Abstract :
‘油榛’是福建省建瓯市广泛种植的优良农家品种之一。本研究利用SSR分子标记对‘油榛’栽培群体进行遗传多样性分析,探索其变异情况,结果表明:由10个SSR引物组合对锥栗农家品种‘油榛’栽培群体的30个植株进行PCR扩增,共计扩增出26个条带,多态性条带26个,多态性条带占比100%,30个植株的遗传相似性系数在0.115 4~1.000 0之间,平均为0.877 2,表明锥栗农家品种‘油榛’具有较高的遗传多样性。为了解‘油榛’栽培群体30个植株的亲缘关系,对30个植株进行聚类分析,其中YZ5、YZ1单独各聚为一类,其余28个植株聚为一类。研究结果为在锥栗农家品种栽培群体中进行优树选择提供了理论依据。

Abstract :
油茶是中国重要的木本油料树种,具有较高的利用价值。本研究利用SRAP分子标记对10个福建省主栽普通油茶优良无性系进行分析,探讨其遗传差异,结果表明:从256对SRAP引物组合中筛选出12对引物对10个优良无性系进行PCR扩增,共获得186个条带,其中多态性条带166个,多态性条带的比率为89.2%。EM-2/ME-4引物组合扩增的条带多态性最高,为100%;多态性信息量最小值和最大值分别为0.280和0.394,多态性信息量平均值为0.330。10个优良无性系遗传相似系数最小值和最大值分别为0.591 4和0.774 2,遗传相似系数平均值为0.670 8。聚类分析表明,遗传相似系数为0.677 5处,10个普通油茶优良无性系可以划为4大类:闽43和闽46聚为第Ⅰ类,闽48、闽49、闽53聚为第Ⅱ类,闽50聚为第Ⅲ类,闽54、闽56、闽60、闽62聚为第Ⅳ类。这10个优良无性系之间存在着较高的遗传多样性水平,研究结果为普通油茶杂交育种和分子育种工作的开展提供参考。

Abstract :
Methyleugenol is widely found in essential oils from aromatic plants, attracting insects for pollination and possessing antimicrobial and antioxidant features. Methyleugenol content in the essential oil from Melaleuca bracteata leaves is 90.46 %, making it an excellent material for assessing methyleugenol's biosynthetic pathway. Eugenol synthase (EGS) and eugenol-o-methyltransferase (EOMT) represent key genes in methyleugenol's biosynthetic pathway but their respective functions are unclear. In this study, overexpression and virus-induced gene silencing (VIGS) techniques were applied to investigate the major regulatory gene among MbEGS1, MbEGS2, MbEGS3, and MbEGS4 in the biosynthesis pathway of methyleugenol. We also aimed to examine the coordination mechanism between EGS and EOMT in methyleugenol production by M. bracteata. The data revealed MbEGS4 transcription and methyleugenol content were most significantly increased after overexpression of the MbEGS1-4 gene in M. bracteata and Fragaria vesca. Conversely, MbEGS4 transcription and methyleugenol content were most significantly reduced after MbEGS1-4 gene silencing by VIGS in M. bracteata. These results underscore a major regulatory role for the MbEGS4 gene in the biosynthetic pathway of methyleugenol in M. bracteata. Remarkably, upon overexpression and silencing of MbEOMTs, the transcription levels of MbEGSs were notably upregulated, with MbEGS4 showing the highest increase. Collectively, these findings suggested the presence of a specific transcription factor bound to the promoters of MbEGS and MbEOMT, inducing their expression for the synergistic and efficient production of methyleugenol in M. bracteata.

Abstract :
Advancement of the oil tea industry requires the development of high-yielding and superior-quality varieties of Camellia oleifera, a major oilseed crop. However, traditional breeding methods, hampered by lengthy cycles and low selection accuracy, significantly constrain the breeding process. Identifying single nucleotide polymorphisms (SNPs) associated with target traits, and applying molecular marker-assisted selection (MAS) for these traits, can thereby shorten the breeding cycles and amplify the breeding efficiency. In this study, we utilized the hexaploid C. oleifera as the reference genome to identify high-quality SNPs and constructed a high-density genetic linkage map of C. oleifera that spanned 1566.733 cM, included 3097 SNPs, and was anchored to 15 linkage groups. Using interval mapping, we localized quantitative trait loci (QTLs) for 11 juvenile traits in C. oleifera, identifying 15 QTLs for growth traits and 24 QTLs for leaf traits, including 4 stable QTLs. The logarithm of odds (LOD) scores for individual QTLs ranged from 3.48 to 14.62, explaining 9.86-48.61% of the phenotypic variance. We further identified 2 SNPs associated with growth traits (marker11-951 and marker12-68) and 10 SNPs associated with leaf traits (marker11-276, marker11-410, marker11-560, marker13-16, marker13-39, marker13-110, marker13-731, marker14-701, marker14-910, and marker14-1331). These results provide valuable insights into the genetic mapping of key traits in C. oleifera and will contribute to the development of new varieties with high yield and superior quality in the future.

为探究‘油榛’栽培群体不同个体间的变异情况,筛选优良单株。以福建省建瓯市农家品种‘油榛’ 30株个体为材料,对其进行农艺性状和栗疫病抗性的测定,进一步作变异分析、方差分析、相关性分析,并对主要性状进行标准化排序,评选优良单株。结果表明:各性状平均变异系数介于6.02%~21.28%之间,坚果高度变异系数最小,果苞厚变异系数最大。方差分析表明各性状都存在显著(P<0.05)或极显著(P<0.01)差异。经标准化排序最终筛选出果实品质优良单株为YZ7、 YZ19、YZ28、 YZ21、 YZ4,抗病性优良单株为YZ5、 YZ30。农家品种‘油榛’栽培群体变异丰富,选择潜力较大,研究结果可为锥栗丰产优质新品种选育提供参考。

Juice sac granulation (a physiological disorder) leads to large postharvest losses of pomelo (Citrus maxima). Previous studies have shown that juice sac granulation is closely related to lignin accumulation, while the molecular mechanisms underlying this disorder remain elusive in pomelo. Our results showed that the lignin content in NC (near the core) and FC (far away from the core) juice sacs overall increased from 157 DPA (days post anthesis) to 212 DPA and reached a maximum at 212 DPA. Additionally, the lignin content of NC juice sacs was higher than that of FC juice sacs. In this study, we used transcriptome-based weighted gene co-expression network analysis (WGCNA) to address how lignin formation in NC and FC juice sacs is generated during the development of pomelo. After data assembly and bioinformatic analysis, we found a most correlated module (black module) to the lignin content, then we used the 11 DEGs in this module as hub genes for lignin biosynthesis. Among these DEGs, PAL (phenylalanine ammonia lyase), HCT (hydroxycinnamoyl-CoA shikimate/quinate hydroxycinnamoyl transferase), 4CL2 (4-coumarate: CoA ligase), C4H (cinnamate 4-hydroxylase), C3 ' H (p-coumarate 3-hydroxylase), and CCoAOMT1 (caffeoyl CoA 3-Omethyltransferase) were the most distinct DEGs in granulated juice sacs. Co-expression analysis revealed that the expression patterns of several transcription factors such as MYB, NAC, OFP6, and bHLH130 are highly correlated with lignin formation. In addition, the expression patterns of the DEGs related to lignin biosynthesis and transcription factors were validated by qRT-PCR, and the results were highly concordant with the RNA-seq results. These results would be beneficial for further studies on the molecular mechanism of lignin accumulation in pomelo juice sacs and would help with citrus breeding.

Abstract :
Actinidiaceae, an economically important plant family, includes the Actinidia, Clematoclethra and Saurauia genus. Kiwifruit, with remarkably high vitamin C content, is an endemic species widely distributed in China with high economic value. Although many Actinidiaceae chloroplast genomes have been reported, few complete mitogenomes of Actinidiaceae have been studied. Here, complete circular mitogenomes of the four kiwifruit species and Saurauia tristyla were assembled. Codon usage, sequence repeats, RNA editing, gene transfers, selective pressure, and phylogenetic relationships in the four kiwifruit species and S. tristyla were comparatively analyzed. This research will contribute to the study of phylogenetic relationships within Actiniaceae and molecular barcoding in kiwifruit.

以多花水仙金盏银台3年生水仙花种球为试材,研究乙烯作用抑制剂1-甲基环丙烯(1-MCP)处理对水仙花开放过程中ACC氧化酶活性的影响,以期为水仙花花期调控研究提供理论依据。试验分别设置处理1 (1-MCP处理):在水仙花抽穗高6~7cm时用1-MCP包裹水仙花剑茎,处理2 (乙烯利+1-MCP处理):在种植前15d将种球用乙烯利熏烟处理后,待水仙花抽穗高6~7cm时用1-MCP包裹水仙花剑茎,处理3 (CK):水仙花直接种植,不作任何处理。在花期采用酶联免疫分析法测定各处理水仙花不同开花时期和不同部位器官ACC氧化酶活性。结果表明:1-MCP处理的水仙花ACC氧化酶活性从花苞期到初花期呈下降趋势,从初花期到盛花期活性稍有上升,后又下降,但相对稳定,初花期、盛花期和衰老期ACC氧化酶活性差异不显著。与1-MCP处理相比,乙烯利+1-MCP处理的水仙花ACC氧化酶活性一直处在相对较高的水平,在盛花期到衰老期呈现显著上升趋势且维持在一个较高水平值;CK处理水仙花的ACC氧化酶活性从花苞期到初花期显著增加,而在后3个时期无显著性变化,但其均比1-MCP处理和乙烯利+1-MCP处理的ACC氧化酶活性高。衰老期乙烯利烟熏后再进行1-MCP处理显著提高水仙花全花及子房部位ACC酶活性,1-MCP处理和乙烯利+1-MCP处理的水仙花花瓣ACC氧化酶活性有所提高。

利用生物信息学方法,从全基因组水平鉴定枇杷MADS-box基因家族成员,并对其系统进化关系、保守结构域、基因结构、基因复制和启动子顺式作用元件进行分析;同时,结合转录组数据和qRT-PCR结果,分析鉴定出的MADS-box基因家族成员在不同品种和不同成熟阶段的表达情况.本研究共鉴定出89个枇杷MADS-box基因(命名为EjMADS1~EjMADS89),其N端都含有MADS结构域.系统发育树显示,Ⅰ型MADS-box基因有31个,Ⅱ型有58个.Ⅱ型MADS-box基因可划分为14个亚族,同属于一个亚家族的基因有着相似的保守基序.MADS-box基因以不同密度分布在第1~17号染色体上,存在12对串联重复基因.转录组分析表明,在果实成熟过程中,有8个MADS-box基因在果实成熟期间差异表达,其中,EjMADS26、EjMADS43和EjMADS41这3个基因序列分别与其他调控果实成熟的SEP亚族基因高度相似,且它们的启动子中都含有赤霉素和脱落酸响应元件.另外,EjMADS37和EjMADS05在不同品种间(特早熟/中晚熟)有明显表达差异.综上,推测EjMADS26、EjMADS43、EjMADS41、EjMADS37和EjMADS05对果实成熟有一定的调控作用.

百香果产量问题是当前百香果产业面临的突出问题,尤其是上半年产量是决定果农收益的关键。本文就如何提高黄金百香果上半年产量,提出了一些栽培方法,涉及大苗定植、科学密植、高垅高墩种植等栽培技术,主要目的是提早增加单位面积的结果枝数量,从而提高早期产量,为广大百香果种植户实现早产丰产提供技术参考。
