Abstract :
BACKGROUNDThe red imported fire ant (RIFA, Solenopsis invicta), a highly destructive invasive pest, has rapidly spread through human trade, posing significant threats to agricultural and forest ecosystems. Due to its preference for high-fat and high-protein foods, ham sausage is commonly used as bait to monitor RIFA populations in invaded areas. However, the presence of volatile organic compounds (VOCs) in such baits may affect their effectiveness because VOCs can act as either attractants or repellents. Identifying VOCs that specifically attract RIFA is essential to improve bait efficacy.RESULTSThis study aimed to identify attractant compounds within bait VOCs for RIFA, leveraging the highly-expressed antennal odorant-binding protein 1 (OBP1) and reverse chemical ecology approach. Additionally, we examined the effects of mixtures of these attractants on RIFA behavior. Our findings revealed that anethole, 1S-(-)-beta-pinene, and beta-caryophyllene individually attracted RIFA at 0.1 mu g/mu L. Notably, a combination of anethole and 1S-(-)-beta-pinene enhanced behavioral activity more than individual compounds, suggesting synergistic effects. Conversely, the addition of beta-caryophyllene to anethole significantly reduced RIFA activity. These results provide a theoretical basis for developing behavioral regulators targeting RIFA.CONCLUSIONThis study demonstrates that the integration of OBP-based in vitro assays with computational simulations can effectively identify behaviorally active compounds for RIFA. Additionally, it clarifies the optimal ratios of active VOCs in baits, offering valuable theoretical guidance for enhancing RIFA population monitoring efforts. (c) 2025 Society of Chemical Industry.

Abstract :
Plums are economically important stone fruits, valued for their nutritional quality and health-promoting properties. However, their quality and marketability are often compromised by a physiological disorder called fruit hollowness and browning (HB). This study employed an integrated physiological, biochemical, and proteomic approach to investigate the molecular mechanisms underlying HB in Huangguan plum. Huangguan plum fruit samples from seven developmental stages were analyzed for lignin content, key enzyme activities, and expression patterns of genes involved in lignin biosynthesis. The results demonstrated that HB-affected fruit accumulated significantly higher lignin levels, particularly during post-veraison stages, accompanied by elevated activities of lignin biosynthesis-related enzymes like phenylalanine ammonia-lyase and laccase. Expression analysis of lignin pathway genes revealed distinct transcriptional dynamics, with notable upregulation of PsPAL1 and PsLAC7 in HB fruit at critical developmental stages. Proteomic profiling identified 941 and 1249 differentially expressed proteins between HB and non-HB fruit at the veraison and ripe stages, respectively. KEGG enrichment analysis highlighted significant alterations in the phenylpropanoid biosynthesis pathway (map00940), which is central to lignification. These findings provide new insights into the physiological and molecular basis of HB in plum fruit and a scientific foundation for developing effective strategies to mitigate or address this disorder and improve fruit quality.

Abstract :
Entomopathogenic nematodes (EPNs) are obligate parasitic "biopesticides" that play a vital role in pest management. A thorough understanding of their pathogenic mechanisms is essential for promoting their widespread use in agricultural pest control. The pathogenicity of EPNs arises from two key factors: the pathogenicity of their symbiotic bacteria and the nematodes' intrinsic pathogenic mechanisms. This review concentrates on the latter, offering an exploration of the excretory/secretory products of EPNs, along with their pathogenic mechanisms and key components. Particular attention is given to specific excretory/secretory proteins (ESPs) identified in various EPN species. The aim is to provide a foundational reference for comprehending the role of these ESPs in pest control. Furthermore, the review discusses the potential of these findings to advance the development of eco-friendly biopesticides, thereby supporting sustainable agricultural practices.

Abstract :
BACKGROUND: In agricultural pest management, especially in combatting the invasive red imported fire ant (RIFA, Solenopsis invicta), significant challenges emerge as a consequence of the constraints of solely depending on chemical insecticides or entomopathogenic nematodes (EPNs). The utilization of chemical insecticides carries environmental and ecological hazards, whereas EPNs, when applied independently, might not offer the immediate effectiveness necessary for adequate RIFA suppression. Acknowledging these hurdles, our study investigates a synergistic method that integrates EPNs with chemical insecticides, aiming to fulfill the urgent demand for more efficient and environmentally friendly pest control solutions. RESULTS: Our evaluation focused on the interaction between the highly pathogenic Steinernema riobrave 7-12 EPN strain and prevalent insecticides, specifically beta-cypermethrin and a mixture of bifenthrin and clothianidin, applied at highly diluted recommended concentrations. The findings revealed a notable increase in RIFA mortality rates when EPNs and these insecticides were used together, outperforming the results achieved with each method individually. Remarkably, this enhanced efficacy was especially evident at lower concentrations of the bifenthrin-clothianidin mixture, indicating a valuable approach to minimizing reliance on chemical insecticides in agriculture. Furthermore, the high survival rates of EPNs alongside the tested insecticides indicate their compatibility and potential for sustained use in integrated pest management programs. CONCLUSION: Our research underscores the effectiveness of merging EPNs with chemical insecticides as a powerful and sustainable strategy for RIFA management. This combined approach not only meets the immediate challenges of pest control in agricultural settings, but also supports wider environmental objectives by reducing the dependency on chemical insecticides. Abstract :
BackgroundEntomopathogenic nematodes (EPNs) in an aqueous suspension treatment showed potential in the biocontrol of the red imported fire ant Solenopsis invicta, whereas colony relocation following this inundative application had restricted overall effectiveness. As a novel strategy, EPN pre-infected insect cadavers carrying the next generation of nematode-infective juveniles (IJs) might be used to efficiently manage insect pests. However, this strategy has not been tested on S. invicta. This study evaluated EPNs emerging from wax moth Galleria mellonella cadavers to facilitate infection of S. invicta compared to those used in aqueous suspensions. ResultsAmong seven EPN species, Steinernema riobrave and Heterorhabditis bacteriophora had the highest insecticidal efficiency in aqueous treatment. Cadavers of G. mellonella colonized by either one of two EPN species were not damaged by worker ants, ensuring that the IJs could develop within and emerge successfully from the cadavers. Likewise, compared to an aqueous suspension treatment with an equivalent number of IJs, treatment with an S. riobrave-infected cadaver increased the mortality of S. invicta by & AP;10%, whereas infection by H. bacteriophora did not differ between treatments. However, the coexistence of S. riobrave- and H. bacteriophora-infected cadavers adversely affected the control of S. invicta, likely as a result of competition resulting from the increased dispersal of each emerging EPN species. ConclusionUsing EPN-infected G. mellonella cadavers as a strategy increased the mortality of S. invicta in the laboratory. This study provides positive evidence for the future applications of S. riobrave-infected cadavers in the biocontrol of red imported fire ants. & COPY; 2023 The Authors. Abstract :
Combinations of entomopathogenic nematode (EPN) species are sometimes more or less effective than individual species for the management of insect pests. We hypothesized that these outcomes are due in part to dispersal rates that differ when EPN species are conspecific or heterospecific. Dispersal rates of three heterorhabditid species, six steinernematid species, and mixtures of some were assessed using image analysis of nematodes on water agar. The dispersal rates between the genera differed significantly and were unrelated to the estimated body mass or the putative foraging strategy, including that of the recently named Steinernema khuongi, characterized here as a cruise forager (actively search for hosts). Heterorhabditis indica dispersed more rapidly on agar when combined with S. diaprepesi, but not with S. glaseri. The presence of S. diaprepesi in soil microcosms also increased the proximity of H. indica to Galleria mellonella host insects, while H. indica presence reduced the number of G. mellonella killed by S. diaprepesi. Nevertheless, increasing the H. indica dispersal rate did not increase its insecticidal effectiveness, likely due to competition with the more virulent S. diaprepesi. Rather, the effect of combining the species on the mortality of G. mellonella was additive. Our results suggest that interspecific EPN communication affects not only orientation but also dispersal rate, with potential impacts on biological control and the subsequent fitness of each species.
