黑米新品种紫两优7206营养品质分析

Abstract :

【目的】对新育成的黑米两系杂交稻紫两优7206进行营养品质分析,为黑稻良种选育及功能食品研发提供参考价值。【方法】以超级稻宜优673作对照,分析紫两优7206与宜优673的氨基酸、脂肪酸、花色苷和总黄酮等含量差异,以及总抗氧化能力与清除自由基能力的差异。【结果】紫两优7206与宜优673在水分、灰分和脂肪含量方面无明显差异,均检测出17种氨基酸,8种脂肪酸。紫两优7206的必需氨基酸含量高于FAO提出的必需氨基酸模式,其蛋白质含量、总氨基酸与必需氨基酸含量分别比宜优673高7.33%、11.08%和9.71%。紫两优7206比宜优673具有较高的各呈味氨基酸TAV值,丰富了黑米食味品质。2种稻米的饱和脂肪酸(SFA)、单不饱和脂肪酸(MUFA)和多不饱和脂肪酸(PUFA)比值比较接近中国营养协会推荐的“<1∶1∶1”的比例,且均含有人体必需脂肪酸亚油酸与亚麻酸。紫两优7206的花色苷、总黄酮含量,以及总抗氧化能力与清除自由基能力均极显著优于宜优673。【结论】杂交配组选育的紫两优7206较宜优673具有更丰富的营养价值,对功能性黑稻的培育与食品研发具有一定参考价值。

Abstract :

Bacterial wilt caused by Ralstonia solanacearum is one of the most important diseases in solanaceous plants, including peppers. It generally tends to be more serious under warm-temperature and moist (WM) conditions than at moist room-temperature (RM) conditions. Although immunity mechanisms at room temperature have been intensively studied, the mechanisms underlying WM conditions remain poorly understood. Herein, the pepper cysteine protease CaZingipain2 was expressed and functionally characterized in pepper immunity against R. solanacearum at WM conditions and at room temperature. The results showed that CaZingipain2 localized to the nucleus and was upregulated at the transcript level in pepper plants upon R. solanacearum infection under WM conditions (RSWM). Virus-induced gene silencing of CaZingipain2 significantly increased the susceptibility of pepper plants to RSWM, and was coupled with the downregulation of CaPRP1 and CaMgst3, which are specifically related to pepper immunity against RSWM, according to our previous studies, while its overexpression significantly reduced the susceptibility of N. benethamiana plants to RSWM compared to that of wild-type plants. In addition, our data showed that CaZingipain2 also acts positively in pepper immunity against R. solanacearum infection at room temperature by upregulating the SA- and JA-responsive PR genes, including CaNPR1 and CaDEF1. All these results indicate that CaZingipain2 improves pepper immunity against R. solanacearum under WM conditions and at room temperature by regulating different PR genes.

Abstract :

Plants need to fine-tune defense responses to maintain a robust but flexible host barrier to various pathogens. Helix-loop-helix/basic helix-loop-helix (HLH/bHLH) complexes play important roles in fine-tuning plant development. However, the function of these genes in plant immunity and how they are regulated remain obscure. Here, we identified an atypical bHLH transcription factor, Oryza sativa (Os)HLH46, that interacts with rice receptor-like cytoplasmic kinase (RLCK) Os BRASSINOSTEROID-SIGNALING KINASE1-2 (OsBSK1-2), which plays a key role in rice blast resistance. OsBSK1-2 stabilized OsHLH46 both in vivo and in vitro. In addition, OsHLH46 positively regulates rice blast resistance, which depends on OsBSK1-2. OsHLH46 has no transcriptional activation activity and interacts with a typical bHLH protein, OsbHLH6, which negatively regulates rice blast resistance. OsbHLH6 binds to the promoter of OsWRKY45 and inhibits its expression, while OsHLH46 suppresses the function of OsbHLH6 by blocking its DNA binding and transcriptional inhibition of OsWRKY45. Consistent with these findings, OsWRKY45 was up-regulated in OsHLH46-overexpressing plants. In addition, the oshlh46 mutant overexpressing OsbHLH6 is more susceptible to Magnaporthe oryzae than is the wild type, suggesting that OsHLH46 suppresses OsbHLH6-mediated rice blast resistance. Our results not only demonstrated that OsBSK1-2 regulates rice blast resistance via the OsHLH46/OsbHLH6 complex, but also uncovered a new mechanism for plants to fine-tune plant immunity by regulating the HLH/bHLH complex via RLCKs.

本发明公开了水稻黑色种皮调控基因Ra的分子标记引物及其应用,本发明方案属于植物生物技术领域,提供了对水稻黑色种皮起决定作用的Ra基因进行基因分型的功能性标记的方案,所述分子标记引物包括一对外引物ZiA‑1‑Ra‑OW、ZiA‑1‑Ra‑OB和一对内引物ZiA‑1‑Ra‑IB、ZiA‑1‑Ra‑IW,一对外引物和一对内引物的引物序列如SEQ ID NO:1~4所示;本方案分子标记引物通过PCR扩增,即可对水稻苗期Ra基因进行分型;其具有功能性标记扩增效率高的优点,且不同等位基因型所扩增的条带差异明显,利用该分子标记引物可以实现对水稻苗期黑色种皮性状的精确跟踪,提高水稻黑米品种的选育效率。

杂交水稻不同栽培方式的产量形成及经济效益分析

以紫两优737、野香优699和野香优676等3个杂交水稻品种为试验材料,设置免耕直播、翻耕直播和人工插秧(CK)3种不同栽培方式,比较研究其生育期、产量及其构成因素的差异,测算生产成本及经济效益,以期找到适宜的栽培方式。结果表明,与人工插秧相比,直播栽培的生育期缩短7~10 d,株高下降7.1%~16.3%,有效穗数增加53.2%~77.8%,每穗总粒数减少27.3%~34.0%,产量显著下降2.6%~8.8%,穗长、结实率和千粒重等性状无显著差异,免耕直播和翻耕直播在农艺性状和产量性状上的差异不显著;免耕直播的收益最高(10 872.0~28 770.0元/hm~2),翻耕直播的收益排第2位(9 064.0~26 700.0元/hm~2),人工插秧的收益最低(7 580.0~25 410.0元/hm~2),免耕直播和翻耕直播的纯收入分别比人工插秧高13.2%~61.4%和5.1%~30.8%。综合比较3种栽培方式,免耕直播的经济效益最高。

【目的】选育和利用优质糯稻不育系。【方法】采用350Gy~(60)Co-γ射线辐照光温敏核不育系6311S干种子,借助淀粉遇KI-I2溶液的显色反应原理,在其M2带胚糙米群体获得糯质基因突变体。经福建、海南异地种植鉴定与选择,育成糯质6311S不育系,命名为闽糯2S。【结果】除直链淀粉含量差异显著外,闽糯2S与6311S具有相同的生育期,十分相似的农艺性状、花器性状、花粉不育特性以及育性转换特性。【结论】闽糯2S于2020年6月通过福建省农作物品种审定委员会审定,用闽糯2S配制成的优质高产杂交糯稻新组合糯两优8号,于2020年9月通过湖北省农作物品种审定委员会审定。
