Abstract :
Pomelo is prone to juice sac granulation during ripening and storage processes, resulting in decreased moisture content, firm flesh, and a diminished flavor, which impacts its market value and consumer experience. Due to the complex reasons for juice sac granulation, combined with the relatively thick peel of pomelo, the detection methods for pomelo are lacking, and the difficulty of detection is high. To address the subjectivity and inability to achieve non-destructive detection in traditional granulation rate calculations, this study proposes a non-destructive detection method for assessing pomelo granulation quality based on nuclear magnetic resonance imaging (MRI) and hyperspectral imaging technology. The granulation rate is calculated using MRI technology to obtain a standardized and consistent granulation value. Combining the granulation value obtained from MRI technology with the spectral information of pomelo obtained through hyperspectral imaging technology, a spectral network called Shuffle_spcc is constructed using the ShuffleNet structure, widely employed in deep learning. Moreover, based on the granulation model Shuffle_spcc, a multi-output model is built using the principles of transfer learning to achieve the simultaneous prediction of multiple internal quality attributes, thus creating a predictive model. The experimental results indicate that, compared to Partial Least Squares Regression (PLSR) and Principal Component Regression (PCR) models, the Shuffle_spcc model achieves the best prediction results for granulation rate. The R in the training and validation sets reach 0.82 and 0.80, respectively. The root mean square error calibration (RMSEC) and root mean square error prediction (RMSEP) are 2.83% and 2.61%, respectively. The prediction of sugar content also yields excellent results, with R of 0.84 and 0.81 achieved on the training and validation sets, respectively. However, the prediction results for acidity and moisture content are comparatively poor. Therefore, the method based on nuclear magnetic resonance imaging and hyperspectral imaging techniques can be applied to detect juice sac granulation in pomelo. In subsequent studies, this provides a valuable reference for non-destructive quality inspection of the internal quality of large-sized fruits. © 2024, The Authors. All rights reserved.

【目的】探究减轻芦柑裂果的生理与分子机制。【方法】以10年生早熟芦柑(易裂品种)为试材,果面喷施0.3 g·L~(-1)螯合钙和10 mg·L~(-1)GA_3为处理,以清水为对照(CK)。测定裂果率和果实外观品质,同时测定果皮ROS含量和抗氧化能力、细胞壁代谢酶活性和结构成分含量,以及抗氧化酶和细胞壁代谢酶相关基因表达差异。【结果】Ca+GA_3处理有效降低了裂果率,增大了芦柑果形指数、单果质量和果顶果皮厚度;提高了芦柑果皮中抗氧化酶SOD、CAT活性,降低了H_2O_2、O_2~■产生速率和MDA含量以及POD、PPO活性。同时,Ca+GA_3处理提高了芦柑果皮果胶、纤维素含量,降低木质素含量以及PME、PL、PG、CX和PAL、4CL、C4H活性;提高了果皮的延展性和韧性,减少了裂果的发生。Ca+GA_3处理显著提高了抗氧化酶相关基因CrSOD、CrCAT在芦柑果皮中的相对表达量,降低了CrPOD、CrPPO以及细胞壁代谢相关基因CrPME、CrPL、CrPG、CrCX、CrPAL、Cr4CL、CrC4H的相对表达量。【结论】在生理与分子水平上明确了Ca+GA_3处理可以通过调控果皮活性氧代谢与细胞壁代谢来降低裂果率。研究结果对生产上预防和减轻芦柑裂果具有一定理论意义和应用价值。

【目的】研究芦柑果园土壤施用豆粕发酵液的应用效果,为芦柑提质增效生产提供参考。【方法】于芦柑果实膨大期,用稀释40倍的豆粕发酵液和等量清水分别进行浇施,分析豆粕发酵液对芦柑果园土壤酶活性和果实品质的影响。【结果】施用豆粕发酵液对芦柑果园土壤pH值无显著影响;土壤脲酶和酸性磷酸酶活性分别提高44.15%和20.86%,土壤蔗糖酶与过氧化氢酶活性无显著差异;芦柑叶片全磷和全钾含量分别增加5.71%和23.42%,全氮含量差异不显著;芦柑果实可溶性固形物含量无显著差异,可滴定酸含量降低7.99%,固酸比提高28.58%;单果质量和果皮厚度分别增加14.42%和16.00%,纵径提高5.56%;果实横径和果实硬度略有增加,但差异不显著。【结论】施用豆粕发酵液可以提高福建泉州永春芦柑果园酸性土壤的脲酶和酸性磷酸酶活性,促进土壤氮磷养分的释放,以及芦柑植株叶片磷钾矿质养分的吸收,并且对芦柑果实外观和内在品质均有改善效果。

Abstract :
Granulation of juice sacs is a physiological disorder, which affects pomelo fruit quality. Here, the transcriptome and ubiquitinome of the granulated juice sacs were analyzed in Guanxi pomelo. We found that lignin accumulation in the granulated juice sacs was regulated at transcription and protein modification levels. In transcriptome data, we found that the genes in lignin biosynthesis pathway and antioxidant enzyme system of the granulated juice sacs were significantly upregulated. However, in ubiquitinome data, we found that ubiquitinated antioxidant enzymes increased in abundance but the enzyme activities decreased after the modification, which gave rise to reactive oxygen species (ROS) contents in granulated juice sacs. This finding suggests that ubiquitination level of the antioxidant enzymes is negatively correlated with the enzyme activities. Increased H2O2 is considered to be a signaling molecule to activate the key gene expressions in lignin biosynthesis pathway, which leads to the lignification in granulated juice sacs of pomelo. This regulatory mechanism in juice sac granulation of pomelo was further confirmed through the verification experiment using tissue culture by adding H2O2 or dimethylthiourea (DMTU). Our findings suggest that scavenging H2O2 and other ROS are important for reducing lignin accumulation, alleviating juice sac granulation and improving pomelo fruit quality.

Pomelo is prone to juice sac granulation during ripening and storage, resulting in decreased moisture content, hard flesh, and bland flavor, which affects market value and consumer experience. Due to the complexity of the causes of juice sac granulation, coupled with the thick peel of pomelo, the detection means of pomelo is lacking and difficult to detect. In order to solve the shortcomings of the traditional granulation rate calculation, which has an error and cannot realize non-destructive detection. This study proposes a non-destructive detection of pomelo juice sac granulation and the calculation of granulation rate based on the nuclear magnetic resonance imaging (MRI) technology combined with the computer vision technology. Based on the strong analytical power of the MRI technology for moisture and the change of moisture in the granulated juice sac of pomelo, in this study, the cross section of pomelo was scanned by MRI scanner to obtain the corresponding MRI images. The MRI images of the cross section of pomelo were segmented using the threshold segmentation method to realize the calculation of the percentage of granulated pulp. The experimental results show that the segmentation threshold T1 = 102 can effectively segment the pulp from the peel, and the segmentation threshold T2 = 175 can effectively segment the granulated pulp from the normal pulp. The granulation rate of pomelo pulp can be calculated by calculating the ratio of the granulated pulp area to the total pulp area. Therefore, based on the MRI technology can be used for the detection of the granulation rate of pomelo juice sac, which provides a certain reference value for the subsequent non-destructive detection of the internal quality of large melons and fruits.

开展脐橙钼缺乏症的诊断及其光合生理响应研究,为柑橘缺钼诊断和合理施用钼肥提供依据。通过田间调查结合形态诊断、叶片分析诊断以及钼酸铵喷施试验,明确脐橙叶片的黄斑型黄化是缺钼引起,采集不同程度缺钼的脐橙叶片,研究缺钼胁迫对脐橙光合生理的影响。结果表明:轻度黄化、重度黄化和正常叶片的氮、钾、镁、硼含量均处于柑橘营养适宜范围,锌、钙含量三者间无显著差异,但轻度黄化和重度黄化叶片的钼含量低于柑橘钼缺乏的临界值(0.10 mg/kg),且均显著低于正常叶片。喷施0.05%钼酸铵可矫治脐橙叶片的黄化症状,进一步证实了缺钼是造成脐橙叶片黄化的原因。新叶出现近圆形、鲜黄色水渍状斑点或斑块,叶扭曲、叶缘向上向内卷曲是诊断脐橙缺钼的主要特征。缺钼显著降低脐橙叶片的光合色素含量、RUBP羧化酶活性、PSⅡ反应中心的潜在活性(F_v/F_o)、最大光化学效率(F_v/F_m)、电子传递能力(ETo/RC)以及量子产额(ETo/CSm),明显提高热耗散系数(DIo/RC),引起光合速率(P_n)的降低,叶片可溶性糖和蔗糖含量减少,光合作用受到抑制。

【目的】探究琯溪蜜柚果实汁胞粒化过程中木质素生物合成途径相关基因在果实发育过程中的表达特征,以揭示汁胞粒化过程中木质素合成的分子调控机制。【方法】选取2018年花后135、165、195、215 d 4个时期的琯溪蜜柚果实,测定汁胞粒化率及木质素含量以及分析转录组数据筛选到的木质素生物合成途径关键基因的差异表达和相关酶活性变化,并进行汁胞细胞壁木质素沉积的显微观察。【结果】在琯溪蜜柚花后195至215 d果实发育成熟期,木质素生物合成途径中CrPAL1、CrPAL3、CrC4H1、CrC4H2、Cr4CL、CrCCR3、CrCAD3、CrPOD2和CrPOD7等9个基因的表达量都显著增强,qRT-PCR验证结果与转录组数据一致。这期间果实汁胞粒化明显加速,木质素合成相关酶苯丙氨酸解氨酶(PAL)、肉桂酸-4-羟基化酶(C4H)、4-香豆酸辅酶A连接酶(4CL)、肉桂醇脱氢酶(CAD)、过氧化物酶(POD)的活性明显上升,木质素含量显著积累,番红染色显示木质素在蜜柚汁胞细胞壁中明显沉积。【结论】琯溪蜜柚木质素生物合成途径相关基因参与调控汁胞粒化过程中细胞壁木质素的合成。研究结果为今后琯溪蜜柚的品种改良和分子育种提供了理论基础。

该研究以‘南丰蜜橘’为试验材料,于高温来临前期(6月份)在果实表面喷施0.14 g/L螯合钙为钙处理,以喷施清水为对照,测定果皮相关基因表达水平和抗氧化酶活性以及活性氧含量变化,探究钙对‘南丰蜜橘’果皮在日灼发生期间的钙信号转导途径相关基因调节抗氧化系统基因的表达特征,以揭示外源钙减轻蜜橘日灼生理病害的分子与生理机制,为实际生产中喷施螯合钙防控蜜橘日灼生理病害提供理论依据。结果显示:(1)经钙处理后‘南丰蜜橘’果实的日灼发生率较对照显著降低了13.76%。(2)实时荧光定量PCR发现,钙处理显著提高‘南丰蜜橘’钙信号转导途径相关基因(CsNADK、CsMPK8、CsCPK、CsRBOH、CsCAMK1、CsGAD),以及抗氧化酶相关基因(CsSOD、CsCAT、CsPOD、CsAPX、CsGSR、CsGPX)在果皮中的相对表达量。(3)钙处理可显著提高‘南丰蜜橘’果皮抗氧化酶(SOD、CAT、POD、APX、GR、GPX)活性,并显著降低O-·2、H2O2和MDA的含量。(4)荧光染色发现,在7-10月高温和强直射光胁迫期,果皮细胞产生了过量的ROS绿色荧光,且随胁迫时间的延长荧光不断漫延积累,但钙处理的ROS荧光强度均显著弱于对照。研究表明,喷施螯合钙处理可通过增强蜜橘果皮钙信号转导途径相关基因的表达,调控抗氧化系统基因表达以及抗氧化酶活性,清除过量的ROS,维持果皮细胞ROS代谢平衡,从而提升蜜橘果皮的抗日灼能力;生产中于高温来临前期在果实表面喷施螯合钙,是一种简便有效的预防蜜橘日灼病害的技术措施。
