Development of bait station to complement attract-and-kill agents of Zeugodacus tau (Diptera: Tephritidae) SCIE

Abstract :

Zeugodacus tau (Walker) (Diptera: Tephritidae) is an important agricultural pest currently managed primarily through the application of insecticides due to limited control strategies. Bait station devices are target specific and have emerged as a behaviorally based alternative to traditional insecticide sprays for managing Z. tau. In this study, we designed a bait station by integrating female-biased olfactory, visual, and gustatory elements, and a killing agent in a wax-matrix. Our results showed that the wax-matrix integrated with spinetoram showed the highest toxicity to immature and mature Z. tau females. Furthermore, the color and shape of the spinetoram bait station significantly influenced its attractiveness and toxicity to female Z. tau. Green sausage-shaped exteriors were the most effective color and shape examined. Subsequent experiments showed a length-dependent effect on mortality and visiting frequencies of Z. tau females when the bait stations were 9-13 cm long. The addition of the olfactory stimulus of 5% ammonium acetate to the bait station attracted a higher number of mature Z. tau females than the control. The killing efficacy of the weathered bait station was similar to that of the fresh station within an 8-week period (over 794 mm of rainfall). The bait station developed herein would provide new insight into the attract-and-kill strategy for Z. tau and alleviate the pressure of the actual management program for this pest.

Abstract :

Background Olfaction plays an important role in host-seeking by parasitoids, as they can sense chemical signals using sensitive chemosensory systems. Psyttalia incisi (Silvestri) (Hymenoptera: Braconidae) is the dominant parasitoid of Bactrocera dorsalis (Hendel) in fruit-producing regions of southern China. The olfactory behavior of P. incisi has been extensively studied; however, the chemosensory mechanisms of this species are not fully understood. Results Bioinformatics analysis of 64,515 unigenes from the antennal transcriptome of both male and female adults P. incisi identified 87 candidate chemosensory genes. These included 13 odorant-binding proteins (OBPs), seven gustatory receptors (GRs), 55 odorant receptors (ORs), 10 ionotropic receptors (IRs), and two sensory neuron membrane proteins (SNMPs). Phylogenetic trees were constructed to predict evolutionary relationships between these chemosensory genes in hymenopterans. Moreover, the tissue expression profiles of 13 OBPs were analyzed by quantitative real-time PCR, revealing high expression of seven OBPs (1, 3, 6, 7, 8, 12, and 13) in the antennae. Conclusion This study represents the first identification of chemosensory genes and the determination of their expression patterns in different tissues of P. incisi. These results contribute to a better understanding of the function of the chemosensory system of this parasitoid species.

Abstract :

Fopius arisanus (Sonan, 1932), an important egg parasitoid of several notorious tephritid pests, plays a key role in biological control programs. In the present study, the whole mitochondrial genome of F. arisanus was sequenced and characterized. The mitogenome of F. arisanus is 16,425 bp in length with 14.94% GC content, and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and two ribosomal RNA genes (rRNAs). The phylogenetic trees demonstrated that F. arisanus is sister group to Psyttalia concolor, P. humilis, P. lounsburyi and Diachasmimorpha longicaudata.

Abstract :

One environmentally friendly method used to manage Bactrocera dorsalis (Hendel), a key agricultural pest of substantial economic importance, is the sterile insect technique (SIT). Nevertheless, several deficiencies related to this strategy impair the success of the SIT, including the inferior performance of released sterile males compared with wild males, which could be partly solved by the utilization of gut symbionts as probiotic dietary components. In this study, a culture-dependent method was used to isolate and characterize gut-associated bacterial species in adult B. dorsalis genetic sexing strain (GSS) males. In addition, three bacterial isolates from the Enterobacteriaceae family, namely, Enterobacter sp., Morganella morganii, and Moellerella wisconsensis, were used as supplements in larval and adult diets to assess their effects on the life-history traits of irradiated males. Consistent with many previous studies, Enterobacter spp. was shown to be beneficial, with some quality control indices, such as adult size, pupal weight, survival rate under stress and nutritionally rich conditions, and mating competitiveness, being significantly increased, while slight nonsignificant increases in emergence rate and flight ability were observed. Conversely, the M. morganii and M. wisconsensis strains both had negative effects on irradiated male fitness and mating competitiveness. Our results, in combination with those of earlier studies, can contribute to improving the effectiveness of SIT application by enhancing the different aspects of augmentative rearing and biological traits of pests under laboratory rearing conditions.

双翅目昆虫触角感受器种类与功能研究进展 CSCD

Abstract :

双翅目是昆虫第四大目,与人类关系较为密切,物种多样,生态类型丰富。触角是昆虫的主要嗅觉感受器官,在昆虫觅食、求偶、交配、聚集、产卵等生活史过程中发挥着获取信息的重要作用。笔者通过检索Web of Science外文数据库和万方、维普和知网中文数据库,发现目前关于双翅目触角感受器超显微结构的观察研究涉及32个科248种昆虫,主要集中在与人类农业生产和生活息息相关的害虫,如蚊科、实蝇科、蠓科、厕蝇科、瘿蚊科、丽蝇科、蝇科、狂蝇科等,而近些年对双翅目天敌昆虫如寄蝇科和食蚜蝇科的研究也倍受关注。此外,笔者汇总整理研究表明,双翅目昆虫的感受器共13种,常见的有毛形感受器、锥形感受器、腔锥形感受器、刺形感受器、棒状感受器5种,还有部分感受器是双翅目某些科特有的,如环丝是瘿蚊科昆虫特有的感受器。另外,双翅目昆虫触角感受器的特征在科、属、种分类阶元中存在差异;对比我国不同地区的橘小实蝇成虫触角感受器类型,发现该害虫不同地理种群的触角感受器类型和特征存在差异。例如,我国台湾的橘小实蝇成虫触角着生栓锥形感受器和棒状感器,然而我国大陆的橘小实蝇未发现以上2种感器,并且大陆的橘小实蝇触角着生台湾种群没有的刺形感受器、腔锥型感受器或柱形感受器;广州的橘小实蝇锥形感受器基部未呈现弯曲状态,然而福建和吉林长春的橘小实蝇锥形感受器存在基部弯曲的亚型。笔者还综述了双翅目昆虫各种感受器类型在形态和功能方面的研究概况,探讨了触角感受器在害虫防治和昆虫系统发育研究中的应用前景。

本实用新型提供一种南亚实蝇幼虫寄生蜂自动分离装置,包括盛放盒及位于盛放盒上部的分离盒,所述盛放盒与分离盒不透光,所述盛放盒内底部置放有用于保湿的保湿层,所述保湿层上铺设有含水细砂层,所述盛放盒的顶部具有第一开口,第一开口处具有第一隔离网,所述分离盒内部中空下部与盛放盒顶部连接且分离盒顶部设有第二开口,所述第二开口处具有第二隔离网,所述第二隔离网的间隙小于第一隔离网,本实用新型中本发明在保湿层上未加隔离板和载物盘,直接以细沙加至保湿层,延缓水分蒸腾的速度,增加保湿效果,利用不同孔径的隔离网从而满足对南亚实蝇和寄生蜂的分离。

Simple Summary Storing insects at low temperature has been an effective approach for mass rearing programs, which can increase the shelf-life of bioagents and flexibly manipulate their pre-release periods. However, introducing insects to temperatures outside of their accustomed range can lead to stress and thereby influence their quality and competitiveness in the field. Bactrocera dorsalis is an economically important pest can devastate many fruits and vegetables, and cold storage has been used in storing, packing and shipping of sterile B. dorsalis males to control the wild population, but the impact of cold storage on the quality of B. dorsalis has never been investigated. Here, we examined the impacts of pupal cold storage on developmental parameters and quality of B. dorsalis adults. Overall, we found that pupal cold storage has a negative effect on some biological parameters of B. dorsalis, especially in fecundity. Thus, heat exposure was further conducted on the post-storage adults, and our results show that short-term heat exposure could partially compensate for the loss in fecundity in B. dorsalis adults resulting from the pupal cold storage. Our findings indicate that the flexibility of the B. dorsalis mass rearing program could be improved by utilizing cold storage and heat exposure. Cold storage and heat exposure are crucial components of tephritid fruit fly mass-rearing programs, as they influence the development and fitness traits of produced flies. This work investigated the effects of cold storage on the pupal developmental parameters and quality of Bactrocera dorsalis (Hendel) genetic sexing strain (GSS) adults. Furthermore, the impact of short-term thermal exposure on the fecundity of B. dorsalis (GSS) that also underwent pupal cold storage was examined. Our results show that pupal development time, emergence rate, partial emergence rate, flight ability and fecundity were significantly affected by low temperature and pupal age and their interaction. Pupal cold storage did not pose negative impacts on the mating competition and response to methyl eugenol (ME) in the males. In addition, compared with the adults that were subjected to the same pupal storage protocol (five-day-old pupae stored at 13 degrees C), adult exposure to 41 degrees C for 1 h showed significant reparative effects on fecundity. In summary, the cold storage procedure of B. dorsalis (GSS) pupae has the potential to improve the flexibility and efficiency of mass-rearing schedules. Furthermore, short-term thermal exposure showed reparative effects on the fecundity costs induced by pupal cold storage in B. dorsalis (GSS).
