复合脂肪酸对骡鸭原代肝细胞脂质沉积及AMPK信号通路的影响

Abstract :

试验旨在探究复合脂肪酸对骡鸭原代肝细胞超微结构、脂质沉积、氧化应激及AMPK信号通路相关基因表达的影响。试验使用不同浓度的复合酸(油酸+棕榈酸)进行处理:500μmol/L+250μmol/L、1 000μmol/L+500μmol/L、1 500μmol/L+750μmol/L和2 000μmol/L+1 000μmol/L,并持续处理36 h构建原代骡鸭肝细胞脂质沉积模型。结果显示:(1)分离的骡鸭原代肝细胞有丰富糖原颗粒和脂滴的存在。(2)与对照组相比,油酸+棕榈酸浓度为1 500μmol/L+750μmol/L时,培养骡鸭原代肝细胞36 h,细胞活力没有显著差异(P>0.05),脂滴数量显著增加(P<0.05)。(3)透射电镜结果显示,复合脂肪酸处理的骡鸭原代肝细胞和线粒体均出现明显水肿,脂滴出现聚合,并且膜和嵴的结构受损。此外,与对照组相比,肝细胞脂滴含量、谷丙转氨酶和谷草转氨酶活性、丙二醛和活性氧水平显著上升(P<0.05)。(4)与对照组相比,复合脂肪酸处理的骡鸭原代肝细胞中AMP活化蛋白激酶(AMPK)、脂蛋白脂肪酶(LPL)和肉碱棕榈转移酶-1A(CPT-1A) mRNA表达量以及AMPK蛋白表达量显著降低(P<0.05),而白细胞分化抗原36(CD36)、乙酰辅酶A羧化酶1(ACC1) mRNA及白细胞分化抗原36(CD36)、胆固醇调节元件结合蛋白1(SREBP1)蛋白表达量显著提高(P<0.05)。因此,油酸+棕榈酸浓度为1 500μmol/L+750μmol/L处理骡鸭原代肝细胞36 h能通过调控AMPK信号通路,有效增加肝细胞脂质沉积,但同时易诱导肝细胞氧化应激。

Abstract :

The aim of this study was to examine the effects of NH3 variations across different positions within a cage-reared duck house on the egg production performance and fecal microbiome in Muscovy ducks. Totals of 3,168 female Muscovy ducks (180 +/- 2 d) were randomly assigned to 1,056 cages. From d 293 to 300, the egg production rate and levels of NH3, H2S, CO2, temperature, humidity, light intensity, and dust particles were recorded. Two spatial distribution-based experimental classification methods were applied, one grouping ducks into Front, Middle, and End categories, and the other into First, Second, and Third layers. On d 300, serum and feces samples were collected for further analysis. The result showed that both the egg production rate and serum PROG level in the Front were higher than End (P < 0.05). However, no significant differences were found among the First, Second, and Third groups. Among the environmental factors, ammonia (NH3) is a major harmful gas in cage-reared duck houses, with observed concentrations showing a gradient from lower levels in the Front group to higher levels in the Middle group (P < 0.05), and even higher levels in the End group (P < 0.05). Gene sequencing of the 16S rRNA gene revealed a higher relative abundance of Firmicutes in both the Front and Middle groups compared to the End group (P < 0.05). Specifically, within the Firmicutes phylum, the relative abundance of Lactobacillus and Romboutsia was notably higher in the Front group compared to both the Middle and End groups (P < 0.05). What's more, the abundance of Lactobacillus had a negative correlation with NH3 concentration and positive correlation with egg production rate. In conclusion, NH3 concentrations showed variations across different areas of the cage-reared duck house, with higher levels detected at the rear. The elevated NH3 level was identified as the main factor negatively impacting egg production in Muscovy ducks and contributing to a decline in the abundance of Firmicutes, specifically Lactobacillus.

Involvement of the AMPK Pathways in Muscle Development Disparities across Genders in Muscovy Ducks SCIE

Abstract :

The differences in muscle development potential between male and female ducks lead to variations in body weight, significantly affecting the growth of the Muscovy duck meat industry. The aim of this study is to explore the regulatory mechanisms for the muscle development differences between genders. Muscovy ducks of both sexes were selected for measurements of body weight, growth traits, hormone levels, and muscle gene expression. The results show that male ducks compared to females had greater weight and growth traits (p < 0.05). Compared to male ducks, the level of serum testosterone in female ducks was decreased, and the estradiol levels were increased (p < 0.05). The RNA-seq analysis identified 102 upregulated and 49 downregulated differentially expressed genes. KEGG analysis revealed that among the top 10 differentially enriched pathways, the AMPK signaling pathway is closely related to muscle growth and development. Additionally, the mRNA and protein levels of CD36, CPT1A, LPL, and SREBP1 were increased and the P-AMPK protein level decreased in the female ducks compared to the male ducks (p < 0.05). In conclusion, muscle development potential difference between male and female ducks is regulated by sex hormones. This process is likely mediated through the activation of the AMPK pathway.

Variations in gut microbiota composition and reproductive hormone levels between laying and broody Muscovy ducks SCIE

Abstract :

High broodiness in Muscovy ducks impedes animal husbandry growth. The interaction between endocrine hormones and gut microbiota has been proven to play a crucial role in reproductive performance, and whether it can regulate the broody behavior of Muscovy ducks requires further research. Nine laying ducks (Laying group) and nine broody ducks (Broodiness group) were selected. Corresponding serum, ileum, and cecum chyme were collected for further research. The results showed that, compared to the laying group, the serum concentration of prolactin decreased, while the levels of Mullerian inhibiting substance, follicle-stimulating hormone, and follistatin increased in the broodiness group (P < 0.05). 16S rDNA sequencing showed that, the broodiness group exhibited lower abundance levels of Rothia, Streptococcus, and Lactobacillus, whereas the abundance of Turicibacter, Aliicoccus, and Facklamia was higher in the ileum compared to the laying group (P < 0.05). In the cecum, the broodiness group exhibits a significant reduction in the abundance of Butyricicoccus and unclassified_f_Rikenellaceae, while the abundance of Christensenellaceae_R-7_group, Ruminococcus_torques_group, Parabacteroides, norank_f_Oscillospiraceae, Cloacibacillus, Sellimonas, Shuttleworthia, norank_f_UCG-010, unclassified_f_Lachnospiraceae, Oscillospira, Synergistes, Family_XIII_AD3011_group and Eubacterium_nodatum_group is higher compared to the laying group. A Spearman correlation analysis reveals that both in the ileum and cecum, serum hormones exhibit significant correlations with the top 20 abundant intestinal microbial genera. Among these, serum follistatin has most entries of significant correlations with the detected microbial genera (P < 0.05). In conclusion, the broody behavior of Muscovy ducks can be modulated by the interaction between hormones and gut microbiota. Notably, the relationship between Follistatin and the composition of gut microbiota, specifically Firmicutes, is the most prominent.

Abstract :

This study investigates the underlying mechanism through which dietary supplementation of pyrroloquinoline quinone disodium (PQQ) alleviates intestinal inflammation and cell apoptosis in piglets challenged with lipopolysaccharide (LPS). Seventy-two barrows were divided into three groups: control (CTRL), LPS challenged (LPS), and LPS challenged with PQQ supplementation (PQQ + LPS). On d 7, 11, and 14, piglets received intraperitoneal injections of LPS or 0.9% of NaCl (80 mu g/kg). After a 4 h interval following the final LPS injection on d 14, blood samples were obtained, and all piglets were euthanized for harvesting jejunal samples. The results showed that dietary supplementation of PQQ improved the damage of intestinal morphology, increased the down-regulated tight junction proteins, and reduced the increase of serum diamine oxidase activity, the intestinal fatty acid binding protein, and TNF-alpha levels in piglets challenged with LPS (p < 0.05). The proteomics analysis revealed a total of 141 differentially expressed proteins (DEPs), consisting of 64 up-regulated DEPs and 77 down-regulated DEPs in the PQQ + LPS group compared to the LPS group. The KEGG pathway analysis indicated enrichment of the tight junction pathway and the apoptosis pathway (p < 0.05). Compared to the LPS group, the piglets in the PQQ + LPS group had increased levels of Bcl-2 protein, reduced positive apoptosis signals, and a decrease in the abundance of MKK 3/6 and p-p38 proteins (p < 0.05). In conclusion, dietary supplementation of PQQ could alleviate jejunal inflammatory damage and cell apoptosis in piglets challenged with LPS through the MKK3/6-p38 signaling pathway.

Abstract :

Muscovy ducks' high broodiness hinders industry growth. Studying broodiness regulation contributes to the theoretical foundation for enhancing reproductive performance in Muscovy ducks. Experiment 1, a total of 18 Muscovy ducks were divided into 2 groups: Laying group (LO) and Broody group (BO). To collect ovaries for morphological and transcriptome analysis. Experiment 2, Primary Muscovy ducks granulosa cells (GC) were isolated and treated with or without GDF9 at appropriate concentrations as indicated. Experiment 3, GC were treated with or without GDF9 in the presence or absence of a receptor inhibitor. The cell viability, cell apoptosis rate and levels of TGF-beta pathway were determined. In vivo, there was a gradual disappearance of follicles in the ovaries and accompanied by follicle atrophy and a concentration of cytoplasm in BO group. The transcriptome expression profile revealed a total of 1,185 up-regulated differentially expressed transcripts (DEs) and 1,258 down-regulated DEs in the BO group compared to the LO group. The up-regulated differentially expressed GDF9 is involved in regulating the TGF-beta pathway, which is among the top 10 pathways identified through the KEGG pathway analysis (P < 0.05). Additionally, the fluorescence intensity of apoptosis is primarily observed in the granulosa layers of the ovary. Compared to the LO group, the mRNA level of TGF-beta pathway and the protein of GDF9 and p-Smad2/3 were increased in ovary of the BO group (P < 0.05). In vitro, GDF9 supplementation demonstrated does-related promotion of GC (P < 0.01). Compared to CTRL group, 12 ng/mL GDF9 supplementation to GC increased the rate of cell apoptosis, the mRNA and protein expression of TGF-beta pathway and the apoptosis-related genes. Pretreatment of GC with GDF9-receptor inhibitor largely abrogated the negative function of GDF9 treatment (P < 0.05). In summary, granulosa cell apoptosis leading to follicle atresia in broodiness of Muscovy ducks is associated with GDF9 activation of the TGF-beta pathway. This discovery lays a solid foundation for understanding duck follicular development and enhancing egg production in Muscovy ducks.

Hindered growth often occurs because of psychological and environmental stress during the weaning period of piglets. This study aimed to compare the effects of growth performance, diarrhea indices, digestibility of nutrients, antioxidant capacity, neurotransmitters levels and metabolism of weaned pigs fed diets supplemented with pyrroloquinoline quinone (PQQ) and zinc oxide (ZnO). Pigs weaned at d 28 (n = 108) were fed with three different diets including: the basal diet (CTRL group), the basal diet supplemented with 3.0 mg/kg PQQ (PQQ group) and the basal diet containing 1,600 mg/kg ZnO (ZNO group). During the first 14 d, weaned pigs fed the diet supplemented with PQQ and ZnO decreased feed to gain ratio and diarrhea rate (P < 0.01). Compared with the CTRL group, average daily gain was increased in weaned pigs in the PQQ group from d 15 to 28 (P = 0.03). Compared with the CTRL group, pigs fed PQQ and ZnO supplemented diets showed improved apparent total tract digestibility (ATTD) of nutrients (P < 0.05). During the overall experimental period, the concentration of malondialdehyde was decreased in plasma of pigs in the PQQ and ZNO groups compared with the CTRL group (P < 0.05). At d 28, the concentration of vasoactive intestinal peptide (VIP) and calcitonin gene-related peptide (CGRP) was lower in plasma of weaned pigs in the PQQ and ZNO groups compared with the CTRL group (P < 0.05). There was no difference between the PQQ and ZNO group in growth performance, ATTD of nutrition, antioxidant capacity and neurotransmitters levels. PQQ increased 3-methoxy-4hydroxymandelate (P < 0.05) compared with the CTRL group. According to metabolomic analysis, erucamide, formononetin and 3-methyl-L-histidine were up-regulated in the PQQ group (P < 0.05). Compared with the CTRL group, aloesin and dibutyl adipate were down-regulated in the PQQ group (P < 0.05). In conclusion, similar to ZnO, PQQ improves growth performance, digestibility of nutrients, antioxidant capacity, neuromodulation and metabolism of weaned pigs. This study is aimed to evaluate the effect and underling mechanism of dietary supplementation with pyrroloquinoline quinone (PQQ) disodium on improving inflammatory liver injury in piglets challenged with lipopolysaccharide (LPS). A total of seventy-two crossbred barrows were allotted into four groups as follows: the CTRL group (basal diet + saline injection); the PQQ group (3 mg/kg PQQ diet + saline injection); the CTRL + LPS group (basal diet + LPS injection) and the PQQ + LPS group (3 mg/kg PQQ diet + LPS injection). On days 7, 11 and 14, piglets were challenged with LPS or saline. Blood was sampled at 4 h after the last LPS injection (day 14), and then the piglets were slaughtered and liver tissue was harvested. The results showed that the hepatic morphology was improved in the PQQ + LPS group compared with the CTRL + LPS group. PQQ supplementation decreased the level of serum inflammatory factors, aspartate aminotransferase and alanine transaminase, and increased the HDL-cholesterol concentration in piglets challenged with LPS; piglets in the PQQ + LPS group had lower liver mRNA level of inflammatory factors and protein level of alpha-smooth muscle actin than in the CTRL + LPS group. Besides, mRNA expression of STAT3/TGF-beta 1 pathway and protein level of p-STAT3(Tyr 705) were decreased, and mRNA level of PPAR alpha and protein expression of p-AMPK in liver were increased in the PQQ + LPS group compared with the CTRL + LPS group (P < 0 center dot 05). In conclusion, dietary supplementation with PQQ alleviated inflammatory liver injury might partly via inhibition of the STAT3/TGF-beta 1 pathway in piglets challenged with LPS.

Abstract :

Hindered growth often occurs because of psychological and environmental stress during the weaning period of piglets.This study aimed to compare the effects of growth performance,diarrhea indices,di-gestibility of nutrients,antioxidant capacity,neurotransmitters levels and metabolism of weaned pigs fed diets supplemented with pyrroloquinoline quinone(PQQ)and zinc oxide(ZnO).Pigs weaned at d 28(n=108)were fed with three different diets including:the basal diet(CTRL group),the basal diet supplemented with 3.0 mg/kg PQQ(PQQ group)and the basal diet containing 1,600 mg/kg ZnO(ZNO group).During the first 14 d,weaned pigs fed the diet supplemented with PQQ and ZnO decreased feed to gain ratio and diarrhea rate(P<0.01).Compared with the CTRL group,average daily gain was increased in weaned pigs in the PQQ group from d 15 to 28(P=0.03).Compared with the CTRL group,pigs fed PQQ and ZnO supplemented diets showed improved apparent total tract digestibility(ATTD)of nutrients(P<0.05).During the overall experimental period,the concentration of malondialdehyde was decreased in plasma of pigs in the PQQ and ZNO groups compared with the CTRL group(P<0.05).At d 28,the concentration of vasoactive intestinal peptide(VIP)and calcitonin gene-related peptide(CGRP)was lower in plasma of weaned pigs in the PQQ and ZNO groups compared with the CTRL group(P<0.05).There was no difference between the PQQ and ZNO group in growth performance,ATTD of nutrition,antioxidant capacity and neurotransmitters levels.PQQ increased 3-methoxy-4-hydroxymandelate(P<0.05)compared with the CTRL group.According to metabolomic analysis,eru-camide,formononetin and 3-methyl-L-histidine were up-regulated in the PQQ group(P<0.05).Compared with the CTRL group,aloesin and dibutyl adipate were down-regulated in the PQQ group(P<0.05).In conclusion,similar to ZnO,PQQ improves growth performance,digestibility of nutrients,antioxidant capacity,neuromodulation and metabolism of weaned pigs.Thus,like ZnO,PQQ can be effectively applied in weaned pigs.

白羽番鸭LIF基因克隆及其组织表达差异分析

【目的】探究白血病抑制因子(leukemia inhibitory factor, LIF)基因在白羽番鸭中的生物学功能,阐明其在下丘脑、垂体、卵巢、输卵管膨大部、子宫组织中的表达模式。【方法】以白羽番鸭卵巢组织为研究对象,采用cDNA末端快速扩增(rapid-amplification of cDNA ends, RACE)技术克隆白羽番鸭LIF基因序列,并进行相似性比对和系统进化树构建;通过生物信息学方法分析LIF蛋白理化性质及结构。利用实时荧光定量PCR技术检测LIF基因在白羽番鸭下丘脑、垂体、卵巢、输卵管膨大部、子宫5个组织中的相对表达量。【结果】克隆得到白羽番鸭LIF基因序列长1 820 bp,其中CDS区长636 bp,编码211个氨基酸。核苷酸序列相似性比对结果表明,白羽番鸭LIF基因序列与凤头潜鸭、绿头鸭、棕硬尾鸭、黑天鹅、金丝雀、鹌鹑、鸡、山雀的相似性分别为99.36%、99.21%、99.21%、98.11%、92.03%、90.68%、90.58%和87.60%。系统进化树分析表明,白羽番鸭与凤头潜鸭的遗传距离最近,与鸡的遗传距离最远。白羽番鸭LIF蛋白是碱性亲水的稳定蛋白,有16个磷酸化位点、7个N-糖基化位点及1个信号肽,主要定位于细胞外和溶酶体;LIF蛋白质二级结构主要由α-螺旋和无规则卷曲构成,蛋白质三级结构预测结果与二级结构一致。白羽番鸭LIF蛋白可能与IL6ST、CNTFR、IL6、OSMR、LIFR、IL6R、STAT3、CNTF、IFNGR1、IFNGR2蛋白具有相互作用。组织表达图谱显示,LIF基因在白羽番鸭5种组织中均有表达,其中就巢期白羽番鸭LIF基因表达量在输卵管膨大部、卵巢组织显著高于产蛋期(P<0.05);两个时期LIF基因在输卵管膨大部的表达量均最高,且显著高于其他组织(P<0.05)。【结论】试验成功克隆了白羽番鸭LIF基因,其在输卵管膨大部的表达量显著高于其他组织,且就巢期表达量高于产蛋期。结果为探究白羽番鸭卵巢发育的调控机制提供理论依据。
