Key Sweet Potato Viruses in Fujian Province and Their Distribution, Harmfulness, and Implications in China SCIE

Abstract :

China, the largest global producer of sweet potatoes, faces significant threats from viral diseases, particularly in Fujian Province, where sweet potatoes are the second most important food crop after rice. This study identified 11 viruses, including sweet potato feathery mottle virus (SPFMV) and sweet potato chlorotic stunt virus (SPCSV), infecting sweet potatoes in Fujian. Sequence comparisons revealed diverse strains from various sources. Virus prevalence varied across regions, with Quanzhou, Fuzhou, and Putian severely affected, detecting 10, 9, and 7 viruses, respectively, compared to only 3 in Sanming and Longyan. In particular, sweet potato virus disease (SPVD) caused the most severe damage during the seeding stages, resulting in dwarfing and leaf deformation, while the damage was lighter during the growth period, manifesting as the yellowing and brittleness of the leaves, ultimately reducing the yield. Compound infestations predominated, with between 0 and 6 viruses infecting different sweet potato varieties. Single-virus infections were observed for sweet potato virus 2 (SPV2), sweet potato symptomless virus 1 (SPSMV-1), and sweet potato pakakuy virus (SPPV), while others, particularly SPCSV, were frequently co-infected with SPFMV, leading to SPVD development. Further analysis showed that the RNase3 expression of SPCSV was correlated with the SPVD severity in sweet potato. These findings provide insights into the epidemiology of sweet potato viruses and serve as a reference for developing targeted disease management strategies.

<正>甘薯作为中国重要的粮食作物,伴随着社会进步与人们生活方式及健康观念的转变,已逐渐成为当代的“宠儿”。随着甘薯产业的持续扩张与深化,其分工日趋精细,专业化程度亦在不断提升。特别是在福建省,甘薯作为首要的粮食与加工作物,其种植规模与产量均位列粮食作物前茅。因此,构建并完善福建省的甘薯产业社会化服务体系,对于推动甘薯产业的稳健发展,提升生产效益,促进农业结构的优化与调整,进而提升福建省农业的整体竞争力具有深远意义。社会化服务在农业领域具体表现在将生产、销售等各个环节外包给专业的社会化服务公司进行操作。农户只需将部分农业生产收益支付给这些公司,便可保留大部分收益。

Abstract :

Researchers often need to synthesize genes of interest in this era of synthetic biology. Gene synthesis by PCR assembly of multiple DNA fragments is a quick and economical method that is widely applied. Up to now, there have been a few software solutions for designing fragments in gene synthesis. However, some of these software solutions use programming languages that are not popular now, other software products are commercial or require users to visit servers. In this study, we propose a Python program to design DNA fragments for gene synthesis. The algorithm is designed to meet the experimental needs. Also, the source code with detailed annotation is freely available for all users. Furthermore, the feasibility of the algorithm and the program is validated by experiments. Our program can be useful for the design of gene synthesis in the labs and help the study of gene structure and function.

A Python program to merge Sanger sequences: an update SCIE

Abstract :

Gene cloning is an important step in investigating gene structure and function. To verify gene sequence, Sanger sequencing is used, which may produce several overlapping sequencing files that need to be merged before alignment to the target gene sequence is performed. Previously, we reported the Python program to Merge Sanger sequences (https://peerj.com/articles/11354/), which ran in command line and relied heavily on EMBOSS suite. In this updated version of the program, we have made several remarkable improvements. It provides a graphical user interface (GUI) written with tkinter, which is convenient and stable. It does not require users to rename the input sequences before performing merging. With regard to the implementation, the updated version utilizes Python function (Align.PairwiseAligner) to align adjacent sequences, which is more flexible (can adjust program parameter i.e., the number of first-time consecutive matching bases). The new version of the program makes merging Sanger sequences much more convenient and facilitates gene study.

本发明公开了一种利用CRISPR‑Cas13a检测甘薯SPVD病毒病的方法,主要内容包括LwaCas13a基因的表达与蛋白纯化、guide RNA的获得、LwaCas13a‑sgRNA复合物组装、甘薯SPVD病毒病检测。主要原理是,构建LwaCas13a‑sgRNA复合物,利用其中的gRNA特异识别病毒RNA后,启动Cas13a剪切RNA的功能,释放荧光信号,经检测荧光信号,可快速检测低浓度的病毒RNA。该方法是继指示植物嫁接法、PCR法和ELISA三种甘薯病毒病检测方法之外的一种新方法,具有检测时间短、不需要逆转录、快速且高效的检测方法,检测的病毒浓度下限与PCR方法相当。

本实用新型涉及一种甘薯切条装置,包括机架,所述机架上设置有水平的作业台面,所述作业台面上设置有网格刀,所述网格刀的正上方设置有经电动机构驱动升降的压板,所述网格刀的外周部设置有甘薯的定位夹具。该甘薯切条装置设置有电动机构,电动机构能驱动压板升降。当把甘薯经定位夹具放置在网格刀上以后,电动机构会驱动压板下降,将甘薯向下压入网格刀,网格刀一次就能将甘薯切成条状。本实用新型采用电动推切的方式不仅省时省力,而且安全高效,使用方便。

Abstract :

为了建立甘薯羽状斑驳病毒(Sweet potato feathery mottle virus, SPFMV)和甘薯矮化褪绿病毒(Sweet potato chlorotic stunt virus, SPCSV)的逆转录环介导等温扩增(RT-LAMP)检测方法,提高检测灵敏度,实现结果的可视化。根据SPFMV外壳蛋白基因(CP)的核苷酸序列和SPCSV的热休克蛋白基因(Hsp 70)的核苷酸序列设计4条RT-LAMP特异性引物,采取单因素优化试验,对RT-LAMP反应体系中的多个因素包括时间、温度、BST聚合酶、Mg~(2+)、RNase抑制剂、dNTPs和Betaine浓度优化,恒温扩增60 min。经琼脂糖凝胶电泳分析,SYBR Green I可视化显色,结果表明:SPFMV的优化反应体系为:FIP/BIP 2μL、F3/B3 0.5μL、BST聚合酶1.0μL、dNTPs 0.6μL、MgSO_4 1.5μL、RNase抑制剂1.0μL、Betaine 7μL,62℃60 min。SPCSV的优化反应体系为:FIP/BIP 2μL、F3/B3 0.5μL、BST聚合酶1.0μL、dNTPs 0.6μL、MgSO_4 1.5μL、RNase抑制剂1.2μL、Betaine 7μL,64℃60 min。进一步利用SPFMV全基因组的4个片段(SPFMV-1、SPFMV-2、SPFMV-3、SPFMV-4)、SPCSV-Hsp 70和RGNNV进行特异性检验,分别建立了SPFMV和SPCSV的特异性RT-LAMP检测方法,扩增出了具有RT-LAMP的典型瀑布状条带,与凝胶电泳和SYBR Green I显色结果一致,SPFMV和SPCSV的灵敏度检测下限分别为:1×10~(-6)、1×10~(-3) ng·μL~(-1),该方法检测灵敏度高,实现了结果的可视化。对田间甘薯苗和离体组织培养甘薯苗进行检测验证,SPFMV-RT-LAMP检测方法成功率为100%,SPCSV-RT-LAMP检测方法的成功率为95%,表明研发的SPFMV和SPCSV的RT-LAMP检测方法适用于SPFMV和SPCSV的快速检测。

薯圆加工专用型甘薯种质资源筛选及工艺优化

随着人们生活水平提高,消费观念转变,以甘薯为主要原料的薯圆,越来越受到年轻人的喜欢。以筛选适合薯圆加工专用型甘薯种质资源和研究薯圆加工专用型甘薯的最佳工艺配方为目的,采用福建农林大学洋中科教基地种植的145份甘薯种质为原料进行薯圆加工,根据感官综合评分,筛选出适合薯圆加工的专用型甘薯种质,并对筛选到的种质开展工艺优化正交试验。结果表明:从145份甘薯种质资源中筛选出薯圆加工感官评价分数大于80分的甘薯种质5份,分别为SP1、SP4、SP35、SP70、SP87,以得分较高的SP1开展薯圆最佳工艺研究,其最佳工艺配方为木薯粉20 g、水6 mL、甜叶菊0.10 g。

Abstract :

Red and blue light-emitting diodes (LEDs) affect the quality of sweet potato leaves and their nutritional profile. Vines cultivated under blue LEDs had higher soluble protein contents, total phenolic compounds, flavonoids, and total antioxidant activity. Conversely, chlorophyll, soluble sugar, protein, and vitamin C contents were higher in leaves grown under red LEDs. Red and blue light increased the accumulation of 77 and 18 metabolites, respectively. Alpha-linoleic and linolenic acid metabolism were the most significantly enriched pathways based on Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses. A total of 615 genes were differentially expressed between sweet potato leaves exposed to red and blue LEDs. Among these, 510 differentially expressed genes were upregulated in leaves grown under blue light compared with those grown under red light, while the remaining 105 genes were expressed at higher levels in the latter than in the former. Among the KEGG enrichment pathways, blue light significantly induced anthocyanin and carotenoid biosynthesis structural genes. This study provides a scientific reference basis for using light to alter metabolites to improve the quality of edible sweet potato leaves.
