福建部分地区犬细小病毒的检测与遗传进化分析

Abstract :

为了解福建省犬细小病毒主要流行趋势及其分子进化特点,对2019-2022年间采集的福建省部分地区疑似感染犬细小病毒的犬粪便样品进行PCR检测,将阳性样品进行VP2全长基因组测序,并对其进行遗传进化分析。结果表明:收集的217份病料中犬细小病毒阳性样品有147份,感染率为67.74%。对其中41份阳性样品进行VP2全长基因组测序,有31株CPV-2c (占75.61%)、8株New CPV-2a (占19.51%)和2株New CPV-2b (占4.88%),说明2019-2022年间福建部分地区流行的犬细小病毒毒株亚型以CPV-2c为主。遗传进化分析表明,分离毒株有7个氨基酸位点发生突变;与我国2008-2022年间各亚型流行毒株属于同一个分支,各自有较高的亲缘关系,与美国、日本和意大利分离株亲缘关系较远,有明显的地域性。该研究丰富了犬细小病毒的基因组信息数据库,可为监测犬细小病毒的变异、地区流行趋势及疫苗研发提供一定参考。

Novel ACE2 binding in bat merbecoviruses expands potential host range SCIE

Abstract :

Coronaviruses often cross species barriers, with receptor binding dictating their host range and zoonotic potential. Merbecoviruses, such as MERS-CoV, typically utilize DPP4 as their receptor, whereas Sarbecoviruses, like SARS-CoV, rely on ACE2. This study explores the receptor usage of four merbecoviruses identified in Vespertilionidae bats: HKU5, BtVs-SC2013, HKU25, and P. khulii-2011. Our findings reveal species-specific binding to bat ACE2: HKU5 binds exclusively to Pipistrellus abramus ACE2, P. khulii-2011 interacts solely with Murina aurata ACE2, BtVs-SC2013 recognizes ACE2 from Murina aurata and Myotis myotis, and HKU25 displays the broadest binding range. Beyond bats, BtVs-SC2013 binds to mink ACE2, while HKU25 interacts with both mink and pangolin ACE2, hinting at potential intermediate hosts for cross-species transmission. We also elucidated the mechanism behind HKU5's selective binding preference for P. abramus ACE2. Structural analysis and mutagenesis revealed that a carbohydrate attached at position 329 play a crucial role. Introducing the N-glycosylation site into P. abramus ACE2 eliminated binding, while its removal from P. pipistrellus ACE2, combined with two additional mutations, restored it. Moreover, we pinpointed key residues in mink ACE2 essential for binding the receptor-binding domain (RBD) of BtVs-SC2013 and HKU25. These findings illuminate the receptor usage and host specificity of bat merbecoviruses, enhancing our understanding of their potential for cross-species transmission and adaptation.

Abstract :

The multibasic furin cleavage site at the S1/S2 boundary of the spike protein is a hallmark of SARS-CoV-2 and plays a crucial role in viral infection. However, the mechanism underlying furin activation and its regulation remain poorly understood. Here, we show that GalNAc-T3 and T7 jointly initiate clustered O-glycosylations in the furin cleavage site of the SARS-CoV-2 spike protein, which inhibit furin processing, suppress the incorporation of the spike protein into virus-like-particles and affect viral infection. Mechanistic analysis reveals that the assembly of the spike protein into virus-like particles relies on interactions between the furin-cleaved spike protein and the membrane protein of SARS-CoV-2, suggesting a possible mechanism for furin activation. Interestingly, mutations in the spike protein of the alpha and delta variants of the virus confer resistance against glycosylation by GalNAc-T3 and T7. In the omicron variant, additional mutations reverse this resistance, making the spike protein susceptible to glycosylation in vitro and sensitive to GalNAc-T3 and T7 expression in human lung cells. Our findings highlight the role of glycosylation as a defense mechanism employed by host cells against SARS-CoV-2 and shed light on the evolutionary interplay between the host and the virus.

Abstract :

为了研究猪链球菌2型(SS2)核糖核酸酶Ⅲ(RNaseⅢ)的催化特性,利用大肠杆菌BL21(DE3)异源表达猪链球菌2型RNaseⅢ(SS-RNaseⅢ)并通过镍柱亲和层析的方法分离纯化。异源表达研究显示,重组蛋白RNaseⅢ大部分为可溶性表达;催化特性研究显示,RNaseⅢ主要依赖Mg~(2+)或Mn~(2+)发挥催化功能,并在不同离子浓度和pH值环境下具有不同的催化活性;底物特异性研究显示,RNaseⅢ特异性切割长度为30 bp的双链RNA,同时对内源性的rnc mRNA和5S rRNA具有良好的特异性。上述结果表明,相较于其他细菌RNaseⅢ,SS-RNaseⅢ具有独特的催化特性,为深入理解RNaseⅢ如何介导SS2致病机制奠定了基础。

猪源Treg细胞白细胞介素-2受体α链单克隆抗体的制备

【目的】制备猪源CD25蛋白单克隆抗体,为猪源Treg细胞的检测及生物学研究提供基础。【方法】将6×His猪源CD25基因片段通过同源重组的方法克隆至原核表达载体pET-28a,构建重组质粒pET28a-CD25。然后利用大肠杆菌表达系统和金属螯合亲和层析介质(Ni-NTA)预装柱对6×His-CD25重组蛋白进行表达和纯化。以纯化的重组蛋白作为抗原免疫雌性BALB/c小鼠,取免疫后小鼠的脾细胞与骨髓瘤细胞(SP2/0)进行细胞融合,经过间接ELISA筛选融合细胞并进行克隆化培养,并利用小鼠腹水法大量制备猪源CD25单克隆抗体。【结果】成功获得一株由单一细胞生长且能够稳定分泌CD25单克隆抗体的杂交瘤细胞株。使用间接ELISA与Western blotting分析显示,CD25单克隆抗体不与其他蛋白产生免疫反应,抗体效价为1∶4 098 000,表明CD25单克隆抗体特异性较强且效价高。间接免疫荧光和免疫印迹分析结果显示,CD25单克隆抗体不仅可以识别真核细胞表达的CD25蛋白,而且可以识别宿主脾脏和肺脏中的内源性CD25蛋白。【结论】成功制备了重组猪源CD25重组蛋白并筛选出特异性单克隆抗体,为定性或定量检测猪源Treg细胞CD25蛋白和研究猪源Treg细胞的功能奠定了基础。

Alphaviruses are arboviruses transmitted by mosquitoes and are pathogenic to humans and livestock, causing a substantial public health burden. So far, several receptors have been identified for alphavirus entry; however, they cannot explain the broad host range and tissue tropism of certain alphaviruses, such as Getah virus (GETV), indicating the existence of additional receptors. Here we identify the evolutionarily conserved low-density lipoprotein receptor (LDLR) as a new cell entry factor for GETV, Semliki Forest virus (SFV), Ross River virus (RRV) and Bebaru virus (BEBV). Ectopic expression of LDLR facilitates cellular binding and internalization of GETV, which is mediated by the interaction between the E2-E1 spike of GETV and the ligand-binding domain (LBD) of LDLR. Antibodies against LBD block GETV infection in cultured cells. In addition, the GST-LBD fusion protein inhibits GETV infection both in vitro and in vivo. Notably, we identify the key amino acids in LDLR-LBD that played a crucial role in viral entry; specific mutations in the CR4 and CR5 domain of LDLR-LBD reduce viral entry to cells by more than 20-fold. These findings suggest that targeting the LDLR-LBD could be a potential strategy for the development of antivirals against multiple alphaviruses. Getah virus (GETV), is a mosquito-borne alphavirus of importance in veterinary medicine. Here, the authors show that evolutionarily conserved low-density lipoprotein receptor (LDLR) functions as a receptor for GETV, which may explain the multi-species spread of this virus.

Abstract :

作为一种应用广泛的木材改性方法,热处理可以显著提高木材的尺寸稳定性和生物耐久性,但也会显著降低木材的韧性,影响了热处理木材在建筑结构领域的应用。因此,深入理解热处理木材韧性变化规律和内在机制对于热处理技术的改良具有重要意义。文中综述了热处理温度、时间、介质和压力等工艺条件对木材韧性的影响,结合断裂过程中的裂纹偏转、纤维桥连、裂纹钝化和微裂纹等韧化机制,从微观结构及吸湿性变化、细胞壁化学反应和分子变形机制等方面总结热处理木材韧性降低的主要原因,讨论半纤维素降解和木质素的缩合反应对热处理木材韧性的影响,分析热处理木材的分子应变与剪切滑移机制,并在此基础上针对热处理木材的韧性研究提出几点建议。

细菌病综合实习是提升动物医学专业预防兽医学实践教学质量的重要环节。针对传统预防兽医学中细菌学实验内容零碎、不系统的现状,本文提出融合细菌学相关实验技术,开展连续性好、综合性强的细菌病综合实习。该实习从混合细菌感染动物人工造病开始,通过一系列实验技术手段分离、纯化和鉴定细菌,进而确诊病因,实施后显著提升了学生的综合实践能力和科研创新水平,有利于新时代高素质动物医学专业人才的培养。

Porcine circovirus-associated disease (PCVAD), caused by porcine circovirus type 2 (PCV2), has ravaged the pig industry, causing huge economic loss. At present, PCV2b and PCV2d are highly prevalent genotypes worldwide, while in China, in addition to PCV2b and PCV2d, a newly emerged PCV2e genotype detected in the Fujian province has attracted attention, indicating that PCV2 genotypes in China are more abundant. A preliminary study was conducted to better understand the genetic diversity and prevalence of PCV2 genotypes in southern China. We collected 79 random lung samples from pigs with respiratory signs, from 2018 to 2021. We found a PCV2-positivity rate of 29.1%, and frequent co-infections of PCV2 with PCV3, Streptococcus suis (S. suis), and other porcine pathogens. All PCV2-positive samples were sequenced and subjected to whole-genome analysis. Phylogenetic analysis, based on the PCV2 ORF2 gene and complete genomes, found that PCV2 strains identified in this study belonged to genotypes PCV2a (1), PCV2b (6), PCV2d (10), and PCV2e (6). Importantly, PCV2e was identified for the first time in some provinces, including Guangdong and Jiangxi. Additionally, we found two positively selected sites in the ORF2 region, located on the previously reported antigenic epitopes. Moreover, codon 63, one of the positively selected sites, has different types of amino acids in different genotypes. In conclusion, this study shows that PCV2e is an emerging genotype circulating in southern China, which warrants urgent, specific surveillance to aid the development of prevention and control strategies in China.
