N6-methyladenosine and poly(A) tail-mediated posttranscriptional regulation in bamboo mosaic virus-Dendrocalamus latiflorus interactions SCIE

Abstract :

The economically important bamboo species Dendrocalamus latiflorus Munro (D. latiflorus) exhibits highly susceptible to Bamboo mosaic virus (BaMV), resulting in severe growth and development impairment. However, the proteomic profiles, transcript processing dynamics, and their coordinated posttranscriptional regulation during bamboo-virus interactions remain uncharacterized. Tandem mass tag (TMT)-based quantitative proteomic revealed suppression of photosynthesis-related proteins and upregulation of protein synthesis and degradation, antioxidant within D. latiflorus during BaMV infection. Moreover, the APR1 protein serviced as a regulatory hub for connecting sulfur metabolism, antioxidant, and photosynthesis. Integration of nanopore direct RNA sequencing (DRS) data revealed reduced D. latiflorus full-length read ratios, consequently attenuating transcriptome and proteome correlation. BaMV-infected bamboos presented preferential usage of distal poly(A) site and lengthened poly(A) tail lengths (PALs) of pathogenesis-related (PR) genes. Epitranscriptome analysis showed increased N6-methyladenosine (m6A) ratios in POR (chlorophyll synthesis) and NCED1 (abscisic acid synthesis), which coupled with reduced transcriptional levels. In total, 122 potential m6A modification sites were found in BaMV, with AAACA representing the predominant consensus motif. Collectively, these results offer insights into posttranscriptional regulation networks during bamboo-BaMV interactions.

Abstract :

Endoplasmic reticulum (ER) is an enclosed three-dimensional eukaryotic membrane network composed of flattened sacs. Fusion of homologous membranes to the ER membrane is essential for the maintenance of this network structure. In yeast, ER membrane fusion is mediated by Sey1p, whose paralogues function distinctly in different species. In this study, we investigated the biological functions of MoSEY1 in the devastating rice blast fungus Magnaporthe oryzae by functional genomic approach. Compared to wild type, deletion of MoSEY1 considerably decreased the growth and conidia production of M. oryzae. Additionally, the absence of MoSEY1 delayed appressorium formation and invasive hyphae growth. The appressorium function was also impaired in Delta Mosey1 mutant. Subcellular localization analysis revealed that MoSey1 is localized at the ER. The Delta Mosey1 mutant showed augmented sensitivity to ER stress. Additionally, we found that MoSey1 regulated the unfolded protein response, autophagy, and protein secretion in M. oryzae. In conclusion, our study unveiled the involvement of MoSey1 in the development, pathogenesis, and ER functions in M. oryzae.

Abstract :

Fokienia hodginsii (F. hodginsii), belonging to the genus Fokienia of the Cupressaceae. F. hodginsii has significant application value due to its wood properties and great research value in evolutionary studies as a gymnosperm. However, the genome of F. hodginsii remains unknown due to the large size of gymnosperms genome. Pacific Bioscience sequencing, Hi-C mapping, whole-genome Bisulfite Sequencing (BS-Seq), long-read isoform sequencing (Iso-Seq), direct RNA sequencing (DRS), quantitative proteomics, and metabonomics analysis are employed to facilitate genome assembly, gene annotation, and investigation into epigenetic mechanisms. In this study, the 10G F. hodginsii genome is assembled into 11 chromosomes. Furthermore, 50 521 protein-coding genes are annotated and determined that 65% of F. hodginsii genome comprises repetitive sequences. It is discovered that transposable element (TE)-including introns is associated with higher expression. The DNA methylome of F. hodginsii reveals that xylem has a higher DNA methylation level compared to callus. Moreover, DRS reveals the significant alterations in RNA full-length ratio, which potentially associated with poly(A) length (PAL) and alternative polyadenylation (APA). Finally, the morphology measurement and metabonomics analysis revealed the difference of 14 cultivars. In summary, the genomes and epigenetics datasets provide a molecular basis for callus formation in the gymnosperm family.

Emerging Roles of Exocyst Complex in Fungi: A Review SCIE

Abstract :

The exocyst complex, an evolutionarily conserved octameric protein assembly, plays a central role in the targeted binding and fusion of vesicles at the plasma membrane. In fungal cells, this transport system is essential for polarized growth, morphogenesis, cell wall maintenance and virulence. Recent advances have greatly improved our understanding of the role and regulation of the exocyst complex in fungi. This review synthesizes these developments and focuses on the intricate interplay between the exocyst complex, specific fungal cargos and regulatory proteins. Insights into thestructure of the exocyst and its functional dynamics have revealed new dimensions of its architecture and its interactions with the cellular environment. Furthermore, the regulation of exocyst activity involves complex signaling pathways and interactions with cytoskeletal elements that are crucial for its role in vesicle trafficking. By exploring these emerging themes, this review provides a comprehensive overview of the multifaceted functions of the exocyst complex in fungal biology. Understanding these mechanisms offers potential avenues for novel therapeutic strategies against fungal pathogens and insights into the general principles of vesicle trafficking in eukaryotic cells. The review therefore highlights the importance of the exocyst complex in maintaining cellular functions and its broader implications in fungal pathogenicity and cell biology.

Most secretory proteins are folded and modified in the endoplasmic reticulum (ER). In Saccharomyces cerevisiae, the absence of Scs2 protein will lead to the separation of the endoplasmic reticulum and plasma membrane, resulting in endoplasmic reticulum dysfunction, but its function is not clear in rice blast fungus or even filamentous fungus. In this study, we report the identification and characterization of MoSCS2 in the pathogenesis of the rice blast fungus Magnaporthe oryzae. Protein subcellular localization showed that MoSCS2 is mainly localized in the endoplasmic reticulum. Compared to the wild-type strain Guy11, the deletion mutant Delta Moscs2 showed a significant reduction in growth and conidiation. MoSCS2 deficiency also resulted in abnormal conidial morphology and septum formation. The Delta Moscs2 mutant shows delayed appressorium formation, and the appressorium of Delta Moscs2 mutant could not form huge turgor pressure to penetrate the host epidermal cell wall. Pathogenicity and plant leave infection assays showed that knockout of MoSCS2 significantly inhibited the expansion of the invasive hyphae in host cells, ultimately leading to the decline of pathogenicity. Moreover, MoSCS2 gene is also involved in the regulation of cell wall and endoplasmic reticulum stress response. In conclusion, MoSCS2 plays an important role in the growth, asexual production, conidia morphogenesis, infection-related morphogenesis and pathogenicity of M. oryzae.

稻瘟病菌蛋白激酶调控因子MoMOB1的功能分析

稻瘟病菌(Magnaporthe oryzae)引起的水稻稻瘟病,每年造成全球水稻减产约10%~30%,对全球粮食生产构成严重威胁。MOB (Mps one binder)蛋白家族成员在物种进化过程中高度保守,并作为重要激酶调控因子与NDR (Nuclear DBF2-related)蛋白激酶等结合,在细胞周期的有丝分裂退出和胞质分离等事件中发挥重要作用。本研究利用功能丧失和获得的方法建立了MoMOB1敲除突变体,对突变体进行了功能回补;并对背景菌株、敲除突变体和回补菌株,在形态特征和致病性等方面进行了生物性状分析,以解析稻瘟病菌中MoMOB1的生物学功能。与背景菌株Ku80相比,MoMOB1缺失后稻瘟病菌生长速率明显减慢,分生孢子萌发延迟,突变体产孢量显著降低,分生孢子出现畸形且隔膜数目异常,部分分生孢子没有隔膜或只有一个隔膜。致病性分析发现,ΔMoMOB1突变体致病性明显减弱。回补菌株的表型与背景菌株Ku80相似。综上所述,MoMOB1在稻瘟病菌营养生长、产孢量、分生孢子发育、附着胞形成和致病性方面发挥关键作用。

BACKGROUND Soil quality assessment is a critical strategy for determining optimum fertilization in intensive pomelo production. In this study, we evaluated the soil quality status and mapped the spatial distribution of 347 soil samples collected from pomelo orchards in Pinghe County, southern China. We analyzed nine chemical parameters and an altitude indicator. RESULTS The mean soil quality index (SQI) was 0.355 in the total data set (TDS) and 0.292 in the minimum data set (MDS). Available Ca (Avail-Ca), pH value, organic matter and altitude were selected as indicators of soil quality in the MDS. The SQI in mature orchards (>10 years) was higher than that in young orchards (<10 years), while no differences between soil types and altitude gradients were identified. We detected a significant positive correlation between the SQI based on TDS (SQI(TDS)) and the SQI based on MDS (SQI(MDS)), and the spatial distribution of soil properties and SQI(TDS) showed a uniform pattern, except for Avail-N, Avail-B and SQI(MDS). Overall, unfavorable soil quality indicators, including rich in Avail-P, deficient in Avail-Ca, -Mg and -B, soil acidification and high altitude, were considered to be limiting factors for pomelo production. CONCLUSION The soil chemical quality in pomelo orchards is generally low, indicating that integrated management by controlling acidification, reducing planting altitude, regulating fertilization and monitoring soil properties is required for sustainable pomelo production. 稻瘟病菌中与NDR激酶Dbf2相互作用的Momob1蛋白的功能研究

蛋白激酶Dbf2属于NDR (Nuclear Dbf2-Related)激酶家族,现有研究表明,真核生物中的NDR激酶序列具有高度保守性,并调控细胞的有丝分裂、细胞生长和发育等过程。真菌的胞质分裂主要包括3个阶段:胞质分裂起始位点的选择;调节蛋白的有序激活和定位;肌动球蛋白环的收缩。该过程需要一个非常保守的信号网络的激活,该信号网络在芽殖酵母中称为有丝分裂退出网络(Mitotic Exit Network,MEN),Dbf2和Mob1是信号通路的一部分。Dbf2与Mob1会形成复合体,并且Mob1为Dbf2的共激活因子。本实验对稻瘟病菌中Mob1基因的功能进行了初步的探索,基于反向遗传学的策略,通过基因敲除技术得到Mob1缺失突变体,再通过对该突变体表型变化的观察来推断该基因的功能。表型分析发现,与野生型Ku80相比,Momob1的缺失使得菌落的生长速率受到明显抑制、分生孢子的形态发生严重畸形、分生孢子及隔膜的数量明显减少、附着胞的形成明显延迟、致病菌的侵染能力显著下降。亚细胞定位表明Momob1定位于菌丝的隔膜处,这与Modbf2在菌丝中的亚细胞定位相同。以上结果表明Momob1对稻瘟病菌的发育和致病侵染过程至关重要。综上所述,稻瘟病菌中Mob1基因可与Dbf2基因相互作用,调控稻瘟病菌的产孢,隔膜和附着胞的形成以及胞质分裂等相关进程,其具体调控机制有待深入研究。

Growth, photosynthesis, and nutrient uptake in wheat are affected by differences in nitrogen levels and forms and potassium supply SCIE

Nitrogen (N) and potassium (K) are essential macronutrients for plants growth; however, the mechanism by which K mediates negative effects on ammonium-sensitive plants is still poorly understood. We hypothesized that K supplies may enhance antagonistic ammonium stress while improving nitrate nutrition function, which wheat seedlings were grown in sand culture in the presence of two N forms (ammonium; nitrate) supplied at two rates (2, 10 mmol L-1) and three K levels (0.5, 5, 15 mmol L-1). We found that a high N rate increased plant biomass under nitrate nutrition, while it had a negative effect under ammonium nutrition. Compared with nitrate, biomass was depressed by 54% or 85% for low or high N rate under ammonium. This resulted in a reduction in gas exchange parameters and a subsequent decrease in growth variables and nutrient uptake, whereas these parameters increased significantly with increasing K levels. Moreover, in principal components analysis, these variations were highly clustered under nitrate nutrition and highly separated under ammonium nutrition. Our study shows a clear positive interaction between K and N, suggesting that high K supply relieves ammonium stress while improving growth vigor under nitrate nutrition by enhancing nutrient uptake and assimilate production in wheat plants.
