Abstract :
为解决青梅品种群品种混淆杂乱、遗传关系复杂难辨的问题,采用简单重复序列区间(ISSR)分子标记技术对福建永泰、仙游、秀屿以及诏安4个地区的11个青梅品种的遗传多样性和亲缘关系进行分析。结果表明,从100条ISSR引物中筛选出22条多态性引物,共扩增得到91个基因位点,其中多态性位点72个,占79.12%。Popgene软件分析结果表明,11个青梅品种的Nei′s遗传多样性指数平均值为0.257 6,有效等位基因数平均值为1.425 7,Shannon′s信息多样性指数平均值为0.393 3。非加权组平均法(UPGMA)聚类分析结果表明,11个青梅品种可以分为3大类,第一大类包括‘灯架山1号’‘灯架山2号’‘小喜村3号’和‘白粉梅’;第二大类包括‘小喜村1号’‘小喜村2号’‘红梅’‘大水梅’‘青梅1号梨子叶’和‘翁种’;第三大类只有‘原生种’1个品种。11个青梅品种的遗传一致度为0.615 4~0.824 2,遗传距离为0.193 4~0.485 5。福建永泰‘小喜村1号’和‘小喜村2号’的遗传一致度最高,遗传距离最小,亲缘关系最近;福建永泰‘灯架山1号’与福建诏安元山‘大水梅’的遗传一致度最低,遗传距离最大,亲缘关系最远。

Abstract :
Phyllanthus emblica L. is a nutritionally and medicinally valuable fruit tree with extensive germplasm diversity. This study evaluated 61 genetically diverse accessions collected from China and South Asia for 27 physicochemical traits, including 13 physical and 14 chemical attributes, to assess genetic diversity and identify elite germplasm for breeding. Substantial phenotypic variation was observed, with coefficients of variation (CV) ranging from 5.13 to 98.09% for physical traits and 1.51-59.82% for biochemical traits. Notably, single fruit weight (CV: 98.09%), flesh stickiness (63.65%), crude fat (59.82%), and polysaccharide content (50.12%) exhibited high variability. Fifteen traits conformed to a normal distribution and were categorized into five probability-based grades. Correlation analysis showed that pericarp strength was significantly positively correlated with soluble solids, titratable acid, total flavonoids, polyphenols, and tannin content, while flesh hardness correlated positively with crude fiber and total sugar. Principal component analysis (PCA) reduced the 27 traits to 12 representative indicators, accounting for 82.54% of total variation. Hierarchical clustering grouped the germplasms into three classes: Class I (19 accessions) with high antioxidant contents and strong textures; Class II (2 accessions) with the largest fruit size and high flesh toughness; and Class III (40 accessions) with high moisture content and lower values for most traits, suitable for juice processing. A comprehensive biochemical evaluation model was constructed using PCA-derived weights, and the top-performing accessions-PEF30, PEF41, PEF35, PEF17, and PEF31-demonstrated superior nutritional quality based on total sugar, polyphenols, vitamin C, and flavonoid contents. These accessions are promising candidates for functional food development, fresh consumption, and cultivar improvement. This study provides a robust analytical framework and theoretical foundation for the classification, conservation, and targeted utilization of P. emblica germplasm in breeding programs.

【目的】NAC转录因子广泛参与植物生长发育和应对逆境胁迫过程,被认为是植物次生细胞壁生物合成转录调控的一级开关。鉴定李PsNAC转录因子家族,探索其与皇冠李果实空腔褐变的关系。【方法】采用生物信息学方法,分析李PsNAC家族成员、理化性质、系统发育和基因结构等,并通过qRT-PCR技术,分析PsNACs在空腔褐变皇冠李果实中的表达模式。【结果】李PsNACs包含115个成员,不均匀地分布在8条染色体上,可分为17个亚族,与植物次生细胞壁合成相关的OsNAC003和OsNAC7亚族分别含6和10个PsNACs。PsNACs启动子区域含有丰富的激素响应元件和MYB结合位点。10个PsNACs在皇冠李空腔褐变果实中的表达量均高于非空腔褐变果实。【结论】本结果为研究PsNAC家族成员与皇冠李果实空腔褐变的具体关联性奠定了重要基础。

Abstract :
Orchid mycorrhizal (OM) fungi form mutualistic relationships with orchids, facilitating the uptake of minerals from the soil in exchange for sugars and lipids provided by the plant. In this study, we explored the role of the DoSWEET14 sugar transporter in Dendrobium officinale during OM symbiosis. Transcriptome sequencing revealed significant changes in gene expression in mycorrhizal roots, particularly the upregulation of DoSWEET14. Subcellular localization studies confirmed that DoSWEET14 is localized to the plasma membrane, suggesting its involvement in nutrient exchange between the orchid and OM fungi. Complementation assays using a yeast strain deficient in hexose transport demonstrated that DoSWEET14 has broad substrate specificity, efficiently transporting multiple monosaccharides. Additionally, overexpression of DoSWEET14 in Arabidopsis increased sugar content without affecting biomass, supporting its role in carbohydrate transport and storage during symbiosis. These findings suggest that DoSWEET14 plays a key role in regulating carbon allocation and stabilizing the mutualistic relationship between Dendrobium officinale and OM fungi.

【目的】明确柰李果实空腔褐变的发生规律,探索柰李果实空腔褐变与木质素积累的关系;并进行木质素生物合成途径的关键基因4CL基因家族鉴定与分析,为后续研究柰李果实空腔褐变的调控、防治和分子机制奠定基础。【方法】以‘皇冠’李盛花后10、25、40、55、70和85 d的果实为试材,研究果实空腔褐变的发生规律。采用石蜡切片观察组织结构变化,气相色谱-质谱联用(gas chromatography-mass spectrometry)技术明确木质素单体类型,紫外分光光度计测定木质素含量和木质素生物合成相关酶活性,qRT-PCR技术分析基因表达。结合NCBI、TBtools、MEME、MEGA11、SOPMA、SWISS-MODEL和PlantCARE等软件进行4CL基因家族的鉴定与分析。【结果】‘皇冠’李果实空腔始于转色期,成熟期空腔褐变率达到37.73%,转色期和成熟期空腔褐变果的果形指数显著高于非空腔褐变果。石蜡切片结果显示,空腔褐变部位的果肉被番红溶液染成红色,空腔褐变面积随果实发育增大。果肉木质素总量随果实发育逐渐增加,空腔褐变果的木质素总量极显著高于非空腔褐变果(P<0.001)。空腔褐变果的木质素单体主要包括愈创木基丙烷(22.6%)和紫丁香基丙烷(10.8%)。果实发育期,空腔褐变果的4CL和PAL酶活性均显著高于非空腔褐变果。空腔褐变果的8个Ps4CL表达均高于非空腔褐变果,2个PsPAL在空腔和非空腔褐变果的表达趋势基本一致,转色期空腔褐变果的PsPAL表达略高于非空腔褐变果。Ps4CL1、Ps4CL2、Ps4CL7和Ps4CL8可能参与李果肉木质素生物合成,Ps4CL基因家族均含内源激素类响应元件。共线性分析显示,李与拟南芥存在2对4CL同源基因,与桃有8对4CL同源基因。通过构建互作网络,鉴定到核心枢纽基因Ps4CL5、Ps4CL7和Ps4CL8,都分别至少有30个共表达基因。qRT-PCR结果表明,Ps4CL基因表达呈现组织特异性和时空特异性。【结论】‘皇冠’李果实空腔褐变发生的关键时期为转色期,明确了木质素总含量和单体类型、木质素生物合成相关酶活性及关键基因表达的变化,WGCNA鉴定到关键基因参与木质素生物合成途径。

The lily is a globally popular cut flower, and managing dormancy in lily bulblets is essential for continuous, year-round production. While nitric oxide (NO) has been shown to influence seed dormancy and germination, its role in dormancy release in lilies was previously unconfirmed. In this study, we investigated the effects of NO on dormancy release in lily bulblets using SNP and c-PTIO. Results showed that SNP treatment promoted dormancy release, while c-PTIO inhibited it. Measurement of endogenous NO levels in the bulbs, along with enzyme activities of NOS-like and NR and gene expression levels of LoNOS-IP and LoNR, confirmed that NO plays a role in promoting dormancy release in lilies. To further elucidate the physiological mechanisms involved, we analyzed H2O2 levels, antioxidant enzyme activities, endogenous hormone levels, and carbohydrate metabolism in the bulbs. Findings demonstrated that NO facilitated dormancy release by increasing H2O2, gibberellins (GAs), indole-3-acetic acid (IAA), zeatin riboside (ZR), reducing sugars, and by accelerating the metabolism of abscisic acid (ABA) and starch. This study provides a foundation for deeper investigation into the mechanisms underlying dormancy release in lily bulbs.

This study investigates the potential of 5 % pullulan (PUL)-based edible coating to extend the shelf life and maintain the postharvest quality of 'Furongli' plums stored at 25 +/- 2 degrees C and 75-80 % relative humidity for 12 days. PUL-coated plums exhibited 31 and 72.7 % lower weight loss rate and disease incidence compared to uncoated plums after 12 days of storage, respectively. The coating significantly reduced oxidative stress markers, including MDA (22 %), H2O2 (25.3 %), and O-2(center dot-) (25.1 %), while improving membrane integrity by reducing electrolyte leakage by 18 %. Additionally, the coating delayed carotenoid degradation by 81.3 % and preserved the fruit's visual appearance by reducing hue and chroma changes by 31.7 % and 30.8 %, respectively. Firmness was better maintained, along with reduced respiration and ethylene production in coated plums. Biochemical analysis revealed that PUL coatings enhanced antioxidant defense by increasing both the activities and gene expression of antioxidant enzymes (SOD, POD, and CAT) and maintained higher levels of ascorbic acid, total phenolics, flavonoids, and overall antioxidant activity. The coating preserved cell wall components like hemicellulose, cellulose, and protopectin while limiting increases in water-soluble pectin, thus delaying fruit softening. Additionally, PUL coating reduced activities and gene expression of pectin-degrading enzymes (PME and PG). Quality parameters, including TSS, TTA, and TSS-acid ratios, remained stable in coated fruits, ensuring better flavor and marketability. Overall, the study highlights the potential of PUL-based coatings for large-scale postharvest application, offering a sustainable and effective solution to extend fruit shelf life and reduce storage losses.

Abstract :
Plums are economically important stone fruits, valued for their nutritional quality and health-promoting properties. However, their quality and marketability are often compromised by a physiological disorder called fruit hollowness and browning (HB). This study employed an integrated physiological, biochemical, and proteomic approach to investigate the molecular mechanisms underlying HB in Huangguan plum. Huangguan plum fruit samples from seven developmental stages were analyzed for lignin content, key enzyme activities, and expression patterns of genes involved in lignin biosynthesis. The results demonstrated that HB-affected fruit accumulated significantly higher lignin levels, particularly during post-veraison stages, accompanied by elevated activities of lignin biosynthesis-related enzymes like phenylalanine ammonia-lyase and laccase. Expression analysis of lignin pathway genes revealed distinct transcriptional dynamics, with notable upregulation of PsPAL1 and PsLAC7 in HB fruit at critical developmental stages. Proteomic profiling identified 941 and 1249 differentially expressed proteins between HB and non-HB fruit at the veraison and ripe stages, respectively. KEGG enrichment analysis highlighted significant alterations in the phenylpropanoid biosynthesis pathway (map00940), which is central to lignification. These findings provide new insights into the physiological and molecular basis of HB in plum fruit and a scientific foundation for developing effective strategies to mitigate or address this disorder and improve fruit quality.

The Lanzhou lily (Lilium davidii var. unicolor) is a variant of the Sichuan lily of the lily family and is a unique Chinese 'medicinal and food' sweet lily. Somatic cell embryogenesis of Lilium has played an important role in providing technical support for germplasm conservation, bulb propagation and improvement of genetic traits. Somatic embryogenesis receptor-like kinases (SERKs) are widely distributed in plants and have been shown to play multiple roles in plant life, including growth and development, somatic embryogenesis and hormone induction. Integrating the results of KEGG enrichment, GO annotation and gene expression analysis, a lily LdSERK1 gene was cloned. The full-length open reading frame of LdSERK1 was 1875 bp, encoding 624 amino acids. The results of the phylogenetic tree analysis showed that LdSERK1 was highly similar to rice, maize and other plant SERKs. The results of the subcellular localisation in the onion epidermis suggested that the LdSERK1 protein was localised at the cell membrane. Secondly, we established the virus-induced gene-silencing (VIGS) system in lily scales, and the results of LdSERK1 silencing by Tobacco rattle virus (TRV) showed that, with the down-regulation of LdSERK1 expression, the occurrence of somatic embryogenesis and callus tissue induction in scales was significantly reduced. Finally, molecular assays from overexpression of the LdSERK1 gene in Arabidopsis showed that LdSERK1 expression was significantly enhanced in the three transgenic lines compared to the wild type, and that the probability of inducing callus tissue in seed was significantly higher than that of the wild type at a concentration of 2 mg/L 2,4-D, which was manifested by an increase in the granularity of the callus tissue.
