Dual Localization and Functional Divergence of V-ATPase Subunit A: Nuclear Shuttling Mediates Distinct Roles in Dark- and MeJA-Induced Leaf Senescence SCIE

Abstract :

Retrograde signaling regulates plant senescence, but the role of vacuoles in this process remains unclear. Here, we demonstrate that rice vacuolar H+-ATPase subunit A (OsVHA-A) localizes to both the cytoplasm and nucleus. Sucrose treatment increased OsVHA-A expression and nuclear accumulation, while darkness reduced it. Methyl jasmonate (MeJA) initially promoted OsVHA-A nuclear translocation but decreased it upon prolonged exposure. Downregulation of OsVHA-A expression accelerated MeJA-induced rice leaf senescence but delayed darkness-induced senescence. MeJA treatment also significantly upregulated the expression of OsMYC2 and OsMAPK6 in OsVHA-A-RNAi plants compared to wild-type plants. Moreover, OsVHA-A downregulation notably increased the level of expression of genes associated with sugar signaling and transport under dark conditions. Immunoprecipitation-mass spectrometry and molecular docking analyses identified interactions between OsVHA-A and OsTPR1, OsMed14, sucrose transporters, and enzymes involved in sucrose metabolism. The binding of OsVHA-A with OsTPR1 and OsSUS1 was confirmed by BiFC. These findings highlight the multifunctional role of OsVHA-A in coordinating organelles and nuclear signaling, providing new insights and potential strategies for manipulating senescence to improve rice yield and quality.

Abstract :

pH treatment promotes single-cell lipid accumulation and significantly affects microalgae growth. This study investigates the correlation between lipid content and environmental pH using the model diatom Phaeodactylum tricornutum (P. tricornutum). We compared three distinct pH treatment strategies-continuous, intermittent, and a two-phase culture-in P. tricornutum. Rigorous analysis of chlorophyll content, cell density, and lipid content indicated that ongoing pH treatment at pH 9.5 (CHES) emerged as the most effective approach for lipid accumulation in P. tricornutum. The CHES buffer treatment significantly boosted total lipid yield and led to a reduction in protein content. Carbohydrate content experienced a slight decline under CHES buffer treatment, but changes were observed in the activities of key enzymes. Specifically, [acyl-carrier-protein] S-malonyltransferase (MAT) activity decreased after 3 days in the control treatment, while no significant change was noted under the CHES buffer treatment. In contrast, diacylglycerol O-acyltransferase (DGAT) activity showed upregulation 2 and 3 days post-CHES buffer treatment. Moreover, the study identified differentially expressed genes enriched in Gene Ontology (GO) terms associated with protein biosynthesis, photosynthesis, nucleoside metabolism, and transferase activity. These outcomes underscore the pivotal role of CHES buffer in orchestrating primary metabolism, potentially steering carbon flux towards lipogenesis. As a result, the potential of microalgae as a sustainable source of biofuels contributes significantly to the transition towards a more environmentally friendly energy landscape.

OsCNGC7 modulates calcium dynamics and accelerates leaf senescence in rice SCIE

Abstract :

Calcium plays a crucial role in regulating plant senescence. However, the specific effects of increased intranuclear calcium versus cytoplasmic calcium on aging remain unclear. Cyclic nucleotide-gated channels (CNGCs), which manage Ca2* levels in plant cells, are particularly significant in this context. These channels are known to relocate between the nuclear envelope and the plasma membrane in response to stress and developmental signals. Through this movement, CNGCs help regulate the balance of cytosolic and intranuclear Ca2*. In this study, we categorized the 16 CNGC genes in rice into five subgroups. OsCNGCs are notably expressed in leaves, especially during the reproductive stage. Both OsCNGC6 and OsCNGC7 exhibit dual localization to the plasma membrane and the nuclear envelope. Knockdown of OsCNGC7 led to reduced levels of Ca2* and K* in plants. Conversely, yeast expressing the OsCNGC7 gene showed increased sensitivity to Ca2*. Additionally, while the [Ca2*]cyt was maintained at relatively low levels in both wild-type and OsCNGC7-RNAi lines, the fluorescence intensity was significantly higher in OsCNGC7-overexpressing lines, particularly in the nucleus of root tips. Overexpression of OsCNGC7 resulted in enhanced stomatal opening and accelerated leaf senescence from the tillering stage to grain filling in rice. Treatment with MeJA rapidly induced OsCNGC7 expression, while knockdown of OsCNGC7 delayed both MeJA-induced and dark-induced leaf senescence. Further analysis revealed that OsCNGC7 interacts with OsKAT2 and OsALMT2. In conclusion, our findings highlight the distinct roles of OsCNGCs in regulating senescence. This knowledge could provide new strategies for manipulating plant senescence and enhancing crop productivity.

实验教学效果不但关系着学生掌握知识、提高实验技能的程度,也关系着学生科研素养和家国情怀的培养,直接影响我国创新型人才培养的成效。该文以植物生理生化实验课——硝酸还原酶活性的测定为例,采用以问题为导向(problembased learning,PBL)教学模式,从教学设计、实施方案和典型教学活动等方面探讨思政教育融入实验课的方法与技巧,使学生的动手和创新能力、发现和解决问题的能力得以有效提高,使学生的家国情怀和“两山”意识、科技助力乡村振兴意识和“三农”意识得以有效提升。该设计将为农林高校创新实验教学提供范式,也为高效培养学生综合素质提供借鉴。

“三农情怀”是做好“三农工作”的重要基础,将厚植“三农情怀”教育与高等教育深度融合是农林院校践行立德树人根本任务的重要途径。福建农林大学开展以教师为主体、学生为中心、厚植三农情怀、培养新农人为目标的植物生物技术课程体系,该课程将思政教育、理论教学、实践教学、文化养成和教学评价五个模块有机融合,引入“三农”系列讲座,创新教学内容和教学模式,将课程建设与基地建设、社会实践巧妙衔接,采用环境熏陶、知识拓展、文化引领、情感体验、宣传感染等形式助力学生三农文化养成,为培养“懂农业、爱农村、爱农民”的新农人夯实了基础,也为同行提供借鉴。

如何通过科研育人促进大学生创新型人才培养已成为全国高校工作的重点,但部分高校比较注重对研究生的科研素质培养,缺乏对本科生创新意识、创新思维、创新方法和创新能力等创新素养的培育。文章系统梳理了创新理念和科研育人的内涵,从高校、教师和培育对象3个层面深入剖析科研育人过程中遇到的问题,并由此提出改变困境的实施路径,为高校创造更加有利的科研育人环境提供借鉴,也为高校更有效的培养创新型人才提供参考。

Plant nutrition status is closely associated with plant defense against insect herbivores. However, the way nitrogen supply regulates rice anti-herbivore is not clear. This study investigated the effects of low (LN, 0.3 mM) and high (HN, 3 mM) nitrate levels on rice resistance against the striped stem borer Chilo suppressalis (SSB), one of the major destructive rice pests. Seven-day-old rice seedlings were cultured with different nitrate levels for 30 days and then inoculated with third instars of SSB. LN significantly enhanced rice anti-herbivore defense and lowered the total nitrogen content in the plants, but increased the content of free amino acids after SSB infestation. Additionally, LN significantly increased the accumulation of phenolic acids and flavonoids, especially lignin, resulting in enhanced constitutive defense in SSB-infested plants. SSB feeding led to a rapid accumulation of secondary metabolites. HN application led to the accumulation of metabolites derived from cinnamic acid, p-coumaric acid, p-coumaric CoA, feruloyl CoA, and apigenin, while LN led to the accumulation of metabolites derived from 3-dehydroquinic acid, phenylalanine, acetyl CoA, and aspartic acid. Collectively, our finding suggests that nitrogen deficiency enhances rice anti-herbivore defense via constitutive defense by the accumulation of phenolic acids and flavonoids.
