【目的】建立兔源支气管败血波氏杆菌(Bb)的快速检测方法,用于兔波氏杆菌病的诊断。【方法】以Bb的fimX基因为靶基因,设计特异性引物并构建质粒标准品。在此基础上建立检测兔源Bb的SYBR GreenⅠ荧光定量PCR方法,对该方法的特异性、敏感性和重复性进行评价,并将该方法初步应用于临床样品的检测。【结果】荧光定量PCR方法仅对兔源Bb的检测结果呈阳性,对其他常见兔源病原菌的检测结果均呈阴性;对质粒标准品的最低检测限为13.8拷贝·μL

Synergistic Enhancement of Eimeria maxima Vaccine Efficacy Through EF-1α Antigen and Chicken XCL1 Chemokine Adjuvant Combination SCIE

Abstract :

Coccidiosis is a major parasitic disease that suppresses poultry productivity and causes significant global economic losses. Currently, controlling Eimeria parasites relies primarily on the use of anticoccidial drugs or live vaccines. However, these conventional control strategies face the dual constraints of escalating drug resistance and unsustainable economic expenditures. In this study, the efficacy of a chimeric subunit vaccine comprising Eimeria maxima Elongation Factor-1 alpha (EmEF1 alpha) and chicken chemokine Ligand-1 (ChXCL1) was assessed for protection against experimental Eimeria maxima infection. The synthetic gene fragment ChXCL1-EmEF1 alpha was ligated to the pET28a vector and expressed in vitro. Western blot analysis confirmed the successful expression of the recombinant ChXCL1-EmEF1 alpha protein. Chickens immunized with the ChXCL1-EmEF1 alpha exhibited a significantly stronger IgY response and higher secretion of IL-2 and IL-17 compared to those vaccinated with recombinant ChXCL1 alone or challenged solely with E. maxima. Furthermore, the ChXCL1-EmEF1 alpha group demonstrated enhanced anticoccidial effects, including reduced intestinal lesions, higher body weight gain, and lower oocyst shedding compared to control groups. Following E. maxima challenge, the EmEF1 alpha and ChXCL1-EmEF1 alpha groups demonstrated robust protective efficacy, achieving high ACI values of 182 and 178, respectively. In contrast, the ChXCL1 and UC groups exhibited significantly lower ACI values (150 and 149, respectively), indicating minimal protection. This improvement was also reflected in the immune response, with significantly elevated levels of CD4+ and CD8+ T cells in the ChXCL1-EmEF1 alpha-treated chickens. Moreover, ChXCL1 acts as an effective adjuvant when fused with EmEF1 alpha, enhancing the vaccine's anticoccidial efficacy. These results suggest that the ChXCL1-EmEF1 alpha chimeric immunogen is a promising candidate for developing subunit vaccines against E. maxima infections.

The Effect and Mechanism of Cinnamaldehyde on Testosterone Secretion in Murine Model SCIE

Abstract :

Cinnamon (Cinnamomum cassia (L.) D. Don) is a commonly used drug in traditional Chinese medicine; it is especially useful for improving the reproductive performance in males and females. Cinnamaldehyde is the main component of cinnamon, which has bactericidal and disinfectant properties, promotes gastrointestinal motility and regulates testosterone secretion in the male. The present study aimed to investigate the mechanisms by which cinnamaldehyde regulates testosterone secretion in male animals. For this purpose, four-month-old male mice (n=25) were randomly divided into five groups (n=5 each) and treated with PBS, DMSO and three doses of cinnamaldehyde (25, 50 and 75mg/kg) by gavage for 2 weeks. Histological (IHC), ELISA and molecular analyses (WB, RTqPCR) were used to assess effects of cinnamaldehyde on murine liver, serum testosterone levels, and testicular enzymes associated with testosterone production. The mechanisms by which cinnamaldehyde affects testosterone secretion in testis Leydig cells were explored using ELISA, flow cytometry, WB, fluorescent quantitative PCR, and dual luciferase reporter systems. The results showed that cinnamaldehyde elevated murine body weight, serum testosterone, testicular interstitial cell density, and steroidogenic enzyme expression (StAR, CYP11A1, 3 beta- HSD) without altering hepatic tissue and testicular indices. Cinnamaldehyde treatment at 5x10-5M increased the expression of steroid hormone synthase in Leydig cells, increased the proportion of cells in the S phase, significantly increased the promoter activity of the steroid hormone synthesis nuclear receptor steroidogenic factor 1 (SF-1), and up-regulated the expression of the nuclear receptor SF-1. The present study showed that cinnamaldehyde can upregulate the expression of steroid hormone synthase by increasing the activity of the steroid hormone synthesis nuclear receptor promoter, which in turn increases the synthesis and secretion of testosterone from testicular interstitial cells.

Immunogenicity and protection against infectious bursal disease via a transgenic Eimeria acervulina expressing IBDV VP2-2C3d fusion protein SCIE

Abstract :

Infectious bursal disease (IBD), caused by the infectious bursal disease virus (IBDV), significantly threatens global poultry health by inducing immunosuppression and causing economic losses. To enhance vaccination efficacy, we engineered a transgenic strain of Eimeria acervulina (Ea-2C3d) expressing a fusion protein composed of IBDV VP2 and three tandem C3d segments (3C3d), utilizing C3d's adjuvant properties to boost immune responses. The transgene was generated by integrating codon-optimized VP2 and 3C3d sequences into the E. acervulina genome using restriction enzyme-mediated transfection. PCR, protein, and genome sequencing confirmed the successful integration and expression of VP2 fusion C3d, but only two copies of C3d were successfully expressed, due to a partial deletion of one C3d copy during the transfection process. In vivo studies demonstrated that Ea-2C3d elicited significantly higher anti-VP2 antibody titers than the parental Ea-VP2 strain (P < 0.05), especially following second immunization. Upon challenge with virulent IBDV, chickens immunized with Ea-2C3d displayed reduced bursal lesions (histopathological score <= 1) and maintained bursal integrity (bursal index >0.7), comparable to those receiving a commercial subunit vaccine. Despite reduced reproductive capacity in the transgenic parasites, Ea-2C3d maintained its immunogenicity and safety. These findings highlight that C3d adjuvant enhances VP2-mediated protection in a coccidial vector, presenting a novel dual-protection strategy against IBD and coccidiosis.

海藻酶解产物对大黄鱼生长、消化吸收及免疫指标的影响

试验旨在探究海藻酶解产物对大黄鱼生长、消化吸收及免疫指标的影响。选取315尾健康状况良好、体重(109.30±2.94) g的大黄鱼幼鱼,随机分为3组,每组3个重复,每个重复35尾大黄鱼。A组在基础饲料中添加0.5%的复合海藻酶解产物,B组在基础饲料中添加0.5%的海带酶解产物,对照组(C组)饲喂基础饲料。试验期56 d。结果显示:A组和B组大黄鱼末重、存活率(SR)、平均增重(AWG)、增重率(WGR)显著高于C组(P<0.05)。A组肌肉中粗蛋白含量显著高于C组(P<0.05),水分和灰分含量显著低于C组(P<0.05)。A组和B组大黄鱼肠道绒毛高度(VH)和隐窝深度(CD)均显著大于C组(P<0.05)。C组大黄鱼血清补体C3水平显著高于A组和B组(P<0.05),补体C4水平显著高于B组(P<0.05)。研究表明,在饲料中添加0.5%海藻酶解产物或0.5%海带酶解产物可提升大黄鱼生长性能及SR,改善肌肉营养成分,促进肠道营养物质的吸收,增加抗病力,以0.5%复合海藻酶解产物添加效果较好。

Abstract :

Microsporum canis (M. cains) skin diseases are zoonotic, which cause serious diseases. Therefore, it is crucial to prevent and treat M. cain's skin diseases in companion animals for public health security and animal welfare. This study analyzed the potential anti-inflammatory mechanisms of Cinnamaldehyde (CA) in treating M. cains skin diseases through network pharmacology and molecular docking. CA was formulated into a dissolving microneedle for application. The findings suggest that CA may interact with Interleukin-1(3 (IL-1(3), Cellular tumor antigen p53 (P53), Microtubule-Associated Protein Kinase 14 (P38 alpha), and Tumor necrosis factor (TNF) to target pathways such as TNF signaling, Toll-like receptor signaling, and IL-17 signaling, ultimately reducing skin inflammation associated with M. cains skin diseases. Microneedles administration significantly enhances efficacy compared to traditional delivery methods. In vivo studies demonstrated that the efficacy of CA-HA/PVA DMNs was comparable to that of TER-HA/PVA DMNs. Compared to the use of DMNs alone, the symptom score decline index (SSDI) in the TER-HA/PVA DMNs group (TM group) and CA-HA/PVA DMNs group (CM group) increased by 25.01 % and 21.1 %, respectively, and both were superior to TER treatment alone. CA-HA/PVA DMNs and TER-HA/PVA DMNs effectively alleviate skin erythema, reduce dander production, and decrease skin keratinization at the superficial level. Moreover, at the histopathological level, they are successful in preventing fungal invasion of hair follicles. In summary, this study introduces a novel approach for treating M. cains skin diseases in companion animals.

Protective role of Lactobacillus plantarum in Eimeria tenella infection: Impact on hematological, inflammatory, and apoptotic gene responses SCIE

Coccidiosis, a major parasitic disease in poultry, causes substantial economic losses worldwide. The rise of drug-resistant Eimeria strains has intensified the need for alternative control measures. This study evaluated the probiotic Lactobacillus plantarum (L. plantarum) as a dietary supplement to protect chickens from Eimeria tenella (E. tenella) infection. We examined its effects on growth performance, disease severity, cecal lesions, oocyst scores, immune response, haematological parameters, and the expression of genes related to inflammation and apoptosis. The levels of pro-inflammatory cytokines (IL-6 and IL-8) and apoptosis-related proteins (Bax, Bcl-2, and caspase-3) were measured by mRNA expression. Probiotic-supplemented groups downregulated the mRNA expression levels of the pro-apoptosis protein Bax. They up-regulated the mRNA expression levels of the anti-apoptosis protein Bcl-2 more than those of the control groups. The findings also showed a statistically higher lymphoproliferative (P < 0.05) response in the probiotics @1.5 x 10(8) CFU)-supplemented group in feed compared to the control group. The probiotics administered in the feed groups showed reduced oocysts shedding, fewer intestinal lesions, and improved hematological parameters than the control (E. tenella infected, non-treated) group. The research showed that probiotic supplementation without anticoccidial drugs could protect chickens against the adverse effects of E. tenella.

This experiment investigated the effects of seaweed polysaccharide (SP) and seaweed enzymatic hydrolysate (SEH) on the growth performance, serum biochemical indices, antioxidant capacity, and intestinal function of Muscovy ducks. A total of 240 healthy 1 day female Muscovy ducks (48.85 +/- 0.45 g) were randomly divided into 3 treatment groups, with 4 replicates per group and 20 ducks per replicate. The control (CON) group received a basic diet supplemented with 20 mL/kg of water, the SP group received a basic diet supplemented with 20 mL/kg of SP, and the SEH group received a basic diet supplemented with 20 mL/kg of SEH. The experimental period lasted for 28 d. The results indicate that, compared to the CON group, the average daily feed intake (ADFI) and feed to gain (F/G) of the SP and SEH groups of ducks significantly decreased at 28 d (p < 0.05). In the SP group, serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST), as well as the concentrations of glucose (GLU), triglycerides (TG), total cholesterol (TCHO), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C), were significantly reduced (p < 0.05). In the SEH group, the activities of ALT and AST were also significantly lower (p < 0.05). Additionally, serum total antioxidant capacity (T-AOC) levels and superoxide dismutase (SOD) activity in the SEH group were significantly higher than those in the CON group (p < 0.05), while the malondialdehyde (MDA) content was significantly reduced (p < 0.05). Compared to the CON group, serum levels of immunoglobulin A (IgA), immunoglobulin G (IgG), interleukin-4 (IL-4), and interleukin-10 (IL-10) in the SP group were significantly increased (p < 0.05), whereas the levels of tumor necrosis factor-alpha (TNF-alpha), interleukin-1 beta (IL-1 beta), and interleukin-6 (IL-6) were significantly decreased (p < 0.05). In the SP and SEH groups, the villus height (VH) and the villus height to crypt depth (V/C) of the Muscovy ducks significantly increased (p < 0.05), while the crypt depth (CD) significantly decreased (p < 0.05). A significant increase in the abundance of Barnesiella was observed in the SP and SEH groups (p < 0.05), whereas the abundances of UCG-005 and Romboutsia significantly decreased (p < 0.05). LEfSe analysis indicated that g__Bacillus and g__Veillonella were significantly abundant in the SP group (p < 0.05), while g__Coriobacteriaceae_UCG_002 was significantly abundant in the SEH group (p < 0.05). In summary, the addition of SP and SEH to the feed can promote the healthy growth of ducks by improving intestinal morphology, regulating the structure of intestinal microbiota, enhancing antioxidant capacity and immune function, and optimizing metabolic indicators. This occurs while reducing feed intake and feed-to-weight ratio, and there is a certain specificity in their mechanisms of action.
