Abstract :
Cryptomeria japonica var. sinensis is a valuable timber and ecological tree species in East Asia. Despite previous research that has revealed significant differences in tree height, diameter at breast height, and wood volume among C. japonica var. sinensis from different geographical sources, their genetic structure and phylogenetic relationship among different populations are still unable to accurately determined. Therefore, we extensively collected different geographical sources of Cryptomeria spp. in China (Fujian, Zhejiang area) and Japan, and performed their genetic structure and affinities by using simplified genome sequencing (GBS). A total of 306.5 Gb of sequence raw data and 875,329 high-quality SNPs were obtained, with an average of 4.26 Gb of raw data per sample. The results showed that the genetic variation of C. japonica var. sinensis populations was lower than that of C. japonica populations, and the genetic diversity of C. japonica var. sinensis in Fujian populations was slightly higher than that of Zhejiang populations. At the same time, molecular analysis of variance showed that the genetic variation of these two populations mainly came from within the population (99.6%). Furthermore, the cluster analysis results showed that C. japonica var. sinensis was clustered into five groups, and the clustering results were still consistent with the actual geographical distribution. All findings clarified the genetic background and structure of Cryptomeria spp. This will be a reference for the conservation, utilization, and genetic breeding improvement of C. japonica var. sinensis germplasm resources.

【目的】探究花绒寄甲(Dastarcus helophoroides)自然种群和商品化种群的发育特性及其雌、雄成虫形态特征之间的差异,为其种群的有效复壮和林间合理释放提供依据。【方法】以花绒寄甲自然种群和商品化种群为研究对象,比较各虫态生长发育特性;在体视显微镜下观察和比较雌、雄成虫的鞘翅端角区、肛板顶角角度、肛板两侧缘拐点位置、肛板长度与宽度之比、肛板刻窝共5个外部形态特征和生殖器构造的差异。【结果】花绒寄甲两个种群的卵—蛹历期存在显著差异,自然种群的幼虫期、预蛹期和蛹期均显著长于商品化种群。自然种群的卵孵化率和幼虫存活率以及蛹长、蛹宽和蛹质量显著大于商品化种群。采用单一特征鉴别性别时,依据肛板长度与宽度之比或肛板顶角角度鉴定的准确率最高,均为86.67%;综合多个特征鉴别性别时,依据肛板顶角角度和肛板长度与宽度之比鉴定的准确率最高,为88.33%。【结论】花绒寄甲商品化种群卵—蛹历期较自然种群缩短,且部分性状出现退化现象。依靠肛板顶角角度和肛板长度与宽度之比对该虫进行无损伤性别鉴定比较可靠。

Abstract :
In this study, a new stag beetle of the genus Dorcus MacLeay, 1819 is described and illustrated, namely Dorcus liyuani Qi & Zhan, new species, from Gansu Province, northwestern China. The new species is very similar to its related species, especially D. tianlongi Wang & Zhou, 2019 and D. liyingbingi Huang & Chen, 2013, in external characters, but can be clearly distinguished by male genital features. There is also a clear geographical isolation between the new species and the closely related species.

Abstract :
A new fossil stag beetle genus, Cretofigulus gen. nov., is identified and described from mid-Cretaceous Kachin amber from northern Myanmar. Two new species, C. caoqiangi sp. nov. and C. hollowayae sp. nov. are herein described and illustrated. Additionally, the morphological differences between Cretofigulus gen. nov. and the related extant genus Figulus Macleay, 1819 are discussed. Cretofigulus gen. nov. is the second fossil record of the subfamily Lucaninae from Burmese amber. Despite an evolutionary divergence of approximately 99 million years, Cretofigulus gen. nov. exhibits a striking morphological resemblance to extant Figulus. Key diagnostic features of Cretofigulus gen. nov., including reduced sexual dimorphism, suggest behavioral and ecological parallels with modern Figulini Burmeister, 1847, such as carnivorous feeding habits and adaptation to a monogamous lifestyle within decaying wood.

Simple Summary The molecular pathogenesis of Bipolaris fujianensis, a novel fungal pathogen causing shoot blight in Chinese fir (Cunninghamia lanceolata), remains unclear. Through comparative transcriptomics, we analyzed its gene expression during infection. Key findings include the identification of differentially expressed genes involved in transport, hydrolysis, carbon metabolism, and biosynthesis of secondary metabolites. Notably, we predicted the involvement of cytochrome P450 and a major facilitator superfamily (MFS) transporter in the production and transport of the diterpenoid toxin ophiobolin F. These insights advance the understanding of B. fujianensis virulence and may facilitate the development of control strategies against this disease.Abstract Bipolaris fujianensis is a novel pathogenic species causing Chinese fir (Cunninghamia lanceolata) shoot blight (CFSB), first discovered in Nanping City, Fujian Province. However, its molecular pathogenic mechanisms remain largely unknown. Elucidating theses mechanisms has the potential of aiding future developments in disease management and resistance breeding in Chinese fir. In this regard, we examined the expression pattern of B. fujianensis grown on PDA (BGPDA), and during infection of Chinese fir at 24 h (BGCF-E), 48 h (BGCF-M), and 5 d (BGCF-L) post inoculation. Comparative transcriptome analysis identified 4133 differentially expressed genes (DEGs), including 1778 upregulated and 2355 downregulated in BGCF compared with BGPDA. During the infection process, Gene ontology (GO) enrichment analysis indicated that transporters and hydrolases in the molecular function categories play essential roles. Kyoto encyclopedia of genes and genomes (KEGG) analyses showed glycolysis/gluconeogenesis, carbon metabolism, and secondary metabolite biosynthesis were the major enriched pathways. Furthermore, this pathogen could produce diterpenoid toxin ophiobolin F, with cytochrome P450 and MFS transport proteins likely involved in its biosynthesis and transport predicted by RT-qPCR.

松毛虫凸腿小蜂Kriechbaumerella dendrolimi是我国南方针叶林重大食叶害虫柳杉毛虫Dendrolimus houi的蛹期优势天敌。通过调查该蜂的自然寄主种类,明确其对不同寄主的选择性与适合度,为其在野外转主寄生情况和潜在的控害能力提供理论依据。为此,在柳杉毛虫为害的柳杉Cryptomeria fortunei林分内展开调查,采集并鉴定大型鳞翅目昆虫,记录松毛虫凸腿小蜂对不同种类蛹的寄生特性及其寄生不同蛹后的子代情况。结果表明,在受害林分中,共获得10种大型鳞翅目昆虫,具有不同的蛹期;松毛虫凸腿小蜂对不同种类的蛹存在选择性差异,优先选择柳杉毛虫蛹(占93.33%),且耗时最短(163.98s)。在寄生阶段,该蜂能准确定位、识别柳杉毛虫蛹等寄主,也能快速分辨非寄主如银杏大蚕蛾Saturnia japonica。从寄生结果来看,该蜂能在柳杉毛虫、乌桕大蚕蛾Attacusatlas、思茅松毛虫D.kikuchii、明目大蚕蛾Antheraea frithi、樗蚕Samia cynthia、樟蚕Saturnia pyretorum和马尾松毛虫D. punctatus这7种蛹内成功繁育子代,但其子代之间的生物学特性存在显著差异。除柞蚕Antheraeapernyi蛹外,柳杉毛虫蛹繁育的子代数量最多且雌蜂体型最大,马尾松毛虫蛹繁育的子代最少且雌蜂最小。研究表明,受害林分中存在着丰富的大型鳞翅目昆虫种类,理论上可供松毛虫凸腿小蜂完成其年生活史,有望实现林间持续防控的效果。

Abstract :
The Japanese fir, Cryptomeria japonica, is ecologically and commercially vital in China. However, infestations by Dendrolimus houi Lajonquiere larvae cause widespread dieback, mimicking mechanical damage effects, with unclear differential impacts and mechanisms. To address this, 6th instar larvae of D. houi were introduced to three-year-old C. japonica seedlings to induce pest infestation, while mechanical damage and non-infested pests were controlled. Then, next-generation sequencing techniques were employed, and transcriptome sequencing was conducted to analyze the distinct damage mechanisms responding to C. japonica. This study revealed 10,412 DEGs between pest infestation and controls and 5535 DEGs comparing pest infestation to mechanical damage. Functional enrichment analysis highlighted the involvement of these DEGs in crucial processes such as photosynthesis, amino acid and nucleotide metabolism, flavonoid biosynthesis, and plant hormone signaling pathways. In particular, 786 unique DEGs were discerned in pest-infested samples. Key enriched pathways, such as pyruvate and propionate metabolism, were pinpointed, with flavonoid biosynthesis potentially closely linked to pest-feeding inductions. These findings provided valuable insights into the molecular responses of C. japonica to D. houi infestation, laying the foundation for future research aimed at developing pest-resistant varieties of Japanese fir.

研究我国林业重要食叶害虫——柳杉毛虫保幼激素结合蛋白(JHBP),对于揭示其生长发育、化蛹、摄食、交配生殖及抗药性等生命活动具有重要意义。参照模式昆虫家蚕的41条JHBP蛋白序列,构建系统进化树,并对这些基因所编码的蛋白质二级结构、疏水性、蛋白质序列基序和三级结构等生物学特性进行分析,利用heatmap可视化工具分析柳杉毛虫JHBP在不同发育时期的表达水平。结果表明,柳杉毛虫JHBP基因共有JHBPa、JHBPc、JHBPd 3个亚家族,分别包含6、26、15个JHBP基因;蛋白序列至少含有1个JHBP结构域,且其中Dho05G001220蛋白序列不仅含有JHBP结构域,还含有Grp_allergen结构域,各蛋白序列的疏水性区域呈明显的聚集分布,亲水性区域的聚集分布则不明显; JHBP基因在柳杉毛虫的低龄和中龄幼虫、蛹和成虫中显著表达,表明JHBP明显参与调控不同虫态的发育或变态过程。研究结果可为进一步明确柳杉毛虫JHBP基因家族的生物学功能提供参考。

Kriechbaumerella dendrolimi (Hymenoptera, Chalcididae) is a dominant pupal parasitoid species of various significant pine caterpillars, including Dendrolimus houi Lajonquiere (Lepidoptera, Lasiocampidae), with great potential for utilization. So far, the mass rearing of K. dendrolimi has been successfully established using Antheraea pernyi (Lepidoptera, Saturniidae) pupae as alternative hosts and released in the forest to suppress D. houi populations. However, the outcome is still expected to be improved due to lower parasitism rates, which might be related to the autonomous immune function of A. pernyi pupae. In our study, we investigated the effects of K. dendrolimi parasitization on the immune responses of A. pernyi pupae by measuring the expression of key immune factors: superoxide dismutase (SOD), polyphenol oxidases (PPOs), Attacin, Lysozymes (LYSs), and serine proteases (PRSSs). Our results show that parasitization significantly upregulated these immune factors, with distinct temporal patterns observable between 4 and 48 h post-parasitization. This upregulation highlights a robust immune response, adapting over time to the parasitic challenge. These findings suggest that specific immune mechanisms in A. pernyi pupae are activated in response to K. dendrolimi, shedding light on potential targets for enhancing host resistance. Metabolomic analyses complemented these findings by illustrating the broader metabolic shifts associated with the immune response. Specifically, Attacin was significantly upregulated twice, hypothesizing that the parasitoid's venom contains at least two parasitic factors. Metabolomics analysis revealed a significant metabolite difference within parasitized A. pernyi pupae. The highest number of differential expression metabolites (DEMs) was observed at 16 h post-parasitism (1184 metabolites), with fewer DEMs at 8 h (568 metabolites) and 32 h (693 metabolites), suggesting a close relationship between parasitism duration and the number of DEMs. These fluctuations reflected the fundamental process of immune interaction. KEGG enrichment results showed that the DEMs were mainly enriched in energy metabolism and immune-related pathways, indicating that parasitism is a process of continuous consumption and immune interaction in the host. These DEMs could also become future targets for regulating the immune functions of A. pernyi pupae and could provide reference data for optimizing mass-rearing techniques.
