Health effects of astaxanthin in the intestinal tract of yellow-feathered broilers SCIE

Abstract :

Astaxanthin (AST), a keto carotenoid, is widely recognized for its antioxidant, anti-inflammatory, and apoptosisregulating properties, but its effects on intestinal health have not been elucidated. Therefore, this experiment aimed to investigate the effects of astaxanthin on intestinal morphology, barrier function, mucosal immunity, and cecal flora in yellow-feathered broilers, and to explore the potential mechanisms. A total of 288 male yellow- feathered broilers (1-day-old) were randomly allocated to four groups with six replicates of 12 birds each. The control group (CON) was fed a basal diet, the test groups were fed a basal diet added with 20 mg/kg, 40 mg/kg, and 80mg/kg of astaxanthin (AST20, AST40, and AST80), respectively. The results showed that compared with the CON group, the villus height and the villus-to-crypt ratio of broiler jejunum in the astaxanthin-added group increased, while the crypt depth decreased (P < 0.05). In addition, IL-1 beta content and gene expression in broiler jejunal mucosa decreased, IL-6 gene expression decreased, and IL-4 content and gene expression increased (P < 0.05). The tight junction protein ZO-1 expression level in the jejunum mucosa of broilers was up-regulated, and the content of serum D-lactic acid was decreased (P < 0.05). Besides, the alpha-diversity and beta-diversity analyses showed that astaxanthin regulated both the diversity and the structure of the intestinal flora of broilers. After multiple comparative analyses, the relative abundance of norank_f__Eubacterium_coprostanoligenes_group, Lachnospiraceae NC2004_group, and unclassified_p_Firmicutes increased in the AST20 group compared to the CON group, while that of Ruminococcus_torques_group in the AST80 group decreased (P < 0.05). Therefore, AST can regulate the immune function of broiler intestinal mucosa, improve intestinal morphology and structure, and then influence intestinal permeability. It also has a considerable regulatory effect on the diversity and structure of broiler intestinal flora and plays a multifaceted role in maintaining broiler intestinal health.

Abstract :

Antibiotics have been commonly added to poultry feed to promote growth and prevent diseases, but overuse raises concerns about human health and environmental safety. Finding safe alternatives is crucial for sustainable poultry production. Astaxanthin (AST) is a red, lipid-soluble carotenoid possessing antioxidant and anti-inflammatory properties. This study investigated the effects of different doses of astaxanthin (20, 40, and 80 mg/kg) on growth performance, meat quality, antioxidant capacity, immune function, and gut microbiota in yellow-feathered broilers. Results showed that dietary astaxanthin supplementation improved the meat quality (increased pH, a* and b* values, decreased L* value and drip loss), enhanced antioxidant capacity (reduced MDA, increased T-AOC and SOD levels) and immune function (decreased IL-1 beta and IL-6, increased IL-4 levels), and regulated gut microbiot (increased Faecalibacterium, norank_f__Ruminococcaceae, decreased Romboutsia and Turicibacter) (P < 0.05). Thus, astaxanthin may serve as a potential novel functional feed additive, beneficial for improving broiler health and meat quality.

两种植物黄酮醇对动物肠道黏膜屏障的调控

Abstract :

肠道黏膜屏障是维持机体肠道稳态平衡和阻碍病原菌入侵的重要屏障,包括物理屏障、化学屏障、微生物屏障和免疫屏障4个部分,其结构和功能的完整性对维持动物肠道健康至关重要。肠道黏膜屏障损伤会导致肠道功能紊乱,影响机体健康。植物黄酮醇是自然界分布最广的类黄酮,越来越多的研究表明,黄酮醇具有调节肠道功能、减轻肠道炎症和抗氧化等作用。针对槲皮素和山奈酚两种典型的植物黄酮醇对动物肠道黏膜屏障生理调控作用及对畜禽动物的影响进行了综述,旨在为植物黄酮醇作为功能性饲料添加剂应用于畜禽养殖提供依据。

二甲酸钾与月桂酸对肉鸡肝脏脂肪酸组成和抗氧化能力的影响

Abstract :

本试验旨在探究饲粮中添加二甲酸钾或月桂酸对白羽肉鸡肝脏脂肪酸组成、肝脏抗氧化能力的影响。选取240羽1日龄、体重(41.88±2.57)g的白羽肉公鸡,分为4个试验组,即空白组(基础饲粮)、抗生素组(基础饲粮+400 mg/kg杆菌肽锌)、二甲酸钾组(基础饲粮+1 000 mg/kg二甲酸钾)、月桂酸组(基础饲粮+500 mg/kg月桂酸),每组5个重复,每个重复12羽。试验期共42 d。结果表明:与空白组相比,抗生素组、二甲酸钾组和月桂酸组21日龄白羽肉鸡肝脏中C16:0、C18:0、C18:1n9c、C18:2n6c、C20:4n6、C20:5n3、C22:6n3含量均降低(P<0.05),抗生素组C22:1n9含量提高(P<0.05);相较于空白组,3个处理组21日龄肉鸡肝脏超氧化物歧化酶(SOD)含量和42日龄肉鸡肝脏过氧化氢酶(CAT)含量均提高(P<0.05),二甲酸钾组21日龄肉鸡肝脏CAT含量提高(P<0.05);月桂酸组42日龄肝脏和血清谷胱甘肽过氧化物酶(GSH-Px)含量均提高(P<0.05);与空白组相比,添加二甲酸钾可促进21日龄肉鸡肝脏CAT基因表达(P<0.05),3个处理组21日龄肉鸡肝脏SOD和42日龄肉鸡肝脏CAT基因表达量均提高(P<0.05),而月桂酸组42日龄肉鸡肝脏GPX基因表达量升高(P<0.05);与对照组相比,添加二甲酸钾可抑制21日龄肉鸡肝脏Kelch样ECH关联蛋白1(Keap1)基因表达(P<0.05),添加抗生素、二甲酸钾或月桂酸均可降低42日龄肉鸡肝脏Keap1基因相对表达量(P<0.05);二甲酸钾组和月桂酸组21日龄与42日龄肉鸡肝脏核呼吸因子2(Nrf2)基因表达水平均较对照组上调(P<0.05)。可见,饲粮中添加二甲酸钾、月桂酸可以改变肝脏脂肪酸组成,且提高肉鸡血清GSH-Px和肝脏SOD、GSH-Px、CAT等抗氧化酶含量,并可能通过调节Keap1-Nrf2信号通路,促进CAT、SOD、GPX基因的表达,进而提高白羽肉鸡的抗氧化能力。

This experiment investigated the effects of dietary supplementation with nano-composites of copper and carbon (NCCC) on antioxidants, immune functions, and the cecum microbiota of weaned Ira white rabbits. A total of 240 weaned 35-day-old Ira white rabbits were randomly allocated to five dietary treatments (n = 6 per treatment, each replicate consisted of eight rabbits) that included the control group (CON) with a basal diet, the SAL group with 60 mg/kg salinomycin (SAL) in addition to the basal diet, and the NCCC I, II, III groups, which were supplemented with 50, 100, and 200 mg/kg NCCC, respectively, in addition to the basal diet. The test lasted for 28 d. The results showed that dietary NCCC supplementation increased the liver Cu/Zn-SOD content and up-regulated the gene expression of Cu/Zn-SOD (p < 0.05), while also reducing the content of MDA in the liver and enhancing the antioxidant capacity of Ira white rabbits. Moreover, the NCCC diet supplementation reduced the content of IL-6 and down-regulated the relative expression of IL-6 and IL-1 beta genes in the jejunum of Ira white rabbits (p < 0.05). In addition, the metagenomic analysis of 16 S rRNA showed significant differences in the cecal microbial structure of weaned Ira white rabbits in the NCCC III group compared with the CON, NCCC I, and NCCC II groups (p < 0.05). Firmicutes and Bacteroidetes were the dominant phyla of cecal microorganisms in weaned Ira rabbits in the NCCC diet groups. The dominant genera included unidentified Eubacteriaceae, unclassified Lachnospiraceae, Christensenellaceae, and Ruminococcus. Furthermore, the relative abundance of Ruminococcus in the NCCC I and II groups was lower than that in the CON group in the cecum of Ira white rabbits (p < 0.05). In summary, our results showed that diet supplementation with NCCC could enhance the antioxidant capacity in the liver, alleviate intestinal inflammation, and regulate the structure of intestinal flora, improving the health of Ira white rabbits.

巨菌草对育肥从江香猪生长性能及肠道健康的影响

本试验旨在探究巨菌草替代不同水平基础饲粮对育肥从江香猪生长性能及肠道健康的影响。选取6月龄体重相近的健康从江香猪30头,随机分为3组(每组5个重复,每个重复2头)。对照组饲喂100%基础饲粮,试验Ⅰ、Ⅱ组分别用新鲜青绿巨菌草替代10%、20%基础饲粮,试验期90 d。通过测定育肥从江香猪生长性能、肠道发育、肠道机械屏障和盲肠菌群结构,并进行经济效益分析,为巨菌草在猪的生产应用中提供理论依据。本试验主要结果如下:1)生长性能:与对照组相比,试验Ⅰ、Ⅱ组育肥从江香猪生长性能及胴体性状各指标无显著差异(P>0.05)。巨菌草不计成本时,与对照组相比,试验Ⅰ组的增重成本降低8.73%,试验Ⅱ组降低11.59%;巨菌草按市场价格0.35元·kg-1计算时,试验Ⅰ组的增重成本降低2.78%,试验Ⅱ组增加1.35%。2)肠道发育:与对照组相比,试验Ⅰ、Ⅱ组育肥从江香猪胃的相对重量显著提高(P<0.05),小肠相对重量有升高的趋势(P=0.092)。与对照组相比,试验Ⅰ组育肥从江香猪回肠隐窝深度显著降低(P<0.05),回肠绒毛高度/隐窝深度(V/C)、空肠GLP-2和回肠IGF-1R基因表达量显著提高(P<0.05);试验Ⅱ组育肥从江香猪回肠绒毛高度、V/C及空肠GLP-2基因表达量显著提高(P<0.05)。3)肠道免疫及机械屏障:与对照组相比,试验Ⅰ、Ⅱ组育肥从江香猪回肠IFN-γ含量显著降低,试验Ⅱ组空肠TGF-β含量显著提高(P<0.05);试验Ⅰ、Ⅱ组回肠ZO-1基因表达量显著提高,试验Ⅱ组空肠Occludin基因表达量显著提高(P<0.05)。4)盲肠微生物区系:与对照组相比,试验Ⅰ、Ⅱ组育肥从江香猪Shannon指数显著提高,链球菌属相对丰度显著下降(P<0.05)。试验Ⅰ组和试验Ⅱ组毛螺菌科未分类菌属有上升趋势(P=0.065),螺旋体门(P=0.085)、密螺旋体属(P=0.085)有下降趋势。综上所述,巨菌草替代部分基础饲粮对育肥从江香猪生长性能、胴体性状无显著影响,但可提高经济效益并促进胃肠道的发育,提升肠道屏障功能,有利于肠道健康。巨菌草不计成本时,替代20%基础饲粮为宜,巨菌草按市场价格0.35元·kg-1计算时,替代10%基础饲粮为宜。

Kaempferol and Vitamin E Improve Production Performance by Linking the Gut-Uterus Axis Through the Reproductive Hormones and Microbiota of Late-Laying Hens SCIE

This study evaluated the effects of kaempferol (KAE), and vitamin E (VE) on the performance, reproductive hormones, and the composition of the cecum and uterus microbiota in late-laying hens. A total of 192 49-week-old Jinghong No. 1 laying hens were randomly divided into four groups, with six replicates in each group and eight laying hens in each replicate, pre-reared for one week and formally tested for ten weeks. The CON group was fed basal diets, the VE group, the KAE group, and the KAE + VE group were fed a basal diet to which was added 0.2 g/kg VE, 0.4 g/kg KAE, and 0.2 g/kg VE + 0.4 g/kg KAE, respectively. The results are as follows. Compared to the CON group, the VE group, the KAE group, and the KAE + VE group significantly increased the egg production rate, average daily egg weight and significantly decreased the feed-to-egg ratio. The VE + KAE group significantly improved the Haugh unit. The VE group, the KAE group, and the KAE + VE group considerably enhanced the eggshell strength, eggshell relative weight, eggshell thickness, yolk color, and relative yolk weight. The serum E2 and LH levels of the KAE group and the KAE + VE group and the serum FSH levels of the KAE + VE group were significantly higher. In the ovary, the KAE group and the KAE + VE group's ESR1 gene expression levels were significantly higher, and the KAE + VE group's FSHR gene expression levels were markedly higher. In the uterus, the KAE group and the KAE + VE group's ESR1 gene expression levels were dramatically higher, and the KAE + VE group's ESR2 and FSHR gene expression levels were significantly higher. 16S rRNA gene sequencing revealed a significant aggregation of cecum and uterus colonies in the Beta diversity PCoA. In the cecum, Firmicutes, Bacteroidetes, and WPS-2 were the dominant phylums. In the uterus, the Firmicutes, Proteobacteria, and Bacteroidetes were the dominant phylums. The KAE + VE group's F/B was significantly higher at the phylum level than in the CON group and the VE group. In summary, the addition of VE and KAE to the diet can improve the production performance of late-laying hens, increase the content of reproductive hormones, and stabilize the cecal and uterus microbiota, which may be related to the hormone and microbiota linkage of the gut-uterus axis.

In vivo and in vitro protective effect of lutein against inflammatory response in lipopolysaccharide-induced broilers SCIE

Abstract :

Lutein, a naturally occurring xanthophyll carotenoid abundant in leafy green vegetables, is known for its antioxidative and anti-inflammatory effects. This research assessed how lutein modulates intestinal inflammation triggered by lipopolysaccharide (LPS) exposure in vivo and in vitro. In vivo trials involved 240 male yellowfeathered broilers (1-day-old) divided into four groups: control (basal diet), lutein-supplemented (40 mg/kg), LPS-challenged (1 mg/kg), and lutein+LPS. Growth parameters, inflammatory cytokines, and TLR4/MyD88 pathway components were analyzed. In vitro, chicken intestinal epithelial cells were exposed to LPS (20 mu g/mL) with or without lutein pretreatment. Results indicated that lutein mitigated LPS-induced growth suppression (ADG: P = 0.09; BWG: P = 0.10) and reduced pro-inflammatory cytokines (IL-1(3, IL-6) and the inflammatory signaling pathway transduction molecules TLR4, MyD88 expression (P < 0.05). In vitro, 80 mol/L lutein optimally attenuated inflammatory markers. In conclusion, lutein prevented the LPS-induced decline in growth performance, attenuated the inflammatory response, and inhibited the TLR4/MyD88 signaling pathway, thereby exerting a protective effect on the intestinal tract of broilers. These findings highlight lutein's potential to counteract LPS-induced inflammation via TLR4/MyD88 modulation, offering a strategy for poultry health management.

Abstract :

Simple Summary: Juncao-substrate Ganoderma lucidum Residue (HWE-JGLR) is a dark brown powder. It is obtained by cultivating G. lucidum using mycorrhizae as culture material and extracting it from the remaining culture substrate after G. lucidum harvesting. The application of HWE-JGLR in animal feeds not only reduces the waste of resources but also promotes the reuse of agricultural resources and sustainable development. In our study, HWE-JGLR not only optimised the growth performance of broilers but also enhanced their immune responses. Specifically, dietary supplementation with HWE-JGLR was associated with increased production and gene expression of the immunomodulatory cytokine interleukin-4 (IL-4) and IL-10. Meanwhile, levels and gene expression of the pro-inflammatory cytokine IL-1 beta were significantly reduced. In addition, HWE-JGLR enhanced the intestinal barrier aspect in poultry. The addition of HWE-JGLR upregulated the gene expression of the broiler jejunal mucosal tight junction proteins Claudin-1 and ZO-1. The purpose of this experiment was to investigate the effects of Hot Water Extract of Juncao-substrate Ganoderma lucidum Residue (HWE-JGLR) on the immune function and intestinal health of yellow-feather broilers. In an animal feeding experiment, 288 male yellow-feather broilers (1 day old) were randomly allocated to four treatment groups with six replicates of 12 birds each. The control (CON) group was fed a basal diet. HJ-1, HJ-2, and HJ-3 were fed a basal diet supplemented with 0.25%, 0.50%, and 1.00% HWE-JGLR, respectively. The feeding trial lasted for 63 d. The results showed increased ADFI (p = 0.033) and ADG (p = 0.045) of broilers in HJ-3, compared with the CON group. Moreover, higher contents of serum IL-4 and IL-10 and gene expression of IL-4 and IL-10 in jejunum mucosa and lower contents of serum IL-1 beta and gene expression of IL-1 beta in jejunum mucosa in HJ-3 were observed (p < 0.05). Additionally, the jejunal mucosal gene expression of Claudin-1 and ZO-1 in HJ-2 and HJ-3 was higher than that in the CON group (p < 0.05). As for the microbial community, compared with the CON group, the ACE index, Shannon index, and Shannoneven index of cecal microorganisms in HJ-2 and HJ-3 were elevated (p < 0.05). PCoA analysis showed that the cecal microbial structure of broilers in HJ-2 and HJ-3 was different from the CON group (p < 0.05). In contrast with the CON group, the broilers in HJ-2 and HJ-3 possessed more abundant Desulfobacterota at the phylum level and unclassified Lachnospiraceae, norank Clostridia vadinBB60 group and Blautia spp. at the genus level, while Turicibacter spp. and Romboutsia spp. were less (p < 0.05). In conclusion, dietary supplementation with HWE-JGLR can improve growth performance, enhance body immunity and intestinal development, and maintain the cecum microflora balance of yellow-feather broilers.

Abstract :

大豆寡糖(SBOS)广泛存在于各种植物中,尤其是豆科植物,具有维护肠道健康、保护肝脏功能、增强机体免疫力、调节脂质代谢、抗氧化等生理功能,在畜禽生产中有广阔的应用前景。因SBOS为抗营养因子,易导致动物机体发生胃肠胀气、腹泻,但其维护肠道健康、抗氧化等生理功能备受关注。本文综述了SBOS的生理功能及不同含量SBOS在畜禽营养方面的作用,为SBOS作为新型饲料添加剂提供理论依据。
