Abstract :
Tea oil is an important high-quality edible oil derived from woody plants. Camellia oleifera is the largest and most widely planted oil-producing plant in the Camellia genus in China, and its seeds are the most important source for obtaining tea oil. In current research, improving the yield and quality of tea oil is the main goal of oil tea genetic breeding. The aim of this study was to investigate the degree of genetic variation in an early crossing population of C. oleifera and identify single nucleotide polymorphisms (SNPs) and genes significantly associated with fruit traits, which can provide a basis for marker-assisted selection and gene editing for achieving trait improvement in the future. In this study, we selected a crossing population of approximately 40-year-old C. oleifera with a total of 330 samples. Then, ddRAD sequencing was used for SNP calling and population genetic analysis, and association analysis was performed on fruit traits measured repeatedly for two consecutive years. The research results indicate that over 8 million high-quality SNPs have been identified, but the vast majority of SNPs occur in intergenic regions. The nucleotide polymorphism of this population is at a low level, and Tajima's D values are mostly greater than 0, indicating that the change in this population was not suitable for the model of central evolution. The population structure analysis shows that the population has seven theoretical sources of genetic material and can be divided into seven groups, and the clustering analysis results support the population structure analysis results. Association analysis identified significant SNPs associated with genes related to the seed number of a single fruit and seed kernel oil content. Our findings provide a basis for molecular breeding and future genetic improvement of cultivated oil tea.

The Fagaceae family, comprising over 900 species, is an essential component of Northern Hemisphere forest ecosystems. However, genomic data for tropical and subtropical genera Castanopsis and Castanea remain limited compared to the well-studied oak. Here, we present chromosome-level genome assemblies of Castanopsis carlesii and Castanea henryi, with assembled genome sizes of 927.24 Mb (N50 = 1.57 Mb) and 780.10 Mb (N50 = 1.07 Mb), respectively, and repetitive sequence contents of 45.79% and 44.88%. Comparative genomic analysis revealed that the estimated divergence time between Castanopsis and Castanea was determined to be 48.3 Mya and provided evidence that both genera experienced only one of the ancient whole genome triplication event (gamma event) shared with most eudicots. The development of C. carlesii flower bracts and cupules was controlled by A- and E-class genes, suggesting that the cupules may originate from the bracts. Additionally, genes involved in sucrose and starch metabolism genes played distinct roles during C. carlesii fruit development. The amplification of the nucleotide-binding leucine-rich repeat (NLR) gene family in Fagaceae exhibited similarities, indicating that this expansion may be an adaptation to similar environmental pressures. This study provides valuable genomic resources for Asian Fagaceae and enhances our understanding of Fagaceae evolution.

Glyptostrobus pensilis is an endangered tree species, and detecting its genetic diversity can reveal the mechanisms of endangerment, providing references for the conservation of genetic resources. Samples of 137 trees across seven populations within Fujian Province were collected and sequenced using double-digest restriction site-associated DNA (ddRAD-seq). A total of 3,687,189 single-nucleotide polymorphisms (SNPs) were identified, and 15,158 high-quality SNPs were obtained after filtering. The genetic diversity in the populations was found to be low (Ho = 0.08630, He = 0.03475, pi = 0.07239), with a high genetic differentiation coefficient (Fst). When K = 4, the coefficient of variation (CV) error value was minimized, suggesting that the 137 individuals could be divided into four groups, with frequent gene flow between them. Principal component analysis (PCA) divided the seven populations into two major categories based on their north-south geographic location. The clustering was consistent with those obtained from the PCA. The main reasons for the endangerment of G. pensilis are likely to be poor natural regeneration, human disturbances, and climatic factors. It is recommended that methods such as in situ conservation, ex situ conservation, and the establishment of germplasm banks be implemented to maintain the genetic diversity of G. pensilis populations.

Tissue culture processes have been established for Dendrobium wilsonii Rolfe, a valued ornamental and medicinal orchid. Pods aged 180 days served as seed sources. These seeds underwent surface sterilization and were then aseptically germinated on Murashige and Skoog (MS) medium supplemented with 0.5 mg L-1 Benzyl adenine (BA), 0.5 mg L-1 Naphthalene acetic acid (NAA), 20 g L-1 Sucrose (Su), 7 g L-1 Agar, 1 g L-1 Activated charcoal (AC), 1 g L-1 Peptone and 1 g L-1 Hyponex No.1 incubated in a growth chamber at 25.2 degrees C with a 12-hour photoperiod and a light intensity of 2200 lx. Multiple shoot induction and proliferation was on MS medium in vitro produced shoots were rooted on MS medium with Hyponex No.1, 25 g L-1 Su, 1 g L-1 AC and 1.5 g L-1 Peptone. Peptone enhanced seedling growth and promoted clumped shoot development. The establishment of Dendrobium wilsonii Rolfe regeneration system provides a methodology for conserving germplasm resources and for commercial propagules production.

‘油榛’是福建省建瓯市广泛种植的优良农家品种之一。本研究利用SSR分子标记对‘油榛’栽培群体进行遗传多样性分析,探索其变异情况,结果表明:由10个SSR引物组合对锥栗农家品种‘油榛’栽培群体的30个植株进行PCR扩增,共计扩增出26个条带,多态性条带26个,多态性条带占比100%,30个植株的遗传相似性系数在0.115 4~1.000 0之间,平均为0.877 2,表明锥栗农家品种‘油榛’具有较高的遗传多样性。为了解‘油榛’栽培群体30个植株的亲缘关系,对30个植株进行聚类分析,其中YZ5、YZ1单独各聚为一类,其余28个植株聚为一类。研究结果为在锥栗农家品种栽培群体中进行优树选择提供了理论依据。

油茶是中国重要的木本油料树种,具有较高的利用价值。本研究利用SRAP分子标记对10个福建省主栽普通油茶优良无性系进行分析,探讨其遗传差异,结果表明:从256对SRAP引物组合中筛选出12对引物对10个优良无性系进行PCR扩增,共获得186个条带,其中多态性条带166个,多态性条带的比率为89.2%。EM-2/ME-4引物组合扩增的条带多态性最高,为100%;多态性信息量最小值和最大值分别为0.280和0.394,多态性信息量平均值为0.330。10个优良无性系遗传相似系数最小值和最大值分别为0.591 4和0.774 2,遗传相似系数平均值为0.670 8。聚类分析表明,遗传相似系数为0.677 5处,10个普通油茶优良无性系可以划为4大类:闽43和闽46聚为第Ⅰ类,闽48、闽49、闽53聚为第Ⅱ类,闽50聚为第Ⅲ类,闽54、闽56、闽60、闽62聚为第Ⅳ类。这10个优良无性系之间存在着较高的遗传多样性水平,研究结果为普通油茶杂交育种和分子育种工作的开展提供参考。

为筛选出生长良好且栗疫病抗性强的优良家系和优良单株,对12个锥栗半同胞家系的栗疫病抗性相关性状及生长性状进行变异分析、方差分析、相关性分析和遗传参数估算。结果表明,12个锥栗半同胞家系各性状的平均变异系数为20.19%,病斑面积的变异系数最大,为39.76%,叶形指数的变异系数最小,为11.14%。病斑宽、叶面积、叶周长、叶长、叶脉数在家系间具有显著差异(P<0.05),感病率、感病指数、叶宽在家系间具有极显著差异(P<0.01)。苗高与感病率呈显著负相关(P<0.05),与感病指数呈极显著负相关(P<0.01);叶长与感病率、感病指数呈极显著负相关(P<0.01)。筛选出JO14(‘白露仔’)、JO8(‘油桐仔’)、JO2(‘大苞榛’) 3个优良家系;筛选出9株(JO2-25、JO13-29、JO14-32、JO2-12、JO8-17、JO3-4、JO5-26、JO2-19、JO12-20)栗疫病抗性强的优良单株,4株(JO2-5、JO7-16、JO8-17、JO14-3)生长良好的优良单株,其中JO8-17抗性强且生长良好。

为研究浙江红花油茶实生群体的表型变异规律及优良单株筛选方法,加强对浙江红花油茶基因资源的保护和育种利用,以浙江红花油茶(Camellia chekiangoleosa)实生群体为研究对象,对其表型性状进行变异分析、相关性分析、回归分析和通径分析,结合主成分分析、模糊隶属函数法和多维空间坐标综合评定值累加法进行优良单株的选择。结果表明:单株之间变异丰富,25个表型性状的变异系数为5.74%~57.16%,多样性指数为1.934 6~2.095 9,产量经济性状的变异幅度最大,多样性指数最小;回归分析与通径分析确定了果实数、单果鲜籽重、单果干仁重和种仁含油率是影响单株产油量的关键表型性状,其中,单果干仁重是主要决策变量,果实数是主要限制变量;主成分分析将25个性状转化为6个主成分因子,累计贡献率为86.557%。根据以上研究结果筛选出9株优良单株,可作为浙江红花油茶丰产优质新品种的选育材料。

【目的】利用基因组和转录组数据鉴定分析花发育相关的MADS-box基因家族成员的结构特征及表达模式,为柳属花被缺失的分子机制研究提供理论依据。【方法】通过生物信息学方法鉴定柳属的长梗柳(Salix dunnii)、欧蒿柳(S.viminalis)、红皮柳(S.purpurea)的MADS-box基因家族成员,并对3种柳树花发育相关的MADS-box基因系统发育关系、基因复制和丢失、基因结构和理化性质进行分析。根据雌雄花芽的转录组数据分析长梗柳花发育相关基因在花芽不同发育阶段的表达模式。【结果】在长梗柳、欧蒿柳和红皮柳中分别鉴定出82、82和98个MADS-box家族基因。3种柳树的花发育相关基因经历了基因复制和丢失,相同亚类基因结构和保守结构域相似,但部分A、B、C和E亚类基因的K-box结构域缺失。长梗柳花发育相关基因的表达结果显示,在雌花和雄花中具有不同的表达模式。其中,B亚类SdMADS26和SdMADS4基因表达量均偏低。【结论】柳属植物花发育相关基因中部分成员的基因结构和表达模式发生了变化,推测这些变化可能与柳属花被缺失相关。

[目的 ]为掌握板栗、锥栗种内嫁接以及板栗和锥栗种间嫁接亲和性差异,筛选广亲和性砧穗组合应用于生产实践。[方法 ]以板栗、锥栗种内和种间4种嫁接类型1年生嫁接苗为材料,对其嫁接口愈合情况、生长性状、叶片表型及光合生理指标进行调查测定,并采用石蜡切片对嫁接口愈合情况进行细胞学观测,统计数据进行综合评价。[结果 ](1)4种嫁接类型中,板栗和锥栗种内嫁接成活率为35.71%~91.33%,且嫁接口愈合情况良好,而板栗和锥栗种间嫁接成活率仅为16.67%~45.00%,且存在部分嫁接口愈合不良现象;(2)板栗或锥栗种内嫁接组合苗高、地径和叶片数量性状表现均显著优于板栗和锥栗种间嫁接组合(p <0.05);(3)不同嫁接类型叶表型性状均存在差异,其中板栗种内嫁接组合叶面积和叶片厚度均显著大于板栗嫁接锥栗组合;锥栗种内嫁接组合叶柄长度显著大于锥栗嫁接板栗组合;(4)细胞学观察发现,板栗或锥栗种内嫁接砧穗间隔离层消失,砧、穗结合为一体,而种间嫁接砧穗间有隔离层出现;(5)各嫁接组合综合评价得分排名由高到低依次为A4>A7>A3>A2>A1>A8>A5>A6。[结论 ]板栗和锥栗种间嫁接存在一定的亲和性,但普遍低于板栗或锥栗种内嫁接组合的亲和性。
