The Global Transcription Factor FvCon7 Plays a Role in the Morphology, FB1 Toxin Production, and Pathogenesis of Fusarium verticillioides SCIE

Abstract :

Fusarium verticillioides, an important global pathogenic fungus, compromises crop quality and yield by infecting maize, sugarcane, and some Solanaceae, endangering food security through contaminated grains and cereals with the fumonisin B1 (FB1) toxin. While Con7 has been reported as a transcription factor involved in the sporulation and pathogenicity of some pathogenic fungi, the function of FvCon7 and its regulatory genes in F. verticillioides remains uncharacterized. Gene deletion mutants of Delta Fvcon7 were constructed through homologous recombination, which exhibited defects in vegetative growth, survival, sporophore development, conidiation, conidial germination, and carbon metabolism. Carbon metabolism defects led to a significant accumulation of glycogen granules in hypha and lipid bodies in conidia. Additionally, Delta Fvcon7 displayed impaired cell wall structure and integrity, along with an altered expression of genes encoding cell wall-degrading enzymes (such as chitinase), as detected by qRT-PCR. Moreover, Fvcon7 also plays a role in the pathogenicity of maize and sugarcane through different splicing, defective conidia, reduced survival viability, differential expression of secreted proteins, and deficiencies in antioxidant stress capacity. Furthermore, using yeast one-hybrid (Y1H) assays, FvCon7 was found for the first time to directly regulate the expression of FvFUMs by binding to the CCAAT box within the promoters of six key FvFUMs, thereby affecting FB1 production. Overall, FvCon7 functions as a global transcription factor regulating multiple phenotypes. This study provides a theoretical basis for elucidating the mechanism of transcription factor FvCon7 regulating toxin production and pathogenesis in F. verticillioides.

Abstract :

Fusarium verticillioides is an important pathogen of maize and causes serious yield losses and food safety issues worldwide. F. verticillioides produces highly toxic mycotoxin Fumonisin B1 (FB1) in infested commodities which makes these food and feeds unsafe for humans and animals. For pathogenic fungi to successfully penetrate its plant hosts, the pathogen secretes hydrolytic enzymes that can facilitate penetration into the plant cutin layer. However, there is limited information on how cutinases transcriptionally regulated to impact F. verticillioides pathogenicity. In this study, our aim is to functionally characterize cutinase transcription factors that regulate key cutinase activities that are directly associated with F. verticillioides pathogenicity and FB1 biosynthesis. Gene deletion of cutinase transcription factor FvCTF1 alpha did not affect the growth and morphology of the fungal mycelia on CMII medium, whereas the conidiation, utilization of sodium acetate and sodium oleate, stress tolerance against cell wall interfering agent(,) and the cutinase and pectinase activities in the Delta Fvctf1 alpha mutant were negatively impacted. FvCtf1 alpha regulates the expression of induced cutinase genes FvCUT1 and FvCUT4 by binding to their GC-rich promoters. In addition, FvCtf1 alpha, containing a novel function in regulating FB1, interacts with the promoter of FvFUM1 and FvFUM6 to down-regulate the expression of FvFUM1 and FvFUM6, resulting in decreased production of FB1 in the Delta Fvctf1 alpha strain. Delta Fvctf1 alpha exhibited decreased pathogenicity in maize due to the down-regulation of pathogenicity-related genes as well as key downstream cutinase genes FvCUT3 and FvCUT4 in F. verticillioides. We also demonstrated that FvCtf1 alpha regulated FvCUT3 and FvCUT4 differently; FvCUT4 via direct regulation while FvCUT3 via indirect regulation by interacting with FvFarB, a homologous protein of FvCtf1 alpha. Moreover, RNA-seq analysis showed that FvCtf1 alpha was associated with many pathways, such as fatty acid metabolism, carbon source utilization, cell wall integrity, oxidative stress, and fumonisin synthesis in F. verticillioides. Our study demonstrated that FvCtf1 alpha was not only involved in the regulation of cutinases but also a broad spectrum of pathways that ultimately affect F. verticillioides virulence and mycotoxin biosynthesis.

Abstract :

Transcriptional regulation mediated by the balance of histone acetylation and deacetylation is fundamental in responding to environmental cues by impacting chromatin remodeling. Histone deacetylases (HDACs) are enzymes that remove acetyl groups from histone and non-histone proteins, thus restoring a tight chromatin structure. In pathogenic fungi, HDACs have been implicated in growth, secondary metabolite biosynthesis, and virulence. However, the role of HDACs in the mycotoxin fumonisin B1 (FB1)-producing fungus Fusarium verticillioides is poorly understood. In this study, we systematically characterized six F. verticillioides HDACs. An increased level of H4K16ac was observed in the deletion mutant of FvHOS2, which was associated with vegetative growth, conidiation, and virulence when infecting sugarcane and maize. FvRpd3 appeared to be essential for vegetative growth, while FvHda1 promoted growth, and both contributed to conidiation and pathogenicity. In contrast, FvSirt4 displayed a negative correlation with these processes. Additionally, the FB1 production was positively affected by FvHos2 and FvRpd3, but negatively impacted by Fvhda1, FvSir2, FvHst2, and FvSirt4 through the regulation of different key fumonisin biosynthetic (FUM) genes. Further findings indicate an association between FvSirt4 and FvSkb1, which is a histone methylase that positively regulates FB1 and pathogenicity. Moreover, as a global transcriptional regulator, over 2365 genes (similar to 15% of the genome) enriched in multiple metabolic pathways were significantly downregulated in the Delta Fvhos2 mutants relative to the wild type. Overall, our results suggest distinct roles of HDACs in regulating the growth, virulence, mycotoxin FB1 production, and gene expression in F. verticillioides.

茶提取物复合方对眼致病菌的抑制作用研究

采用滤纸圆片法检测尤溪绿茶、尤溪红茶、尤溪白茶、尤溪乌龙茶、菊花、蒲公英的浸提液,互叶白千层精油及山茶油的单方与复合方对眼科常见致病菌金黄色葡萄球菌、大肠杆菌、青霉菌、镰刀菌的抑制效果,以筛选有较好抑制效果且耐贮存的茶提取物草本复合配方。结果表明:在所选用的复配方中,WT∶T∶EOMA-2(白茶、蒲公英、互叶白千层精油的质量浓度比为3∶1∶1)、WT∶C∶T∶EOMA-2(白茶、菊花、蒲公英、互叶白千层精油的质量浓度比为3∶1∶1∶1)对眼科常见致病菌金黄色葡萄球菌和大肠杆菌的综合抑菌作用较好;互叶白千层精油和个别复配液(WT∶T∶EOMA-2、WT∶T∶EOMA-1、WT∶C∶T∶EOMA-2、WT∶EOMA∶CO-1)对眼致病真菌青霉菌和镰刀菌有一定作用;复合方综合抑菌作用与水浸出物含量、茶多酚含量高度相关。此外,优化的复配方液体试剂显示对热稳定性较差,但有一定程度的抗紫外线能力,表明这些方剂宜低温贮藏且不必严格避光保存。本研究初步寻找到对眼睛亲和性较好的草本复合配方,可为干眼病患者的雾化熏蒸治疗提供依据。

Abstract :

The peroxisomal matrix oxidase, catalase and peroxidase are imported peroxisomes through the shuttling receptors, which regulates the cellular oxidative homeostasis and function. Here, we report that PTS1 shuttling receptor FvPex5 is involved in the localization of PTS1, utilization of carbon sources and lipids, elimination ROS, cell wall stress, conidiation, fumonisin B1 (FB1) production, and virulence in maize pathogen Fusarium verticillioides. Significantly, differential expression of PTS1-, PTS2-, PEX-and FB1 toxin-related genes in wild type and Delta Fvpex5 mutant were examined by RNA-Seq analyses and confirmed by RT-PCR assay. In addition, different expression of PTS1 and PTS2 genes of the Delta Fvpex5 mutant were enriched in diverse biochemical pathways, such as carbon metabolism, nitrogen metabolism, lipid metabolism and the oxidation balance by combining GO and KEGG annotations. Overall, we showed that FvPex5 is involved in the regulation of genes associated with PTS, thereby affecting the oxidation balance, FB1 and virulence in F. verticillioides. The results help to clarify the functional divergence of Pex5 orthologs, and may provide a possible target for controlling F. verticillioides infections and FB1 biosynthesis.

Fusarium verticillioides, a well-known fungal pathogen that causes severe disease in maize and contaminates the grains with fumonisin B1 (FB1) mycotoxin, affects the yield and quality of maize worldwide. The intrinsic roles of peroxisome targeting signal (PTS)-containing proteins in phytopathogens remain elusive. We therefore explored the regulatory role and other biological functions of the components of PTS2 receptor complex, FvPex7 and FvPex20, in F. verticillioides. We found that FvPex7 directly interacts with the carboxyl terminus of FvPex20 in F. verticillioides. PTS2-containing proteins are recognized and bound by the FvPex7 receptor or the FvPex7-Pex20 receptor complex in the cytoplasm, but the peroxisome localization of the PTS2-Pex7-Pex20 complex is only determined by Pex20 in F. verticillioides. However, we observed that some putative PTS2 proteins that interact with Pex7 are not transported into the peroxisomes, but a PTS1 protein that interacts with Pex5 was detected in the peroxisomes. Furthermore, Delta Fvpex7pex20 as well as Delta Fvpex7pex5 double mutants exhibited reduced pathogenicity and FB1 biosynthesis, along with defects in conidiation. The PTS2 receptor complex mutants (Delta Fvpex7pex20) grew slowly on minimal media and showed reduced sensitivity to cell wall and cell membrane stress-inducing agents compared to the wild type. Taken together, we conclude that the PTS2 receptor complex mediates peroxisome matrix proteins import and contributes to pathogenicity and FB1 biosynthesis in F. verticillioides.

Abstract :

Fusarium verticillioides is a key maize pathogen and produces fumonisins, a group of mycotoxins detrimental to humans and animals. Unfortunately, our understanding on how this fungus interacts with maize to trigger mycotoxin biosynthesis is limited. We performed a systematic computational network-based analysis of largescale F. verticillioides RNA-seq datasets to identify gene subnetwork modules associated with virulence and fumonisin regulation. F. verticillioides was inoculated on two different maize lines, moderately resistant line hybrid 33K44 and highly susceptible line maize inbred line B73, to generate time-course RNA-Seq data. Among the highly discriminative subnetwork modules, we identified a putative hub gene FvLCP1, which encodes a putative a type-D fungal LysM protein with a signal peptide, three LysM domains, and two chitin binding domains. FvLcp1 is a unique protein that harbors these domains amongst five representative Fusarium species. FvLcp1 is a secreted protein important for fumonisin production with the LysM domain playing a critical role. The chitin-binding domain was essential for in vitro chitin binding. Using Magnaporthe oryzae, we learned that FvLcp1 accumulates in appressoria, suggesting that FvLcp1 is involved in host recognition and infection. Full length FvLcp1 suppressed BAX-triggered plant cell death in Nicotiana benthamiana. This unique type-D LysM secreted protein with a chitin-binding domain in F. verticillioides was shown to be potentially involved in suppressing host cell death and promoting fumonisin biosynthesis while the pathogen colonizes maize kernels.
