杉木ClHSP70基因的克隆及表达模式分析

Abstract :

本研究通过克隆、生物信息学和表达分析方法对杉木热激蛋白70(heat shock protein 70,HSP70)基因进行研究,为深入理解杉木HSP70基因的功能和杉木遗传改良以及其可持续栽培提供科学依据。本研究选用杉木优良无性系“洋061”一年生苗作为试验材料,利用逆转录聚合酶链式反应(RT-PCR)克隆得到ClHSP70基因。运用Expasy软件等在线软件预测并分析ClHSP70蛋白的理化性质、跨膜螺旋域、信号肽、二级结构和三级结构;运用Cell-PLoc 2.0在线软件预测蛋白的亚细胞定位;运用Mega 11软件构建系统发育树;克隆ClHSP70基因并构建到pCAMBIA35s-EGFP载体中,分析ClHSP70蛋白的亚细胞定位;并利用实时荧光定量PCR(quantitative real-time PCR)对其表达水平进行分析。克隆得到的ClHSP70基因编码670个氨基酸,ClHSP70蛋白的分子式为C

Abstract :

为了解不同林分密度对杉木Cunninghamia lanceolate人工林土壤特性的影响,以福建省洋口国有林场的不同林分密度的实验林为研究对象,对不同林分密度下杉木人工林的土壤含水量、化学计量比、酶活及林下植被多样性进行研究,结果表明:1)各个密度对杉木人工林林下植被多样性中的物种丰富度指数和Shannon-Wiener指数无显著性影响(P>0.05),对Simpson指数和Pielou均匀度指数却有较大影响。2)不同林分密度对土壤化学性质有显著差异(P<0.05),随着林分密度的上升,土壤全碳含量先增加后减少,土壤全氮先下降后上升。3)不同林分密度对于土壤酶活性具有显著影响(P<0.05),其中,随着林分密度的上升,土壤的多酚氧化酶、酸性磷酸酶活性降低,而土壤脲酶和土壤蔗糖酶活性增加。4)不同林分密度对土壤微生物优势菌群存在显著影响(P<0.05),低林分密度一定程度上可以提高土壤微生物细菌群落结构多样性。在土壤细菌群落与理化性质相关性分析中,全磷(TP)、有效磷(AP)含量与土壤优势菌群Actinobacteria呈极显著相关(P<0.01)。全碳(TC)、全氮(TN)与优势菌群Zoopagomycota呈极显著相关(P<0.01)。总体而言,低林分密度是杉木人工林种植的最佳选择。

Effects of Different Stand Densities on the Composition and Diversity of Soil Microbiota in a Cunninghamia lanceolata Plantation SCIE

Abstract :

As sustainable forest management gains increasing attention, comprehending the impact of stand density on soil properties and microbial communities is crucial for optimizing forest ecosystem functions. This study employed high-throughput sequencing in conjunction with soil physicochemical analysis to assess the effects of stand density on soil physicochemical properties and microbial community characteristics in Chinese fir plantations, aiming to elucidate the influence of density regulation on ecosystem services. Our results suggested that changes in soil physicochemical properties and microenvironmental conditions were key drivers of soil microbial diversity. Total carbon (TC), soluble nitrogen (SN), and light fraction organic matter decreased with increasing stand density, while total potassium (TK) and available phosphorus (AP) concentrations increased. The plot with a density of 900 trees ha-1 exhibited the highest bacterial diversity, in contrast to the plot with 1500 trees ha-1, which showed the lowest. The dominant microbial taxa were similar across different stand retention densities, with Acidobacteria, Proteobacteria, and Chloroflexi being the predominant bacterial phyla and Ascomycota and Basidiomycota being the main fungal groups. Significant positive correlations were observed between soil microbial community structures and environmental factors, particularly with respect to soil phosphorus and nitrogen content. The present study demonstrated that reduced stand densities modulated soil nutrient content and enhanced bacterial diversity, thereby contributing to a more complex and stable soil ecosystem structure. These insights provide a scientific foundation for optimizing the management of Chinese fir plantations, thereby supporting the sustainable development of forest ecosystems.

Abstract :

Chinese fir (Cunninghamia lanceolata) is an important fast-growing tree species for timber production and ecological protection in China. Yet, its tissue culture for seedling propagation is hampered by low proliferation and poor quality. Light quality is vital for seedling proliferation and growth, but the regulatory mechanisms remain poorly understood. In this study, a transcriptome and metabolome were integrated to explore light quality's effects on adventitious shoot proliferation of tissue-cultured Chinese fir seedlings. The seedlings were grown under red, green, blue, and composite light-emitting diode conditions, with white light as the control. Results showed that blue and blue-dominant composite light enhanced proliferation by promoting auxin and cytokinin and increased biomass. Red light promoted shoot height, leaf area, and carotenoid content due to elevated gibberellin and reduced auxins and cytokinin levels but inhibited proliferation due to hormonal imbalances. Green light increased abscisic acid levels and suppressed growth. Transcriptome and metabolome analyses identified key pathways including plant hormone signal transduction, photosynthesis, and flavonoid and carotenoid biosynthesis. Weighted gene co-expression network analysis (WGCNA) identified four key genes regulated by light quality that further modulated hormone biosynthesis and signaling transduction. This research provided insights for optimizing Chinese fir seedling proliferation and growth, contributing to sustainable plantation management.

Genotype by environment interaction for growth traits of families of Acacia melanoxylon based on BLUP and GGE biplot SCIE

Abstract :

Acacia melanoxylon is a high-quality timber species renowned for its superior heartwood and wide global use. To assess its stability, adaptability, and productivity in Fujian Province, China, a regional trial involving 47 A. melanoxylon families was conducted across four sites. The study utilized best linear unbiased prediction (BLUP) breeding values were estimated using 5-year-old diameter at breast height (DBH) and tree height data, and genotype plus genotype-by-environment (GGE) biplots were used to evaluate families and trial sites. The biplot generated based on BLUP values for DBH revealed that the four trial sites clustered into two groups. Group 1 comprised sites ZX (Zhangpu Zhongxi State-owned Forest Farm, Zhangpu County), LX (Quanzhou Luoxi State-owned Forest Farm, Quanzhou City), and XQ (Nanping Xiqin Teaching Forest Farm, Nanping City), while Group 2 included site LS (Nan'an Luoshan State-owned Forest Farm, Nan'an City). Sites LX and XQ showed strong positive correlations with each other and moderate correlations with ZX, indicating similar family rankings. In contrast, LX was negatively correlated with LS, reflecting opposing trends in genotype performance. Site ZX exhibited high discriminative power and representativeness, identifying it as the most effective location for evaluating families. Families 38, 35, 9, 17, and 24 demonstrated a combination of high yield and stability, underscoring their potential for extensive cultivation. The measurement of DBH is more cost-effective and accurate compared to tree height, and it aligns with our breeding objectives. Therefore, this study primarily focused on DBH for selection, and the results provide a theoretical foundation for promoting Acacia melanoxylon in Fujian Province.
