土壤含水量对玫瑰茄花青素含量及DFR基因表达水平的影响

Abstract :

【目的】分析土壤含水量对玫瑰茄萼片花青素含量以及二氢黄酮醇-4-还原酶(DFR)基因表达水平的影响,为玫瑰茄栽培时的水分管理以及进一步明确DFR基因在花青素合成中的功能提供参考。【方法】以玫瑰茄品种‘广东早熟’为试验材料,分别设置了33%(极度干旱)、55%(轻度干旱)、80%(渍涝)3个土壤含水量处理,分别测定各处理下开花后10(前期)、20(中期)、30 d(后期)玫瑰茄萼片中花青素的含量以及5个DFR基因的表达量,并对花青素含量与DFR基因表达量的相关性进行了分析。【结果】(1)不同土壤含水量下,随着开花时间的推移,玫瑰茄萼片花青素含量均呈下降趋势,且开花前期与开花后期的差异显著;含水量为80%时,花青素含量下降幅度最大;开花前、中期,土壤含水量为33%的花青素含量明显低于土壤含水量为55%、80%的含量;开花后期,3个土壤含水量处理的花青素含量差异不显著。(2)不同土壤含水量下,DFR基因的表达量随着开花时间的推移呈现不同的变化趋势。土壤含水量为33%时,5个DFR基因的表达量高于其他两个含水量处理,且在开花后30 d显著上调,达到最高值,其中,DFR4表达量最高,DFR5表达量最低;土壤含水量为55%、80%时,5个DFR基因的表达量均较低。(3)土壤含水量为55%时,DFR4、DFR5表达量与花青素含量显著相关,表明其为调控花青素合成的关键基因。【结论】严重的干旱和渍涝都不利于玫瑰茄萼片花青素的合成;干旱胁迫促进了DFR基因的表达,而渍涝情况下DFR基因的表达受到了抑制;DFR4、DFR5基因为调控玫瑰茄花青素合成的关键基因。

Crop sample prediction and early mapping based on historical data: Exploration of an explainable FKAN framework SCIE

Abstract :

Accurate and timely crop mapping is essential for food security assessment, and high-quality feature factors are the core foundation for accurate mapping. However, deep learning model crop classification algorithms have achieved some success, while the models themselves struggle to explain the specific contribution and impact of different features on the results. In this study, a self-adaptive Feature-attention Kolmogorov-Arnold Network (FKAN) is proposed for interpretable and scalable crop mapping. The model integrated the adaptive weighted feature attention module (AWFA) and the interpretable KAN network, which can visualize the complex associations between features and target crops and automatically capture and filter effective key spatiotemporal features, thus enhancing the interpretability of the model. Experimental results demonstrate that integrating optical, radar, and terrain features yields superior performance in both sample prediction and crop mapping, surpassing existing methods. The proposed FKAN achieves an overall accuracy and F1 score exceeding 0.90. Optical and radar features contribute the most significantly to classification accuracy, while terrain data provides complementary enhancement. By aligning with key crop phenology and leveraging the Google Earth Engine (GEE), FKAN establishes the first operational platform for global winter wheat identification, enabling accurate and scalable crop mapping. The migrated model achieves over 85% accuracy across different regions and years, demonstrating strong robustness and generalization capability. The study identifies optimal phenological periods and feature indices for different crops, providing scientific guidance for future mapping efforts. The FKAN model demonstrated robustness, scalability, and interpretability, was able to automatically extract high-confidence pixels and generate crop planting probabilities, providing an efficient and scalable solution for large-scale crop monitoring. This study generated the first global winter wheat map GlobalWinterWheat10m dataset by the FKAN algorithm. The code and demo link is accessible at https://github.com/FZUcheng123/FKAN.

水稻籼粳交后代茎秆抗倒性状的遗传效应分析

Abstract :

水稻抗倒伏性状是一个复杂性状,剖析其遗传效应,对提高水稻抗倒伏能力具有参考价值。本文旨在通过植物数量性状的加性-显性遗传模型,探讨水稻抗倒伏性状的遗传效应。以籼粳杂交后代为研究对象,分析了16个抗倒伏性状的遗传方式和遗传参数。研究发现,9个性状的加性方差占遗传主效应比率变幅为49.9%~72.1%,7个性状的显性方差占遗传主效应比率变幅为48.2%~87.1%,9个性状的加性方差和显性方差占表现型方差均达显著或极显著水平,受基因加性效应和显性效应共同控制。基部抗折力、鞘叶厚度等8个性状与倒伏指数呈负相关,有利于提高基部抗折力和水稻抗倒能力。籼粳中间型亲本的抗倒伏性状杂种优势强,育种价值高,有效利用各性状的遗传特点,对培育抗倒伏的籼粳杂种具有重要指导意义。

【目的】对新育成的黑米两系杂交稻紫两优7206进行营养品质分析,为黑稻良种选育及功能食品研发提供参考价值。【方法】以超级稻宜优673作对照,分析紫两优7206与宜优673的氨基酸、脂肪酸、花色苷和总黄酮等含量差异,以及总抗氧化能力与清除自由基能力的差异。【结果】紫两优7206与宜优673在水分、灰分和脂肪含量方面无明显差异,均检测出17种氨基酸,8种脂肪酸。紫两优7206的必需氨基酸含量高于FAO提出的必需氨基酸模式,其蛋白质含量、总氨基酸与必需氨基酸含量分别比宜优673高7.33%、11.08%和9.71%。紫两优7206比宜优673具有较高的各呈味氨基酸TAV值,丰富了黑米食味品质。2种稻米的饱和脂肪酸(SFA)、单不饱和脂肪酸(MUFA)和多不饱和脂肪酸(PUFA)比值比较接近中国营养协会推荐的“<1∶1∶1”的比例,且均含有人体必需脂肪酸亚油酸与亚麻酸。紫两优7206的花色苷、总黄酮含量,以及总抗氧化能力与清除自由基能力均极显著优于宜优673。【结论】杂交配组选育的紫两优7206较宜优673具有更丰富的营养价值,对功能性黑稻的培育与食品研发具有一定参考价值。

[目的]选育优良的光温敏两系不育系,以提高其杂种后代的产量和品质。[方法]采用光温敏两系不育系‘RGD-7S’为母本与三系保持系‘中浙B’杂交,经5年9代的系谱法选育,育成不育性稳定、稻米品质优、异交特性好、株叶形态优良的光温敏两系不育系‘稆S’,并分析了其主要特征特性及杂种优势。[结果]‘稆S’不育性稳定,育性转换临界温度<23℃,稳定不育期127 d左右。该不育系制种产量高,异交特性好,柱头外露率78.62%,其中双边外露率39.74%。采用‘稆S’配组的高产、优质两系杂交稻组合‘稆两优质美’先后通过了福建省和海南省农作物品种审定。[结论]光温敏两系不育系‘稆S’不育性稳定,育性转换临界温度低,稻米品质优,配合力强,应用前景广泛。

优质抗病糯稻种质的性状鉴定及其杂种优势利用

【目的】为有效改良传统糯稻品种的产量、品质及抗性等综合性状,满足当前专用型糯稻市场的发展需求。【方法】采用不完全双列杂交(NC II)方法,构建亲本及20个杂种一代的遗传群体,评价糯稻亲本及杂交组合产量和品质性状的育种利用价值,鉴定出高产、抗病、低直链淀粉含量、高碱消值的糯稻新种质。【结果】7个优良品系的实割产量变幅7 744.35~9 216.45 kg·hm~(-2),其中X-104实割产量最高,达9 216.45 kg·hm~(-2),X-27实割产量达8 960.40 kg·hm~(-2),明显优于3个对照;两个株系植株矮壮,且分别携带了Pi-1、Pi-k~h和Pi-1、Pi-9、Pi-k~h的稻瘟病抗性基因,表现为中抗稻瘟病;直链淀粉含量(amylose content, AC)分别为1.71%和1.68%;碱消值(alkali spreading value,ASV)达6.75级和6.13级,育种价值高。在杂种优势利用上,X-27除了株高性状一般配合力为正效应外,该株系有利提高杂种后代的产量与稻米品质。X-57降低后代的株高和AC,提高后代产量和ASV。X-104虽产量高,品质好,但不利于杂种优势利用。闽糯2S/X-41、闽糯2S/X-107杂交一代在产量、株高、AC及碱消值等性状上具有明显的超亲优势和竞争优势,在杂交糯稻上应用价值高。【结论】杂交糯稻的产量与品质均为复杂性状,其中有效穗数、穗长、千粒重、长宽比、单株产量、AC、ASV主要受糯稻恢复系的控制。各性状的广义遗传率(h~2B)变幅14.72%~53.99%,狭义遗传率(h~2N)变幅10.25%~43.07%,遗传率较低,易受环境影响。育种上需大量配组,方可培育出不同用途的专用型糯稻杂交稻。

Abstract :

Bulked-segregant analysis by deep sequencing (BSA-seq) is a widely used method for mapping QTL (quantitative trait loci) due to its simplicity, speed, cost-effectiveness, and efficiency. However, the ability of BSA-seq to detect QTL is often limited by inappropriate experimental designs, as evidenced by numerous practical studies. Most BSA-seq studies have utilized small to medium-sized populations, with F-2 populations being the most common choice. Nevertheless, theoretical studies have shown that using a large population with an appropriate pool size can significantly enhance the power and resolution of QTL detection in BSA-seq, with F-3 populations offering notable advantages over F-2 populations. To provide an experimental demonstration, we tested the power of BSA-seq to identify QTL controlling days from sowing to heading (DTH) in a 7200-plant rice F-3 population in two environments, with a pool size of approximately 500. Each experiment identified 34 QTL, an order of magnitude greater than reported in most BSA-seq experiments, of which 23 were detected in both experiments, with 17 of these located near 41 previously reported QTL and eight cloned genes known to control DTH in rice. These results indicate that QTL mapping by BSA-seq in large F-3 populations and multi-environment experiments can achieve high power, resolution, and reliability. Map-based cloning of LPD, a major gene positively regulates leaf prickle development in eggplant SCIE

Prickles on eggplant pose challenges for agriculture and are undesirable in cultivated varieties. This study aimed to uncover the genetic mechanisms behind prickle formation in eggplant. Using the F2 and F2:3 populations derived from a cross between the prickly wild eggplant, YQ, and the prickle-free cultivated variety, YZQ, we identified a key genetic locus (LPD, leaf prickle development) on chromosome E06 associated with leaf prickle development through BSA-seq and QTL mapping. An auxin response factor gene, SmARF10B, was predicted as the candidate gene as it exhibited high expression in YQ's mature leaves, while being significantly low in YZQ. Downregulating SmARF10B in YQ through RNAi using a simple and efficient Agrobacterium-mediated genetic transformation method named Seedling Apical Cut Infection (SACI) developed in this study substantially reduced the size and density of leaf prickles, confirming the role of this gene in prickle development. Besides, an effective SNP was identified in SmARF10B, resulting in an amino acid change between YQ and YZQ. However, this SNP did not consistently correlate with prickle formation in eight other eggplant materials examined. This study sheds light on the pivotal role of SmARF10B in eggplant prickle development and introduces a new genetic transformation method for eggplant, paving the way for future research in this field.

水稻种质资源茎杆抗倒伏性状的评价与分析

【目的】抗倒伏是当前水稻育种的重要关注性状,评价种质资源抗倒伏特性对培育抗倒品种具有重要意义。【方法】以国内200份种质资源为材料,系统调查9个与水稻抗倒伏相关性状,采用形态学聚类及通径分析,评价上述种质资源抗倒伏性状的育种价值。【结果】以倒伏指数为标准,将供试材料分为高抗倒型、抗倒型和倒伏型3种类型,其中81.5%的种质资源具有较好的抗倒能力。不同抗倒类型种质资源的茎秆物理性状与倒伏指数相关系数存在明显差异。如恢复系等材料的株高、弯曲力矩和基部第2节长与倒伏指数呈极显著正相关,其中弯曲力矩与倒伏指数的相关系数最高,达0.789;而基部抗折力、鞘叶厚度与倒伏指数呈负相关,相关系数分别为-0.754~(**)和-0.659~(**)。不育系的株高和基部第2节长与倒伏指数呈极显著正相关,其中株高与倒伏指数的相关系数达0.681~(**);基部抗折力与倒伏指数相关系数为-0.841~(**)。通径分析表明,恢复系等材料的弯曲力矩对倒伏指数为直接正向效应(Py=0.519),不育系的株高对倒伏指数呈直接正向效应(Py=0.737),双亲基部抗折力对倒伏指数均呈直接负向效应,直接通径系数分别为-0.331和-0.702。【结论】水稻抗倒伏性状是一个复合性状,不同种质资源具有不同抗倒特性,科学利用育种资源是水稻抗倒伏育种的关键。
