The genetic structure and selective sweep analysis of meat pigeon breeds raised in Hainan using whole genome resequencing technology SCIE

Abstract :

To investigate the genetic relationships and domestication status of Hainan meat pigeon populations, wholegenome sequencing was performed on 70 individuals across seven breeds: Shiqi (SQ), Silver King (SK), Shenwang (SW), Dabao (DB), Tianxiang No.1 (TX), Hybrid King (HK), and White Carneau (WC), with each breed represented by 10 individuals (5 males and 5 females). From 824 Gb of high-quality sequencing data, we identified 13,347,452 high-confidence single nucleotide polymorphisms (SNPs). Population structure analysis revealed divergent genetic clustering: WC, DB, TX, and HK individuals demonstrated extensive genomic admixture, while SK, SQ, and SW breeds formed distinct clusters with minimal interbreed genetic exchange. Selective sweep analysis highlighted genes under strong positive selection (TAFA2, FNDC3B, TFAP2B, PDCL3, EPC1 and LHX2). Functional annotation linked TAFA2 to accelerated growth in SK pigeons through muscle development modulation. The genes FNDC3B and TFAP2B were associated with fat deposition capacity in SQ pigeons. Concurrently, PDCL3, NEK6, LHX2, and EPC1 were implicated in SW pigeons' large body size and superior production traits. This study provides the first genomic compendium for Hainan meat pigeons, delineating their genetic diversity, adaptive evolution, and domestication patterns. The identified selection signatures establish a foundation for precision breeding strategies targeting growth, reproduction, and climate resilience in commercial pigeon production.

姜黄素对热应激肉鸡盲肠菌群的影响

Abstract :

试验旨在研究不同水平姜黄素对热应激肉鸡盲肠菌群的影响。选择180只体重相近、健康的1日龄雄性艾拔益加(Arbor Acres,AA)白羽肉鸡,随机分为5个组,每组3个重复,每个重复12只鸡。对照组和热应激组饲喂基础饲粮,试验组在基础饲粮中分别添加75mg/kg、150mg/kg、300mg/kg的姜黄素。热应激试验于第21天开始,第42天结束。对照组的环境温度控制在(22±1)℃,24h;热应激组和试验组的环境温度控制在(34±1)℃,8h/d,其余时间控制在(22±1)℃。结果表明:(1)添加75mg/kg的姜黄素能够显著提升肉鸡盲肠菌群的多样性(P<0.05);(2)添加150mg/kg的姜黄素能够增加大肠杆菌属(Colidextribacter)比例;(3)KEGG功能富集分析结果显示,添加75mg/kg和150mg/kg的姜黄素能够提高氨基酸合成与代谢、碳代谢、糖酵解和糖异生、半胱氨酸和蛋氨酸代谢等通路的相对丰度。综上所述,饲粮中添加姜黄素在一定程度能够调节热应激肉鸡盲肠微生物平衡,维持肠道健康。

Abstract :

为了降低嘉积鸭饲粮中豆粕使用量,开发新的饲料资源,试验选用360只40日龄健康嘉积鸭母鸭,按照体重相近原则随机分成4组,每组6个重复,每个重复15只鸭。对照组饲喂基础饲粮,试验组在基础饲粮基础上分别用6%、12%和18%木薯叶粉部分替代豆粕,各组营养水平一致。试验期30 d。结果显示:与对照组相比,6%和12%木薯叶粉对嘉积鸭终末体重和平均日增重无显著影响(P>0.05),18%木薯叶粉显著降低嘉积鸭终末体重和平均日增重(P<0.05),并显著增加料重比(P<0.05);6%和12%木薯叶粉显著提高嘉积鸭胸肌率(P<0.05),18%木薯叶粉显著降低嘉积鸭腹脂率(P<0.05),6%、12%和18%木薯叶粉显著降低嘉积鸭皮脂率(P<0.05);18%木薯叶粉显著降低嘉积鸭血清天门冬氨酸氨基转移酶活性和尿素含量(P<0.05),12%木薯叶粉显著降低嘉积鸭血清总蛋白含量(P<0.05),6%、12%和18%木薯叶粉显著降低嘉积鸭血清尿酸含量(P<0.05)。研究表明,嘉积鸭饲粮中使用6%~12%木薯叶替代豆粕可行,其中12%木薯叶粉在维持正常生长性能基础上,比6%木薯叶粉具有更高的终末体重和胸肌率,建议作为嘉积鸭饲料中木薯叶粉的适宜添加水平。

Effects of curcumin on jejunal morphology and intestinal integrity of broiler under heat stress SCIE

Abstract :

Heat stress adversely affects poultry production, intestinal health, and antioxidant levels. This study investigated the effects of curcumin supplementation on the jejunal morphology of broilers under heat stress. A total of 108 one-day-old male Arbor Acres broilers were divided into three groups: the control group (C), the heat stress group (HS), and the curcumin group (CUR). The C and HS groups were fed a basal diet, while the CUR group received 50 mg/kg of curcumin, supplemented from days 8-42. From days 21-42, the HS and CUR groups were exposed to 34 f 1 degrees C for 8 h daily (9:00-17:00), with the remaining time spent at 22 f 1 degrees C. The C group was maintained at 22 f 1 degrees C throughout the experiment. The results revealed that curcumin supplementation alleviated jejunal mucosal damage. under heat stress. It increased the villus height to crypt depth ratio (V/C ratio) and significantly upregulated the expression of TJP1, ZO-1, MLCK, and BCL-2. Additionally, curcumin downregulated the expression of CASP3 and CASP9 in the jejunum. Furthermore, curcumin activated autophagy by enhancing the expression of SIRT1, PGC-1 alpha, and Beclin1. In summary, curcumin supplementation can help maintain gut health in broilers under heat stress and exert protective effects at the cellular level.

福建省猪源葡萄球菌多重耐药基因的分子流行病学调查

【目的】明确多重耐药基因cfr、optr A和poxt A在福建省猪源葡萄球菌中的分子流行特征,为耐药葡萄球菌的控制提供科学依据。【方法】通过PCR扩增16S r RNA对2020-2022年分离自福建省福州市、厦门市、漳州市、三明市、龙岩市、南平市、宁德市不同猪场的107株菌(样品为鼻拭子、唾液、粪便和肺脏)进行鉴定。对分离菌株进行cfr、optr A和poxt A多重耐药基因检测;采用MEGA软件对cfr和optr A阳性菌株进行系统发育分析;采用琼脂稀释法或微量肉汤稀释法对cfr和optr A基因阳性菌株进行3种抗菌药物(酰胺醇类抗菌药物氟苯尼考和氯霉素,恶唑烷酮类抗菌药物利奈唑胺)最小抑菌浓度(minimum inhibitory concentration,MIC)测定。【结果】经鉴定107株菌均为葡萄球菌,分属18个种,其中以科氏葡萄球菌占比最高。流行病学调查结果显示,107株葡萄球菌主要来自猪鼻拭子样品(占67.3%),并且在福州市猪场的样品中分离率最高(36.4%)。PCR筛查结果显示,在107株葡萄球菌中未检出poxt A基因阳性菌株;检测出14株cfr阳性菌和6株optr A阳性菌,检出率分别为13.1%和5.6%,其中葡萄球菌FJNP2209-1和FJCT2212-1同时携带cfr及optr A基因。系统发育分析结果显示,14株cfr阳性葡萄球菌处于同一个分支内,其中菌株FJFQ2211与所研究的其他菌株亲缘关系稍远;6株optr A基因阳性葡萄球菌属于2个分支。药敏试验结果表明,携带多重耐药基因cfr或optr A的18株葡萄球菌对氯霉素和氟苯尼考100%耐药,对利奈唑胺的耐药率为83.33%。【结论】福建省不同猪场来源的葡萄球菌中多重耐药基因cfr和optr A阳性率较高,但poxt A基因未检出。福建省猪源葡萄球菌对氟苯尼考、氯霉素、利奈唑胺均表现出较高的耐药率,应加强抗菌药物在养猪场中的规范使用。

Mechanism of fibroblast growth factor 1 regulating fatty liver disorder in mule ducks SCIE

Abstract :

Mule ducks tend to accumulate abundant fat in their livers via feeding, which leads to the formation of a fatty liver that is several times larger than a normal liver. However, the mechanism underlying fatty liver formation has not yet been elucidated. Fibroblast growth factor 1 ( FGF1 ), a member of the FGF superfamily, is involved in cellular lipid metabolism and mitosis. This study aims to investigate the regulatory effect of FGF1 on lipid metabolism disorders induced by complex fatty acids in primary mule duck liver cells and elucidate the underlying molecular mechanism. Hepatocytes were induced by adding 1,500:750 mmol/L oleic and palmitic acid concentrations for 36 h, which were stimulated with FGF1 concentrations of 0, 10, 100, and 1000 ng/mL for 12 h. The results showed that FGF1 signi ficantly reduced the hepatic lipid droplet deposition and triglyceride content induced by complex fatty acids; it also reduced oxidative stress; decreased reactive oxygen species fluorescence intensity and malondialdehyde content; upregulated the expression of antioxidant factors nuclear factor erythroid 2 related factor 2 ( Nrf2 ), HO -1, and NQO-1; signi ficantly enhanced liver cell activity; promoted cell cycle progression; inhibited cell apoptosis; upregulated cyclin-dependent kinase 1 ( CDK1 ) and BCL-2 mRNA expression; and downregulated Bax and Caspase-3 expression. In addition, FGF1 promoted AMPK phosphorylation, activated the AMPK pathway, upregulated AMPK gene expression, and downregulated the expression of SREBP1 and ACC1 genes, thereby alleviating excessive fat accumulation in liver cells induced by complex fatty acids. In summary, FGF1 may alleviate lipid metabolism disorders induced by complex fatty acids in primary mule duck liver cells by activating the AMPK signaling pathway.

Abstract :

Mule ducks accumulate a large amount of fat in their livers when fed high-energy feed, which is predominantly used for producing fatty livers. Nevertheless, there is limited research on the molecular mechanisms underlying the formation of fatty liver in mule ducks. Fatty acid translocase (CD36) is a sensor for fatty acids and lipid metabolism regulator, which may play a crucial role in the accumulation of fat in the liver of mule ducks. In this study, Overexpression and CD36 gene interference for 24 h was followed by induction of liver cells with 400 mmol/L palmitic acid (PA) for 24 h. The results demonstrated that CD36 overexpression increased hepatic triglyceride content, lipid droplet deposition, oxidative stress, and cell apoptosis. However, interference with CD36 had the opposite effect. CD36 overexpression suppressed the expression of AMPK and CPT-1A genes but enhanced the expression of ACC1 and LKB1 genes, with interference yielding contrasting results. Additionally, the expression of CD36 inhibited the AMPK pathway, reduced AMPK phosphorylation, downregulated AMPK protein expression, and upregulated SREBP1 protein expression. This promoted palmitic acid-induced hepatocyte fat accumulation. In summary, CD36 promotes palmitic acid-induced fat accumulation in primary mule duck liver cells through the AMPK signaling pathway.

Abstract :

To explore differences in the breast muscle meat quality and their potential regulatory mechanisms between 50d-old (D50) and 90-d-old (D90) Jiaji ducks, we investigated the development of the breast muscle tissue, contents of fatty acids (FAs) and amino acids (AAs), as well as metabolite and gene profiles in the breast muscle of both D50 and D90 ducks. The breast muscle rates at D90 were significantly higher than those at D50 (P < 0.05). The contents of docosahexaenoic acid; arachidonic acid; and most AAs at D90 were extremely significantly higher than those at D50 (P < 0.01). Metabolomics and transcriptomics analysis identified several meat quality-related differential metabolites (DMs) and differentially expressed genes (DEGs). Specifically, 1-acylglycerol-3-phosphate O-acyltransferase 2 ( AGPAT2 ) was significantly positively correlated with lysophosphatidylcholine (LPC). Considering that LPC is a crucial regulator of meat quality, LPC and AGPAT2 contents were significantly higher at D90 than at D50. We hypothesized that AGPAT2 improved meat quality at D90 by increasing LPC content. Similarly, adenosine monophosphate deaminase 1 ( AMPD1 ) was significantly positively correlated with uric acid (a negative regulator of meat quality), and both AMPD1 and uric acid were significantly higher at D50. Thus, we concluded thatAMPD1 decreased the meat quality of D50 Jiaji ducks by increasing uric acid content. Overall, the meat quality of D90 Jiaji ducks was better than that of D50 ducks. The results of this study provide a foundation for investigating the mechanisms regulating duck meat quality.

Development and application of a cGPS 20K liquid-phase SNP microarray in Jiaji ducks SCIE

Abstract :

In order to provide a low-cost, high efficient, and highly accurate tool for molecular breeding of Jiaji ducks, we constructed a cGPS(Genotyping by Pinpoint Sequencing of captured targets) 20 K liquid-phase microarray using resequencing data from this valuable poultry breed for the first time. The microarray contains 20,327 highquality snp loci, mainly from the 30 Jiaji duck resequencing samples collected in this study, and some loci were supplemented from the 135 duck resequencing data from KUNMING INSTITUTE OF ZOOLOGY.CAS. This microarray showed excellent performance in two production tests. The microarray was used to genotype a population of 236 Jiaji ducks, and the genotyping data were then used for population structure analysis and genome-wide association studies (GWAS) of plumage color phenotypes. According to the population structure analysis, the population of Jiaji ducks could be divided into four subpopulations using genetic distance matrices. Using GWAS analysis, 38 significant SNP loci were identified within a region on chromosome 14 that contained 30 genes. Among them, EDNRB2 and VAMP7 were identified as strong candidate genes for the regulation of plumage color in Jiaji ducks. Two mutations upstream of EDNRB2 were identified as tightly linked to the colorless phenotype. In addition, two KASP markers were designed for the SNP loci associated with EDNRB2 (HIC_SCAFFOLD_14_14984620, HIC_SCAFFOLD_14_15016766). The KASP genotyping results showed strong correlations between different genotypes on the SNP locus HIC_SCAFFOLD_14_15016766 and the plumage phenotype. In conclusion, this independently designed microarray will be useful for large-scale genotyping and can lay the foundation for future screening of mutation loci and functional genes.

Abstract :

本发明公开了白碘酚在制备蛋鸡输卵管炎造模剂中的应用,白碘酚为含碘药物、白芨、甘油和纯水的混合物,一种蛋鸡输卵管炎造模方法,通过输卵管灌注法进行蛋鸡输卵管炎造模,包括以下步骤:步骤一、配制白碘酚;步骤二、将配置好的试剂存储于锥形瓶中,做好密封保存,备用;步骤三、开始造模前,每天进行早晚两次饲喂,直到蛋鸡的产蛋率能够稳定在90%以上,之后开始进行造模。本发明的有益效果为:将自制的造模制剂,通过输精管灌注到蛋鸡输卵管子宫部的方法,成功制造的单机输卵管炎的病理模型;具有操作简单、可重复性好、炎性特征明显等优点,可为蛋鸡输卵管炎的相关研究提供试验手段。
