灵芝优良品种全基因组分子标记的建立

Abstract :

灵芝Ganodorma lingzhi的优良品种灵芝13具有农艺性状较好、子实体产量较高、孢子粉产量较高的优良性状。本研究首先以其单核菌株灵芝13-5为材料,通过三代HiFi测序及Hi-C测序组装成完整的基因组;然后选取5个重复的双核菌株通过Illumina高通量测序后,以灵芝13-5基因组作为参考进行SNP calling,建立了基于灵芝13全基因组的分子标记。基因组包含13条染色体,共45.73 Mb。5个重复菌株SNP calling去重合并后,获得319 074个共有的异核等位差异位点,作为用于灵芝13菌株鉴别的分子标记库。从实验室菌种库中随机抽取7个灵芝菌株,与灵芝13菌株共有的等位差异位点占比介于22.20%–45.18%之间,远低于阈值,说明这些菌株与灵芝13差异较大,不是相同菌株。本研究建立了基于灵芝13全基因组及其异核等位差异位点的分子标记,能够准确区分灵芝13这一优良品种。

Abstract :

粗毛纤孔菌是一种天然的药用真菌,具有多种生物活性,其子实体颜色在成熟后期可由黄褐色转变为黑色,即为黑色素。通过探究其理化性质发现,粗毛纤孔菌黑色素(IHFM)属于真黑色素,具有良好的热、光和及金属稳定性等。黑色素是粗毛纤孔菌的活性成分之一,具有保肝、抗癌、调节肠道菌群结构、抗氧化等活性。首先,在保肝方面的研究显示,IHFM一方面通过抑制Keap-1、激活Nrf2信号通路显著提高下游GCLC、HO-1、NQO1等的表达;另一方面显著抑制MyD88介导的TLR4/NF-κβ信号通路并降低下游炎症因子IL-1β、TNF-α、IL-6等的表达水平,进而缓解小鼠酒精性引发的肝损伤。在调节肠道菌群方面,IHFM可以调节酒精性肝损伤小鼠中肠道菌群的结构,上调肠道菌群中有益菌的相对丰度,降低有害菌的相对丰度。综上,我们的研究可为IHFM开发为保肝、抗氧化剂等相关产品奠定理论基础。

本发明为一种粗毛纤孔菌黑色素的制备方法和应用,粗毛纤孔菌黑色素的制备方法,包括如下步骤:S1,收集粗毛纤孔菌子实体并烘干粉碎后经筛网筛选过滤;S2,称取粉末并加入NaOH溶液,并置于超声波中充分反应;S3,在离心机上经离心处理后,收集上清液;S4,在S3收集的上清液中加入酸性溶液并加热;S5,再次离心后收集沉淀物;S6,将沉淀物进行纯化;S7,离心收集上清液,最终通过流水透析后,冷冻干燥后获得纯化后的粗毛纤孔菌子实体黑色素样品。所述粗毛纤孔菌黑色素能够显著抑制人乳腺癌细胞生长、增殖、迁移的能力;并诱导人乳腺癌细胞的凋亡和人乳腺癌细胞焦亡。因此本发明粗毛纤孔菌黑色素具有开发为抗乳腺癌药物的潜能。

银耳附属染色体的多态性与起源研究:对基因组分化与结构变异的深入解析

Abstract :

附属染色体虽然对真菌的正常生长和发育并非必需,但在子囊菌门真菌中的研究已较为深入,而在担子菌门中的报道则相对较少。本研究对来自不同来源的16株银耳(Tremella fuciformis)进行了全基因组测序,获得了27条完整的单倍型基因组序列,用于比较分析。结果显示这些基因组在大小上存在显著差异,变异幅度超过三分之一,主要由附属染色体和核心染色体中的重复序列引起。银耳基因组包含8至10条核心染色体,其多态性主要源于染色体之间的融合或分离。每个基因组含有2至10条附属染色体,共计107条,这些附属染色体的分布模式与系统发育关系及其与Annulohypoxylon stygium的配对特异性密切相关。附属染色体通常较小,富含转座子或重复序列,基因密度较低,但它们之间的同源性明显高于核心染色体。附加染色体表现出快速而多样的结构变异(SV),导致各系统发育分支内共享基因比例低于核心染色体。在银耳两种形态细胞类型的生长和转化过程中,核心和附属染色体均表现出高频的染色体拷贝数变异。大多数附属染色体上的基因与核心染色体上的基因不具同源性,且其编码的蛋白质在NCBI nr数据库中缺乏匹配序列。本研究首次揭示了担子菌门中附属染色体的多样性,并对其起源提供了新的见解,认为这些染色体可能起源于银耳物种分化之前尚未被探索的其他物种。

Exploration of rational donor-acceptor adjustment strategies to achieve ACQ to AIE transformation and their potential applications as multi-functional AIEgens SCIE

In this work, a new series of rofecoxib analogues were designed and synthesized by employing the migration of the donor (D) group and donor (D)-acceptor (A) switching principle to achieve ACQ-AIE conversion. Among them, when the D unit dimethylamino was anchored on the para-position of the benzene ring D, the corresponding compound L1, namely (Z)-5-(4-(dimethylamino)benzylidene)-4-(4-(methylsulfonyl)phenyl)-3-phenylfuran-2(5H)-one, was obtained exhibiting obvious aggregation-caused quenching (ACQ) behavior. Subsequently, after migrating the D group from ring D to ring B, the resulting L2 exhibits an aggregation-induced emission (AIE) effect. Meanwhile, by switching the position of the D and A units based on the structure of L1, the corresponding compound L3 also shows AIE behavior. These results demonstrate that migrating the position of the donor group and switching the positions of the donor and acceptor have significant effects on the fluorescent properties of the compounds. Moreover, single crystal X-ray analysis of L2 and L3 revealed that the stronger molecular C-H & ctdot;pi interactions contribute to greater structural rigidification, which in turn enhances their bright emission in the solid state. In addition, L2 and L3 exhibited solvatochromic, acidichromic, and trace water properties with reversible multi-stimulus behaviors. More importantly, L2 and L3 demonstrated highly specific fluorescent imaging ability (RL2 = 0.97, RL3 = 0.98) toward lipid droplets in living MCF-7 cells, with low cytotoxicity. In summary, the creative design concepts of adjusting the position of D and switching the positions of D and A units, as well as the ACQ to AIE conversion provide alternative ways for developing new multifunctional AIEgens, L2 and L3.

Domestication Cultivation and Nutritional Analysis of Hericium coralloides SCIE

Abstract :

Hericium coralloides is a valuable medicinal and edible mushroom renowned for its unique bioactive compounds. This study focuses on the isolation of a wild strain (SH001) exhibiting promising cultivation potential and health promoting properties. A wild fungal strain from the Tibetan Plateau was isolated and identified as a novel H. coralloides based on its morphological and molecular characteristics. The optimal growth conditions were found to be 30 degrees C, pH 7.0, fructose as the preferred carbon source, and yeast extract as the optimal nitrogen source. Nutritional analysis revealed that the fruiting bodies were rich in protein (15.4 g/100 g dry weight), dietary fiber (34.7 g/100 g dry weight), and minerals, while being low in fat (3.5 g/100 g dry weight). The most abundant amino acids were glutamic acid, followed by aspartic acid. The polysaccharides exhibited significant antioxidant activity, with ABTS+ scavenging comparable to that of Vitamin C (Vc), achieving a clearance rate of 96.95% at concentrations between 0.25-5.00 mg/mL. At a concentration of 5 mg/mL, the DPPH and OH radical scavenging activities reached their peak (83.77% and 67.31%, respectively), along with the highest iron ion reducing capacity (FRAP value: 4.43 mmol/L. Polysaccharides also exhibited notable anticancer activity, inhibiting HepG2 liver cancer cells and MDA-MB-468 breast cancer cells, with IC50 values of 3.896 mg/mL and 2.561 mg/mL, respectively. This study demonstrates that wild H. coralloides can be successfully cultivated in vitro. In conclusion, the fruiting bodies possess substantial nutritional value, and the polysaccharides extracted from them show promising antioxidant and anticancer activities, particularly against HepG2 liver cancer cells and MDA-MB-468 breast cancer cells.

Abstract :

Hepatocellular carcinoma (HCC) is one of the common most malignant tumors. This study aimed to determine the in vitro and in vivo anticancer activity of cordycepin and elucidate its mechanism of action. The results of in vitro and in vivo studies revealed that cordycepin inhibited proliferation and migration in HepG-2 cells and inhibited the growth of HepG-2 xenograft-bearing nude mice by inducing apoptosis. Transcriptome sequencing analysis revealed a total of 403 differential genes, which revealed that cordycepin may play an anti-HCC role by regulating Hippo signaling pathway. The regulatory effects of cordycepin on the Hippo signaling pathway was further investigated using a YAP1 inhibitor. The results demonstrated that cordycepin upregulated the expression of MST1 and LAST1, and subsequently inhibited YAP1, which activated the Hippo signaling pathway. This in turn downregulated the expression of GBP3 and ETV5, and subsequently inhibited cell proliferation and migration. Additionally, YAP1 regulated the expression of Bax and Bcl-2, regulated the mitochondrial apoptotic pathway, and induced apoptosis by upregulating the expression of the caspase-3 protein. In summary, this study reveals that cordycepin exerts its anti-hepatocarcinoma effect through regulating Hippo signaling pathway, and GBP3 and ETV5 may be potential therapeutic targets for hepatocarcinoma.(c) 2024 Beijing Academy of Food Sciences. Abstract :

A fungal strain (X21156) collected in Tibet was used as the material, identified based on its morphological characteristics and internal transcribed spacer (ITS) sequence; its optimal culture conditions were analyzed by single-factor experiments; artificial domestication and cultivation were carried out; its nutrient composition was determined; and the bioactivities of its polysaccharides were detected using chemical antioxidant assays and MTT assays. The results showed that the strain was Pleurotus citrinopileatus Sing. Its optimal culture conditions were a pH of 7, a temperature of 25 degrees C, glucose (20 gL-1) as the carbon source, and yeast powder (20 gL-1) as the nitrogen source. The fresh weight of a single domesticated fruiting body was 41.16 g. The strain had high protein (28.5%), high fiber (34%), and low fat (1.4%) contents, with high proportions of fresh and sweet amino acids. Polysaccharides had good scavenging ability on ABTS+, DPPH, and OH free radicals (EC50 0.06 mg/mL, 1.21 mg/mL, and 3.62 mg/mL, respectively), and the cytotoxicity of polysaccharides to hepatocellular carcinoma cells (HepG2) (IC50: 1.69 mg/mL) was higher than that of triple-negative breast cancer cells (MDA-MB-468) (IC50: 1.76 mg/mL). In conclusion, the study provides a reference on the optimal culture conditions, domestication and cultivation, and dietary and medicinal values of wild P. citrinopileatus Sing.

Abstract :

Liquid fermentation is an efficient culture for obtaining polysaccharides from edible mushrooms. In this study, the polysaccharide content and biomass were examined by introducing microorganisms into the Wolfiporia cocos fermentation system. Three edible mushroom co-fermentation systems were established, among which the Wolfiporia cocos-Ganoderma lucidum co-fermentation system significantly increased the mycelial biomass of the system by 57.71% compared to Wolfiporia cocos alone and 91.22% compared to Ganoderma lucidum alone, and the intracellular polysaccharide content was significantly increased. Physiological activities of polysaccharides showed that mycelial polysaccharides in the Wolfiporia cocos-Ganoderma lucidum system had stronger anti-tumor cell value-adding and anti-tumor cell migration activities compared with Wolfiporia cocos and Ganoderma lucidum fermentation alone. The transcriptomic study of Wolfiporia cocos mycelium induced by exogenous substances suggested that the exogenous substances could enhance the intracellular polysaccharide content of Wolfiporia cocos through the upregulation of the expression of alpha-glycosyltransferase encoded by ALG10 and the downregulation of alpha-glycosidases encoded by MAN1B in the glycolytic metabolism of Wolfiporia cocos. This study provides a new direction for the transformation of polysaccharides from Wolfiporia cocos and Ganoderma lucidum into functional foods and new product development, and provides an experimental basis.
