复合脂肪酸对骡鸭原代肝细胞脂质沉积及AMPK信号通路的影响

Abstract :

试验旨在探究复合脂肪酸对骡鸭原代肝细胞超微结构、脂质沉积、氧化应激及AMPK信号通路相关基因表达的影响。试验使用不同浓度的复合酸(油酸+棕榈酸)进行处理:500μmol/L+250μmol/L、1 000μmol/L+500μmol/L、1 500μmol/L+750μmol/L和2 000μmol/L+1 000μmol/L,并持续处理36 h构建原代骡鸭肝细胞脂质沉积模型。结果显示:(1)分离的骡鸭原代肝细胞有丰富糖原颗粒和脂滴的存在。(2)与对照组相比,油酸+棕榈酸浓度为1 500μmol/L+750μmol/L时,培养骡鸭原代肝细胞36 h,细胞活力没有显著差异(P>0.05),脂滴数量显著增加(P<0.05)。(3)透射电镜结果显示,复合脂肪酸处理的骡鸭原代肝细胞和线粒体均出现明显水肿,脂滴出现聚合,并且膜和嵴的结构受损。此外,与对照组相比,肝细胞脂滴含量、谷丙转氨酶和谷草转氨酶活性、丙二醛和活性氧水平显著上升(P<0.05)。(4)与对照组相比,复合脂肪酸处理的骡鸭原代肝细胞中AMP活化蛋白激酶(AMPK)、脂蛋白脂肪酶(LPL)和肉碱棕榈转移酶-1A(CPT-1A) mRNA表达量以及AMPK蛋白表达量显著降低(P<0.05),而白细胞分化抗原36(CD36)、乙酰辅酶A羧化酶1(ACC1) mRNA及白细胞分化抗原36(CD36)、胆固醇调节元件结合蛋白1(SREBP1)蛋白表达量显著提高(P<0.05)。因此,油酸+棕榈酸浓度为1 500μmol/L+750μmol/L处理骡鸭原代肝细胞36 h能通过调控AMPK信号通路,有效增加肝细胞脂质沉积,但同时易诱导肝细胞氧化应激。

龙岩山麻鸭蛋壳质量与元素组成、基质含量和超微结构关系的研究

Abstract :

本试验旨在研究蛋壳元素组成、超微结构和钙化壳基质含量对龙岩山麻鸭蛋壳质量的影响。试验选用300日龄龙岩山麻鸭所产白壳蛋300个,根据蛋壳强度平均分为高、中、低3个试验组,每组5个重复,每个重复20个蛋,测定蛋壳质量、元素组成和超微结构。结果表明:高、中、低蛋壳强度组蛋壳强度分别为(4.67±0.09) kg/cm2、(3.78±0.02) kg/cm2和(2.86±0.03) kg/cm2,组间差异显著(P<0.05);各组间的蛋重、蛋形指数、蛋壳厚和蛋壳重无显著差异(P>0.05)。3组间钙化壳的基质含量差异不显著(P>0.05),各组元素相对含量在蛋壳超微结构的不同层次差异不显著(P>0.05)。对蛋壳超微结构观察发现,与中、低强度蛋壳组相比,高强度蛋壳组横断面致密性更好,乳突顶部连接更紧密;并且各组间的乳突层有效厚度差异显著(P<0.05),高、低蛋壳强度组的乳突间隙差异显著(P<0.05)。综上所述,厚度相似的蛋壳,其蛋壳各层元素相对含量和基质含量对龙岩山麻鸭蛋壳强度无显著影响,蛋壳超微结构致密度对蛋壳质量具有重要作用,其中乳突层有效厚度和乳突间隙是影响蛋壳质量的重要因素。

Abstract :

【目的】以鸭疫里默氏菌(Riemerella anatipestifer)噬菌体CRP2为对象,研究尾丝蛋白(Tail fiber protein,TFP)对噬菌体吸附功能的影响。【方法】克隆ORF70假定的TFP基因,构建重组表达载体pET28a(+)-TFP,在大肠杆菌E.Coli BL21(DE3)中诱导表达,通过Ni-NTA亲和层析纯化。酶联免疫吸附试验(ELISA)测定重组蛋白与CRP2噬菌体的宿主菌外膜蛋白(Outer membrane protein, OMP)的结合作用。竞争吸附试验测定重组蛋白对CRP2噬菌体吸附率的影响。【结果】构建了pET28a(+)-TFP重组表达质粒,在大肠杆菌中得到了可溶性表达。纯化的重组蛋白可以与宿主菌的OMP结合,也可以抑制CRP2对其宿主菌的吸附。【结论】ORF70编码的TFP是噬菌体CRP2的一种受体结合蛋白,其对应的受体是宿主菌的OMP。

Mechanism of fibroblast growth factor 1 regulating fatty liver disorder in mule ducks SCIE

Abstract :

Mule ducks tend to accumulate abundant fat in their livers via feeding, which leads to the formation of a fatty liver that is several times larger than a normal liver. However, the mechanism underlying fatty liver formation has not yet been elucidated. Fibroblast growth factor 1 ( FGF1 ), a member of the FGF superfamily, is involved in cellular lipid metabolism and mitosis. This study aims to investigate the regulatory effect of FGF1 on lipid metabolism disorders induced by complex fatty acids in primary mule duck liver cells and elucidate the underlying molecular mechanism. Hepatocytes were induced by adding 1,500:750 mmol/L oleic and palmitic acid concentrations for 36 h, which were stimulated with FGF1 concentrations of 0, 10, 100, and 1000 ng/mL for 12 h. The results showed that FGF1 signi ficantly reduced the hepatic lipid droplet deposition and triglyceride content induced by complex fatty acids; it also reduced oxidative stress; decreased reactive oxygen species fluorescence intensity and malondialdehyde content; upregulated the expression of antioxidant factors nuclear factor erythroid 2 related factor 2 ( Nrf2 ), HO -1, and NQO-1; signi ficantly enhanced liver cell activity; promoted cell cycle progression; inhibited cell apoptosis; upregulated cyclin-dependent kinase 1 ( CDK1 ) and BCL-2 mRNA expression; and downregulated Bax and Caspase-3 expression. In addition, FGF1 promoted AMPK phosphorylation, activated the AMPK pathway, upregulated AMPK gene expression, and downregulated the expression of SREBP1 and ACC1 genes, thereby alleviating excessive fat accumulation in liver cells induced by complex fatty acids. In summary, FGF1 may alleviate lipid metabolism disorders induced by complex fatty acids in primary mule duck liver cells by activating the AMPK signaling pathway.

番鸭不同质地蛋壳超微结构及其与蛋壳强度的关联分析

本试验旨在研究番鸭正常蛋、沙壳蛋和条纹蛋等3种不同质地蛋壳在超微结构性状上的异同,并分析蛋壳超微结构性状与蛋壳强度之间的关联性。试验选用300日龄笼养白羽番鸭所产白色的正常蛋、沙壳蛋和条纹蛋厚度相近的蛋壳各12枚,分别作为3种不同质地蛋壳的试验组,每组设12个重复,每个重复1枚蛋壳。使用S3400N型扫描电镜观察蛋壳的超微结构,测定乳突层厚度和乳突间隙,计算有效厚度和平均乳突大小。结果表明:1)蛋壳强度在3种蛋之间差异显著(P<0.05),表现为正常蛋>沙壳蛋>条纹蛋;3种蛋间蛋形指数和钙化壳重差异均不显著(P>0.05)。2)在超微结构上,沙壳蛋和条纹蛋蛋壳的外表面皲裂较正常蛋蛋壳多而深,气孔较多,表观不平整;与正常蛋蛋壳相比,沙壳蛋蛋壳内表面纤维疏松凌乱,纤维间空隙较多,而条纹蛋蛋壳内表面纤维分布较沙壳蛋更为稀少,纤维间有明显间隙;沙壳蛋和条纹蛋蛋壳的横断面平整性和致密度都更差,乳突层与壳膜层联系也更加不紧密。在超微结构性状中,3种蛋壳间的钙化壳厚度和平均乳突大小差异不显著(P>0.05);沙壳蛋和条纹蛋蛋壳的乳突间隙均显著大于正常蛋蛋壳(P<0.05),而沙壳蛋和条纹蛋蛋壳之间差异不显著(P>0.05);乳突层厚度在3种蛋壳之间均存在显著差异(P<0.05),表现为正常蛋<沙壳蛋<条纹蛋;3种蛋壳间有效厚度也存在显著差异(P<0.05),但表现为正常蛋>沙壳蛋>条纹蛋。3)3种蛋壳的乳突层厚度与蛋壳强度呈显著负相关(P<0.05),其相关系数分别为-0.868、-0.856和-0.837;乳突间隙与蛋壳强度也呈显著负相关(P<0.05),其相关系数分别为-0.736、-0.764和-0.781;有效厚度与蛋壳强度呈显著正相关(P<0.05),其相关系数分别为0.693、0.743和0.657;钙化壳厚度和平均乳突大小与蛋壳强度的相关性均不显著(P>0.05)。由此可见,厚度相似的番鸭正常蛋、沙壳蛋和条纹蛋蛋壳其蛋壳强度和超微结构均存在显著差异,其中乳突层厚度和乳突间隙与蛋壳强度均呈显著负相关,有效厚度与蛋壳强度呈显著正相关;在超微结构层面上,乳突层厚度、乳突间隙及有效厚度是影响蛋壳强度的主要因素。

Ammonia negatively impacted egg production and altered the diversity of microbial communities in cage-reared Muscovy ducks SCIE

Abstract :

The aim of this study was to examine the effects of NH3 variations across different positions within a cage-reared duck house on the egg production performance and fecal microbiome in Muscovy ducks. Totals of 3,168 female Muscovy ducks (180 +/- 2 d) were randomly assigned to 1,056 cages. From d 293 to 300, the egg production rate and levels of NH3, H2S, CO2, temperature, humidity, light intensity, and dust particles were recorded. Two spatial distribution-based experimental classification methods were applied, one grouping ducks into Front, Middle, and End categories, and the other into First, Second, and Third layers. On d 300, serum and feces samples were collected for further analysis. The result showed that both the egg production rate and serum PROG level in the Front were higher than End (P < 0.05). However, no significant differences were found among the First, Second, and Third groups. Among the environmental factors, ammonia (NH3) is a major harmful gas in cage-reared duck houses, with observed concentrations showing a gradient from lower levels in the Front group to higher levels in the Middle group (P < 0.05), and even higher levels in the End group (P < 0.05). Gene sequencing of the 16S rRNA gene revealed a higher relative abundance of Firmicutes in both the Front and Middle groups compared to the End group (P < 0.05). Specifically, within the Firmicutes phylum, the relative abundance of Lactobacillus and Romboutsia was notably higher in the Front group compared to both the Middle and End groups (P < 0.05). What's more, the abundance of Lactobacillus had a negative correlation with NH3 concentration and positive correlation with egg production rate. In conclusion, NH3 concentrations showed variations across different areas of the cage-reared duck house, with higher levels detected at the rear. The elevated NH3 level was identified as the main factor negatively impacting egg production in Muscovy ducks and contributing to a decline in the abundance of Firmicutes, specifically Lactobacillus.

Mechanism of fatty acid transposase (CD36) promoting fat accumulation in mule ducks SCIE

Abstract :

Mule ducks accumulate a large amount of fat in their livers when fed high-energy feed, which is predominantly used for producing fatty livers. Nevertheless, there is limited research on the molecular mechanisms underlying the formation of fatty liver in mule ducks. Fatty acid translocase (CD36) is a sensor for fatty acids and lipid metabolism regulator, which may play a crucial role in the accumulation of fat in the liver of mule ducks. In this study, Overexpression and CD36 gene interference for 24 h was followed by induction of liver cells with 400 mmol/L palmitic acid (PA) for 24 h. The results demonstrated that CD36 overexpression increased hepatic triglyceride content, lipid droplet deposition, oxidative stress, and cell apoptosis. However, interference with CD36 had the opposite effect. CD36 overexpression suppressed the expression of AMPK and CPT-1A genes but enhanced the expression of ACC1 and LKB1 genes, with interference yielding contrasting results. Additionally, the expression of CD36 inhibited the AMPK pathway, reduced AMPK phosphorylation, downregulated AMPK protein expression, and upregulated SREBP1 protein expression. This promoted palmitic acid-induced hepatocyte fat accumulation. In summary, CD36 promotes palmitic acid-induced fat accumulation in primary mule duck liver cells through the AMPK signaling pathway.

This study investigates the underlying mechanism through which dietary supplementation of pyrroloquinoline quinone disodium (PQQ) alleviates intestinal inflammation and cell apoptosis in piglets challenged with lipopolysaccharide (LPS). Seventy-two barrows were divided into three groups: control (CTRL), LPS challenged (LPS), and LPS challenged with PQQ supplementation (PQQ + LPS). On d 7, 11, and 14, piglets received intraperitoneal injections of LPS or 0.9% of NaCl (80 mu g/kg). After a 4 h interval following the final LPS injection on d 14, blood samples were obtained, and all piglets were euthanized for harvesting jejunal samples. The results showed that dietary supplementation of PQQ improved the damage of intestinal morphology, increased the down-regulated tight junction proteins, and reduced the increase of serum diamine oxidase activity, the intestinal fatty acid binding protein, and TNF-alpha levels in piglets challenged with LPS (p < 0.05). The proteomics analysis revealed a total of 141 differentially expressed proteins (DEPs), consisting of 64 up-regulated DEPs and 77 down-regulated DEPs in the PQQ + LPS group compared to the LPS group. The KEGG pathway analysis indicated enrichment of the tight junction pathway and the apoptosis pathway (p < 0.05). Compared to the LPS group, the piglets in the PQQ + LPS group had increased levels of Bcl-2 protein, reduced positive apoptosis signals, and a decrease in the abundance of MKK 3/6 and p-p38 proteins (p < 0.05). In conclusion, dietary supplementation of PQQ could alleviate jejunal inflammatory damage and cell apoptosis in piglets challenged with LPS through the MKK3/6-p38 signaling pathway.

Muscovy ducks' high broodiness hinders industry growth. Studying broodiness regulation contributes to the theoretical foundation for enhancing reproductive performance in Muscovy ducks. Experiment 1, a total of 18 Muscovy ducks were divided into 2 groups: Laying group (LO) and Broody group (BO). To collect ovaries for morphological and transcriptome analysis. Experiment 2, Primary Muscovy ducks granulosa cells (GC) were isolated and treated with or without GDF9 at appropriate concentrations as indicated. Experiment 3, GC were treated with or without GDF9 in the presence or absence of a receptor inhibitor. The cell viability, cell apoptosis rate and levels of TGF-beta pathway were determined. In vivo, there was a gradual disappearance of follicles in the ovaries and accompanied by follicle atrophy and a concentration of cytoplasm in BO group. The transcriptome expression profile revealed a total of 1,185 up-regulated differentially expressed transcripts (DEs) and 1,258 down-regulated DEs in the BO group compared to the LO group. The up-regulated differentially expressed GDF9 is involved in regulating the TGF-beta pathway, which is among the top 10 pathways identified through the KEGG pathway analysis (P < 0.05). Additionally, the fluorescence intensity of apoptosis is primarily observed in the granulosa layers of the ovary. Compared to the LO group, the mRNA level of TGF-beta pathway and the protein of GDF9 and p-Smad2/3 were increased in ovary of the BO group (P < 0.05). In vitro, GDF9 supplementation demonstrated does-related promotion of GC (P < 0.01). Compared to CTRL group, 12 ng/mL GDF9 supplementation to GC increased the rate of cell apoptosis, the mRNA and protein expression of TGF-beta pathway and the apoptosis-related genes. Pretreatment of GC with GDF9-receptor inhibitor largely abrogated the negative function of GDF9 treatment (P < 0.05). In summary, granulosa cell apoptosis leading to follicle atresia in broodiness of Muscovy ducks is associated with GDF9 activation of the TGF-beta pathway. This discovery lays a solid foundation for understanding duck follicular development and enhancing egg production in Muscovy ducks.

Abstract :

Leukaemia inhibitory factor (LIF), a member of the interleukin-6 (IL-6) family, is a multifunctional cytokine. The maturation-to-ovulation process of poultry follicles is determined by granulosa cell proliferation and differentiation. Granulosa cell apoptosis and degeneration lead to follicular atresia, which reduces the number of normally developing follicles and leads to a decrease in the poultry egg production rate, thus affecting the large-scale development of poultry breeding. In this study, the LIF gene overexpression vector pCDH-CMV-LIF and a siRNA that inhibits LIF gene expression were transfected into primary granulosa cells from white Muscovy duck ovaries for functional study. Compared with that in the control group, LIF gene expression was confirmed to be significantly decreased or increased in the transfection groups (P P < 0.01). After LIF overexpression, the expression of the cell cycle-related genes CCND1, CDK-1 and PCNA was decreased (P P < 0.05); apoptosis was promoted; the proapoptotic genes Bax and caspase-3 were significantly upregulated (P P < 0.01); and the antiapoptotic gene Bcl-2 was significantly downregulated (P P < 0.01). After LIF interference, the expression of the cell cycle-related genes CCND1, CCNE1, CDK-1 and PCNA and the antiapoptotic gene Bcl-2 significantly increased (P P < 0.01), whereas the expression of the proapoptotic genes Bax, caspase-3 and caspase-9 significantly decreased (P P < 0.01). In summary, the LIF gene is involved in regulating the biological function of ovarian granulosa cells in white Muscovy ducks. LIF gene expression promotes granulosa cell apoptosis and inhibits cell cycle progression. These experimental results provide insights into the follicular development mechanism of white Muscovy ducks.
