Optimization of Bougainvillea tissue culture system by response surface methodology SCIE

Abstract :

Bougainvillea is valued for its ornamental and medicinal properties, but its current propagation primarily relies on cuttings, which suffer from low efficiency. Although tissue culture offers faster propagation and is not limited by seasonal properties, the technical system remains underdeveloped. This study applies the Response Surface Methodology (RSM) to optimize the axillary bud induction and rooting stages of Bougainvillea tissue culture. By fitting a multivariate quadratic regression model to determine the relationship between factors and response values, the study aimed to refine the tissue culture system for Bougainvillea. The main findings of this study are as follows: (1) In the axillary bud induction stage, the optimal induction medium was MS (Murashige and Skoog) + 6-BA (6-Benzylaminopurine) 2.534 mg L- 1 + NAA (1-Naphthaleneacetic acid) 0.1 mg L-1, achieving a bud induction rate of 93.3 %. (2) The ideal rooting medium consists of 0.66 MS + IBA (Indole-3-butyric acid) 2.0 mg L- 1 + NAA 0.09 mg L-1. Upon optimizing the culture system, the bud induction rate during the primary culture stage rose to 93.3 %, and the rooting rate during the rooting stage reached 66.7 %. In conclusion, the optimization of culture media using a multifactorial approach proved more effective than the traditional twofactor method, and more accurately optimizes the media of different stages, which provides valuable insights for improving the Bougainvillea tissue culture system.

Abstract :

As a dioecious fruit tree, Kiwifruit requires pollination for female plants to bear fruit. This study used 2,4-Dichlorophenoxyacetic acid (2,4-D), a plant growth regulator, to induce fruit set, improve quality and increase kiwifruit yield in Actinidia chinensis var. 'Donghong' without pollination. Exogenous application of 2,4-dichlorophenoxyacetic acid (2,4-D) effectively triggered parthenocarpic fruit development in 'Donghong' kiwifruit (Actinidia chinensis var. 'Donghong'), while significantly improving fruit quality. The fruit set rate of parthenocarpic fruits reached 16.95% with 2,4-D treatment at 140 days after flowering; female flowers treated with 2,4-D showed differences in pollen structure compared to those treated with water. Also, no viability was detected in female flower pollen treated with 2,4-D. At the maturity stage, the fruits from the 2,4-D treatment were slightly smaller than those from the control group. The 2,4-D treatment group showed significant differences from the control group in skin hardness, flesh firmness and core hardness. There was no significant difference in the soluble solids content between the fruits treated with 2,4-D and those from the control group at maturity. However, the dry matter content of the 2,4-D treated fruits was lower than that of the pollinated fruits. The soluble sugar content of mature fruits from 2,4-D treatment reached 18.59 g/kg, and organic acid content was 18.10 g/kg, resulting in a significantly higher sugar-acid ratio than the control group. The seeds from 2,4-D treated fruits failed to germinate normally. This technology can facilitate the induction of fruit sets in actual production, which has significant practical value and potential in kiwifruit processing. From these results, important data were obtained regarding the establishment and production of kiwifruit orchards without including pollinator varieties, which take up a significant amount of space in the orchard establishment and cause a decrease in the yield obtained from the total area.

Abstract :

Kiwifruit, as a functionally dioecious fruit crop, necessitates pollination for female plants to achieve fruit set. In this study, exogenous application of the plant growth regulator 2,4-D successfully induced parthenocarpy, resulting in significantly higher fruit set rates compared to natural pollination while producing seedless fruits containing only empty seed coats. Through transcriptome analysis of the flower and young fruit, 10 key candidate genes were identified. We demonstrated that AcNAC056 and AcMADS1 transcriptionally activate AcGA2ox1 expression by binding to its promoter region, thereby elucidating the molecular mechanism underlying 2,4-D-induced seedless fruit formation. Cross-species validation in Arabidopsis revealed conserved expression patterns of AcGA2ox1, AcNAC056 and AcMADS1 following 2,4-D treatment, suggesting conservation of this regulatory mechanism. The study demonstrated that the application of 2,4-D-induced parthenocarpy technology in kiwifruit not only effectively addresses the fruit drop problem caused by insufficient pollination, but also enhances fruit yield. It provided effective technical support for kiwifruit cultivation, demonstrating broad application prospects and significant value in kiwifruit industry. The study contributed valuable insights into the molecular mechanism regulating parthenocarpy in kiwifruit.

De novo transcriptomes of floral bracts for 22 Bougainvillea accessions SCIE

Abstract :

Bougainvillea glabra is an ornamental tree or shrub with nearly 200 years of application in gardening and landscapes globally. Recently, the growing research interest in the applications of B. glabra extracts, such as medicinal applications, and synthetic materials for nutraceuticals, has led to the development of new techniques to be utilized for studying B. glabra. Moreover, the formations of polymorphic coloration and the mechanism of metamorphic bracts in B. glabra cultivars are worthy of study. However, the multi-omics information for B. glabra cultivars is lacking which hinders the progress of gene-level research and genetic applications. We sequenced the bracts transcriptomes of 22 B. glabra accessions and generated more than 80 Gb clean data. After de novo assembly and optimization, 174,758 unigenes (E90N50 = 2,473 bp) and annotation data were obtained. In addition, a total of 100,115 CDSs were detected. On average, each variety has 69,990 unigenes containing SNPs, among which 35,682 were annotated per variety. These transcriptome data are valuable for gene mining and expression experiments or other scientific areas.

Transcriptomic Insights into Higher Anthocyanin Accumulation in 'Summer Black' Table Grapes in Winter Crop Under Double-Cropping Viticulture System SCIE

Anthocyanins are responsible for grape (Vitis vinifera L.) skin color. To obtain a more detailed understanding of the anthocyanin regulatory networks across' the summer and winter seasons in grapes under a double-cropping viticulture system, the transcriptomes of 'Summer Black' grapes were analyzed using RNA sequencing. The average daily temperature during the harvest stage in the summer crop, ranging from 26.18 degrees C to 32.98 degrees C, was higher than that in the winter crop, ranging from 11.03 degrees C to 23.90 degrees C. Grapes from the winter crop accumulated a greater content of anthocyanins than those from the summer crop, peaking in the harvest stage (E-L38) with 207.51 mg100 g-1. Among them, malvidin-3-O-glucoside (Mv-3-G) had the highest monomer content, accounting for 32%. The content of Cy-3-G during winter increased by 55% compared to summer. KEGG analysis indicated that the flavonoid biosynthesis and circadian rhythm-plant pathways are involved in the regulation of anthocyanin biosynthesis during fruit development. Pearson's coefficient showed significant positive correlations between anthocyanin content and the VvDFR, VvUFGT, VvOMT, VvMYB, and VvbHLH genes in the winter crop; at full veraison stage, their expressions were 1.34, 1.98, 1.28, 1.17, and 1.34 times greater than in summer, respectively. The higher expression of VvUFGT and VvOMT led to higher contents of Cy-3-G and Mv-3-G in the winter berries, respectively.

Environmental airflow is an important factor affecting withering, primarily influencing the development of white tea's aroma. To examine how the aroma of white tea develops under the effect of environmental airflow during withering, tea shoots exposed to airflow (0.5 +/- 0.2 m/s) were sampled and analyzed for water distribution, antioxidant enzyme activity, and volatiles content. The results indicated that environmental airflow hastened the evaporation of free water from witheing leaves during the pre-withering stage, facilitated the transformation of free water to bound water, stimulated antioxidant enzyme activity. Additionally, volatiles associated with green and grassy notes in white tea were dramatically reduced, including 3-hexen-1-ol, methyl salicylate, and (Z)-3- hexen-1-ol acetate. In contrast, volatiles responsible for floral and fruity scents, including linalool and nerolidol, became more concentrated. These findings provide important theoretical insights into increasing white tea manufacturing efficiency and quality.

This review explores the evolving role of the tea green leafhopper, Empoasca onukii, in the tea industry, transitioning from a recognized pest to a significant enhancer of tea quality. Recent research highlights how its feeding behavior stimulates the production of desirable secondary metabolites, thereby improving the flavor profiles and market value of premium teas, particularly varieties like Taiwan's "Oriental Beauty". As consumer demand for unique and artisanal teas rises, the economic benefits associated with E. onukii are becoming increasingly evident, prompting farmers to adopt sustainable agricultural practices that often involve reduced pesticide use. Furthermore, the dynamic interplay between climatic factors, E. onukii population dynamics, and tea cultivation practices necessitates integrated pest management strategies that balance the beneficial and detrimental impacts of this leafhopper. Understanding these complexities not only fosters sustainable production methods but also opens niche markets, benefiting local economies and promoting both economic viability and environmental sustainability in the tea industry.

Abstract :

Dormancy is a critical stage for the development of floral organs in grapevine buds. In regions with insufficient low temperatures, the dormancy period may be prolonged, leading to delayed germination, irregular budbreak, and reduced yields. This indicates the imperative to formulate scientific and sustainable strategies to effectively address grape dormancy and enhance early budbreak quality, while also undertaking comprehensive studies on the critical factors affecting secondary inflorescence formation, thus ensuring consistent yield and quality in grape cultivation systems. This review summarizes recent advances in understanding stress-induced secondary flowering in grapevine, with a focus on postharvest practices like leaf-degreening, pruning, and hydrogen cyanamide application, as well as key mechanisms involving gene regulation, ROS signaling, hormonal crosstalk, and carbohydrate metabolism. Among these practices, leaf degreening promotes nutrient backflow and the differentiation of secondary flower buds; pruning stimulates budburst and increases the proportion of floral buds; and hydrogen cyanamide (HC) application improves the uniformity of budbreak while enhancing both the budbreak and flowering rates in grapevines. Incorporating a variety of grape varieties further facilitates yearround cultivation, thereby presenting a potential pathway for sustainable production. In conclusion, the review draws attention to current challenges and provides insights into future applications, to optimize viticultural practices in response to evolving market and environmental demands.
