Abstract :
Background: Type 2 diabetes mellitus (T2DM) is a serious metabolic syndrome with high mortality and disability rates globally, which usually caused by unhealthy dietary patterns. Cinnamomum camphora leaf is a traditional Chinese medicinal herb used for attenuating hyperglycemia and digestive disorder, and high level of lignans has been found in C. camphora leaf. Purpose: This study aimed to examine the chemical composition of lignans extracted from C. camphora leaf (LCCL), and illustrate its therapeutic effect and mechanism on T2DM and its concomitant glycolipid metabolic disorder. Methods: The components of LCCL were separated and purified by silica gel and macroporous adsorption resin, and were distinguished through LC/MS and NMR. The antioxidant activity of LCCL was determined by free radical scavenging assay in vitro; the hypoglycemic and hypolipidemic abilities were evaluated by alpha-glucosidase, alpha-amylase and pancreatic lipase inhibition trials, respectively. T2DM model mice were established by high-sugar and high-fat (HSHF) feed together with streptozotocin (STZ) infection, and then grouped to assess the effect of LCCL treatment. Hematoxylin-eosin (H&E), Periodic Acid-Schiff (PAS) and oil red O staining were employed to analyze the histopathology. qRT-PCR assay, 16S rRNA analysis, and western blot were conducted to illuminate the anti-diabetic mechanism of LCCL. Results: 6 sesamin lignans were identifed from LCCL. The in vitro assays showed strong inhibitive abilities of LCCL with low IC50 on DPPH (33.68 +/- 0.54 mu g/ml),O-2(-) (39.25 +/- 0.61 mu g/ml), OH center dot (45.72 +/- 0.72 mu g/ml), alpha-glucosidase (0.82 +/- 0.14 mg/ml), alpha-amylase (0.86 +/- 0.11 mg/ml) and pancreatic lipase (0.91 +/- 0.12 mg/ml). LCCL treatment (100, 200 and 400 g kg(-1mg kg-1)) gradually decreased the fasting blood glucose (FBG) and fasting insulin (FINS), improved the glucose and insulin tolerance, down-regulated the homeostasis model assessment insulin resistance (HOMA-IR) indexes, alleviated the hepatic inflammatory response and oxidative stress, promoted the glycogen storage and depleted the fat accumulation in the liver. Besides, LCCL administration alleviated the glycolipid metabolism disorder in T2DM mice with a gut microbiota dependent manner, that significantly increased biodiversity, altered the composition of gut microbiota and increased the proportion of Lactobacillus. Conclusion: The lignan-rich extract of C. camphor leaf (LCCL), containing at least 6 lignans compounds, displayed promising antioxidant, hypoglycemic and hypolipidemic activities. The treatment of LCCL alleviated the glycolipid metabolism disorder in T2DM mice with a gut microbiota dependent manner. These finding suggested that LCCL should be further investigated to develop its complementary therapeutic effect on T2DM.

本发明涉及一种来源于圆齿野鸦椿编码色原酮糖基转移酶的基因和蛋白序列及其重组蛋白的应用,属于生物技术领域。所述基因序列如SEQ ID NO.1所示,序列全长945bp;编码蛋白的氨基酸序列如SEQ ID NO.2所示,编码了315个氨基酸残基,可用于制备黄酮、色原酮碳苷化合物;将所述基因序列连接至表达载体,转入受体细胞,可在植物体外表达所述糖基转移酶;该酶以UDP‑葡萄糖为糖基供体,催化底物2‑羟基柚皮素及去甲丁香色原酮,分别合成黄酮及色原酮碳苷化合物。

【目的】研究栀子花多糖的提取工艺及其脂代谢调节作用,开发栀子花多糖资源。【方法】采用单因素试验和响应面法进行多糖提取工艺优化,并采用油酸钠诱导建立HepG2细胞脂肪积累模型,评价各栀子花多糖提取物对HepG2脂肪积累细胞中脂代谢指标的影响。【结果】在液料比25mL·g~(-1)、超声功率550 W、超声时间140 min、提取温度63℃的条件下,栀子花多糖的提取率高达17.48%,且栀子花多糖可下调高脂HepG2细胞的甘油三酯(TG)、总胆固醇(TC)和低密度脂蛋白(LDL-C)含量,上调细胞内高密度脂蛋白(HDL-C)含量,不影响HepG2细胞的存活率。【结论】栀子花多糖具有良好的降血脂作用。

Abstract :
为探究不同贮藏年份政和白茶的香气特征,采用定量描述分析结合顶空固相微萃取-气相色谱-质谱联用技术,对贮藏0、5、10、15年的政和白牡丹白茶(分别编号BMD0、BMD5、BMD10和BMD15)的香气特征与挥发性成分进行分析。结果表明,不同贮藏年份政和白牡丹的香气特征差异明显,BMD0以毫香、清香、甜香、花香为主,随贮藏时间延长,毫香、清香、甜香、花香减弱,陈香、木香增强。不同贮藏年份的政和白牡丹共检测出66种挥发性成分,以醇类、酯类、酸类为主,BMD0和BMD5醇类含量较高,BMD10和BMD15酯类含量较高,随贮藏年份增加,挥发性成分总量显著降低,相对含量的组成与占比变化显著。所建立的正交偏最小二乘判别分析模型,可将不同贮藏年份政和白牡丹进行有效区分,分别以相对香气活性值(Relative odor activity value,rOAV)>1、变量投影重要性(Variable important for the projection,VIP)>1为依据,筛选出14种和30种差异挥发性成分;以rOAV>1且VIP>1为依据,筛选出β-紫罗兰酮、α-紫罗兰酮、反式-橙花叔醇、苯甲醇、苯乙醛、α-亚麻酸共6种关键差异挥发性成分。本研究为白茶科学储藏、陈年白茶风味解析提供理论参考与依据。

为开发栀子花的营养与药用价值,采用单因素法和均匀设计法分析由超声波法获得的栀子花提取物,通过测定各提取物中环烯醚萜苷、黄酮、三萜的含量及各提取物对α-葡萄糖苷酶活性的抑制率来优化栀子花的提取工艺,并采用胰岛素抵抗的人肝癌HepG2细胞模型评价采用优化工艺获得的提取物对糖代谢的调节作用。结果显示,优化后的栀子花提取工艺为:乙醇体积分数80%、液料比25∶1(mL∶g)、时间120 min、温度60℃。此优化工艺对环烯醚萜苷、黄酮和三萜的提取率分别为2.61%、5.37%、2.84%,对α-葡萄糖苷酶活性的抑制率可达78.2%,能有效提高胰岛素抵抗细胞HepG2的糖代谢能力,降低细胞氧化应激和炎症反应,从而改善细胞胰岛素抵抗状态。

Abstract :
Ethnopharmacological relevance: Euscaphis konishii Hayata is a traditional medicinal plant in China, and its leaves are usually used to make dishes for hepatic or gastrointestinal issues by Chinese She nationality. Pharmacological analysis showed that E. konishii leaves contain high levels of flavonoids and chromones with favorable antihepatoma effect. Aim of the study: The extract from E. konishii leaves was detected to evaluate its chemical composition, and the alcoholic liver injury mice model was adopted to elucidate its hepatoprotective effects. Materials and methods: The total leaf extract from E. konishii was separated by polyamide column to get the flavonoid and chromone-rich extract (FCE). Single compounds from FCE was purified by gel and Sephadex LH-20 chromatography and analyzed by nuclear magnetic resonance (NMR). The chemical component of FCE was confirmed and quantified by HPLC-MS. The OH., O2- , DPPH and ABTS + free radical assays were adopted to estimate the antioxidant activity of FCE in vitro. The alcohol-fed model mice were established to assess the hepatoprotective capacity of FCE in vivo, through biochemical determination, histopathological analysis, mitochondrial function measurement, quantitative Real-Time Polymerase Chain Reaction (qRT-PCR) detection and Western blot determination. Results: 8 flavonoids and 2 chromones were recognized in the FCEextract by both NMR and HPLC-MS. FCE represented strong free radicals scavenging activity in vitro. With oral administration, FCE (50, 100 and 200 mg/ kg) dose-dependently decreased the serum levels of alanine aminotransferase (ALT), alkaline phosphatase (ALP) and aspartate aminotransferase (AST) in alcohol-fed mice. FCE gradually reduced the malondialdehyde (MDA) content, increased the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in the alcohol-treated liver tissues. FCE also alleviated the hepatic inflammation, inhibited the hepatocyte apoptosis and lessened the alcohol-induced histological alteration and lipid accumulation in the liver tissues. FCE administration inhibited the overexpression of endoplasmic reticulum (ER) chaperones signaling and unfolded protein response (UPR) pathways to defense the ER-induced apoptosis. Pretreatment with FCE also restored the mitochondrial membrane potentials andadenosine triphosphate (ATP) levels, which in turn suppressed the Cytochrome C release and mitochondria-induced apoptosis. Conclusions: FCE conferred great protection against alcoholic liver injury, which might be associated with its viability through suppressing reactive oxygen species (ROS) stress and hepatocyte apoptosis.

通过自由基清除试验检测Biflorin和Isobiflorin的体外抗氧化活性;构建H_2O_2诱导的L02人肝细胞氧化损伤模型,以MTT法和化学荧光法测定两者对细胞存活率和ROS消除率的影响;运用RTq-PCR和Western blot分析两者对抗氧化信号通路中关键基因表达的影响.结果显示Biflorin和Isobiflorin有明显去除DPPH、·OH和ABTS~+自由基的能力,提升了被H_2O_2氧化后L02细胞的生存率和ROS去除能力,并能促使肝细胞Nrf2蛋白通过胞浆向核内转移,进而显著上调了抗氧化反应信号途径中HO-1、NQO-1的mRNA和蛋白的表达.结果表明Biflorin和Isobiflorin具有显著的抗氧化活性,通过调控抗氧化信号相关的通路基因和蛋白的表达来抑制细胞的氧化损伤.

本发明涉及一种常绿与落叶类野鸦椿分子标记分类与鉴别方法,收集全国常绿类野鸦椿、落叶类野鸦椿、建宁野鸦椿等群体进行简化基因组测序,获得SNPs;基于SNPs,利用MEGA X软件构建Neighbor‑joining;进化树的结果表明:常绿类野鸦椿和落叶类野鸦椿为姐妹类群,其中建宁野鸦椿群体归并到落叶类野鸦椿,位于落叶类最末尾的进化分支,结果可证实之前认为建宁野鸦椿是野鸦椿的变种是正确的;常绿类野鸦椿就是圆齿野鸦。为野鸦椿属科学分类或生产实践中科学种植区划提供好方法,并能有效防止混种造成的严重损失。

Malvids is one of the largest clades of rosids, includes 58 families and exhibits remarkable morphological and ecological diversity. Here, we report a high-quality chromosome-level genome assembly for Euscaphis japonica, an early-diverging species within malvids. Genome-based phylogenetic analysis suggests that the unstable phylogenetic position of E. japonica may result from incomplete lineage sorting and hybridization event during the diversification of the ancestral population of malvids. Euscaphis japonica experienced two polyploidization events: the ancient whole genome triplication event shared with most eudicots (commonly known as the gamma event) and a more recent whole genome duplication event, unique to E. japonica. By resequencing 101 samples from 11 populations, we speculate that the temperature has led to the differentiation of the evergreen and deciduous of E. japonica and the completely different population histories of these two groups. In total, 1012 candidate positively selected genes in the evergreen were detected, some of which are involved in flower and fruit development. We found that reddening and dehiscence of the E. japonica pericarp and long fruit-hanging time promoted the reproduction of E. japonica populations, and revealed the expression patterns of genes related to fruit reddening, dehiscence and abscission. The key genes involved in pentacyclic triterpene synthesis in E. japonica were identified, and different expression patterns of these genes may contribute to pentacyclic triterpene diversification. Our work sheds light on the evolution of E. japonica and malvids, particularly on the diversification of E. japonica and the genetic basis for their fruit dehiscence and abscission.
