Abstract :
Context The occurrence of apoptosis due to transient oxidative damage to spermatozoa presents a significant challenge in semen preservation. At present, the addition of protective agents is the primary method for mitigating this damage and involves the addition of protective agents.Aims This study aims to elucidate the mechanism of action of cryopreservation and to identify high-quality combinatorial cryopreservation dilutions.Methods In this study, two protective agents, bovine serum albumin (BSA) and sodium alginate (SA), were combined to evaluate the effectiveness of these novel additives in protecting porcine sperm from damage during cryopreservation. The mechanism of action of the SA and BSA combination was further elucidated at the molecular level, with key proteins being identified through proteomic analysis.Key results The findings indicated that sperm treated with 2 mg/mL SA and 5 mg/mL BSA exhibited optimal motility parameters, superior functional integrity and the most effective ability to alleviate oxidative stress. Combined with proteomic data, the results suggested that these additives regulate improvements in sperm quality by mediating the activity of the PI3K-AKT pathway.Conclusions This study found that the combination of SA and BSA provides an effective protective effect for frozen sperm preservation.Implications The findings offer theoretical and technical support for the use of composite additive to protect sperm from damage, which is crucial for enhancing the quality of pig semen and improving reproductive outcomes.

Abstract :
The ACSL5 (acyl-CoA synthetase 5) gene plays a crucial role in the biosynthesis of triglycerides, phospholipids, and cholesterol as well as the metabolism of fatty acids, and goats derive approximately 80 % of their energy from volatile fatty acids. However, there is a limited number of studies on the impact of InDel (insertion/deletion) mutations in the ACSL5 gene on goat traits. Therefore, This study investigated the spatiotemporal expression pattern of ACSL5 in goats and the association between its polymorphism and growth traits, aiming to identify useful DNA markers and provide a basis for the application of marker-assisted selection (MAS) in goat breeding. The qPCR technique was employed in the expression profiles of the ACSL5 gene in Fuqing (FQ) goats. The PCR technique was employed for type detection of the InDel locus of the ACSL5 gene in 528 goats. We analyzed the genotype frequency, allele frequency, polymorphism information content (PIC), and Hardy-Weinberg equilibrium (HWE) of the InDel locus of the ACSL5 gene in goats. A general linear model was used to analyze the relationship between the genotypes and body size traits of goats. qPCR analysis indicated that the ACSL5 gene was expressed ubiquitously in the nine tested tissues of FQ goats. The expression level of ACSL5 in fat tissue at birth was higher than in adult (P<0.01) and weaning (P<0.05) goats. An InDel polymorphism termed del41712 was detected within the fifth intron of the ACSL5 gene. Genetic screening revealed only two genotypes, ID and II, present in the three studied goat breeds. Association analyses conducted on 528 goats linked this InDel polymorphism to body measurement traits, showing significant associations with chest depth (ChD) in FQ goats (P<0.05) and with body height (BH), body length index (BLI), and chest circumference index (ChCI) in Nubian (NB) goats (P<0.05). These findings suggest that InDel mutations in the goat ACSL5 gene may serve as a valuable molecular genetic marker for breeding programs aimed at improving production traits.

This study evaluated the synergistic effects of essential oils (EOs), Saccharomyces cerevisiae (SC), and isomaltooligosaccharides (IMO) on intestinal health and systemic immunity in weaned piglets using a randomized block design across two commercial farms. Within each farm, 108 piglets weaned at 24 d of age were randomly divided into 3 groups (3 pens per group, 12 piglets per pen). The piglets were fed a basal diet (CON) and a basal diet supplemented with antibiotics (0.5 kg/t colistin sulfate + 0.5 kg/t tilmicosin) (ANTI) or additive mixture (0.1 kg/t EOs, 0.3 kg/t SC and 1 kg/t IMO) (ESI) for 28 days. The results showed that both ESI and ANTI treatments significantly increased the ratio of villus height to crypt depth (P < 0.05). Specifically, ESI supplementation elevated serum IgA, hepatic IgA, and jejunal mucosal sIgA levels (P < 0.05), while upregulating hepatic IL1B, IL2, IL6, and IFNG expression alongside jejunal mucosal IL1B, IL6, and PPARG expression-contrasting with its downregulation of jejunal mucosal IFNG mRNA (P < 0.05). Conversely, compared with the CON group, ANTI supplementation increased serum IgA level and jejunal mucosal concentrations of sIgA, IgG, IgM, IFN-gamma, and IL-10, while also upregulating jejunal mucosal gene expression of IGF1 and TGFB1 (P < 0.05). In addition, compared with Farm I, piglets at Farm II exhibited higher jejunal pH, elevated serum levels of IgA and IL-1 beta, and upregulated gene expression of IL1B, IL2, IL6, and IFNG in the liver as well as TGFB1, IL1B, IL6, and PPARG in the jejunal mucosa (P < 0.05). Moreover, the robust diet x farm interaction effects on immune gene expression in the liver (IL1B, IL2, and IL6) and jejunal mucosa (IL1B, IL2, IL6, IFNG, IL10, and PPARG) were observed (P < 0.05). Collectively, ESI improved jejunal epithelial maturation, nutrient absorption, and immune function in weaned piglets, with efficacy partly dependent on farm environmental conditions.

Abstract :
The coloration of plumage in poultry species has substantial economic significance. Putian black ducks encompass 2 distinct strains characterized by black and white plumage variations resulting from selective breeding. This study aimed to identify the molecular mechanisms responsible for plumage coloration in these 2 distinct strains. A comprehensive genome-wide association study was conducted using DNA data sourced from a F2 segregating population, consisting of 71 individuals with black plumage and 39 individuals with white plumage, derived from these distinct 2 strains. This analysis revealed that 894 nucleo-tide polymorphisms and identified 58 candidate genes. Subsequent Gene Ontology and Kyoto Encyclopedia of Genes and Genomes coenrichment analyses identified MITF as a key candidate gene implicated in melanin biosynthesis. Furthermore, extensive screening of significant polymorphic loci within MITF was carried out via mass spectrometry in 3 distinct populations: 100 individuals with black plumage and 100 individuals with white plumage from the F0 generation; and 50 with black plumage form the F1 generation). Eighteen candidate polymorphic loci were identified demonstrating significant associations with variations in black and white plumage. Notably, 8 of these loci were located within the 2,000 bp region upstream of MITF-M. To validate the critical regulatory role of MITF-M in black and white plumage formation, a dual-fluorescence reporter system was constructed, and dual-fluorescence activity was assessed. The results revealed that the fluorescence activity at wild-type sites (corresponding to black plum-age) was significantly higher than that at the mutant -type sites (associated with white plumage) (P < 0.01). To corroborate the pivotal role of MITF-M in black and white plumage formation, qPCR was employed to evalu-ate the expression levels of various MITF variants in black and white feather bulbs. This analysis demonstrated that only MITF-M exhibited specific expression in black feather bulbs. These results elucidate the central role of polymorphic mutations within the MITF promoter region in the regulation of black and white plumage coloration in Putian black ducks. This study extends our understanding of mechanisms governing duck plumage coloration and provides valuable molecu-lar markers for future research in duck production and breeding based on plumage coloration.

Abstract :
The antioxidant properties of crocin are attracting interest, yet the underlying mechanisms by which crocin mitigates oxidative stress-induced intestinal damage have not been determined. This study aimed to elucidate the effects of crocin on oxidative stress, apoptosis, and intestinal epithelial injury in intestinal epithelial cells (IPEC-J2). Using an H2O2-induced oxidative stress model in IPEC-J2 cells, crocin was added to assess its effects. Cell viability and apoptosis were evaluated using methyl thiazolyl tetrazolium assays and flow cytometry. Additionally, oxidative stress markers, such as superoxide dismutase (SOD), catalase (CAT), reactive oxygen species (ROS), and malondialdehyde (MDA), were quantified. We investigated, in which cell oxidation and apoptosis were measured at the gene and protein levels and employed transcriptome analysis to probe the mechanism of action and validate relevant pathways. The results showed that crocin ameliorates H2O2-induced oxidative stress by reducing ROS and MDA levels and by countering the reductions in CAT, total antioxidant capacity, and SOD. Crocin also attenuates the upregulation of key targets in the Nrf2 pathway. Furthermore, it effectively mitigated IPEC-J2 cell apoptosis caused by oxidative stress, as evidenced by changes in cell cycle factor expression, apoptosis rate, mitochondrial membrane potential, and apoptosis pathway activity. In addition, crocin preserves the integrity of the intestinal barrier by protecting tight junction proteins against oxidative stress. Transcriptome sequencing analysis suggested that the mitochondrial pathway may be a crucial mechanism through which crocin exerts its protective effects. In summary, crocin decreases oxidative stress molecule formation, inhibits Nrf2 pathway activity, prevents apoptosis-induced damage, enhances oxidative stress resistance in IPEC-J2 cells, and maintains redox balance in the pig intestine.

紫锥菊(Echinacea)是备受欢迎的天然植物之一,具有抗炎、抗氧化和抑菌等多重生物活性。作为一种安全且具有强免疫活性的天然药物,紫锥菊及其提取物成为不同领域科学家的研究热点。从紫锥菊物种中分离出了多组具有药理活性的化合物,如多糖、糖蛋白、烷酰胺类化合物和咖啡酸类衍生物等,大量研究证明它们可能通过多种信号通路发挥作用,如通过丝裂原活化蛋白激酶(MAPK)、c-Jun N-末端激酶(JNK)等发挥免疫作用,以及通过核因子E2-相关因子2-Kelch样ECH关联蛋白1(Nrf2-Keap1)、核因子kappa B(NF-κB)等发挥抗氧化作用。近年来,在天然原料需求不断增加的背景下,紫锥菊及其提取物在动物养殖方面也显示出良好的应用前景。动物试验表明,紫锥菊及其提取物在一定程度上可促进动物生长,缓解氧化应激,提高繁殖性能,并调节机体免疫反应,提高抗病能力,降低疾病的发生率和传播风险,从而保障动物健康和生产效益。这种天然的功能性添加剂符合现代消费者对安全、健康食品的需求,也为畜禽养殖业的可持续发展提供了可行的选择。作者就紫锥菊的主要化学组分及其生物活性、作用机制,以及紫锥菊及其提取物在畜禽和水产养殖中的应用现状进行综述,旨在为规范紫锥菊有效成分的标准化提取、深入探究紫锥菊及其提取物在动物生产中的作用机制以及进一步拓宽其在畜禽和水产领域的应用提供理论参考。

酵母培养物是活酵母在特定工艺下经过充分厌氧发酵后形成的微生态制品,主要由酵母内容物、酵母细胞壁、酵母细胞代谢物和变性培养基等构成,富含β-葡聚糖、甘露寡糖、维生素、有机酸等多种营养物质,具有调节肠道微生态平衡、吸附霉菌毒素、增强机体免疫力和抗氧化等作用,从而促进肠道健康并提高动物的生产性能。酵母培养物作为一类饲料原料在猪、禽、反刍动物等养殖领域得到了广泛应用。本文从酵母培养物的主要组成、作用机理以及在畜禽生产中的应用进行阐述,以期为酵母培养物在畜禽养殖领域上的深入开发与合理应用提供参考。

【目的】检测福清山羊和努比亚山羊群体细胞介素15受体亚基α(IL15RA–interleukin 15 receptor subunit alpha, IL15RA)基因的表达谱以及突变位点多态性,分析不同位点与山羊生长性状的相关性,进一步分析IL15RA基因对山羊生长性状的影响。【方法】随机挑选福清山羊和努比亚山羊12月龄羯羊各3只,分别采集心、肝、脾、肺、肾、头颈半棘肌、背最长肌、股四头肌和臀股二头肌共9种组织,采用qRT-PCR方法构建该基因在两个品种不同组织中的表达谱,Sanger测序检测福清山羊(n=108)和努比亚山羊(n=286)群体中IL15RA基因的SNP位点,并分析其与生长性状的相关性。【结果】(1)IL15RA基因在肌肉组织中的表达水平显著低于内脏组织(P<0.05),福清山羊背最长肌表达水平显著低于努比亚山羊(P<0.05),肺部组织表达量显著高于努比亚山羊(P<0.05)。(2)福清山羊和努比亚山羊IL15RA基因内含子3存在1个InDel位点,为g.22224_22 225 Del A,Del22224。内含子6存在1个SNP位点,为g.33428A> G。(3)SNP-g.33428位点显著影响(P<0.05)福清山羊的体重、胸围、胸宽、胸深、尻宽等指标。(4)在福清山羊中,单倍型组合DD/AA、ID/GG为优势单倍型组合;在努比亚山羊中,单倍型组合ID/AA、II/GG为优势单倍型组合。【结论】IL15RA基因多态性与福清山羊和努比亚山羊生长性状显著相关,IL15RA基因可作为福清山羊和努比亚山羊生长性状选育的候选基因。

Adipose tissue is the most important energy storage organ in the body, maintaining its normal energy metabolism function and playing a vital role in keeping the energy balance of the body to avoid the harm caused by obesity and a series of related diseases resulting from abnormal energy metabolism. The dysfunction of adipose tissue is closely related to the occurrence of diseases related to obesity metabolism. Among various organelles, mitochondria are the main site of energy metabolism, and mitochondria maintain their quality through autophagy, biogenesis, transfer, and dynamics, which play an important role in maintaining metabolic homeostasis of adipocytes. On the other hand, mitochondria have mitochondrial genomes which are vulnerable to damage due to the lack of protective structures and their proximity to sites of reactive oxygen species generation, thus affecting mitochondrial function. Notably, mitochondria are closely related to other organelles in adipocytes, such as lipid droplets and the endoplasmic reticulum, which enhances the function of mitochondria and other organelles and regulates energy metabolism processes, thus reducing the occurrence of obesity-related diseases. This article introduces the structure and quality control of mitochondria in adipocytes and their interactions with other organelles in adipocytes, aiming to provide a new perspective on the regulation of mitochondrial homeostasis in adipocytes on the occurrence of obesity-related diseases, and to provide theoretical reference for further revealing the molecular mechanism of mitochondrial homeostasis in adipocytes on the occurrence of obesity-related diseases.

功能性低聚糖作为一种新型的生理活性物质,具有绿色、天然环保、无残留的特点,在仔猪生产中可替代部分抗生素。功能性低聚糖可被动物肠道微生物发酵利用,产生短链脂肪酸,调节肠道菌群结构,具有抗炎及免疫调节作用,维持断奶仔猪肠道屏障完整性,进而改善仔猪肠道健康。文章总结分析了功能性低聚糖在断奶仔猪生产中的作用机制及其应用,为其在仔猪生产中的应用提供参考。
