Abstract :
Streptococcus suis (S. suis) is considered as one of the most crucial bacterial pathogens that leads to serious economic losses to the swine industry. Different S. suis serotypes exhibit diverse characteristics in population structure and pathogenicity. Epidemiology data underscore the importance of S. suis serotype 3 (SS3). However, except for a few epidemiological information, limited study information is available on this serotype. Herein, a pathogenic SS3 (the S. suis strain YA) was isolated from infected piglets in clinical practice, and then whole genome sequencing and analysis, hemolytic activity, antimicrobial susceptibility, pathogenicity to mice and piglets were conducted. The results of the whole genome sequencing of the S. suis strain YA showed that the complete genome was 2,167,682 bp in length with a G + C content of 41.2% and exhibited a unique sequence type (ST1801). The result of phylogenetic tree showed that it was most closely related to strain DNC15 and 6407 (ST54) from Denmark. The tet(W) and erm(B) resistant genes were identified in the S. suis strain YA by inserting into rum locus, in accordance with the result of resistance to tetracyclines and macrolide-lincosamide-streptogramin antibiotics. Twenty-seven key virulence factors were detected in the S. suis strain YA, including sly, ef and mrp, which contribute to pathogenicity in mice and piglets, causing bleeding and congestion in multiple tissue organs especially in the brains. And the LD50 value for mice was 1.54 x 107 CFU. Therefore, our research emphasizes the importance of understanding SS3, and provides valuable information for the scientific prevention and control of S. suis.

【目的】兔出血症2型(rabbit hemorrhagic disease type 2, RHdV-2)于2020年在我国被发现,并呈地方性流行。本研究旨在建立一种适用于生产一线的快速、简便且无需昂贵仪器的检测方法。【方法】采用生物信息学软件(Vector NTI alignment)分析兔出血症1型(rabbit hemorrhagic disease type 1, RHdV-1)和兔出血症2型RHdV-2基因组,筛选出RHdV-2特异且保守的VP60基因片段。基于此片段,设计重组酶介导等温核酸扩增技术-侧流层析试纸条检测(recombinase aided amplification-lateral flow dipstick, RAA-LFD)的探针与特异性引物。然后在RAA扩增产物的基础上,设计并筛选最适CRISPR衍生RNA(CRISPR-derived RNA, crRNA),并对簇状规律间隔性成簇短回文重复序列相关蛋白13a(clustered regularly interspaced short palindromic repeats associated protein 13a, CRISPR-Cas13a)的反应体系进行优化。筛选针对RHdV-2的RAA-CRISPR/Cas13a (cpf1)-LFD诊断方法的最适反应时间,并对该方法的敏感性、特异性进行评价。最后使用该方法对临床31份疑似样品进行检测。【结果】同源性分析结果表明,RHdV-2的VP60基因上存在特异且保守的序列,合成VP60特异性引物进行RAA扩增。以RAA产物作为反应模板,发现crRNA1特异性最好。反应时间结果表明,在RAA基础反应后仅再需10 min即可在试纸条上出现明显阳性结果。敏感性试验结果显示,以重组质粒为模板,最低检出质粒拷贝数为6.7×10~1 copies·μL

Abstract :
Porcine epidemic diarrhea (PED) virus (PEDV) is a highly contagious intestinal infection that primarily affects suckling pigs. The interaction about the innate immune evasion of PEDV in intestinal epithelium and microRNA (miRNA) remains unclear. A strain of PEDV belonging to the G2a genotype, designated FJND 2022, was successfully isolated and confirmed. Then, the miRNA profile in exosomes-derived from intestinal porcine epithelial cell line (IPEC) infected with PEDV FJND 2022 for 48 h was evaluated. In exosomes from PEDV-infected IPECs, 34 miRNAs showed differential expression relative to blank cells. A total of 7762 target genes of those differentially expressed miRNAs were forecast, and the miR-193a-5P and its target mRNA interleukin (IL)22 and porcine beta-defensin 1 (pBD1) attracted our interest. After infection with PEDV for 48 h, the mRNA levels and protein levels of IL22 and pBD1 were both notably downregulated, while the mRNA level of miR-193a-5P was significantly decreased. When IPECs were pretreated with the mimic of miR-193a-5P and then infected with PEDV, the mRNA levels of IL22 and pBD1 were significantly increased while the viral load of PEDV was significantly reduced. However, siRNA-mediated knockdown of IL22 abrogated the capacity of miR-193a-5p mimic pretreatment to restore pBD1 expression. Furthermore, the inhibitor of miR-193a-5P was pretreated with IPECs infected with PEDV, resulting in a notable downregulation of IL22 and pBD1 expression, and a significant upregulation of the virus load of PEDV. Finally, we also found that the expression levels of IL22, pBD1, and miR-193a-5P were notably reduced in the small intestinal epithelium of suckling piglets infected with PEDV for 48 h. Therefore, in this study we reveal that PEDV downregulates the miR-193a-5P expression in the intestinal epithelium to evade the antivirus of IL22/pBD1, which provides new insights into PEDV molecular pathogenesis and immune evasion mechanisms.

【目的】旨在研究口服太子参皂苷(RP)对鸡免疫功能和炎症反应的影响,为使用RP作为免疫增强剂提供科学依据,对家禽养殖业的疾病预防和健康维护具有实际应用价值。【方法】试验以鸡毒支原体(MG)抗原、抗体均为阴性的30日龄健康河田鸡为研究对象,根据初始体质量平均分为5组,空白组不给药及疫苗,疫苗对照组口服灭菌生理盐水7 d后免疫MG弱毒疫苗,100,20,5 mg/kg RP试验组分别连续口服RP 7 d后免疫MG弱毒疫苗,一免后28 d进行二免,试验期49 d。采用间接ELISA法检测血清中MG特异性抗体消长规律及抗体阳性率和细胞因子(IFN-γ、IL-4、IL-5、IL-1α、CCL4、IL-17A、IL-6)含量。【结果】(1)RP组与疫苗对照组相比,口服RP能有效提高一免及二免后7 d的MG特异性抗体滴度及抗体阳性率(P<0.05),其中以RP高剂量(100 mg/kg)效果最佳。(2)细胞因子表达水平中,与疫苗对照组相比,RP高剂量(100 mg/kg)和中剂量(20 mg/kg)组血清中免疫因子(IFN-γ、IL-4、IL-5)的含量在一免后3 d呈现显著上升(P<0.05),一免后7 d无明显差异;(3)与疫苗对照组相比,RP组炎症细胞因子(IL-1α、CCL4、IL-17A、IL-6)血清中的含量显著降低(P<0.05),高剂量组(100 mg/kg)下降最为显著。【结论】首免前口服太子参皂苷(RP)能够提高河田鸡的免疫功能,并降低由MG弱毒疫苗引起的炎症反应,其中100 mg/kg RP效果最佳。

为探究番鸭呼肠孤病毒(Muscovy duck reovirus,MDRV)活疫苗对番鸭肝脏源外泌体中microRNA(miRNA)的影响,选择40羽1日龄雏番鸭,随机分为试验组和对照组,每组各20羽。试验组肌肉注射MDRV活疫苗1羽份,对照组肌肉注射等量的灭菌生理盐水。免疫48 h后,采集肝脏制备肝原代细胞培养2 d,取上清用试剂盒法提取外泌体送生物公司进行miRNA测序。接着进行miRNA注释,差异表达miRNA分析,靶基因预测,并对靶基因涉及的生物功能进行GO和KEGG富集分析。结果表明:MDRV活疫苗免疫组与对照组相比,共筛选到显著差异miRNA 39个,预测到369个靶基因,这些靶基因的GO功能注释主要集中在单有机体过程、刺激应答、多细胞有机体过程、定位、细胞器、膜、胞外区、绑定分子功能等;KEGG通路富集分析集中在肝脏中氨基糖和核苷酸糖的代谢、糖酵解/糖异生、溶酶体、磷脂酰肌醇信号传导系统、磷酸肌醇代谢、细胞凋亡、细胞自噬作用等。结果表明,MDRV活疫苗免疫雏番鸭会影响肝细胞外泌体的miRNA表达水平,这些miRNA参与肝脏代谢等多种功能。研究结果可为MDRV感染的科学防控提供理论基础。

In this study, we investigated how Radix pseudostellariae polysaccharide (RPP) enhances the immune response of the inactivated porcine reproductive and respiratory syndrome virus (PRRSV) vaccine through interactions with the microbiome and metabolome. We pretreated sows with 10 mg/kg body weight of RPP via drinking water for 7 days prior to intramuscular injection of the PRRSV vaccine. This significantly increased the concentrations of PRRSV GP5 protein antibody, interleukin (IL)-2, IL-4, IL-10, and interferon (IFN)-gamma. Oral administration of RPP also significantly improved the abundance of beneficial bacteria in the stool, such as Parabacteroides distasonis, Prevotella_copri, Eubacterium_sp., and Clostridium_sp._CAG:226, and decreased the levels of potentially pathogenic bacteria, such as Paraeggerthella and [Clostridium] innocuum, compared to the vaccine alone. These bacterial changes were confirmed using quantitative real-time polymerase chain reaction (Q-PCR). Moreover, RPP treatment significantly increased the blood concentrations of L-theanine, taurodeoxycholic acid (TDCA), and N-arachidonoyl proline, and decreased the levels of L-glutamine, oclacitinib, lipoxin C4, and leukotriene C5 in sows after immunization (p< 0.05). The concentrations of various blood metabolites were validated using sandwich enzyme-linked immunosorbent assay (ELISA), confirming the accuracy of the metabolomics data. Intriguingly, the integration of microbiome and metabolome analyses highlighted the significance of Prevotella_copri and TDCA. We consequently developed a mouse immunity model using GP5 protein and discovered that oral administration of RPP significantly enhanced the levels of GP5 protein antibodies, IL-2, IL-4, IL-10, and IFN-gamma in mouse serum. It also increased the number of CD3+ and CD3+CD4+ cells in the spleen. Additionally, Prevotella_copri was administered into the large intestine via the anus for 7 days prior to the intramuscular injection of the PRRSV GP5 protein. The results demonstrated a significant increase in TDCA and GP5 antibody concentration in the mouse serum, indicating that RPP modulates Prevotella_copri to elevate its metabolite TDCA, thereby enhancing the GP5 antibody level. In conclusion, oral administration of 10 mg/kg RPP optimizes gut flora diversity and blood metabolites, particularly Prevotella_copri and TDCA, thereby improving the immune response to the inactivated PRRSV vaccine.

This study aimed to prepare Gardenia fructus carbon dots (GF-CDs) and examine their efficacy in mitigating oxidative stress and apoptosis in intestinal porcine epithelial cells from the jejunum (IPEC-J2 cells) induced by lipopolysaccharide (LPS). The GF-CDs were synthesized using a one-step hydrothermal method. The oxidative damage model of IPEC-J2 cells was induced through LPS treatment. The potential mechanism by which GF-CDs affect cellular oxidative damage was examined through the perspectives of apoptosis, reactive oxygen species level, antioxidant-related enzyme index, mRNA transcription of antioxidant-related genes, and the expression of antioxidant proteins. The results revealed that GF-CDs, characterized by particle sizes<7 nm, abundant functional groups, and good water solubility, were synthesized using a one-step hydrothermal method. The carbon spots of Gardenia fructus at concentrations of 50, 100, and 200 mu g/mL exhibited protective effects, as evidenced by their ability to enhance viability (P<0.01) and restore cellular morphology after oxidative damage. The GF-CDs decreased oxidative damage and reduced the apoptosis rate of cells by upregulating AKT1 expression and downregulating the expression of Caspase 3, STAT3, TNF-alpha, and JNK. These results indicate that GF-CDs have the characteristic physicochemical properties of CDs, exhibit biological activities related to antioxidation and cellular damage mitigation, and may serve as a potential healthcare product in swine raising.

A comparative study was performed to investigate the differences in plasma pharmacokinetics (PKs) and tissue residues of trimethoprim (TMP) between silky fowls and 817 broilers. The 2 breeds of chickens received compound sulfadiazine suspension by gavage at 20 mg/kg (measured as TMP). Blood and tis-sue samples were collected at predetermined time points. The concentrations of TMP in plasma and tissue samples were determined by a validated high-performance liquid chromatography (HPLC) method. The plasma concentration-time data were subjected to noncompartment analysis by WinNonlin program (Pharsight Co., Mountain View, CA). The mean plasma concentrations of TMP in silky fowls were significantly lower than those in 817 broilers at all time-points. Significant differences were also observed between silky fowls and 817 broilers in maximum concentration (C-max), area under the curve from time 0 to 24 h (AUC(0 -> 24 h)), apparent volume of distribution (V-d), and total body clearance (Cl-B). Silky fowls had significantly higher muscle TMP concentrations and longer tissue residual time than 817 broilers. The tissue concentration of TMP followed the order of leg muscle > breast muscle > liver, which was obviously different from that of 817 broilers. The half-lives of TMP in the leg muscle, breast muscle, and liver of silky fowls were 31.42, 10.78, and 0.38 d, respectively. The current withdrawal time (WDT) was not sufficient to prevent violative residues of TMP in the edible tissues of silky fowls, and a WDT much longer than 8 d might be required.
