西藏血红密孔菌的驯化栽培及营养评价

Abstract :

【目的】对采自西藏地区的野生菌株X21020进行鉴定,探究其最佳培养条件及驯化栽培条件,并测定其驯化栽培子实体的常规营养成分,为血红密孔菌的进一步开发利用提供参考。【方法】采用组织分离法对野生菌株X21020进行分离纯化,采用分子测序法和形态学特征观察法对其进行菌种鉴定,探究其生物学特性并开展驯化栽培,测定驯化栽培成功子实体的常规营养成分。【结果】结合形态学特征观察和ITS测序,将野生菌株X21020鉴定为血红密孔菌(Fabisporus sanguineus)。该菌株菌丝生长的最适碳源为葡萄糖,最适氮源为蛋白胨,p H为5,最适培养温度为40℃。该菌株在以杂木屑为主的栽培料中生长良好,菌丝满袋时间为30 d,原基形成时间为10 d,80 d后采收第一潮菇,单朵子实体鲜质量为32.82 g。该菌株子实体中膳食纤维含量高达7.110 g/kg,粗蛋白含量达1.190 g/kg,脂肪含量仅为0.020 g/kg。含有15种氨基酸,氨基酸总含量为0.486 g/kg。其中药效氨基酸含量为0.109 g/kg,占氨基酸总含量的22.43%,且以天冬氨酸含量最高(0.061 g/kg)。【结论】西藏来源的血红密孔菌X21020可在以杂木屑为主的栽培料中良好生长,其驯化子实体含有丰富的蛋白质和膳食纤维,氨基酸种类多,脂肪含量低,该结果为其在医学和保健品领域的开发利用提供了参考。

Abstract :

粗毛纤孔菌是一种天然的药用真菌,具有多种生物活性,其子实体颜色在成熟后期可由黄褐色转变为黑色,即为黑色素。通过探究其理化性质发现,粗毛纤孔菌黑色素(IHFM)属于真黑色素,具有良好的热、光和及金属稳定性等。黑色素是粗毛纤孔菌的活性成分之一,具有保肝、抗癌、调节肠道菌群结构、抗氧化等活性。首先,在保肝方面的研究显示,IHFM一方面通过抑制Keap-1、激活Nrf2信号通路显著提高下游GCLC、HO-1、NQO1等的表达;另一方面显著抑制MyD88介导的TLR4/NF-κβ信号通路并降低下游炎症因子IL-1β、TNF-α、IL-6等的表达水平,进而缓解小鼠酒精性引发的肝损伤。在调节肠道菌群方面,IHFM可以调节酒精性肝损伤小鼠中肠道菌群的结构,上调肠道菌群中有益菌的相对丰度,降低有害菌的相对丰度。综上,我们的研究可为IHFM开发为保肝、抗氧化剂等相关产品奠定理论基础。

本发明为一种粗毛纤孔菌黑色素的制备方法和应用,粗毛纤孔菌黑色素的制备方法,包括如下步骤:S1,收集粗毛纤孔菌子实体并烘干粉碎后经筛网筛选过滤;S2,称取粉末并加入NaOH溶液,并置于超声波中充分反应;S3,在离心机上经离心处理后,收集上清液;S4,在S3收集的上清液中加入酸性溶液并加热;S5,再次离心后收集沉淀物;S6,将沉淀物进行纯化;S7,离心收集上清液,最终通过流水透析后,冷冻干燥后获得纯化后的粗毛纤孔菌子实体黑色素样品。所述粗毛纤孔菌黑色素能够显著抑制人乳腺癌细胞生长、增殖、迁移的能力;并诱导人乳腺癌细胞的凋亡和人乳腺癌细胞焦亡。因此本发明粗毛纤孔菌黑色素具有开发为抗乳腺癌药物的潜能。

Abstract :

The selection and breeding of high-quality wild edible fungal strains can bring significant economic and social benefits. A wild fungal strain (X21185) from the Tibetan Plateau was isolated and identified as a novel Pleurotus pulmonarius (P. pulmonarius) based on its morphological and molecular characteristics. The appropriate culture conditions for P. pulmonarius were determined. The nutrient contents of P. pulmonarius fruiting bodies were analyzed. Compared with the conventional nutritional contents of the representative edible fungi (Pleurotus ostreatu and Pleurotus eryngii) and egg, the protein, ash, and dietary fiber contents of P. pulmonarius were higher. Four types of essential amino acids, seven types of nonessential amino acids, the total essential and nonessential amino acids of P. pulmonarius were present in considerably higher quantities than those of representative edible fungi (Pleurotus ostreatus and Pleurotus citrinopileatus) and egg, respectively. P. pulmonarius polysaccharides (PPPs) had strong ABTS+, DPPH, and hydroxyl free radical scavenging activities (EC50: 0.051, 3.322, and 2.87 mg/mL, respectively), and the cytotoxicity was higher against HepG2 hepatocellular carcinoma cells (IC50: 1.501 mg/mL) than against MDA-MB-468 triple-negative breast cancer cells (IC50: 2.183 mg/mL). This study provides a foundation for the development of the novel wild P. pulmonarius strain.

Abstract :

Natural melanin biopolymers exhibit a variety of biological activities, but their commercial development is constrained by numerous factors, including high costs, unsustainable sources, the use of harmful solvents during extraction, and low extraction efficiency. Notably, existing research indicates that synthetic melanin differs from natural melanin in nature, and this difference may directly impact its application efficacy. Additionally, the extraction process itself is highly challenging, primarily due to the diversity and complexity of melanin biopolymer structures. The melanin produced by edible fungi primarily belongs to the eumelanin category. Given its outstanding sustainability and accessibility, it is regarded as an ideal raw material for industrial production. To deepen our understanding of edible fungus-derived melanin and promote its effective application across various fields, a comprehensive review of research on melanin isolated from edible fungi is urgently needed. Such a review will help researchers from different disciplinary backgrounds recognize the importance of edible fungus melanin and provide reference information for their research planning. With this objective in mind, this report reviews the latest research progress in recent years regarding extraction methods, structural characterization, biological activity, and application areas of edible fungus-derived melanin. Additionally, the report explores key characteristic parameters for distinguishing different types of melanin and emphasizes the importance of deepening our understanding of the biosynthetic mechanisms of edible mushroom melanin, aiming to lay the foundation for its efficient production and application in the future.

Characteristics, Domestication and Nutrient Analysis of Wild Pleurotus placentodes SCIE

Abstract :

To enhance the systematicity and integrity of mushroom resource research by supplementing the mushroom germplasm bank through the collection of wild mushroom specimens, this project focused on a wild mushroom strain collected from Tibet Autonomous Region. The strain was identified and characterized through morphological observation, ITS sequencing, biological trait analysis, and cultivation experiments. The results indicated that the strain belongs to Pleurotus placentodes. The optimal growth conditions for this strain were determined as follows: fructose and sucrose as the preferred carbon source, yeast powder and beef powder as the optimal nitrogen source, pH 5.0, and a temperature of 20 degrees C. In cultivation experiments, the mycelium fully colonized the substrate in 23 days, primordia formation occurred within 10 d, and the fruiting period lasted 7 d, with an average fresh weight of 39.40 g per fruiting body. Nutritional analysis revealed that P. placentodes has a high protein content (30.30%), high soluble sugar content (6.60%), low fat content (1.00%), and a rich profile of essential amino acids (7.96%). This study successfully isolated, identified, and conducted domestication cultivation and nutritional analysis of a wild P. placentodes, providing scientific references for its potential industrial-scale production.

Domestication Cultivation and Nutritional Analysis of Hericium coralloides SCIE

Abstract :

Hericium coralloides is a valuable medicinal and edible mushroom renowned for its unique bioactive compounds. This study focuses on the isolation of a wild strain (SH001) exhibiting promising cultivation potential and health promoting properties. A wild fungal strain from the Tibetan Plateau was isolated and identified as a novel H. coralloides based on its morphological and molecular characteristics. The optimal growth conditions were found to be 30 degrees C, pH 7.0, fructose as the preferred carbon source, and yeast extract as the optimal nitrogen source. Nutritional analysis revealed that the fruiting bodies were rich in protein (15.4 g/100 g dry weight), dietary fiber (34.7 g/100 g dry weight), and minerals, while being low in fat (3.5 g/100 g dry weight). The most abundant amino acids were glutamic acid, followed by aspartic acid. The polysaccharides exhibited significant antioxidant activity, with ABTS+ scavenging comparable to that of Vitamin C (Vc), achieving a clearance rate of 96.95% at concentrations between 0.25-5.00 mg/mL. At a concentration of 5 mg/mL, the DPPH and OH radical scavenging activities reached their peak (83.77% and 67.31%, respectively), along with the highest iron ion reducing capacity (FRAP value: 4.43 mmol/L. Polysaccharides also exhibited notable anticancer activity, inhibiting HepG2 liver cancer cells and MDA-MB-468 breast cancer cells, with IC50 values of 3.896 mg/mL and 2.561 mg/mL, respectively. This study demonstrates that wild H. coralloides can be successfully cultivated in vitro. In conclusion, the fruiting bodies possess substantial nutritional value, and the polysaccharides extracted from them show promising antioxidant and anticancer activities, particularly against HepG2 liver cancer cells and MDA-MB-468 breast cancer cells.

Abstract :

A fungal strain (X21156) collected in Tibet was used as the material, identified based on its morphological characteristics and internal transcribed spacer (ITS) sequence; its optimal culture conditions were analyzed by single-factor experiments; artificial domestication and cultivation were carried out; its nutrient composition was determined; and the bioactivities of its polysaccharides were detected using chemical antioxidant assays and MTT assays. The results showed that the strain was Pleurotus citrinopileatus Sing. Its optimal culture conditions were a pH of 7, a temperature of 25 degrees C, glucose (20 gL-1) as the carbon source, and yeast powder (20 gL-1) as the nitrogen source. The fresh weight of a single domesticated fruiting body was 41.16 g. The strain had high protein (28.5%), high fiber (34%), and low fat (1.4%) contents, with high proportions of fresh and sweet amino acids. Polysaccharides had good scavenging ability on ABTS+, DPPH, and OH free radicals (EC50 0.06 mg/mL, 1.21 mg/mL, and 3.62 mg/mL, respectively), and the cytotoxicity of polysaccharides to hepatocellular carcinoma cells (HepG2) (IC50: 1.69 mg/mL) was higher than that of triple-negative breast cancer cells (MDA-MB-468) (IC50: 1.76 mg/mL). In conclusion, the study provides a reference on the optimal culture conditions, domestication and cultivation, and dietary and medicinal values of wild P. citrinopileatus Sing.
