Abstract :

This study optimized the extraction of sesamin and 4-hydroxy-sesamin from Cinnamomum camphora residue, yielding 0.3804 % under conditions of 86 degrees C, 43 min, and a 1:8.39 solid-liquid ratio. The purified compounds showed 67.86 % and 78.87 % recovery rates via recrystallization. In LPS-induced BV-2 microglia, 4hydroxy-sesamol (10-40 mu mol/L) activated autophagy, reduced iNOS, COX-2, IL-6, IL-1 beta, TNF-alpha mRNA expression, and demonstrated neuroprotection without cytotoxicity, suggesting its potential in mitigating neuroinflammation.

Abstract :

研究三七内生真菌Nigrospora sp.的化学成分及其生物活性。应用硅胶柱色谱、高效液相色谱等多种色谱分离技术,从该菌株土豆液体发酵产物的乙酸乙酯提取物中分离得到3个聚酮类化合物,根据NMR、HR-ESI-MS、ECD等方法确定其结构,分别鉴定为(R)-2-methyl-4-(2-oxobutyl) cyclopent-2-en-1-one (1)、multiplolide A(2)、8-O-acetylmultiplolide(3)。其中,化合物1是一个新的环戊烯酮类衍生物,化合物2和3是十元大环内酯类衍生物,均为首次从Nigrospora sp.分离得到。抗菌活性实验结果表明,化合物1对大肠杆菌(Escherichia coli)具有微弱的抗菌活性,最小抑菌浓度为64μg/mL。

本发明公开了药物提取技术领域的一种福建柏叶提取物的制备方法及其应用,包括以下步骤:1)取收集的新鲜福建柏叶,经50‑60℃烘干后粉碎,过20目‑50目筛,得干燥的粉末;2)步骤1)所得粉末1000g,加入体积浓度为55‑95%乙醇溶液14500mL,提取温度为70‑90℃并回流提取40‑80min;3)将步骤2)所得物在超声功率为300‑500W条件下提取0.5‑3H,然后过滤,收集提取液,减压浓缩,得福建柏叶浸膏;4)将步骤3)所得福建柏叶浸膏与等量硅藻土拌样后用乙酸乙酯萃取,收集该部分萃取液,减压浓缩回收溶剂,得福建柏叶提取物;福建柏叶制备所得的提取物在体外有显著的抑制α‑葡萄糖苷酶活性。

Abstract :

This study systematically investigated the optimization of total flavonoid extraction from the leaves of Bougainvillea glabra Choisy (B.glabra) and evaluated its antioxidant potential. The impact of three critical parameters (ultrasound time, heating reflux time, and ethanol concentration) on extraction efficiency was investigated using ultrasonication-assisted heat reflux extraction. A three-factor, three-level Box-Behnken design (BBD) was implemented to optimize the extraction procedure, using total flavonoid yield (TFY) as the response variable. The extracted flavonoids' antioxidant potential was measured quantitatively in vitro. The ideal conditions were 53% ethanol concentration, 10 minutes of ultrasonication, and 41 minutes of reflux. This procedure produced a TFY of (6.21 +/- 0.26)%. The extracts exhibited significant free radical scavenging action against 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical. When the concentration exceeded 1.0 mg/ mL, the scavenging rate of DPPH free radicals surpassed 84.1%. When the concentration was more than 0.6 mg/ mL, the ability of total flavonoids to scavenge ABTS free radicals was over 93.8%, which was very similar to how well vitamin C worked. This optimized protocol proves effective for flavonoid extraction, with the obtained products exhibiting significant antioxidant properties in standardized assays.

Abstract :

为优化福建柏叶总二萜提取工艺并研究其抑菌活性,通过响应面法对福建柏叶总二萜提取工艺进行优化,并采用菌丝生长速率法探究福建柏叶总二萜对6种植物病原真菌的抑制活性,以胶孢炭疽菌作为供试菌,进一步对菌丝生理生化指标的变化及保护酶活性进行测定。结果表明,福建柏叶总二萜的最佳工艺条件如下:使用无水乙醇作为提取溶剂,提取温度为79℃,提取时间为53 min,液料比为14.5 mL∶1 g。在此条件下,福建柏叶总二萜得率为36.8%。福建柏叶总二萜对胶孢炭疽菌抑制效果较好,EC50值为1.466 mg·mL

Sustainable Antifungal Potential of the Essential Oil of Litsea cubeba: Multifaceted Mechanisms Targeting Fusarium verticillioides SCIE

Abstract :

The essential oil of Litsea cubeba, extracted from its fruits, demonstrates significant antifungal activity against Fusarium verticillioides, with an EC50 value of 0.122 mu L/mL, comparable to the synthetic fungicide methyl thiophanate (EC50: 0.108 mu L/mL). Notably, at equivalent concentrations, the essential oil of L. cubeba exhibits 31.13% higher inhibitory efficiency than methyl thiophanate, highlighting its potential as an eco-friendly alternative. At 8 mu L/mL, the essential oil effectively controls mycelial growth on tomato surfaces through combined preventive and curative actions. Mechanistic studies reveal that the essential oil induces fungal cell death via multiple pathways, including disruption of cellular integrity and membrane architecture, ROS accumulation, DNA damage, elevated MDA levels, and altered membrane permeability. Transcriptomic analysis further indicates that the essential oil of L. cubeba disrupts fungal oxidative homeostasis and modulates protective enzymes, leading to programmed cell death. These findings underscore the potential of the essential oil of L. cubeba as a sustainable antifungal agent with multifaceted mechanisms superior to conventional fungicides, providing a scientific basis for developing plant-based alternatives in agriculture.

独蒜兰提取物对植物病原真菌的抑菌活性

【目的】研究独蒜兰假鳞茎乙醇提取物对植物病原真菌的抑菌作用,为植物源杀菌剂的开发提供依据。【方法】采用菌丝生长速率法,研究独蒜兰假鳞茎乙醇提取物对15种植物病原菌的抑制活性;以西瓜尖孢镰刀菌作为供试菌,进一步研究该提取物对病原真菌的菌丝干重、细胞膜、过氧化氢酶(CAT)、过氧化物酶(POD)和超氧化物歧化酶(SOD)等的影响。【结果】独蒜兰假鳞茎提取物对辣椒疫霉病菌、西瓜尖孢镰刀菌、番茄灰霉病菌和非洲隐地疫霉菌的抑菌效果明显,其EC50值分别为0.849、0.782、0.813和1.161 mg·mL~(-1);经独蒜兰提取物处理后的西瓜尖孢镰刀菌菌丝干重随着药剂浓度的增加而减少;细胞膜丙二醛含量和相对电导率增加;菌丝体细胞内CAT、POD和SOD 3种保护酶活性增加。【结论】独蒜兰假鳞茎提取物对植物病原真菌具有较好的抑菌活性,其抑菌作用可能与其干扰菌丝生长、使菌丝细胞膜正常功能受损等有关。

Background: Type 2 diabetes mellitus (T2DM) is a serious metabolic syndrome with high mortality and disability rates globally, which usually caused by unhealthy dietary patterns. Cinnamomum camphora leaf is a traditional Chinese medicinal herb used for attenuating hyperglycemia and digestive disorder, and high level of lignans has been found in C. camphora leaf. Purpose: This study aimed to examine the chemical composition of lignans extracted from C. camphora leaf (LCCL), and illustrate its therapeutic effect and mechanism on T2DM and its concomitant glycolipid metabolic disorder. Methods: The components of LCCL were separated and purified by silica gel and macroporous adsorption resin, and were distinguished through LC/MS and NMR. The antioxidant activity of LCCL was determined by free radical scavenging assay in vitro; the hypoglycemic and hypolipidemic abilities were evaluated by alpha-glucosidase, alpha-amylase and pancreatic lipase inhibition trials, respectively. T2DM model mice were established by high-sugar and high-fat (HSHF) feed together with streptozotocin (STZ) infection, and then grouped to assess the effect of LCCL treatment. Hematoxylin-eosin (H&E), Periodic Acid-Schiff (PAS) and oil red O staining were employed to analyze the histopathology. qRT-PCR assay, 16S rRNA analysis, and western blot were conducted to illuminate the anti-diabetic mechanism of LCCL. Results: 6 sesamin lignans were identifed from LCCL. The in vitro assays showed strong inhibitive abilities of LCCL with low IC50 on DPPH (33.68 +/- 0.54 mu g/ml),O-2(-) (39.25 +/- 0.61 mu g/ml), OH center dot (45.72 +/- 0.72 mu g/ml), alpha-glucosidase (0.82 +/- 0.14 mg/ml), alpha-amylase (0.86 +/- 0.11 mg/ml) and pancreatic lipase (0.91 +/- 0.12 mg/ml). LCCL treatment (100, 200 and 400 g kg(-1mg kg-1)) gradually decreased the fasting blood glucose (FBG) and fasting insulin (FINS), improved the glucose and insulin tolerance, down-regulated the homeostasis model assessment insulin resistance (HOMA-IR) indexes, alleviated the hepatic inflammatory response and oxidative stress, promoted the glycogen storage and depleted the fat accumulation in the liver. Besides, LCCL administration alleviated the glycolipid metabolism disorder in T2DM mice with a gut microbiota dependent manner, that significantly increased biodiversity, altered the composition of gut microbiota and increased the proportion of Lactobacillus. Conclusion: The lignan-rich extract of C. camphor leaf (LCCL), containing at least 6 lignans compounds, displayed promising antioxidant, hypoglycemic and hypolipidemic activities. The treatment of LCCL alleviated the glycolipid metabolism disorder in T2DM mice with a gut microbiota dependent manner. These finding suggested that LCCL should be further investigated to develop its complementary therapeutic effect on T2DM.

The bioactive compounds of the pseudobulbs of Pleione bulbocodioides were isolated, purified, and identified using modern spectroscopic techniques, which identified seven butanedioic acid compounds. Compounds 1-3, 5, and 6 were isolated for the first time from Pleione bulbocodioides. The antifungal activity of the seven compounds was evaluated through cytotoxic activity assays, revealing that compound 4, militarine, exhibited moderate inhibitory activity against Phoma herbarum, with an effective concentration (EC50) of 68.575 mu g/mL. We get further deep insight into the antifungal mechanism of militarine against Phoma herbarum through electron microscopy, transcriptomics, and changes in physiological and biochemical indicators. Transcriptomic sequencing revealed 216 significantly upregulated differentially expressed genes and 277 significantly downregulated differentially expressed genes. Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis showed that differentially expressed genes were involved in pathways related to secondary metabolism, biosynthesis, membrane components, nitrogen metabolism, and ATP -binding cassette (ABC) transporter, suggesting that militarine may inhibit the normal growth of hyphae through these pathways.
