A comprehensive proteomic map revealing the regulation of the development of long-duration, red butterfly-shaped fruit in Euscaphis japonica SCIE

Abstract :

Fruit features are crucial for plant propagation, population growth, biodiversity preservation, and evolutionary survival. However, the synergistic regulatory mechanisms underlying the development of fruit traits such as color, shape and duration are unclear. Euscaphis japonica, whose fruits have a red-winged pericarp and persist for a long period of time, is an important ornamental plant in eastern Asia. In this study, we present a complete proteome spanning multiple time points and the phosphoproteome landscape of E. japonica fruit during the maturation and ripening phases. Quantitative evaluation via proteome and phosphoproteome analysis revealed three distinct phases that are consistent with the fruit maturation and ripening stages on a longitudinal time scale. The two-way proteomics analysis revealed functionally important biological events, including anthocyanin accumulation and phytohormone and light signal transduction. Importantly, our integrated analysis, along with experimental validation and phytohormone treatments, suggested that alterations in EjPHYBS21/S37 and EjPHOT1S394/S429 phosphorylation may lead to auxin accumulation and the inhibition of ethylene biosynthesis, thereby initiating the development of long-duration, red butterfly-shaped E. japonica fruit. Our study reveals a mechanism of E. japonica fruit formation that highlights plant adaptive strategies that potentially evolved through interactions with frugivores.

圆齿野鸦椿ARF基因家族鉴定和功能分析

Abstract :

【目的】为进一步探究ARF基因在圆齿野鸦椿生长与发育过程中的作用机制提供科学依据。【方法】通过圆齿野鸦椿全基因组,利用生物信息学,鉴定其ARF基因家族成员,分析EkARF基因的理化性质、染色体定位、进化关系、基因结构、所包含的结构域以及顺式作用元件。探究EkARF基因在不同组织中的表达模式并进行功能预测。【结果】共鉴定出29个EkARF基因(EkARF1~EkARF29),分为4个亚族;除LG02染色体外,其余染色体上均有分布。29个ARF基因的氨基酸数量介于432~1110个,相对分子质量在48 159.66~122 724.62 u,均为亲水性蛋白,其亚细胞定位预测均在细胞核内。29个EkARF蛋白均含有DBD、Auxin_resp结构域,其中21个含有CTD结构域,其基因启动子顺式作用元件与植物光响应、激素的调节、防御机制和生长发育密切相关。EkARF基因在不同组织中表达存在差异,EkARF10在种子中高表达,可能参与种子发育调控;EkARF5随着果实的成熟表达量逐渐降低,可能在果实发育过程中起重要调控作用;EkARF16、EkARF20随着果实发育表达量上调,尤其是EkARF20在果实脱落期呈高表达,与果实成熟变红后仍挂果超130 d不脱落的现象一致,可能是果实发育的关键调控基因。【结论】本研究揭示了EkARF基因在圆齿野鸦椿不同组织部位中的表达模式,为阐明EkARF基因在圆齿野鸦椿生长发育过程中的功能提供理论基础。

Abstract :

Anthocyanins, crucial water-soluble pigments in plants, determine coloration in floral and fruit tissues, while fulfilling essential physiological roles in terms of plant growth, development, and stress adaptation. The biosynthesis of anthocyanins is transcriptionally regulated by WRKY factors, one of the largest plant-specific transcription factor families. Euscaphis japonica is an East Asian species, prized for its exceptionally persistent butterfly-shaped fruits that undergo pericarp dehiscence, overturning, and a color transition to scarlet red. This species represents an ideal system for studying anthocyanin regulation. However, the mechanisms by which WRKY transcription factors orchestrate anthocyanin accumulation during this process remain unknown. In this study, we identified 87 WRKY genes (EjaWRKYs) from the E. japonica genome. Phylogenetic analysis was used to classify these genes into three primary groups, with five subgroups, revealing conserved gene structures and motif compositions, supported by collinearity and comparative synteny analyses. Crucially, ten EjaWRKYs exhibited peak expression during the mature fruit stages, showing positive correlations with key anthocyanin biosynthesis genes. Functional validation through the use of transient transactivation assays in Nicotiana benthamiana confirmed that the five selected EjaWRKYs bind W-box elements and strongly activate reporter gene expression. Our results reveal EjaWRKYs' regulation of anthocyanin accumulation in E. japonica fruit, provide the first comprehensive WRKY family characterization of this species, and establish a foundation for manipulating ornamental traits in horticultural breeding.

本发明涉及一种延长芳樟新稍观赏期并提升芳樟醇含量的方法,其是在芳樟油用林投产后,在每年植株地上部分采收后及春季抽稍前各喷施0.50‑1.85 mmol·L‑1茉莉酸甲酯一次;或是在芳樟观赏林或休闲林或康养林在春季抽稍前喷施0.50‑1.85 mmol·L‑1茉莉酸甲酯二次;发明方法可以促进抽梢和展叶,延长芳樟新稍观赏期,提升成熟叶片芳樟醇相对含量。喷施用量每次每株50‑100 mL。芳樟植株的枝叶产量增长率在13%-23%,芳樟枝叶芳樟醇的相对含量提高11%‑22.3%,抽梢时间提前5‑10 d,新梢转色观赏时间延长5‑10 d;不但可以提高芳樟枝叶产量,也极大提高了新梢转色观赏期,可以吸引更新人康养、休闲提升了效益。

以栀子YABBY基因家族为研究对象,利用TBtools、MEGA、ITOL等软件对栀子YABBY基因家族成员进行鉴定与生物信息学分析,包括栀子YABBY成员鉴定、理化性质、染色体定位、蛋白保守基序、系统进化树、顺式作用元件、共线性和栀子花不同时期表达量进行分析,以期为栀子YABBY基因鉴定及分子功能提供参考依据。结果表明:栀子YABBY基因家族共有5个成员,分布在Chr2、Chr5、Chr9和Chr11染色体上,其结构域保守,均为低亲水性蛋白;顺式元件分析表明,栀子YABBY基因家族含有光响应、非生物胁迫和激素类响应元件;表达模式分析表明,栀子GjYABBY1~GjYABBY3在花不同时期不同部位都有表达,GjYABBY3在雄蕊中持续高表达,有特异性表达;GjYABBY4在萼片发育的Ⅰ和Ⅲ期有高表达,而在不同时期花瓣、花管、雄蕊、雌蕊上均不表达。综上所述,YABBY基因在栀子花和果实生长过程中起重要调控作用。

Abstract :

为明确栀子TCP基因家族及盐胁迫响应,利用生物信息学手段对栀子TCP基因家族进行鉴定、分析和盐胁迫表达试验分析。结果表明,在栀子中共鉴定出16个TCP基因,分为PCF、CYC和CIN共3个亚家族。保守基序、基因结构和理化性质表明,同亚家族基因保守基序相似,不同亚家族的基因保守基序差异较大,16个栀子TCP基因均为亲水蛋白。染色体定位表明,16个栀子TCP基因不均匀地分布在8条染色体上,且包含多个片断段复制事件。顺式作用元件分析发现,栀子TCP基因中含有多种与植物生长发育、应激反应及激素调节相关的作用元件,以胁迫响应元件MYB和MYc最多;盐胁迫表达量显示,栀子TCP基因均能响应盐胁迫,部分基因在重度盐胁迫特异性表达,推测GjTCP13、GjTCP12和GjTCP6可能为栀子耐盐调控的关键基因。综上所述,栀子TCP基因家族成员能够积极响应盐胁迫,增强栀子的耐盐抗性。

Effect of Biochar Using N, P, and K Fertilisers on Growth and Quality of Lithocarpus litseifolius SCIE

Objective: In this study, we aimed to investigate the effects of facile combinations of biochar and N, P, and K fertilisers on the growth and physiological characteristics of Lithocarpus litseifolius seedlings, and to optimise the biochar/NPK ratio of Lithocarpus litseifolius. Method: A four-factor three-level orthogonal method was used to conduct a field experiment using 2-year-old Lithocarpus litseifolius. Nine fertiliser treatments and one control treatment (CK, i.e., no fertiliser) were established in a completely randomised block group with six replications of ten treatments. The plants were planted in October 2020 and harvested in November 2021, and fertiliser was applied once in April 2021 and once in July 2021. Results: Rational application could effectively promote the growth of Lithocarpus litseifolius, and T4(C2N1P2K3) produced the highest increase in plant height growth (17.03 cm), diameter growth (5.47 mm), soluble sugar (94.60 mg/g), soluble protein (4.59 mg/g), and phlorizin (old leaf: 16.00%; tender leaf: 15.13%); T8(C3N2P1K3) resulted in the highest chlorophyll a content (1.46 mg/g), chlorophyll b content (0.62 mg/g), and total chlorophyll content (2.08 mg/g), and T1(C1N1P1K1) resulted in the highest contents of starch (11.60 mg/g) and trilobatin (old leaf: 0.29%; tender leaf: 2.28%). The indicators corresponding to the above three treatments were significantly higher than those under the other treatments (p < 0.05). The results as analysed by the affiliation function method show that the highest mean value of the affiliation function was 0.645 after T4(C2N1P2K3) treatment. Conclusions: The combination of biochar and nitrogen, phosphorus, and potassium fertilisers can effectively increase the biomass and active components of Lithocarpus litseifolius while reducing the amount of chemical fertiliser applied. A comprehensive analysis of the results showed that the T4 treatment (biochar: 20 g/plant; urea: 10 g/plant; superphosphate: 9 g/plant; potassium chloride: 12 g/plant) resulted in the highest comprehensive score, with the highest increase in plant height growth, ground diameter growth, root-crown ratio, soluble sugar, soluble protein, and phlorizin, as well as other indicators.

【目的】为进一步研究WRKY基因家族在调控栀子盐胁迫中的功能作用、培育栀子抗盐新品种。【方法】利用生物信息学方法对栀子WRKY基因家族成员(GjWRKY)进行全基因组鉴定和相关分析。【结果】共鉴定出47个WRKY基因,非均匀地分布在10条染色体上,通过系统发育树将其分为3大组:GroupⅠ、Ⅱ和Ⅲ。GjWRKY基因以串联重复为主要扩增方式,且多数基因成员含有3个外显子。GjWRKY基因家族启动子2000 bp区域含有大量激素类和胁迫类响应顺式作用元件。46个GjWRKY基因在中度(MS)和重度盐胁迫(SS)下具有不同程度的高表达,部分GjWRKY基因产生特异性表达,17个GjWRKY基因表现为上调表达,推测这部分GjWRKY基因可能起正调控作用。【结论】GjWRKY基因在栀子抗盐胁迫中发挥了重要作用。

[目的]探讨圆齿野鸦椿(Euscaphis konishii Hayata)SPL家族成员的组成及在花和果中的表达模式。[方法]利用生物信息方法系统鉴定和分析圆齿野鸦椿EjSPLs基因家族。[结果](1)在基因组中共鉴定到26个具有SBP保守结构域的EjSPLs基因,非均匀地分布在12条染色体上,并有13对基因发生了片段复制。(2)顺式元件分析表明,EjSPLs基因受光、植物激素的调控。(3)系统发育分析表明,EjSPLs可分为8个亚家族,同一亚家族成员具有相似的基因结构和保守基序。(4)表达模式分析表明,EjSPLs基因存在组织特异性,但同一亚家族大部分成员的表达模式相似;此外,20个EjSPLs基因主要在花器官或果中表达,其中7个仅在果的绿果期、变色期、红果期表达。[结论]本研究结果提供了圆齿野鸦椿SPL基因家族的全面信息,揭示了EjSPL在其花和果中的表达模式,为阐明EjSPLs基因的生物学过程奠定了理论基础。

Transcriptome Analyses Reveal Differences in the Metabolic Pathways of the Essential Oil Principal Components of Different Cinnamomum Chemotypes SCIE

Abstract :

The genus Cinnamomum exhibits a rich variety of chemotypes and is an economically important essential oil (EO)-producing plant belonging to the family Lauraceae. Here, we aimed to explore the potential differences in the terpenoid (the principal components of EOs) biosynthesis pathways of different chemotypes at the molecular level in four Cinnamomum species-C. camphora var. linaloolifera, C. kanehirae, C. longipaniculatum, and C. micranthum. Gas chromatography-mass spectrometry (GC-MS) was employed to elucidate the discrepancies in the chemical profiles and compositions of leaf EO terpenoids among the four Cinnamomum species. The results revealed significant variations in leaf EO yields. The main constituents of the leaf EOs from C. camphora var. linaloolifera and C. kanehirae were the acyclic monoterpene linalool, and those of C. longipaniculatum and C. micranthum were the monoterpene eucalyptol and the sesquiterpene beta-caryophyllene, respectively. Furthermore, a comparative transcriptome analysis of the leaves from the four Cinnamomum species revealed that differentially expressed genes (DEGs) were significantly enriched in terpene-related entries. Specifically, 42 and 24 DEGs were significantly enriched to the mevalonate (MVA)/2-methylerythritol 4-phosphate (MEP) pathways and terpene synthase (TPS) activity, respectively. Most genes encoding proteins involved in the terpenoid precursor MVA and MEP pathways exhibited differential expression across the four species, which correlated with the distinct terpenoid profiles observed in their leaf EOs. Four acyclic monoterpene linalool synthase genes-Maker00024100, Maker00014813, Maker00014818, and Maker00018424-were highly expressed in C. camphora var. linaloolifera and C. kanehirae. A monoterpene eucalyptol synthesis gene, Maker00001509, was highly expressed in C. longipaniculatum, and a sesquiterpene beta-stigmasterol synthesis gene, Maker00005791, was highly expressed in C. micranthum. These expression levels were subsequently validated through quantitative real-time polymerase chain reaction (qRT-PCR). In conclusion, the combined results of the GC-MS and transcriptome analyses revealed a strong correlation between the metabolite content of the EOs and gene expression. This research contributes to a better understanding of the differences in terpene accumulation in various chemotypes of Cinnamomum at the molecular and mechanistic levels, laying a solid foundation for the cultivation of an ideal Cinnamomum variety.
