Structural characterization and immune activity evaluation of polysaccharide purified from Viola phillipica SCIE

Abstract :

This study investigated the structure of Viola phillipica polysaccharide (VPP) and its effect as an immune adjuvant for the Newcastle disease vaccine (ND Vaccine). The results showed that VPP comprised of the galacturonic acid, galactose, rhamnose, arabinose with the molar ratio of 36.20: 23.41: 19.72: 11.54 and a molecular weight (Mw) of 1080 kDa. Additionally, the composition of VPP was assessed via FT-IR, methylation, AFM and SEM analyses, indicating that the backbone of the VPP was consisted of T-Ara (f), T-Gal (p), 4-Gal (p)-UA, and T-Rha (p) linkages. Furthermore, VPP was used as an immune adjuvant in combination with ND vaccine in chickens. Remarkable enhancements were observed in the thymus, spleen, and bursa indices of the chickens, the serum levels of ND antibodies and the expression of IL-4 and IFN-gamma cytokines, thereby fostering the proliferation of T and B lymphocytes within the spleen. Notably, histopathological analysis indicated that the VPP groups showed increased lymphoid follicles and splenic nodules, as well as greater splenic red pulp density and bursa of Fabricius area. In summary, these findings suggest that VPP could serve as an adjuvant to enhance the immunogenicity of NDV.

Abstract :

Recent pharmacological studies have demonstrated that Viola philippica polysaccharide (VPP) exhibits a modulating effect on immune activity. However, its utilization has been hampered by its large particle size and complex spatial structure. Polylactic-co-glycolic acid (PLGA) copolymer is recognized as an effective drug delivery carrier, exhibiting excellent biochemical properties. In this experiment, VPP was encapsulated with PLGA to form VPP PLGA nanoparticles (VPP-PLGA NPs). The morphological structure and immunomodulatory effects of VPP-PLGA NPs were evaluated. The particle size of VPP-PLGA NPs was reduced compared to VPP, and the optimal preparation conditions were as follows: The ratio of the organic phase to the internal aqueous phase was 8:1, the ratio of the external aqueous phase to the primary emulsion was 7:1, and the concentration of PLGA was 20 mg/mL. Additionally, VPP-PLGA NPs significantly increased the nitric oxide (NO) content, IL-4, and IFN-gamma levels in RAW264.7 cells, as well as enhanced their phagocytic activity. Furthermore, VPP-PLGA NPs were found to increase NO content and IFN-gamma secretion in bone marrow-derived dendritic cells (BMDCs). These findings suggest that VPP-PLGA NPs could enhance the immune activity and may be utilized as a VPP delivery system for inducing strong immune responses.

Selenium Myricaria germanica Polysaccharides-based Nanoparticles to Improve Immune Activity SCIE

Abstract :

In this study, a nano-drug delivery system based on poly(lactic-co-glycolic acid) (PLGA) that modified selenated Myricaria germanica polysaccharide (sMGP) was developed to enhance immune responses. At the O to W1 ratio of 8.29, the W2 to primary emulsion ratio of 7.62, and the concentration of PLGA was 37.94 mg/mL, the entrapment efficiency of sMGP-PLGA was 90.27%. In vitro, at the concentration of 3.125-25 mu g/mL, sMGP-PLGA significantly enhanced the phagocytosis of RAW264.7 macrophages, facilitated the upregulation of surface molecules CD40 and CD86, and increased the secretion of cytokines interferon-gamma (IFN-gamma) and interleukin (IL)-4, as well as the gene expression levels of toll-like receptor 4 and nuclear factor kappa B. In vivo sMGP-PLGA significantly increased the Newcastle disease antibodies, IFN-gamma, and IL-6 levels, activated the immune functionality of spleen, thymus, and bursa of Fabricius, and improved the organ index. Meanwhile, sMGP-PLGA promoted the proliferation of T and B lymphocytes. These findings indicate that sMGP-PLGA could enhance the immune activity.

Abstract :

In this study, we developed biomimetic Hericium erinaceus polysaccharides PLGA nanoparticles (NPs) coated with macrophage membrane (MM-PLGA-HEP) and examined their immune activity in vitro and in vivo. Nanoparticle characterization revealed that from HEP to MM-PLGA-HEP, there was an increase in negative surface charge (with enhanced stability), structural changes, and a morphological transition from porous HEP sheets to spherical MM-PLGA-HEP. In vitro evaluations confirmed that MM-PLGA-HEP exhibited no cytotoxicity, significantly enhanced macrophage proliferation, dose-dependently stimulated nitric oxide secretion, and uptake studies confirmed its superior internalization efficiency in macrophages. In vivo administration of MM-PLGA-HEP in mice revealed significant improvements in spleen and liver indices, splenic lymphocyte proliferation, macrophage phagocytosis, and CD40/CD86 expression, indicating robust immune activation. Furthermore, MM-PLGA-HEP induced sustained antigen release, resulting in elevated IgG levels and enhanced humoral immune responses compared to controls. These findings underscore the potential of MM-PLGA-HEP as a novel nanocarrier for targeted drug delivery and vaccine adjuvant applications, providing a promising platform for enhanced immunotherapy and antigen-specific delivery.

Abstract :

The present Porcine circovirus type 2 virus (PCV2) vaccine adjuvants suffer from numerous limitations, such as adverse effects, deficient cell-mediated immune responses, and inadequate antibody production. In this study, we explored the potential of a novel nanoparticle (CS-Au NPs) based on gold nanoparticles (Au NPs) and chitosan (CS) that modified Viola philippica polysaccharide (VPP) as efficient adjuvants for PCV2 vaccine. The characterization demonstrated that CS-Au-VPP NPs had a mean particle size of 507.42 nm and a zeta potential value of -21.93 mV. CS-Au-VPP NPs also exhibited good dispersion and a stable structure, which did not alter the polysaccharide properties. Additionally, the CS-Au-VPP NPs showed easy absorption and utilization by the organism. To investigate their immune-enhancing potential, mice were immunized with a mixture of CS-Au-VPP NPs and PCV2 vaccine. The evaluation of relevant immunological indicators, including specific IgG antibodies and their subclasses, cytokines, and T cell subpopulations, confirmed their immune-boosting effects. The in vivo experiments revealed that the medium-dose CS-Au-VPP NPs significantly elevated the levels of specific IgG antibodies and their subclasses, cytokines, and T cell subpopulations in PCV2-immunized mice. These findings suggest that CS-Au-VPP NPs can serve as a promising vaccine adjuvant due to their stable structure and immunoenhancement capabilities.

本试验旨在探寻聚乳酸-羟基乙酸共聚物(poly lactic-co-glycolic acid,PLGA)加载对党参多糖(Codonopsis pilosula polysaccharide,CPP)结构特征和免疫调控作用的影响。采用乳化溶剂挥发法制备党参多糖PLGA纳米颗粒(CPP-PLGA-NPs),应用响应曲面法优化包封条件,通过透射电镜、Zeta电位与纳米粒度仪分析其结构,并利用MTT法、流式细胞术、ELISA法测定CPP-PLGA-NPs和CPP对RAW264.7细胞的免疫调控作用。结果显示,制得的纳米颗粒表面光滑、形态均匀、分散性良好、状态稳定、粒径小。最佳制备条件为:有机相与内水相之比为8.51,外水相与初乳比为7.12,F68的浓度为0.33%,PLGA的浓度为30 mg/m L,制备的纳米颗粒的包封率为93.54%±0.56%。与空白组相比,CPP-PLGA-NPs和CPP都显著提高了巨噬细胞的吞噬活性、NO分泌量、CD40和CD86的表达量、细胞因子(TNF-α、IL-6、IL-1β)的分泌量(P<0.05),在浓度为0.97μg/m L时CPP-PLGA-NPs调控作用达到最高。结果表明,CPP-PLGA-NPs和CPP都有较好的免疫调节活性,为党参多糖的开发与利用提供了理论依据。

Abstract :

Oxidative stress plays an important role in various diseases. miR-221 has been reported to regulate oxidative stress. However, the mechanism of miR-221 in regulating oxidative stress induced by sCPPS(5) remains unclear. This study aimed to investigate the protective effects and mechanisms of miR-221 on oxidative stress induced by sCPPS(5). The expression of SOD, CAT, MDA, LDH, MMP, caspase-3 activity and apoptosis were measured. In addition, the key signaling factors in the Keap1-Nrf2-ARE signaling pathway were determined by real-time PCR and Western blot. Mice were employed to evaluate the effects of sCPPS(5) and the possible mechanism in vivo. sCPPS(5) promoted the expression of SOD and CAT and activated Keap1-Nrf2-ARE signaling pathway inhibit the MDA content, MMP, caspase-3 activity, apoptosis and LDH release rate after transfection with miR-221 mimics and inhibitors. Consistently, sCPPS(5) has the potential to enhance the expression of antioxidant enzymes as well as upregulate mRNA expression of crucial signal proteins in vivo. miR-221 on oxidative stress protection induced by sCPPS(5) possibly through regulating the Keap1-Nrf2-ARE signaling pathway in macrophages.

为探究壳聚糖猴头菇多糖PLGA纳米颗粒(CS-PLGA-HEP)对免疫活性调节的影响,通过制备CS-PLGA-HEP纳米颗粒,采用响应曲面法优化制备工艺,傅里叶红外光谱检测、Zeta电位检测等方法对CS-PLGA-HEP纳米颗粒的结构进行表征检测,并通过鸡新城疫疫苗免疫佐剂注射试验来探究其免疫效果。结果显示,CS-PLGA-HEP的最优条件:F68浓度为0.38%,有机相(O)与内水相(W_1)之比为5.75,外水相(W_2)与初乳(PE)之比为7.81,在此条件下其包封率的实测值为(91.32±0.13)%,接近预测值91.60%。纳米颗粒均匀,表面光滑且状态稳定。当CS-PLGA-HEP组浓度为20 mg/kg时,与猴头菇多糖(HEP)组相比更能有效提高鸡血清新城疫特异性抗体浓度,促进IL-4和IFN-γ的分泌水平,增强鸡脾淋巴细胞的成熟分化,提升机体免疫调节能力。结果表明,采用响应曲面法优化壳聚糖猴头菇多糖PLGA纳米颗粒制备条件合理,且纳米修饰可以增强HEP的免疫调节活性,为免疫增强剂的设计与应用提供了参考。

In this study, a novel nano-drug delivery system (CS-Au NPs) based on gold nanoparticles (Au NPs) and chitosan (CS) that modified Myricaria germanica polysaccharide (MGP) was developed to enhance immune responses. At a MGP to CS Au ratio of 5:1, CS-Au-MGP NPs had a loading capacity of 78.27 %. The structure of CS-Au-MGP NPs were characterized by Transmission electron microscope, TEM-energy dispersive spectroscopy mapping, Fourier transform infrared spectroscopy, X-ray photoelectron spectrometer, particle size and zeta-potential distribution analysis. Under weakly acidic conditions, in vitro CS-Au-MGP NPs release was most effective. In vivo showed that co-immunization with CS-Au-MGP NPs and PCV2 significantly increased the organ index of the thymus, spleen, and liver in mice. Additionally, CS-Au-MGP NPs significantly increased the levels of IgG, IgG1, and IgG2a antibodies, as well as IFN-gamma and IL-6 levels. Furthermore, the CS-Au-MGP NPs promoted proliferation of spleen T and B lymphocytes, increased the number of CD3+, CD4+, and CD8+ cells, and increased the CD4+/CD8+ T cell ratio. Meanwhile, CS-Au-MGP NPs remarkably TLR2/IRAK4 pathway activation and mRNA levels of cytokines (IFN-gamma and IL-6). These results indicated that CS-Au-MGP NPs could enhance the immune activity, and it could be potentially used as an MGP delivery system for the induction of strong immune responses.

本试验旨在利用响应曲面法优化硒化猴头菇多糖纳米微粒的制备工艺条件,研究其对脾淋巴细胞免疫活性的影响。在单因素试验的基础上,采用响应曲面法研究有机相(O)和内水相(W1)之比、外水相(W2)和初乳(PE)之比以及PLGA浓度对硒化猴头菇多糖纳米微粒包封率的影响,并通过透射电镜以及粒径与Zeta电位的表征,检测模拟最优工艺条件制备硒化猴头菇多糖纳米微粒,并通过鸡脾淋巴细胞增殖和细胞因子IFN-γ、IL-4分泌量的检测,探究其免疫效果。结果显示,硒化猴头菇多糖纳米微粒的最佳制备工艺:有机相和内水相之比为4.66,外水相与初乳之比为7.23,PLGA浓度为21.19 mg/m L,在此条件下硒化猴头菇多糖纳米微粒包封率的实测值为(90.21±0.01)%,接近于预测值91.48%。纳米微粒的粒径小,表面光滑,粒子间有明显的界限,无黏连现象。硒化猴头菇多糖纳米微粒浓度为0.49μg/m L时,与硒化猴头菇多糖相比,更能促进鸡脾淋巴细胞增殖(P<0.05)。浓度为0.25μg/m L~0.98μg/m L时,与硒化猴头菇多糖相比,硒化猴头菇多糖纳米微粒更能促进鸡脾淋巴细胞分泌IFN-γ(P<0.05)。结果表明,采用响应曲面法优化硒化猴头菇多糖纳米微粒制备条件是合理可行的,且硒化猴头菇多糖纳米微粒具有免疫增强活性。
