Control efficacy and persistence of fluopyram dust against pine wilt disease SCIE

Abstract :

The effectiveness of fluopyram suspension concentrate against pine wilt disease (PWD) is limited by spraying efficiency and water dependence. A traditional dust formulation with strong dispersibility can overcome these shortcomings. However, its efficacy against PWD remains uncertain. This study evaluated the translocation of fluopyram dust within tree tissues, soil and water degradation, and its effective control against PWD. Nursery tests showed effective prevention; field tests showed dust absorption and translocation into pine tissues. Thirty days following application, residual concentrations in soil were low at 0.09 mg kg-1; no detectable residues were found in water samples. Three years after applying fluopyram, its effectiveness increased to approximately 87%. Based on this study, fluopyram had a half-life of 346 d with persistence lasting up to three years. This provides valuable insight for managing PWD through dust applications.

Abstract :

Cryptomeria japonica var. sinensis is a valuable timber and ecological tree species in East Asia. Despite previous research that has revealed significant differences in tree height, diameter at breast height, and wood volume among C. japonica var. sinensis from different geographical sources, their genetic structure and phylogenetic relationship among different populations are still unable to accurately determined. Therefore, we extensively collected different geographical sources of Cryptomeria spp. in China (Fujian, Zhejiang area) and Japan, and performed their genetic structure and affinities by using simplified genome sequencing (GBS). A total of 306.5 Gb of sequence raw data and 875,329 high-quality SNPs were obtained, with an average of 4.26 Gb of raw data per sample. The results showed that the genetic variation of C. japonica var. sinensis populations was lower than that of C. japonica populations, and the genetic diversity of C. japonica var. sinensis in Fujian populations was slightly higher than that of Zhejiang populations. At the same time, molecular analysis of variance showed that the genetic variation of these two populations mainly came from within the population (99.6%). Furthermore, the cluster analysis results showed that C. japonica var. sinensis was clustered into five groups, and the clustering results were still consistent with the actual geographical distribution. All findings clarified the genetic background and structure of Cryptomeria spp. This will be a reference for the conservation, utilization, and genetic breeding improvement of C. japonica var. sinensis germplasm resources.

Sugar-inducible promoters mitigate the fitness cost of engineered Serratia marcescens in the control of Monochamus alternatus SCIE

Abstract :

Pine wilt disease is a devastating disorder of pine trees, vectored and transmitted by Monochamus alternatus Hope. However, vector control strategies for this disease are hampered by the resistance mechanisms of the pathogen, which hinders the long-term survival of control agents in nature. Serratia marcescens, isolated from Pinus massoniana and the gut of M. alternatus, was selected as the carrier strain for engineered bacteria in this study owing to its symbiotic nature. To realize the possibility of the long-term survival of transgenic bacteria in the gut of M. alternatus and the wild, four sugar-inducible promoters were used to construct transgenic strains. The S. marcescens strains with sugar-inducible promoters exhibited higher fitness than the constitutive strain expressing the Cry3Aa-T toxin, while maintaining a growth curve similar to that of the wild-type strain. The conditional expression of the Cry3Aa-T toxin in S. marcescens was triggered by the presence of sugar, and the lactose-induced transgenic symbiotic strain significantly reduced the survival rate of M. alternatus. Thus, this strategy may enable the engineered bacteria to effectively colonize and spread in the wild, indicating a feasible and sustainable vector control approach against M. alternatus.

Transcriptomic and metabolomic insights into pine wood nematode resistance mechanisms in Pinus massoniana SCIE

Abstract :

Pine wilt disease, caused by the pine wood nematode (PWN), is a devastating systemic disease with significantly impacts on pine species, particularly Pinus massoniana (Masson pine) in South China. This study integrated transcriptomic and metabolomic analyses to identify differentially expressed genes (DEGs) and differentially accumulated metabolites associated with PWN resistance. By comparing the gene expression and metabolic profiles of healthy, mechanically wounded and PWN-infected Masson pine trees at 28 days post-inoculation, we identified that 1310 DEGs were specifically associated with PWN infection after excluding mechanical damage effects. Notably, combined KEGG analysis of transcriptomic and metabolomic data revealed significant enrichment of the alpha-linolenic acid metabolism pathway. Within this pathway, genes such as AOS, LCAT3 and DAD1 exhibited differential expression patterns, highlighting its pivotal role in PWN resistance. Metabolomic analysis revealed that key genes involved in jasmonic acid (JA) biosynthesis and plant hormone signaling showing strong regulation. Additionally, Quantitative Real-Time PCR (qRT-PCR) validation of selected DEGs confirmed the expression patterns observed in the transcriptomic data. Physiological assays also validated changes in key hormone levels, such as JA and methyl jasmonate, which are upregulated in the early stages of plant infection. These results highlight the importance of JA-mediated defence responses and provide novel insights for breeding strategies to improve P. massoniana's resistance to PWN infection.

融合中心损失和焦点损失的蝴蝶自动识别

【目的】针对蝴蝶样本存在类间和类内分布不平衡导致识别性能下降的问题,探索一种多损失融合的蝴蝶自动识别方法。【方法】利用开源的Butterfly-200图像数据集作为实验数据。该数据集包括200种蝴蝶,每种蝴蝶的图像数量从30~885不等。以交叉熵损失(cross-entropy loss)为基准损失,分别叠加对比损失(contrastive loss)、焦点损失(focal loss)、类平衡损失(class-balanced loss)、采样(sampling)、logit调整(logit adjustment),比较算法的识别性能。在此基础上,利用中心损失(center loss)有助于缓解类内不平衡而焦点损失有助于缓解类内和类间不平衡的特点,开展消融实验分析叠加中心损失和焦点损失对识别性能的影响,提出了融合上述这两种损失的蝴蝶自动识别新方法。【结果】交叉熵损失与其他单一损失(对比损失除外)结合时,算法的识别性能基本上呈现不同程度的下降。我们的算法在交叉熵损失基础上结合中心损失和焦点损失后,其识别性能均超过交叉熵损失及其与其他损失的组合,准确率、F1分值、查准率和召回率分别91.67%, 90.68%, 91.68%和90.38%。消融试验进一步证实了中心损失和焦点损失的互补性,同时使用这两种损失能明显提升识别性能。此外,不同权重的损失组合对识别性能也有明显影响。【结论】研究结果证明融合中心损失和焦点损失在一定程度上缓解了类间和类内分布不均衡的问题,能够有效提高蝴蝶识别的准确性,为生态环境监测提供了一种有效的辅助手段。

Herbivore-induced plant volatiles (HIPVs) play a pivotal role in mediating tritrophic interactions between plants, herbivores, and their natural enemies. Paracarophenax alternatus, a parasitic mite targeting the egg stage of Monochamus alternatus, has emerged as a promising biocontrol agent. However, its ability to detect Pinus massoniana-derived HIPVs for host insect localization remains unclear. G-protein-coupled receptors (GPCRs) may play a role in mediating the perception of HIPVs and associated chemosensory signaling pathways in mites. In this study, a total of 85 GPCRs were identified from P. alternatus. All GPCRs exhibited conserved transmembrane domains and stage-specific expression patterns, with 21 receptors significantly upregulated in viviparous mites. Combined with two previously identified odorant receptors (ORs), six candidate chemosensory receptors were selected for molecular dynamics simulations to validate their binding stability with key volatile compounds. The results demonstrate that specific GPCRs likely facilitate HIPV detection in mites, enabling precise host localization within dynamic ecological niches. Our findings provide critical insights into the molecular basis of mite-host interactions and establish a framework for optimizing P. alternatus-based biocontrol strategies against pine wilt disease vectors.

Phototaxis Characteristics of Lymantria xylina (Lepidoptera: Erebidae) SCIE

Abstract :

Lymantria xylina Swinhoe (Lepidoptera: Erebidae) is considered a potentially internationally invasive forest pest with limited knowledge about its phototactic behavior. This study investigated the phototaxis of L. xylina males and females using various insecticidal lamps in the field. The results showed that all lamps attracted both males and females, but females were captured in a very low numbers, with a female-to-male ratio of 1:322. The insecticidal lamp with a peak wavelength of 363 nm was most effective for male trapping. Males exhibited a distinct light-trapping rhythm, peaking around midnight (23:00-0:00), with 29.5% capture, while females were most active at dusk (19:00-20:00), with 44.4% capture. Light-trapped females were smaller and lighter than indoor-emerged females and had lower egg-carrying capacity. Females, when laying eggs, did not exhibit phototactic behavior. Only 14.6% of non-ovipositing females showed phototactic behavior, and only 0.6% flew directly toward the lamp. These findings suggest that while non-ovipositing females can exhibit phototaxis, only a small proportion will, potentially reducing the risk of long-distance dispersal of L. xylina via ocean-going freighters. The results showed that the non-ovipositing females could fly under phototaxis, but their phototaxis was limited. This study provides a basis for the risk assessment of the long-distance dispersal of L. xylina via ocean-going freighters through female moth phototactic flight.

[Objective] Paracarophenax alternatus is a newly identified parasitic mite exhibiting parasitic efficacy against Monochamus alternatus eggs. This study aims to investigate the effects of environmental factors on the parasitism efficiency of P. alternatus, to evaluate its potential in biological control programs, to optimize artificial rearing systems, and to refine field application strategies.[Method] An indoor feeding experiment under controlled laboratory conditions was conducted to assess the impacts of M. alternatus egg age (0-4 days post-oviposition), mite-to-egg ratios (1∶1-8∶1), temperature (17-33 ℃), relative humidity (RH: 60%-95%), and photoperiod (24L, 12L∶12D, 24D) on the parasitism rates of P. alternatus. A predictive model was developed using random forest regression analysis to quantify parasitism rates under varying environmental conditions. [Result]1) Under the conditions (mite-to-egg ratio 5∶1, 25°C, 85% RH, 12L∶12D), the parasitism rate of P. alternatus on newly laid eggs of M. alternatus was the highest, reaching 74.44%, significantly exceeded the parasitism rate of 44.44% on 4-day-old eggs of M. alternatus P2 = 0.83), and predicted the maximum parasitism rates (>90%) under conditions of freshly laid eggs, 5∶1 mite-to-egg ratio, 20-31 ℃, RH >93%, and 24D photoperiod.[Conclusion]Egg age, temperature, humidity, photoperiod, and mite-to-egg ratio significantly influence the parasitism efficiency of P. alternatus on the eggs of M. alternatu. For optimal biological control or mass rearing, conditions should include mite-to-egg ratios ≥5∶1, temperatures of 21-29 ℃, RH ≥90%, and complete darkness. These findings provide critical insights for optimizing mass-rearing protocols and field deployment strategies to enhance the efficacy of P. alternatus in suppressing M. alternatus populations. © 2025 Chinese Society of Forestry.
