Pig bedding biochar enhances the Cd extraction capacity of Perilla frutescens by mitigating the toxicity of Cd-contaminated soil SCIE

Abstract :

Pig bedding biochar (PBBC) and Cd-enriched Perilla frutescens were used for joint remediation of Cd-contaminated soil. Cd-contaminated soil was treated with different concentration of PBBC. The physiological and biochemical indicators of P. frutescens was evaluated under different Cd stress, including biomass, antioxidant system. Meanwhile Cd bioavailability, enzyme activity and nutrient bioavailability of the soil were monitored. Results revealed that PBBC at 1 % and 5 % levels led to decreased diethylenetriaminepentaacetic acid-extractable Cd (DTPA-Cd) content in soil by 19.09 %-20.05 % and 30.10 %-47.08 %, respectively. Moreover, PBBC promoted the transformation of exchangeable Cd (EXC-Cd) into a more stable form, enhanced soil enzymes (peroxidase, acid phosphatase, urease, and sucrase) activities, and alleviated P. frutescens 's oxidative stress. PBBC increased its biomass, consequently enhancing Cd accumulation in the plant's, thereby improving Phytoextraction rate (PER). 1 % PBBC showed the best effect, with a total biomass increased 21.42 %-26.94 %, PER was enhanced by 39.83 %-54.82 %. This study justifies that the combining PBBC with P. frutescens enhances Cd removal from soil, making the PBBC-P. frutescens a promising choice for treating Cd-polluted soil. 紫苏分支酸合成酶PeCS基因克隆、生物信息学及原核表达分析

Abstract :

本研究旨在探究紫苏(Perilla frutescens L.)药用活性成分酚酸生物合成途径中的关键基因。基于前期紫苏转录组测序数据,筛选出酚酸合成途径中的差异表达基因分支酸合成酶(chorismate synthase, CS)基因,获得PeCS基因开放阅读框(ORF),利用生物信息学技术对Pe CS基因功能做出初步预测,构建pET30a-PeCS原核表达载体,并转化大肠杆菌BL21 (DE3),IPTG诱导表达后进行SDS-PAGE检测。通过实时荧光定量PCR(RT-qPCR)技术检测紫苏不同组织(根,茎,叶)中PeCS表达水平。克隆得到紫苏PeCS基因开放阅读框(ORF)长度为877 bp,编码291个氨基酸,生物信息学预测为亲水性蛋白,定位于叶绿体。该蛋白相对分子质量为31 659.16。Pe CS基因编码蛋白不含信号肽且无跨膜区,亚细胞定位于叶绿体。所构建pET30a-PeCS原核表达载体在大肠杆菌BL21 (DE3)中表达的最适条件为15℃、0.5 mmol/L IPTG培养16 h,PeCS重组蛋白主要以包涵体形式存在,蛋白分子量与预测相符。RT-qPCR分析显示PeCS基因在紫苏不同器官组织中均有表达,且在叶中的表达量最高,在茎中表达量最低。本研究成功克隆紫苏分支酸合成酶PeCS基因,有助于更深入地阐释紫苏酚酸合成途径和调控机制。

孑遗植物水松不同季节根际微生物群落结构和功能特征

Abstract :

[目的]探索水松根际微生物群落结构和功能及随季节变化的规律。[方法]采用PCR-DGGE和Biolog-ECO微平板法测定不同季节水松根际土壤理化性质、微生物群落结构和功能,通过主成分分析和冗余分析等方法解析根际微生态各环境因子的内在规律。[结果]水松根际微生物群落中优势细菌为α-变形菌(Alphaproteobacteria)、酸杆菌(Acidobacteria)和δ-变形菌(Deltaproteobacteria),优势真菌为粪壳菌(Sordariomycetes)。夏季水松根际微生物量和微生物多样性最高。根际微生物的碳代谢能力在夏季最强,冬季最弱,大致表现为夏季>秋季>春季>冬季,速效氮可能是微生物群落碳代谢功能的主要驱动因子。[结论]水松根际微生物群落多样性和碳代谢能力在夏季最优,在冬季最差。季节因素显著影响着根际微生物群落结构和功能,开展相关研究时应充分考虑季节因素造成的波动。

Abstract :

Sulfate radical-based advanced oxidation processes exhibit great potential for the degradation of organic pollutants. In this study, P-doped biochar (PBC500) was successfully synthesized by the pyrolysis of H3PO4-impregnated cow manure waste and was employed to activate peroxymonosulfate (PMS) for the elimination of 17 beta-estradiol (E2). The characterization results showed that the surface area, defective structure, and functional groups (C=O and phosphorus-containing groups) of biochar increased after H3PO4 modification. PBC500 exhibited high PMS activation activity and excellent E2 degradation capacity; 97.91% of 3 mg/L E2 can be removed within 90 min using 0.2 g/L PBC500 and 1 mM PMS. Based on the quenching experiments and X-ray photoelectron spectroscopy (XPS) analysis, defective structures, C=O, and P-C groups on biochar act as active sites to promote the catalytic oxidation of E2 by generating O2- and 1O2. In addition, PBC500 displayed excellent reusability, achieving 65.15% E2 degradation after three reuse cycles. Overall, this study presented a new technique that supports a high efficiency, environmentally friendly, and low cost treatment method for E2 wastewater and simultaneously provided a new option for the resource utilization of livestock waste.

Abstract :

Nitrogen (N), phosphorus (P) and potassium (K) are three macroelements in agriculture production, but their combined effects on arsenic (As) toxicity and its translocation in rice plants are not clear. In this study, an orthogonal rotation combination based on different N, P and K (NPK) concentration was first designed to examine their combined effect on the As toxicity, its transformation and migration in rice plants based on the hydroponic culture and pot soil culture. The results showed that 2.0 mg/L arsenite (As(III)) had obvious toxicity on the growth of indica LuYouMingZhan (LYMZ) and the optimal NPK concentration was 28.41, 6 and 50 mg/L based on the quadratic regression of the recovery rate of chlorophyll SPAD value of indica LYMZ. The optimal NPK combination significantly alleviated the physiological toxicity of As(III) on indica LYMZ rice seedling and decreased the accumulation of inorganic As in their roots and shoots by 23.8+1.8 % and 33.4+2.4 % respectively; further pot culture from different As(III) polluted soil showed that the optimal NPK combination significantly increased the dry weight of roots, stems, sheaths and leaves of indica LYMZ rice plants as well as yield indicators by 6.4 %-61.7 % and 7.1 %-89.8 % respectively, decreased the accumulation of As(III) and arsenate by 6.25 %-100 % and 12.36 %-100 % respectively in their roots, stems, sheaths, leaves, brans and kernels except As (III) concentration in their sheaths, decreased the accumulation of dimethylarsenate in their sheaths, leaves, brans and kernels, and had the best repair effect on the translocation of As species in 50 mg/kg As(III)-added soil. Our study provided a desirable strategy for alleviating As toxicity in paddy soil and reducing As pollution in rice plants.

Abstract :

The issue of soil contamination by heavy metals (HMs) has attracted extensive attention. In the present study, the effects of four remediation measures combined with rice intercropping on the quality of soils were evaluated in a mildly HM-contaminated paddy field. The results showed that better comprehensive remediation effects were found in the intercropping system with high and low Cd-tolerant rice than in the monoculture system. Both foliar spraying of sodium selenite and inoculation with Pseudomonas TCd-1 significantly reduced the Nemerow comprehensive pollution index (NCPI) of the soils. The application of biochar and lime significantly increased the soil fertility index. Among all the treatments, the application of 30 t center dot hm-2 biochar and 3600 kg center dot hm-2 lime improved soil fertility the most. The lowest single-factor pollution indices (SFPIs) of Cd, Cu, Zn, Ni and Pb and the NCPI of the soils were observed in the treatment with foliar spraying of sodium selenite at 45 mg center dot L-1, showing the greatest comprehensive reduction in soil HMs. The application of 1200 kg center dot hm-2 lime and 30 t center dot hm-2 biochar and foliar spraying of 45 mg center dot L-1 sodium selenite effectively improved the soil quality. Overall, the soil quality of paddy fields dramatically influenced the cleaner production of rice and is of great significance to the maintenance of food security.

Abstract :

The gap between serious soil heavy metals pollution and inefficient soil remediation threatens human health. This study proposed a method to improve the phytoremediation efficiency using bamboo vinegar (BV) solution and the potential mechanism was discussed. The results demonstrated that the application of BV increases the content of cadmium (Cd) in vacuole and cell wall hemicellulose 2 in leaves of Perilla frutescens. Simultaneously, it enhanced enzyme activities of superoxide dismutase and catalase in leaves. Therefore, this process alleviated the damage of Cd to functional tissues of Perilla frutescens, thus improving the tolerance of plants to Cd. Moreover, the BV application reduced the Cd content bound by root cell wall pectin fractions and insoluble phosphate, subsequently improving the ability of oxalic acids to carry Cd to the aerial parts. Consequently, the aerial parts obtained a larger amount of Cd enrichment. Overall, the Transfer Factor of Cd from roots to stems and enrichment of Cd in Perilla frutescens were maximally increased by 57.70 % and 54.03 % with the application of 50-fold and 300-fold diluted BV under 2 mg & sdot;L- 1 Cd stress, respectively. The results can provide a theoretical basis for the promotion of phytoremediation of Cd-contaminated soil treatment technology.

Spatial analysis and risk assessment of heavy metal pollution in rice in Fujian Province, China SCIE

Abstract :

To comprehensively understand the spatial characteristics and pollution hazards of heavy metals in rice in Fujian Province, the contents of Cd, Hg, As, Pb, Cr, Ni, Cu, and Zn in 260 rice samples were measured based on the field investigation. Their spatial distribution patterns and clustering characteristics were explored by geostatistical methods, and their pollution-health risks were revealed by the pollution index method and exposure evaluation model. The results showed that the average contents of eight heavy metals in rice were below the food safety limits, but the excess rates of As, Cu, and Cd samples reached 33.08%, 22.69%, and 18.85%, respectively. In terms of spatial characteristics, the contents of Cd, Hg, and Ni were higher in the southeastern and northwestern regions, while the contents of As, Cu, and Cr were higher in the southwestern parts of Fujian Province. The hotspots of the corresponding heavy metals were also concentrated in the above-mentioned areas, showing an obvious spatial clustering effect. The pollution risk assessment showed that the single factor pollution index of heavy metals in rice decreased in the order of As (0.536) > Hg (0.505) > Cu (0.421) > Cd (0.378) > Cr (0.318) > Zn (0.304) > Pb (0.286) > Ni (0.116), and the Nemerow comprehensive pollution index was 0.623, which was generally at a safe level with low pollution risk. Furthermore, the non-carcinogenic risk index of heavy metals for adults and children were 3.558 and 6.014, and the carcinogenic risk index were 0.0050 and 0.0084, respectively, with extremely serious health hazards. In contrast, the health damage of heavy metals was higher in children than that in adults, with As and Cd in rice as the pivotal elements. Therefore, attention should be paid to the future monitoring and agricultural management of heavy metal pollution in rice in the region.

不同镉耐性水稻品种资源的筛选

Abstract :

为筛选出不同镉耐性的水稻品种,收集了238份我国南方4省主栽的水稻种质资源.通过分析不同浓度镉处理的种子萌发及幼苗生长指标,发现镉污染显著抑制水稻种子萌发和幼苗生长,不同水稻品种的镉耐性存在显著差异;通过系统聚类,初步筛选出高耐性水稻品种43个、低耐性水稻品种67个;通过复筛获得了稳定的高镉耐性水稻品种4个(金丝软占、宜优673、福龙优两优6387和特优671),低镉耐性水稻品种8个(T两优明占、Y两优837、百香139、丰田优553、广8优165、过山香、两优688、隆两优1377).研究结果为水稻镉污染适应机制研究及镉低积累水稻育种提供一定基础.

[目的]本文选择不同化学生态型紫苏,从叶片类黄酮与总酚含量角度讨论评估镉污染对不同化学生态型紫苏活性成分的影响,为不同化学生态型紫苏资源的药用价值提供参考.[方法]以刻度尺测量株高、根长.以电子天平测定鲜重、干重,采用NaNO2-Al(NO3)3-NaOH比色法测定类黄酮含量,采用福林酚法测定总酚含量.[结果]镉胁迫下,PK型、EK型生长发育情况随镉处理浓度上升总体表现出"低促高抑"的现象.镉胁迫总体上抑制PKPA型生长发育,抑制效果随镉浓度升高显著提高.PK型叶片类黄酮、总酚含量、PKPA型叶片类黄酮含量、EK型叶片总酚含量随镉处理浓度升高呈先升后降的趋势;PKPA型叶片总酚含量随镉处理浓度升高而升高,EK型叶片类黄酮含量在低浓度镉胁迫下变化不显著,高浓度镉胁迫下叶片类黄酮含量显著降低.[结论]低浓度镉环境能够促进PK型、PKPA型叶片类黄酮、总酚物质生成;EK型类黄酮、总酚合成受镉污染影响较小.
