Abstract :
It is urgent to mine novel blast-resistant genes in rice and develop new rice varieties with pyramiding blast-resistant genes. In this study, a new blast-resistant gene, OsBRW1, was screened from a set of rice near-isogenic lines (NILs) with different blast-resistant ability. Under the infection of Magnaporthe oryzae (M. oryzae), OsBRW1 in the resistant NIL Pi-4b was highly induced than that in the susceptible NIL Pi-1 and their parent line CO39, and the blast-resistant ability of OsBRW1 was further confirmed by using CRISPR/Cas9 knockout and over-expression methods. The protein encoded by OsBRW1 was a typical NBS-LRR with NB-ARC domain and localized in both cytoplasm and nucleus, and the transient expression of OsBRW1 was capable of triggering hypersensitive response in tobacco leaves. Protein interaction experiments showed that OsBRW1 protein directly interacted with OsSRFP1. At the early infection stage of M. oryzae, OsBRW1 gene induced OsSRFP1 to highly expression level and accumulated H2O2, up-regulated the defence responsive signalling transduction genes and the pathogenesis-related genes and increased JA and SA content in the resistant NIL Pi-4b. By contrary, lower content of endogenous JA and SA in osbrw1 mutants was found at the same stage. After that, OsSRFP1 was down-regulated to constitution abundance to balance the growth of the resistant NIL Pi-4b. In summary, OsBRW1 solicited OsSRFP1 to resist the infection of blast fungus in rice by inducing the synergism of induced systemic resistance (ISR) and system acquired resistance (SAR) and to balance the growth of rice plants.

本发明公开了一个水稻OsLRG2基因及其在水稻抗瘟病中的应用。所述OsLRG2基因的核苷酸序列如SEQ ID NO:1所示,其编码的蛋白质的氨基酸序列如SEQ ID NO:2所示。该基因的CDS序列长2742bp,编码914个氨基酸的蛋白,该基因编码的蛋白具有CC、NB‑ARC和LRR结构域,属于植物中最典型的NBS‑LRR类抗病蛋白家族成员之一。通过CRISPR/Cas9的方法敲除OsLRG2基因,发现降低了水稻对稻瘟菌的抗性。本发明弥补了现有水稻抗病基因的不足,可应用于抗病育种提高水稻的稻瘟病抗性,因而为高产优质的作物新品种选育提供了新的基因资源和理论基础。

Meteorological factors have a significant impact on tea production; therefore, it is crucial to investigate the meteorological factors that influence tea production for the purpose of optimizing tea production layout, increasing farmers' income, and addressing other related aspects. However, few studies have closely examined the meteorological factors that contribute to the comparative advantage of tea production at the county level. This study aims to explore the spatio-temporal pattern of tea production in Fujian Province from 2001 to 2020 by spatial analysis. Furthermore, we analyzed the comparative advantage of tea production and its spatio-temporal pattern using the resource endowment coefficient. Additionally, we investigated the meteorological factors that influence the comparative advantage of tea production in Fujian Province in 2020 by employing geographic detectors. The results revealed that the spatio-temporal pattern of tea production in Fujian Province during 2001-2020 initially exhibited a scattered distribution, which gradually transformed into a "dual core" agglomeration pattern. The highest tea production was 75,613 t in Anxi in 2020, followed by 73,428 t in Anxi in 2019. Moreover, the spatio-temporal pattern of comparative advantage of tea production in Fujian Province displayed significant differences among counties during 2001-2020. The highest comparative advantage of tea production in Fujian Province was 23.80 in Zhenghe in 2006, with Anxi following closely at 23.18 in 2011. The lowest comparative advantage of tea production in Fujian Province was July, had an obvious impact on the comparative advantage of tea production. Meanwhile, the interaction between any two meteorological factors demonstrated a dual factor enhancement effect. The highest q value was 0.666, observed in the interaction between atmospheric pressure and sunlight intensity in January, followed by 0.587 in the interaction between air temperature and sunlight intensity in June. These findings provide insights for optimizing the spatial layout of tea production, increasing tea yield, and promoting rural revitalization strategies.

Rice blast disease, caused by Magnaporthe oryzae (M. oryzae), is a significant threat to global rice production. Conventional methods for disease management face limitations, emphasizing the importance of sustainable alternatives. In this study, two rice cultivars with different blast resistant abilities, the susceptible variety CO39 and the resistant variety Pi4b, were used as materials to study the effects of Piriformospora indica (Pi) on the resistance to M. oryzae infection and rice growth. The in vitro tests revealed no direct antagonistic interaction between Pi and M. oryzae. However, the in vivo experiments showed that Pi promoted plant growth by increasing root and shoot length, chlorophyll content, and nitrogen uptake, particularly in CO39 during pathogen infection. Pi inoculation also significantly reduced disease severity, which was indicated by smaller lesion areas and shorter lesion lengths in both cultivars but a more pronounced effect in CO39. This occurred due to the decreasing levels of MDA and the modulating activity of antioxidant enzymes in Pi-inoculated rice plants. At the early stage of M. oryzae infection, the expression of the ethylene signaling gene OsEIN2 and the gibberellin biosynthesis gene OsGA20ox1 in Pi-inoculated CO39 decreased but significantly increased in both rice cultivars at the later stage. The reverse was found for the pathogenesis-related (PR) genes OsPR10 and OsPBZ1 and the blast-resistant genes OsBRG1, OsBRG2, and OsBRW1, suggesting early growth suppression for rice resilience to blast followed by a later shift back to growth. Meanwhile, Pi inoculation increased OsCesA9 expression in rice to strengthen cell walls and establish the primary defense barrier against M. oryzae and upregulated the expression of OsNPR1 without a significant difference in CO39 but downregulated it in Pi4b to activate PR genes to enhance plant resistance. In summary, these results underscore the potential of Pi as a sustainable biological control agent for rice blast disease, which is particularly beneficial for blast-susceptible rice cultivars.

Abstract :
Crop production plays an important role in social and economic development. Exploiting the comparative advantages of major crop types has a profound effect on improving crop production. However, studies on these comparative advantages and their influencing factors are few. Here, the temporal variation characteristics of main crop types in Chongqing City from 1985 to 2021 were analyzed. The comparative advantages of these crop types were also evaluated, and the main factors that influenced them were determined using ordinary least square model. Results demonstrated that the temporal variation characteristics of sown area and comparative advantages in scale of six crop types in Chongqing City from 1985 to 2021 had obvious difference. The sown area of hemp crops showed the most variation among the six crop types. Tobacco, vegetable, and hemp crops had comparative advantages in scale in Chongqing City. GDP, pesticide usage, per capita disposable income of rural residents, agricultural film used, relative humidity, and atmospheric pressure were the major factors influencing the comparative advantages of the six crop types in Chongqing City from 1985 to 2021. These findings are useful for adjustment and optimization of planting structure, crop production management, and sustainable development of agriculture.
