Calcium and Boron Foliar Fertilizer to Relieve Cracking of 'Liuyuezao' Pummelos SCIE

Abstract :

'Liuyuezao' pummelo is highly prone to cracking, which seriously affects its quality. The aim of this study was to illustrate the effect of foliar sprays of calcium (Ca) and boron (B) and their combined treatments on the fruit cracking and quality of 'Liuyuezao' pummelos during the fruit expansion period (40-55 days after flowering). Analysis of 12 mineral elements of the pericarp by ICP-MS revealed that the three treatments significantly increased the content of calcium and boron in the corresponding pericarp. These treatments effectively reduced the enzyme activities of pectin methylesterase (PME), polygalacturonase (PG), pectin lyase (PL), beta-galactosidase (beta-Gal), and cellulase (Cx) in the peel and down-regulated the expression of corresponding cell wall-degrading enzyme genes. Calcium, boron, and their combination treatments reduced water-soluble pectin (WSP) in the peel. Simultaneously, they inhibited the degradation of CDTA-soluble pectin (CSP) and Na2CO3-soluble pectin (NSP), thereby stabilizing the cell wall structure. Additionally, these treatments enhanced fruit skin break force (Bf) and elasticity (Ela), ultimately decreasing the fruit cracking rate. Diversification analysis showed that Ca and B elements significantly increased the sugar and vitamin C (Vc) content of 'Liuyuezao' pummelo fruits and reduced their organic acid content, thus improving fruit quality. The study provides new ideas on the use of fertilizer interactions to control fruit cracking and improve the quality of the pummelo fruit.

Abstract :

To explore the dynamic changes in 'Round leaf' jasmine tea (RT) during the scenting process, targeted metabolomics and HS-SPME-GC-MS were used to identify the non-volatile and volatile compounds, and compare them with double-petal jasmine tea (DT). The analysis of taste activity values indicated that Asp, Thea, and Glu played significant roles in shaping the flavor profile of RT. The volatile compounds methyl anthranilate, benzyl acetate, linalool, indole, methyl benzoate and methyl salicylate were the key aroma compounds of jasmine tea. The volatiles (E)-3-hexenyl acetate, (E)-beta-ocimene and benzeneacetaldehyde significantly contributed to the elegance and freshness of RT. Additionally, the aroma compounds in RT reached saturation during the fourth scenting process (S4). Notably, RT reached better quality in S4 while DT reached better quality in S5. It can provide a scientific basis for the optimization of RT quality and its application in production.

壳聚糖-柑橘精油复合处理对蓝莓果实采后贮藏保鲜效果的影响

以鲜食蓝莓为试材,采用浸泡方法研究了不同浓度壳聚糖与柑橘精油复合保鲜剂处理对蓝莓果实采后4℃贮藏过程中腐烂率、呼吸强度、可滴定酸含量、丙二醛含量、谷胱甘肽含量以及过氧化物酶活性、抗坏血酸过氧化物酶活性的影响,筛选壳聚糖与柑橘精油的最佳浓度组合。结果表明:与对照(蒸馏水浸泡)相比,壳聚糖与柑橘精油复合保鲜剂处理可有效降低蓝莓果实在贮藏期间的腐烂率和呼吸强度峰值,抑制呼吸作用和丙二醛的积累,减缓果实可滴定酸含量的下降,保持较高的谷胱甘肽含量,提高过氧化物酶和抗坏血酸过氧化物酶的活性。其中15 g/L壳聚糖和1.0%柑橘精油复合处理的保鲜效果最佳。该研究可为绿色、健康的天然防腐剂在蓝莓保鲜上的应用提供了参考。

A High-Quality Assembly and Comparative Analysis of the Mitogenome of Actinidia macrosperma SCIE

Abstract :

The mitochondrial genome (mitogenome) of Actinidia macrosperma, a traditional medicinal plant within the Actinidia genus, remains relatively understudied. This study aimed to sequence the mitogenome of A. macrosperma, determining its assembly, informational content, and developmental expression. The results revealed that the mitogenome of A. macrosperma is circular, spanning 752,501 bp with a GC content of 46.16%. It comprises 63 unique genes, including 39 protein-coding genes (PCGs), 23 tRNA genes, and three rRNA genes. Moreover, the mitogenome was found to contain 63 SSRs, predominantly mono-nucleotides, as well as 25 tandem repeats and 650 pairs of dispersed repeats, each with lengths equal to or greater than 60, mainly comprising forward repeats and palindromic repeats. Moreover, 53 homologous fragments were identified between the mitogenome and chloroplast genome (cp-genome), with the longest segment measuring 4296 bp. This study represents the initial report on the mitogenome of the A. macrosperma, providing crucial genetic materials for phylogenetic research within the Actinidia genus and promoting the exploitation of species genetic resources.

振动对三角梅苞片脱落和乙烯合成的影响

Abstract :

为探究振动对盆栽三角梅苞片脱落和乙烯生成的影响,对盆栽‘马尼拉小姐’三角梅(Bougainvillea×buttiana‘Miss Manila’)在振动、密闭环境和振动+密闭环境下苞片的脱落率及处理后4~24h的乙烯合成相关指标进行测定。结果表明,振动+密闭环境和振动处理后7 d三角梅苞片脱落率显著高于对照(无处理),苞径长0.5~1.4 cm的苞片(S1)和苞径长1.5~2.4 cm的苞片(S2)的乙烯释放量、1-氨基环丙烷-1-羧酸(ACC)含量、ACC合成酶(ACS)和ACC氧化酶(ACO)活性均显著高于对照,随着处理后时间的延长,总体呈先上升后下降的变化趋势。苞片S1和S2的BsSAMS、BsACS和BsACO基因相对表达量显著上升,与ACS活性和ACO活性的变化一致,总体呈先上升后下降的变化趋势。因此,盆栽三角梅在受到振动后苞片的脱落率、乙烯生成、乙烯生物合成关键酶活性、乙烯合成酶基因BsSAMS、BsACS、BsACO的相对表达量均显著上升,且密闭环境对振动的效应具有促进作用。

Abstract :

Jasminum sambac (L.) Aiton exists in single -petal (SP), double -petal (DP), and multi -petal (MP) phenotypes, which have a sweet floral scent and release aroma when the flowers open. A total of 103 volatiles were discovered from SP and DP jasmines. 16 volatiles were identified as their key odorants. Caryophyllene, geraniol, linalool, alpha-farnesene,beta-ocimene, methyl anthranilate, methyl salicylate, benzeneacetaldehyde and hexanal may form the overall aroma of those two jasmines during flowering. High -resolution transcriptional profiles of those two jasmines at four flowering stages were generated from our recently reported chromosome -level genome assembly, from which we identified 35 differentially expressed genes (DEGs) involved in terpenoid metabolic pathways and 10 DEGs involved in the phenylpropane/benzene metabolic pathway, which may shape the characteristic aroma of those two jasmines at the molecular level. Through co -expression network analysis of key odorants, we found that bZIP8, bHLH6, MYB4, and AP2/ERF3 may contribute to the regulation of aroma volatiles in those two jasmines. These results offer an important theoretical foundation for studying the regulation of aroma formation for SP and DP jasmines.

Abstract :

In this study, Shine Muscat (YG), Gaoqiansui (GQS) and Moldova (ME) were used to study the changes of anthocyanin and related genes expression in grape skin during fruit growth and development. CHS, F3'H, ANS and UFGT were the main genes with relatively high expression levels. Significant differences were observed in the expressions of CHS, F3'H, ANS, and UFGT among YG, GQS, ME in most fruit developmental stages. Large differences were discovered in the anthocyanin contents and components among three grape cultivars. During fruit development YG only contained trace anthocyanins. Peonidin, malvidin and cyanidin derivates are the main components in GQS, while the main components of ME are malvidin, petunidin, and delphinidin. Almost all the genes tested were positively correlated with anthocyanins between GQS and ME but irrelevant in YG during fruit development. High expression of CHS, F3'H, ANS, and UFGT and other genes probably promote the synthesis of anthocyanins. Overall CHS, F3'H, ANS, and UFGT were proposed as prominent candidate genes for anthocyanin synthesis in grape skin. This study provides reference for further understanding the physiological and biochemical mechanisms of anthocyanin synthesis in grape fruits which could be better for breeding programs.

三红蜜柚肉桂酰辅酶A还原酶基因的克隆与表达分析

Abstract :

【目的】肉桂酰辅酶A还原酶(cinnamoyl-CoA reductase,CCR)基因是木质素合成的关键基因,本研究旨在探明该基因在柚汁胞粒化过程中的作用机制。【方法】通过生物信息学、基因克隆与亚细胞定位、荧光定量PCR技术,筛选克隆三红蜜柚CmCCR基因,及其在两个品种(三红蜜柚和六月早1号柚)不同发育时期果实汁胞中的表达情况,并利用乙酰溴法测定柚汁胞木质素含量。【结果】从三红蜜柚汁胞转录组数据中筛选得到5个CmCCR基因,根据果实汁胞成熟期木质素含量与CmCCRs基因的转录水平变化的相关性,初步明确CmCCR1可能参与汁胞木质素代谢;克隆所得CmCCR1基因序列,其开放阅读框长度为1 017 bp;生物信息学分析发现,该基因编码的CmCCR1蛋白分子量为37.5 kDa,等电点(pI)7.06,属于稳定亲水蛋白,含有NAD(P)H/NAD(P)(+)结合(NADB)域,与甜橙CcCCR1有3个相同的Motif,分布情况也大致相同;进化树分析结果表明,CmCCR1与甜橙CcCCR1的亲缘关系最近,且都归于双子叶植物分组;亚细胞定位结果表明CmCCR1定位于内质网(膜)上,与软件预测的结果相吻合;实时定量PCR结果显示CmCCR1基因的表达量中三红蜜柚汁胞成熟阶段显著上升,在六月早1号柚汁胞发育过程中的相对表达量一直处于较低水平,且显著低于同一发育阶段三红蜜柚汁胞的相对表达量;将CmCCR1瞬时转化至六月早1号柚汁胞,发现瞬时转化的汁胞中CmCCR1基因的表达量显著上升,间苯三酚染色程粉红色,说明汁胞中出现了木质素的积累。【结论】CmCCR1基因可能参与调控三红蜜柚汁胞木质素等次级代谢产物的生物合成,其表达量的上升与柚汁胞粒化的发生有关。

Investigation of Effects of Cushioning Packaging on the Physiological and Quality Changes in Chinese Olive Fruits During Cold Chain Transportation SCIE

Abstract :

To gain a deeper understanding of the mechanisms by which cushioning packaging preserves the quality of Chinese olive fruits during cold chain transportation and extends their shelf life, this study simulated cold chain conditions and investigated the effects of cushioning packaging on the physiology, antioxidant capacity, and secondary metabolites of fruits during a 20-day shelf life. The results indicated that the decay rate in cushioning-packaging-treated fruit was 75% lower than that in the unbuffered packaging fruit at day 20 of shelf life. Simultaneously, cushioning packaging treatment mitigated the damage severity of the cell membrane structure and kept the cell membrane permeability at a low level, which was 15.34% lower than that in the unbuffered packaging fruit at day 20 of shelf life. Additionally, cushioning packaging effectively restrained the increases in malondialdehyde (MDA) content and alleviated the decline in chlorophyll and total flavonoid contents. It kept a balance among reactive oxygen species (ROS), antioxidant levels, and antioxidant enzyme activities, thereby reducing mechanical-damage-induced decay rates in Chinese olive fruits during the shelf life. Furthermore, metabolome analysis of Chinese olives during the shelf life was performed comparing those without buffered packaging to those with buffered packaging. The metabolome analysis found that the flavonoid biosynthetic pathway exhibited a higher accumulation of chrysin, neohesperidin, naringenin chalcone, sakuranetin, quercetin, catechin, and naringenin metabolites in cushion-packaging treatment compared to those without cushioning treatment. Furthermore, within the phenylalanine metabolic pathway, the accumulation of phenylalanine, p-coumaraldehyde, p-coumaric acid, coniferin and caffeoyl quinic acid metabolites was significantly higher in buffered-packaging groups compared to those without buffering. Together, these findings suggest that cushioning packaging can effectively sustain the integrity of cell membranes and enhance the shelf-life quality of Chinese olive fruits by regulating the balance of ROS and mitigating oxidative stress during cold chain transportation.

Abstract :

To explore the role of alleles in the aroma biosynthesis pathways of the tea plant, this study was conducted to identify and analyze the alleles of structural genes in the MVA, MEP, LOX and shikimate pathways based on Camellia sinensis 'Jinguanyin' haplotype-resolved genomes. The results showed that 69 pairs of alleles were identified in the four pathways, of which 24 genes showed allele-specific expression (ASE) in at least one tissue. TGY-allele and HD-allele correlations of buds, young leaves, and stems were higher in the six tissues of JGY. KEGG analysis revealed that 24 ASEGs were significantly enriched in GPI-anchor biosynthesis and monoterpenoid biosynthesis pathways. The results suggest that the specifically expressed alleles of CsMCT, CsGGPPS2, CsHMGS2, CsHMGR3, CsTPS10, CsADH4, CsDAHPS, CsCM3, and CsAS2 may play critical roles in the aroma formation of JGY cultivars. The qRT.PCR results showed that the bias of CsTPS1, CsMVD, CsDHQS, and CsAS2 alleles was basically consistent with the expression of the parents (TGY and HD) of JGY in young leaves. In addition, qRT.PCR was used to detect the expression of aroma-related genes during the manufacturing process of JGY and parents (TGY and HD) oolong tea. It showed that CsMVD, CsDXS2, CsTPS3, CsTPS9, CsTPS10, CsTPS13 and CsTPS14 play an essential role in the formation of the characteristic floral and fruit aromas of oolong tea.
