一种猕猴桃属植物体细胞有丝分裂中期染色体制片及其制备方法 ipsunlight

Abstract :

本发明涉及生物技术领域,特别是涉及一种猕猴桃属植物体细胞有丝分裂中期染色体制片及其制备方法。本发明提供了一种猕猴桃属植物体细胞有丝分裂中期染色体制片的制备方法,利用该方法制备得到的制片染色体形态较好且分散均匀。在本发明提供的制备方法中,首先采用0.002mol/L 8‑羟基喹啉溶液预处理猕猴桃属植物根尖3h。随后,采用特定酶液酶解100min,使猕猴桃属植物根尖细胞去壁效果很好,背景干净,材料分裂相较多,易观察到染色体,获得的染色体形态较好,分散均匀,便于猕猴桃属植物后续染色体计数及核型分析(如染色体荧光原位杂交)等染色体遗传研究。

Abstract :

【目的】NAC转录因子广泛参与植物生长发育和应对逆境胁迫过程,被认为是植物次生细胞壁生物合成转录调控的一级开关。鉴定李PsNAC转录因子家族,探索其与皇冠李果实空腔褐变的关系。【方法】采用生物信息学方法,分析李PsNAC家族成员、理化性质、系统发育和基因结构等,并通过qRT-PCR技术,分析PsNACs在空腔褐变皇冠李果实中的表达模式。【结果】李PsNACs包含115个成员,不均匀地分布在8条染色体上,可分为17个亚族,与植物次生细胞壁合成相关的OsNAC003和OsNAC7亚族分别含6和10个PsNACs。PsNACs启动子区域含有丰富的激素响应元件和MYB结合位点。10个PsNACs在皇冠李空腔褐变果实中的表达量均高于非空腔褐变果实。【结论】本结果为研究PsNAC家族成员与皇冠李果实空腔褐变的具体关联性奠定了重要基础。

‘皇冠’李果实空腔褐变与木质素积累的关系及其4CL基因家族鉴定

【目的】明确柰李果实空腔褐变的发生规律,探索柰李果实空腔褐变与木质素积累的关系;并进行木质素生物合成途径的关键基因4CL基因家族鉴定与分析,为后续研究柰李果实空腔褐变的调控、防治和分子机制奠定基础。【方法】以‘皇冠’李盛花后10、25、40、55、70和85 d的果实为试材,研究果实空腔褐变的发生规律。采用石蜡切片观察组织结构变化,气相色谱-质谱联用(gas chromatography-mass spectrometry)技术明确木质素单体类型,紫外分光光度计测定木质素含量和木质素生物合成相关酶活性,qRT-PCR技术分析基因表达。结合NCBI、TBtools、MEME、MEGA11、SOPMA、SWISS-MODEL和PlantCARE等软件进行4CL基因家族的鉴定与分析。【结果】‘皇冠’李果实空腔始于转色期,成熟期空腔褐变率达到37.73%,转色期和成熟期空腔褐变果的果形指数显著高于非空腔褐变果。石蜡切片结果显示,空腔褐变部位的果肉被番红溶液染成红色,空腔褐变面积随果实发育增大。果肉木质素总量随果实发育逐渐增加,空腔褐变果的木质素总量极显著高于非空腔褐变果(P<0.001)。空腔褐变果的木质素单体主要包括愈创木基丙烷(22.6%)和紫丁香基丙烷(10.8%)。果实发育期,空腔褐变果的4CL和PAL酶活性均显著高于非空腔褐变果。空腔褐变果的8个Ps4CL表达均高于非空腔褐变果,2个PsPAL在空腔和非空腔褐变果的表达趋势基本一致,转色期空腔褐变果的PsPAL表达略高于非空腔褐变果。Ps4CL1、Ps4CL2、Ps4CL7和Ps4CL8可能参与李果肉木质素生物合成,Ps4CL基因家族均含内源激素类响应元件。共线性分析显示,李与拟南芥存在2对4CL同源基因,与桃有8对4CL同源基因。通过构建互作网络,鉴定到核心枢纽基因Ps4CL5、Ps4CL7和Ps4CL8,都分别至少有30个共表达基因。qRT-PCR结果表明,Ps4CL基因表达呈现组织特异性和时空特异性。【结论】‘皇冠’李果实空腔褐变发生的关键时期为转色期,明确了木质素总含量和单体类型、木质素生物合成相关酶活性及关键基因表达的变化,WGCNA鉴定到关键基因参与木质素生物合成途径。

Abstract :

Plums are economically important stone fruits, valued for their nutritional quality and health-promoting properties. However, their quality and marketability are often compromised by a physiological disorder called fruit hollowness and browning (HB). This study employed an integrated physiological, biochemical, and proteomic approach to investigate the molecular mechanisms underlying HB in Huangguan plum. Huangguan plum fruit samples from seven developmental stages were analyzed for lignin content, key enzyme activities, and expression patterns of genes involved in lignin biosynthesis. The results demonstrated that HB-affected fruit accumulated significantly higher lignin levels, particularly during post-veraison stages, accompanied by elevated activities of lignin biosynthesis-related enzymes like phenylalanine ammonia-lyase and laccase. Expression analysis of lignin pathway genes revealed distinct transcriptional dynamics, with notable upregulation of PsPAL1 and PsLAC7 in HB fruit at critical developmental stages. Proteomic profiling identified 941 and 1249 differentially expressed proteins between HB and non-HB fruit at the veraison and ripe stages, respectively. KEGG enrichment analysis highlighted significant alterations in the phenylpropanoid biosynthesis pathway (map00940), which is central to lignification. These findings provide new insights into the physiological and molecular basis of HB in plum fruit and a scientific foundation for developing effective strategies to mitigate or address this disorder and improve fruit quality.

Magnesium's impact on fruit quality of loquat: Insights into sugar and acid dynamics SCIE

Abstract :

The flavor and taste of loquat fruits are influenced by soluble sugars (fructose, glucose, and sucrose) and organic acids, with malic acid being crucial. This study explored the effect of foliar magnesium sulfate (MgSO4) application on the sugar -acid profile and associated physiological and molecular factors in loquat fruits. Varied concentrations of MgSO4 (1 %, 2 %, 3 %) and a control (water spray) were tested. The application of 2 % MgSO4 notably enhanced fruit weight and size, increased the sugar -acid ratio by 5.58 -fold, elevated total soluble solids by 53.81 %, and reduced titratable acidity by 72.45 %. The increase in fructose and glucose (by 107.20 % and 77.50 %, respectively) and decrease in malic acid (by 43.65 %) were significant with 2 % MgSO4 treatment. Enzymatic and gene expression analyses revealed significant upregulation of SPS, SS, HK, and FK activities, and corresponding genes, particularly with 2 % MgSO4, while PEPC activity and its associated genes were downregulated. The study also identified a positive correlation of NAD-MDH with malic acid content and varied expression of NAD-ME and NAD-MDH genes, indicating their roles in malic acid biosynthesis. These findings demonstrate that MgSO4 application substantially modified the enzymatic and genetic profile related to sugar and acid metabolism in loquats, leading to improved fruit quality with higher sugar content and reduced acidity. This study offers valuable insights for enhancing loquat fruit quality through magnesium -based horticultural practices.

Citrus is a globally grown fruit crop because of the nutritional benefits. Climate change drastically affects plant physiology, morphology, and anatomy. Physiological alterations within plants allow them to adapt to changing environments, however, the degree and quantification of these modifications differs with varieties. Ten exotic and local citrus rootstocks were evaluated based on morphology, physiology, and anatomical response to different heat stress conditions to identify best suitable rootstocks in climate change scenarios. Brazilian sour orange and Gadha dahi showed heat stress tolerance through maintaining plant growth, retention of root and shoot moisture contents, higher photosynthetic pigments and leaf water potential. Higher moisture contents of root and shoot were recorded (66.00 +/- 1.1% and 73.00 +/- 0.94%) in Brazilian sour orange at moderate levels of stress respectively. Which significantly reduces in Rangpur poona nucellar to (33.33 +/- 0.6% and 32.67 +/- 1.45%) when the level of stress was intensified. Under moderate and severe stress conditions the lowest chlorophyll a content was recorded in Kirrumakki nucellar (2.20 +/- 0.05 mg/g) and Brazilian sour orange (1.32 +/- 0.05 mg/g) respectively, due to reduced photosynthetic pigment, thylakoid membrane damage and alteration of chloroplast. Higher H2O2 and proline production were indication of tolerant rootstock to heat stress in Brazilian sour orange and Gadha dahi. Two highly tolerant (Brazilian sour orange and Gadha dahi) and two highly sensitive rootstocks (Rangpur poona nucellar and Sunki Bentake) from the experiment were selected for stem and leaf anatomical observations. Brazilian sour orange under heat stress was characterized with thicker cell epidermis which reduce the transpiration and evaporation losses in addition to having higher vascular bundle, xylem, and phloem length. These anatomical modifications ensured the transport of water and nutrients across the plant body which helped in maintaining plant vigor and growth. This study provides valuable knowledge in tolerant citrus rootstock to mitigate global warming.

A high-quality chromosome-scale genome assembly of blood orange, an important pigmented sweet orange variety SCIE

Abstract :

Blood orange (BO) is a rare red-fleshed sweet orange (SWO) with a high anthocyanin content and is associated with numerous health-related benefits. Here, we reported a high-quality chromosome-scale genome assembly for Neixiu (NX) BO, reaching 336.63 Mb in length with contig and scaffold N50 values of 30.6 Mb. Furthermore, 96% of the assembled sequences were successfully anchored to 9 pseudo-chromosomes. The genome assembly also revealed the presence of 37.87% transposon elements and 7.64% tandem repeats, and the annotation of 30,395 protein-coding genes. A high level of genome synteny was observed between BO and SWO, further supporting their genetic similarity. The speciation event that gave rise to the Citrus species predated the duplication event found within them. The genome-wide variation between NX and SWO was also compared. This first high-quality BO genome will serve as a fundamental basis for future studies on functional genomics and genome evolution.

Background Guava is a fruit prone to rapid spoilage following harvest, attributed to continuous and swift physicochemical transformations, leading to substantial postharvest losses. This study explored the efficacy of xanthan gum (XG) coatings applied at various concentrations (0.25, 0.5, and 0.75%) on guava fruits (Gola cultivar) over a 15-day storage period.Results The results indicated that XG coatings, particularly at 0.75%, substantially mitigated moisture loss and decay, presenting an optimal concentration. The coated fruits exhibited a modified total soluble soluble solids, an increased total titratable acidity, and an enhanced sugar-acid ratio, collectively enhancing overall quality. Furthermore, the XG coatings demonstrated the remarkable ability to preserve bioactive compounds, such as total phenolics, flavonoids, and antioxidants, while minimizing the levels of oxidative stress markers, such as electrolyte leakage, malondialdehyde, and H2O2. The coatings also influenced cell wall components, maintaining levels of hemicellulose, cellulose, and protopectin while reducing water-soluble pectin. Quantitative analysis of ROS-scavenging enzymes, including superoxide dismutase, peroxidase, catalase, and ascorbate peroxidase, revealed significant increases in their activities in the XG-coated fruits compared to those in the control fruits. Specifically, on day 15, the 0.75% XG coating demonstrated the highest SOD and CAT activities while minimizing the reduction in APX activity. Moreover, XG coatings mitigated the activities of fruit-softening enzymes, including pectin methylesterase, polygalacturonase, and cellulase.Conclusions This study concludes that XG coatings play a crucial role in preserving postharvest quality of guava fruits by regulating various physiological and biochemical processes. These findings offer valuable insights into the potential application of XG as a natural coating to extend the shelf life and maintain the quality of guava fruits during storage.

本发明提供了一种昆虫病原线虫与化学农药联合防治红火蚁的方法,将昆虫病原线虫与联苯·噻虫胺或高效氯氰菊酯联合使用用于防治红火蚁。本申请技术方法对红火蚁的防治效果明显提升,能快速对红火蚁起到防治效果,并且在施放相同数量的Steinernema riobrave(简称Sr)昆虫病原线虫的情况下,加入高稀释倍数化学药剂的处理对红火蚁工蚁的致死率明显高于单独使用线虫的处理。相比单一使用昆虫病原线虫,克服其对红火蚁短期防治效果不佳的局限性。相比化学药剂防治不存在高用量和环境污染问题,大幅减少化学药剂用量,更加绿色安全。相比沸水法处理不用携带加热设备,更加便捷。

BACKGROUND: In agricultural pest management, especially in combatting the invasive red imported fire ant (RIFA, Solenopsis invicta), significant challenges emerge as a consequence of the constraints of solely depending on chemical insecticides or entomopathogenic nematodes (EPNs). The utilization of chemical insecticides carries environmental and ecological hazards, whereas EPNs, when applied independently, might not offer the immediate effectiveness necessary for adequate RIFA suppression. Acknowledging these hurdles, our study investigates a synergistic method that integrates EPNs with chemical insecticides, aiming to fulfill the urgent demand for more efficient and environmentally friendly pest control solutions. RESULTS: Our evaluation focused on the interaction between the highly pathogenic Steinernema riobrave 7-12 EPN strain and prevalent insecticides, specifically beta-cypermethrin and a mixture of bifenthrin and clothianidin, applied at highly diluted recommended concentrations. The findings revealed a notable increase in RIFA mortality rates when EPNs and these insecticides were used together, outperforming the results achieved with each method individually. Remarkably, this enhanced efficacy was especially evident at lower concentrations of the bifenthrin-clothianidin mixture, indicating a valuable approach to minimizing reliance on chemical insecticides in agriculture. Furthermore, the high survival rates of EPNs alongside the tested insecticides indicate their compatibility and potential for sustained use in integrated pest management programs. CONCLUSION: Our research underscores the effectiveness of merging EPNs with chemical insecticides as a powerful and sustainable strategy for RIFA management. This combined approach not only meets the immediate challenges of pest control in agricultural settings, but also supports wider environmental objectives by reducing the dependency on chemical insecticides.
