Non-Targeted Metabolomic Analysis of Ethanol Extract of Propolis and Its Anti-Inflammatory Effects in LPS-Induced BV2 Microglial Cells via the TLR4 Signaling Pathway SCIE

Abstract :

Propolis contains abundant flavonoid and phenolic compounds, whose composition and concentration vary significantly in different geographical origins, thereby affecting its bioactive properties including anti-inflammatory, antioxidant, and antimicrobial activities. In this study, the flavonoid and phenolic content in the ethanol extract of propolis (EEP) from Henan (HN) and Shandong (SD) provinces was quantitatively analyzed, and the results showed that concentrations of both bioactive components in HN were slightly higher than those in SD. The non-targeted metabolomics technology was further employed to analyze the components of EEP, and a total of 10683 metabolites were detected. In the comparison between the samples of HN and SD, there were a total of 1436 differential metabolites, with 553 decreased and 883 increased in the HN sample. Among them, there were 205 differential metabolites related to flavonoids and phenols, with 108 decreased and 97 increased in the HN sample. However, a greater number of carboxylic acids and derivatives, fatty derivatives and organooxygen metabolites were found at higher relative levels in the HN sample. As a result, the EEP of the HN sample was selected to investigate its inhibitory effect on inflammation in lipopolysaccharide (LPS)-induced BV2 microglia cells. The results showed that LPS promoted the M1 polarization of BV2 microglia. However, treatment with EEP at concentrations of 10 mu g/mL, 5 mu g/mL, and 2.5 mu g/mL could partially restore the cell morphology to its non-activated state. Meanwhile, LPS stimulation increased the protein levels of IL-1 beta, IL-6 and TNF-alpha significantly, as well as the relative gene expression levels of IL-1 beta, IL-6, TNF-alpha, COX-2, iNOS and TLR4. After treatment with the EEP, the expression levels of these three proteins and six genes were significantly decreased. These findings revealed that EEP effectively inhibited the M1 polarization of LPS-induced BV2 cells and decreased the expression of inflammatory factors, indicating its potential as a therapeutic agent for neuroinflammation.

Azoxystrobin and Picoxystrobin Lead to Decreased Fitness of Honey Bee Drones (Apis mellifera ligustica) SCIE

Abstract :

Honey bees (Apis mellifera ligustica) are essential pollinators in both ecosystems and agricultural production. However, their populations are declining due to various factors, including pesticide exposure. Despite their importance, the reproductive castes, particularly drones, remain understudied in terms of pesticide effects. To investigate the effects of azoxystrobin and picoxystrobin on honey bee drones, the drones were exposed to different concentrations of azoxystrobin and picoxystrobin for 14 days; the drone survival, body weight, nutrient content, reproductive organs, and sperm concentration were assessed. Results showed that exposure to both fungicides caused a significant reduction in drone survival rates, with survival rates decreasing progressively as the duration of exposure increased. Compared to the control group, the body weights of drones in all treatment groups were significantly lower on days 7 and 14. Nutrient analysis revealed that low concentrations of azoxystrobin and picoxystrobin increased protein levels, while free fatty acid content decreased significantly in all treatment groups. No significant changes were observed in the total carbohydrate content. Morphological examination of reproductive organs showed that the lengths of the mucus glands and seminal vesicles in drones were significantly shorter in the treatment groups compared to the control group. Furthermore, exposure to azoxystrobin and picoxystrobin resulted in a significant decline in sperm concentration in the drones. These findings indicate that azoxystrobin and picoxystrobin have adverse effects on the health and reproductive capacity of honey bee drones. The present study highlights the need to reassess the risks posed by these fungicides to pollinators, particularly given the critical role of drones in maintaining the genetic diversity and resilience of honey bee colonies. Further research is warranted to elucidate the underlying mechanisms of these effects and explore potential mitigation strategies.

Abstract :

Ganoderma lucidum is a traditional Chinese medicinal fungus, and ganoderma triterpenoids (GTs) are one of the main bioactive compounds. These compounds have various pharmacological functions, including anti-tumor, antioxidant, anti-inflammatory, liver-protective, and immune-regulating effects. However, the manner in which they accumulate, and their biosynthesis mechanisms remain unclear. To screen for the genes that are involved in the biosynthetic pathway of GTs, this study analyzed the differential metabolites and differentially expressed genes (DEGs) among different growth stages of G. lucidum, including the primordia (P), the matured fruiting body (FM), and the post-spore fruiting bodies (FP) using targeted metabolomics and transcriptomics analysis, respectively. The results showed that a total of 699 components were detected, including lignans, terpenoids, amino acids and derivatives, and phenolic acids, among others. Among them, a total of 112 types of triterpenes were detected. Compared with the primordia, there were eight differential metabolites of triterpenoids, with three decreasing and five increasing in the FM stage. A comparison between the FM stage and the FP stage revealed that there were 13 differential metabolites of triterpenoids. A transcriptomics analysis showed that there were 371 DEGs in the P_vs_FM group, including 171 down-regulated genes and 200 up-regulated genes. In the FM_vs_FP group, 2567 DEGs were identified, with 1278 down-regulated genes and 1289 up-regulated genes. Through targeted metabolomics and transcriptome correlation analysis, six TFs and two CYP450s were significantly associated with four triterpenoid components. The results showed that these TFs and CYP450s were positively or negatively correlated with the four triterpenoid components. In addition, interestingly, some flavonoids and phenolic compounds, which have been reported in plants, were also detected in G. lucidum, indicating that it has the potential to be engineered into a strain capable of synthesizing flavonoid compounds. This study provides useful information about key genes involved in GT biosynthesis, but further exploration and in-depth research are needed to better understand the functions of these genes.

Abstract :

[目的]探究灵芝(Ganoderma lucidum)不同生长阶段的成分差异。[方法]使用气相色谱-质谱检测法(gas chromatographymass spectrometry,GC-MS)和超高效液相串联四极杆飞行时间质谱法(ultra performance liquid chromatography-quadrupole time-offlight mass spectrometry,UPLC-QTOF/MS)技术,选用ACQUITY超高效液相色谱仪HSS T3色谱柱在Waters超高效液相色谱(ultra performance liquid chromatography,UPLC)系统上对灵芝原基、产孢前和产孢后的子实体进行成分分析与鉴定。[结果] GC-MS共检测到24种成分,其中醛类8种、酯类4种、烃类1种、酸类1种、醇类1种、酮类2种、胺类2种、吡嗪类1种、咪唑类1种、吡咯类1种、呋喃类1种、酚类1种。UPLC-QTOF/MS共检测90种成分,最主要的成分三萜类共有67种,此外还包括脂肪酸类8种、氨基酸及其衍生物类3种、苷类2种、维生素1种、生物碱类2种、有机酸类2种、糖类2种、萜1种、杂萜1种、甾体1种。共有73种成分在原基中检出,其中三萜类成分54种;72种成分在产孢前子实体中检出,其中三萜类成分57种;52种成分在产孢后子实体中检出,其中三萜类成分有42种。[结论]灵芝不同生长阶段成分存在明显差异,本研究结果为灵芝不同时期的化学成分的分析与鉴定,以及化学物谱图的完善提供了基础数据,为相关药理、药效分析提供了有效参考。

The first dikaryotic genome of Ganoderma cultivar Zizhi S2 (56.76 Mb, 16,681 genes) has been sequenced recently. 98.15% of complete BUSCOs were recovered in this genome assembly and high-confidence annotation rate improved to 91.41%. Collinearity analysis displayed the nuclear genome were 80.2% and 93.84% similar to reference genome of G. sinense at nucleotide and amino acid levels, which presented 8,521 core genes and 880 unique orthologous gene groups. Among that, at least six functional genes (tef1-alpha, beta-tubulin, rpb2, CaM, Mn-SOD and VeA) and a newly discovered fip gene were highly similar 99.27%similar to 100% to those in reference genome. And the mt-LSU, mt-SSU and 13 PCGs in their mitogenome were also highly conserved with 99.27%-99.87% and 99.08%-100% identity, respectively. So that, this cultivar Zizhi S2 is confirmed conspecific with Ganoderma sinense (NCBI: txid1077348). The new fip gene (MN635280.1_336bp) existing a novel mutation which can be reflected on the phylogenetic tree and 3-dimensional model topology structure.

Comparative Mitogenomics Provides Valuable Insights for the Phylogeny and New DNA Barcodes of Ganoderma SCIE

Ganoderma is the most important genus in the family Ganodermataceae; many species have attracted much attention and widely cultivated because of their medicinal values, but so far, not a sequenced mitogenome derived from dikaryon strains has been explicitly recorded. Herein, four novel mitogenomes of commonly cultivated Ganoderma (G. leucocontextum H4, G. lucidum G6, G. sinense MZ96 and G. tsugae SS) were de novo assembled and given detail functional annotations. Collinearity analysis revealed that the four mitogenomes shared 82.93-92.02% similarity with their corresponding reference mitogenomes at the nucleotide level. A total of 15 core protein-coding genes (PCGs), along with rrnL and rrnS (mtLSU and mtSSU) were chosen as potential candidates for constructing their individual phylogenetic trees. These trees were compared with those derived from the concatenated sequences of 15 core PCGs. And finally, we found that the atp9 and nad4L were the most reliable markers for the phylogenetic analysis of Ganoderma and chosen as standard sequences to generate new DNA barcodes. This finding was further verified by comparing it against almost all available Ganoderma mitogenomes in the NCBI, with Trametes versicolor (Polyporaceae) and Rigidoporus microporus (Meripilaceae) as two outgroups. A total of 52 mitogenomes from three families were highly conserved, with identical gene lengths for atp9 (222 bp) and nad4L (267 bp). These genes were capable of distinguish distinctly different various species, which are grouped into separate clades within the phylogenetic trees. The closest related clades (I and II), including at least 30 samples of the three classical taxonomic species (G. lingzhi, G. sichuanense and G. lucidum), differed in only one SNP. The single base mutation rate increased with the evolutionary divergence of the phylogenetic clades, from two to three SNPs in earlier clades (e.g., clade IV containing G. leucocontextum) to five to six SNPs in later clades (e.g., clade X containing G. sinense). Despite these variations between species, the atp9 and nad4L genes of Ganoderma mitogenomes consistently encoded the same ATP synthase F0 subunit c (73 aa) and NADH dehydrogenase subunit 4L (88 aa). These two genes have been identified as reliable markers of new DNA barcodes, offering valuable insights and contributing significantly to understanding the evolutionary relationships and phylogeny of the Ganoderma genus and even the Ganodermataceae family.

紫芝基因组rDNA序列特征及ITS2二级结构比较

紫芝栽培品种‘紫芝S2’(武芝2号)的ITS序列与NCBI数据库中5个紫芝菌株/分离株相似度高达99.79%–100%,在系统进化树上相聚成一类。本研究预测‘紫芝S2’基因组与参考基因组中的rRNA基因簇,分析rDNA结构及各构件序列间的多态性。从高质量‘紫芝S2’基因组中挖掘得到完整rDNA,序列全长40.377kb,由4组串联重复的(18S、5.8S、28S、5S)rRNA基因簇组成,并含有完整的基因内间隔区(ITS1、ITS2)和基因间间隔区(IGS1、IGS2)。在紫芝S2的rDNA中,高度保守的28S rRNA基因间出现3个SNP和2个插入(1 bp,10 bp)位点;虽然第4条ITS2中有1个SNP位点,但紫芝S2的4条ITS2在二级结构上的分子形态高度一致,与ITS2数据库中其他紫芝菌株仅存在螺旋区间夹角的微小差异。由‘紫芝S2’基因组rDNA的ITS2生成的DNA条形码与二维码,可以作为该栽培品种鉴定与同源物种其他菌株鉴别的分子标记。

本实用新型公开的属于菌种采集技术领域,具体为用于采集灵芝孢子的装置,包括罩筒,顶部与底部外侧均开设有螺纹槽,上旋盖螺纹连接于罩筒顶端,下旋盖螺纹连接于罩筒底端,下旋盖内卡接有培养皿,隔层部件置于罩筒内,包括与连接于罩筒内侧的卡环,及连接于卡环内的隔层,将罩筒内灭菌,然后将去柄消毒的灵芝背面置入隔层上,旋紧上旋盖,灵芝孢子弹射进入下旋盖内置的培养皿内,解决了野外实体组织块分离菌种时易污染的问题,并且,由于隔层卡接在卡环内,方便对隔层进行更换,改变缺口的大小,同时,卡环通过卡座与滑槽卡装配合,连接在罩筒内,方便改变卡环自罩筒内的距离,进而改变隔层距离培养皿之间的距离,便于对弹射的菌种进行收集。
