AIEgen-based metal-organic gels for rapid and ultrasensitive discrimination of piceatannol from core-isostructural analogues SCIE

Abstract :

A highly stable and strongly fluorescent metal-organic gel, Zr-TCBPE-MOG, was successfully synthesized through the combination of aggregation-induced emission luminogens (AIEgens) 1, 1, 2, 2-tetra(4-carboxylbiphenyl) ethylene (H4TCBPE) with Zr (IV) cations. The fluorescence of Zr-TCBPE-MOG exhibits a sensitive response to piceatannol (3,3 ',4,5 '-tetrahydroxy trans-stilbene, PCT), whereas its structural analogues, such as resveratrol, pterostilbene, rhapontigenin, and pinosylvin, which share the same core skeleton, have no effect on it. Therefore, Zr-TCBPE-MOG can function as a fluorescent probe for rapid differentiation and detection of PCT among these structural analogues. A simple and rapid fluorescence method was established via the selective fluorescence quenching effect of PCT on Zr-TCBPE-MOG through the photoinduced electron transfer effect. Under optimal measurement conditions, the proposed method achieves high sensitivity for PCT with a linear detection concentration range from 3.5 to 250 nM and a low detection limit of 2.8 nM. It also can realize selective determination of PCT in serum samples. This work reveals the great potential of MOG for the rapid and highly specific differentiation of structurally similar drugs.

Abstract :

Rapid, sensitive and specific detection of foodborne pathogens is of great significance to the early warning and prevention of foodborne diseases and environmental pollution. Here, an electrochemiluminescent (ECL) biosensor with the cascaded reaction between two nanozymes was developed for the detection of E. coli O157:H7. Cobalt single-atom catalyst (Co-SAC@NC) with oxidase (OD) -like activity, as a co-reaction accelerator of O-2, catalyzed oxygen to reactive oxygen species (ROSs) and boosted ECL of luminol-O-2 system. Another nanozyme CeO2@mrGO with excellent phosphatase-like activity by the introduction of Fe was use to label Ab. CeO2@mrGO captured E. coli O157:H7, and then combined with Ap on the electrode surface through the affinity of aptamer and antibody to form a sandwich structure on the gold electrode (CeO2@mrGO-Ab/E. coli O157:H7/Ap-AuE). CeO2@mrGO converted AAP into AA, which consumed ROS and quenched ECL emission of luminol-O-2 system. As a consequence, this ECL sensor had a linear response range of 17-1.7 x 10(5) CFU/mL with LOD of 2.78 CFU/mL. This aptamer-antibody birecognized system was able to be applied to assay E. coli O157:H7 in untreated contaminated milk samples with satisfactory results.

Effect of inorganic arsenic in paddy soil on the migration and transformation of selenium species in rice plants SCIE

Abstract :

Selenium (Se) in paddy rice is one of the significant sources of human Se nutrition. However, the effect of arsenic (As) pollution in soil on the translocation of Se species in rice plants is unclear. In this research, a pot experiment was designed to examine the effect of the addition of 50 mg As/kg soil as arsenite or arsenate on the migration of Se species from soil to indica Minghui 63 and Luyoumingzhan. The results showed that the antagonism between inorganic As and Se was closely related to the rice cultivar and Se oxidation state in soil. Relative to the standalone selenate treatment, arsenite significantly ( p < 0.05) decreased the accumulation of selenocystine, selenomethionine and selenate in the roots, stems, sheaths, leaves, brans and kemels of both cultivars by 21.4%-100.0%, 40.0%-100.0%, 41.0%-100%, 5.4%96.3%, 11.3%-100.0% and 26.2%-39.7% respectively, except for selenocystine in the kemels of indica Minghui 63 and selenomethionine in the leaves of indica Minghui 63 and the stems of indica Luyoumingzhan. Arsenate also decreased ( p < 0.05) the accumulation of selenocystine, selenomethionine and selenate in the roots, stems, brans and kernels of both cultivars by 34.9%-100.0%, 30.2%-100.0%, 11.3%-100.0% and 5.6%-39.6% respectively, except for selenate in the stems of indica Minghui 63. However, relative to the standalone selenite treatment, arsenite and arsenate decreased ( p < 0.05) the accumulation of selenocystine, selenomethionine and selenite only in the roots of indica Minghui 63 by 45.5%-100.0%. Our results suggested that arsenite and arsenate had better antagonism toward Se species in selenate-added soil than that in selenite-added soil; moreover, arsenite had a higher inhibiting effect on the accumulation of Se species than arsenate. Abstract :

纳米酶是一类既有纳米材料的独特性能又有催化功能的模拟酶,它既有天然酶的高催化活性,又有模拟酶稳定经济的特点,在生物传感检测领域的应用十分广泛。本研究采用水热法合成具有辣根过氧化物酶样活性的氧化铈纳米酶。该纳米氧化铈同时具有+4价和+3价两种形态,其中Ce~(4+)能与过氧化氢反应,使3,3',5,5’-四甲基联苯胺(TMB)氧化,溶液显蓝色。由于抗坏血酸(AA)能还原Ce~(4+),抑制其催化活性以及TMB的氧化,从而使溶液的颜色较浅。而抗坏血酸氧化酶可催化氧化AA,阻碍了其对Ce~(4+)的还原,使纳米氧化铈的催化性能恢复。利用这种方法,实现对抗坏血酸氧化酶的定量检测,在12 U/L~400 U/L范围内呈良好的线性关系(R~2=0.9704),检测限为3.2 U/L。该研究为基于氧化铈纳米酶的比色方法在抗坏血酸氧化酶活性检测及相关应用提供基础。

Abstract :

Nitrogen (N), phosphorus (P) and potassium (K) are three macroelements in agriculture production, but their combined effects on arsenic (As) toxicity and its translocation in rice plants are not clear. In this study, an orthogonal rotation combination based on different N, P and K (NPK) concentration was first designed to examine their combined effect on the As toxicity, its transformation and migration in rice plants based on the hydroponic culture and pot soil culture. The results showed that 2.0 mg/L arsenite (As(III)) had obvious toxicity on the growth of indica LuYouMingZhan (LYMZ) and the optimal NPK concentration was 28.41, 6 and 50 mg/L based on the quadratic regression of the recovery rate of chlorophyll SPAD value of indica LYMZ. The optimal NPK combination significantly alleviated the physiological toxicity of As(III) on indica LYMZ rice seedling and decreased the accumulation of inorganic As in their roots and shoots by 23.8+1.8 % and 33.4+2.4 % respectively; further pot culture from different As(III) polluted soil showed that the optimal NPK combination significantly increased the dry weight of roots, stems, sheaths and leaves of indica LYMZ rice plants as well as yield indicators by 6.4 %-61.7 % and 7.1 %-89.8 % respectively, decreased the accumulation of As(III) and arsenate by 6.25 %-100 % and 12.36 %-100 % respectively in their roots, stems, sheaths, leaves, brans and kernels except As (III) concentration in their sheaths, decreased the accumulation of dimethylarsenate in their sheaths, leaves, brans and kernels, and had the best repair effect on the translocation of As species in 50 mg/kg As(III)-added soil. Our study provided a desirable strategy for alleviating As toxicity in paddy soil and reducing As pollution in rice plants.

A simple and stable dual-emission fluorescent system based on R6G/Eu-MOF for the rapid and sensitive determination of orientin SCIE

Abstract :

A stable dual-emission fluorescent system, R6G/Eu-MOF, was obtained by simple mixing Eu-MOF with fluorescent rhodamine 6G (R6G). This system not only achieved robust dual luminescence performance at 430 nm (Eu-MOF) and 555 nm (R6G) under single-wavelength excitation, but also endowed it with significantly improved fluorescence stability. Orientin (OT), a bioactive traditional Chinese medicine (TCM) component, was found to efficiently quench the dual fluorescent emissions via the synergistic effect of dynamic quenching, photo-induced electron transfer and molecular interactions. Therefore, a rapid fluorescent assay for OT was developed based on the variations in fluorescence signals at 430 nm and 555 nm, as well as the alterations in the ratio between these two signals. This method can achieve highly sensitive determination of OT in the linear range of 1.1-220 mu M, with a LOD of 0.26 mu M at 430 nm and 0.16 mu M at 555 nm (3 sigma/N). The R6G/Eu-MOF hybrid exhibits significantly lower detection limits and a broader linear range for OT than either R6G or Eu-MOF alone. This work presents a simple strategy to improve the water stability of Eu-MOF and also provides a rapid analysis of small molecule drugs using a fluorescence approach based on a dual-emission MOF hybrid system.

A Sensitive and Quick Fluorescent Sensor for the "Turn-On" Detection and Imaging of Glutathione Based on Sulfur Quantum Dots and MnO2 Nanosheets SCIE

Abstract :

Glutathione (GSH) is one of the most abundant bioethanol antioxidants in living cells. Here, a fluorescent probe based on MnO2 nanosheets and sulfur quantum dots (SQDs) was fabricated. Because of the synergistic effect of IFE and FRET, the fluorescence from SQDs was quenched by MnO2 nanosheets. In the presence of GSH, the fluorescence of SQDs could be recovered because of the reduction of MnO2 nanosheets by GSH. The method can detect GSH in the concentration range of 5 similar to 1000 mu M with the detection limit as low as 1.26 mu M. This quick, easy, and cost-effective sensor could be used for the quantification of GSH in serum samples and the imaging of GSH in Escherichia coli O157:H7.

本发明公开了硫量子点材料在缓解苗期水稻铅毒害中的应用,硫量子点材料的制备方法包括:将预设重量份的升华硫粉、氢氧化钠加入到水中混合,再加入预设量PEG‑400,然后在预设温度和搅拌速度下,反应预设时长,反应所得的产物在剧烈搅拌下,加入预设浓度的H2O2进行反应,制得硫量子点材料;本发明方案合成的硫量子点材料的成本低且性质稳定,原料只需要升华硫粉、氢氧化钠、蒸馏水、PEG‑400、过氧化氢;除此之外,本发明方案硫量子点具有纳米材料缓释性的特点,能够调节缓解水稻生长周期中的铅毒害效应。

Abstract :

Many traditional Chinese herbs are susceptible to ochratoxin A (OTA), a potent mycotoxin, which causes serious effects on the quality of the herb and on people's health. The development of methods to detect OTA is extremely important. Most methods for detecting OTA are based on a single-signal output mode, which might be easily influenced by complex environmental conditions. In this research, by taking advantage of the cleavage of DNA by target-induced CRISPR-Cas12a activity and the difference in electrostatic force of DNA to different charge electrochemiluminescent (ECL) and electrochemical (EC) probes, a biosensor is developed for the detection of OTA. First, the CRISPR-Cas12a system consists of a well-designed crRNA, its complementary strand (also as an aptamer for OTA), and Cas12a. Without the target, this CRISPR-Cas12a system is in the "activated stage", which digests hairpin DNA on the electrode, resulting in a weak ECL signal and strong current response. With the introduction of OTA bound with the aptamer, CRISPR-Cas12a activity is inhibited ("locked stage"). Thus, hairpin DNA remained intact on the electrode, resulting in recovery of the ECL signal and attenuation of the current intensity. As a result, this label-free dual-mode sensing platform realizes an assay for OTA in Morinda officinalis. This target-regulated CRISPR-Cas12a activity-sensing platform with dual-mode output not only provides high sensitivity (due to the CRISPR-Cas12a system), but also has good anti-interference ability against complex substrates (due to dual-mode output), and exhibits a broad range of prospects for application.

Abstract :

Zearalenone (ZEN), a widespread mycotoxin, can cause great harm to people's health. In order to assay ZEN, an immobilization-free electrochemical sensor has been developed. A multifunctional hairpin DNA has been carefully designed, including three functions: the aptamer for zearalenone (ZEN), primer, and template sequence. This hairpin DNA can anchor on polydopamine nanospheres (PDANSs), which can protect DNA against the digestion of enzymes and prevent the occurrence of strand displacement amplification (SDA). In the presence of ZEN, the hairpin DNA is dissociated from PDANSs due to the interaction between ZEN and the aptamer, and the SDA reaction is initiated with the help of endonuclease and polymerase. During the SDA process, substantial amounts of negatively charged dsDNA are generated. The MB molecules are embedded into the dsDNA grooves to obtain the complex with a negative charge. The confined MB is repelled on the surface of the negatively charged ITO electrode, leading to the decline of the current.
