction for two-dimensional gel electrophoresis (2-DE) analysis in kenaf leaf tissues, three extraction methods (trichloroacetic acid/acetone, urea/thiourea, and phenol extraction methods) were applied to the extraction of kenaf leaf protein. The results were compared in regard to protein extraction efficiency, sodiumdodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), and 2-DE gels. Furthermore, the 2-DE system was optimized for four aspects: the pH range of IPG (immobilized gradient) stripes, sampling methods, sample volumes, and concentration of polyacrylamide gels. The data presented showed that the phenol extraction method is the best method to perform 2-DE analysis of kenaf leaf protein. The protein extracted from phenol extraction method reached the purity of (26.40 +/- 0.859)%, showed (25.67 +/- 1.53) protein bands in one dimension SDS-PAGE gels, and (1 374 +/- 54.44) protein spots on 2-DE gels. The research also indicates that kenaf leaf protein spots were distributed mainly within the pH range of 4-8. More clear background with a better distribution effect and many protein spots could be obtained on 2-DE gels under the conditions of active rehydration loading, 24 cm IPG strips (linear pH gradient of 4-7), 1.4 mg samples, and 12% SDS-PAGE gels.

Two-dimensional gel electrophoresis (2-DE), coupled with mass spectroscopy, was used to study seed abortion in Dimocarpus longan Lour. (cv. Minjiao 64-1) by comparing normal and aborted seeds at three developmental stages. More than 1,000 protein spots were reproducibly detected in 2-DE gels, with 43 protein spots being significantly altered in their intensity between normal and aborted seeds at least at one stage. Thirty-five proteins were identified by matrix-assisted laser desorption ionization-time of flight-tandem mass spectrometry (MALDI-TOF-MS/MS) analysis and protein database searching. Most of the identified proteins were associated with a variety of functions, including energy and metabolism (30%), programed cell death (9%), antioxidative processes (14%), chaperonin (23%), cell division, amino acid metabolism, secondary metabolism, and other functional classes. Furthermore, the expression patterns of HSP70 and cytosolic ascorbate peroxidase (cAPX) were validated by immunoblotting analysis. This study provides a novel, global insight into proteomic differences between normal and aborted seeds in longan. We anticipate that identification of the differentially expressed proteins may lead to a better understanding of the molecular basis for seed abortion in longan.

A proteomic approach was taken to compare the proteomes of normal flowering buds and flowering reversion buds in longan (Dimocarpus longan Lour. cv. Longyou). Two-dimensional gel electrophoresis (2-DE), coupled with mass spectroscopy and protein database searching, recognized 18 proteins that were differentially expressed in flowering reversion buds. Eleven of these were down-regulated, whereas seven were up-regulated. A subset of 13 proteins was identified by MALDI-TOF-TOF/MS and classified into regulatory proteins (kinase, 20S proteasome alpha 6 Subunit, putative alpha 7 proteasome subunit, auxin-induced protein, and abscisic stress ripening-like protein), antioxidant-related proteins (Chain A, GDP-mannose-3',5'-epimerase, putative lactoylglutathione lyase, and Chain A, ascorbate peroxidase), pollen fertility-related proteins (putative leucoanthocyanidin reductase 2, and putative isoflavone reductase), photosynthesis-related proteins (large subunit, ribulose-bisphosphate carboxylase-oxygenase), and molecular chaperones (disulfide isomerase). Among them, regulatory and antioxidant-related proteins accounted for almost two-thirds of these proteins, suggesting that they may play a more important role in bud differentiation. Identification of these proteins provides insights that may lead to a better understanding of the molecular basis for flowering reversion in longan. (C) 2009 Elsevier B.V. All rights reserved.
