Abstract :
Sugar-mediated regulation of hormone signaling is crucial for optimizing growth under normal conditions and ensuring survival during environmental stress. Previous studies have shown that sugar starvation induces the degradation of BRASSINAZOLE RESISTANT 1 (BZR1), the master transcription factor of the brassinosteroid (BR) signaling pathway, thereby inhibiting growth. However, the molecular mechanism linking sugar signaling to BZR1 degradation remains unknown. To identify proteins that mediate starvation-induced BZR1 degradation, we performed a quantitative proteomic analysis of the BZR1 interactome under starvation conditions and identified UBIQUITIN PROTEIN LIGASE 3 (UPL3) as a sugar-regulated protein that promotes BZR1 degradation and regulates growth and survival in response to sugar availability. upl3 mutants showed increased BZR1 accumulation and larger seedling size compared to the wild type under sugar-limiting conditions, but not when grown on sugar-containing medium, which indicates that UPL3 mediates BZR1 degradation and growth inhibition under sugar-limited conditions. Although upl3 mutations promoted growth under short-term starvation, they substantially reduced survival under long-term starvation. The enhanced growth phenotype of upl3 was also observed when target of rapamycin (TOR) was inactivated, but not when BR biosynthesis was blocked, suggesting that UPL3 acts downstream of sugar-TOR signaling to regulate BZR1 degradation. Furthermore, UPL3 protein levels increased post-transcriptionally in response to starvation and TOR inhibition, and decreased upon sugar treatment. Our study identifies UPL3 as a key molecular link between sugar signaling and BR signaling. We propose that sugar-TOR signaling inhibits UPL3 to promote BZR1 accumulation and growth, thereby optimizing plant growth and survival in response to sugar availability.

Abstract :
Plant-associated microbes play a crucial role in host growth, health, and stress resistance. Host plants significantly influence the assembly of their microbial communities. To examine the influence of plant provenance on bacterial composition and plant growth promoting (PGP) properties, we compared readily culturable bacteria isolated from the root interior and rhizosphere of Tripterygium wilfordii Hook f., a woody Chinese medicinal plant, across three provenances grown in different regions. Regardless of the host growing environment, the abundance of endophytic bacterial colonies followed the trend: Fujian provenances > Hunan provenances > Hubei provenances. A total of 227 isolates were classified into 21 genera from the root interior and 40 genera from the rhizosphere, with Pseudomonas and/or Bacillus as the dominant genera. The taxonomic composition varied across plant provenances and host growing environments. Plants from the same provenance but cultivated in different locations exhibited greater similarity in their endophytic bacterial composition. The majority of the assayed strains displayed one or more PGP traits. Local plants harbored a higher proportion of PGP strains than non-local plants. Our findings indicate that plant provenance significantly influences the composition of root-associated culturable bacteria, particularly endophytic communities. Plants in their native environments may recruit PGP bacteria to enhance their fitness.

Deep learning networks have been widely used for vegetation detection in field images to monitor environmental information, but network structures optimized by manual methods require extensive experience and trial and error (T&E). So, this paper proposes an intelligent agent to auto-optimize networks using a Directed Evolutionary Genetic Algorithm (DEGA) and an auto-transfer learning method (FMTPI) for vegetation detection in dry thermal valley. Firstly, the basic conditions to build the agent were determined, showing that the optimal basic conditions were a block type (BT) of ResNetB , an input size (IS) of 753, a intermediate block number (M) of 4, and a kernel size (X) of 5. Secondly, the operation conditions to drive the agent were obtained with the improved FMTPI, showing that the fitting combination of exponential and inverse functions in FMTPI can reduce the time cost by 90.1%. Finally, the agent auto-optimized the pre-trained network, showing that the model size (MS) could be reduced by 87.3% while maintaining a detection accuracy of 92.3%. The agent can auto-optimize networks and improve network efficiency, providing a meaningful reference for vegetation and environment detection.

分析了雷公藤(Tripterygium wilfordii Hook. f.)11株内生真菌的促生潜力,并将11株内生真菌与雷公藤组培苗共生培养,研究了其对组培苗生长、叶片叶绿素含量和叶绿素荧光参数、主要营养元素含量以及雷公藤红素和雷公藤甲素含量的影响。结果表明:雷公藤11株内生真菌均具有分泌吲哚乙酸、产铁载体和溶磷能力,其中,菌株NS6、NS7、NS14和NS32的吲哚乙酸分泌量较大(大于17.00 mg·L~(-1)),菌株NS1、NS4、NS6、NS25和NS33的产铁载体能力很高[80%<铁载体相对产量(SP)≤100%],菌株NS1、NS4、NS6、NS13和NS31溶磷量较大(大于0.70 mg·L~(-1))。与对照相比,菌株NS1、NS6、NS31、NS32和NS33总体显著(P<0.05)增加雷公藤组培苗的单株鲜质量和干质量,其中,菌株NS6、NS32和NS33显著增加组培苗株高,菌株NS1和NS6显著促进组培苗最长根长,菌株NS6显著增加组培苗单株叶片数。与对照相比,除菌株NS13外,其余10株菌株均显著提高组培苗叶片叶绿素含量;菌株NS1、NS7、NS18、NS31和NS32显著提高组培苗叶片最大荧光,菌株NS14显著提高组培苗叶片PSⅡ最大光化学效率。与对照相比,菌株NS1、NS7、NS14、NS31、NS32和NS33显著提高组培苗C、P和K含量,菌株NS1、NS4、NS32和NS33显著提高组培苗N含量。与对照相比,11株内生真菌均能显著提高组培苗根和茎中雷公藤红素含量,仅菌株NS33显著提高叶片中雷公藤红素含量;与11株内生真菌共生培养的组培苗茎和叶片中未检出雷公藤甲素,菌株NS6、NS7、NS13、NS25、NS32和NS33显著提高根中雷公藤甲素含量。综合研究认为,菌株NS1、NS6、NS31、NS32和NS33具有多种促生潜力且能改善雷公藤组培苗生长,促进根和茎中雷公藤红素积累;菌株NS6、NS32和NS33促进根中雷公藤甲素积累,具有较好的应用潜力。

针对有机磷农药废水的处理,本研究通过对牡蛎壳粉末(Oyster shell photocatalyst, OSP)进行TiO_2负载,制备出Zn/TiO_2-牡蛎壳光催化剂(Zn/TiO_2-OSP).在此基础之上,对光催化材料的形貌结构与光学性能进行表征,并探究其对有机磷农药废水的降解性能.结果表明,所制备的Zn/TiO_2-OSP材料检测出TiO_2的衍射峰,并在478 cm~(-1)处出现Ti—O的伸缩振动峰,同时EDS中也检测出Ti的信号,证明了TiO_2的成功负载.此外,对于初始浓度10 mg·L~(-1)的两种有机磷废水,Zn/TiO_2-OSP对敌敌畏的降解效率可达100%,对辛硫磷的降解率达到90.1%;光催化剂降解效果与光照时间、溶液初始浓度及催化剂用量等显著相关,具有较好的光催化活性,且多次回收再利用后仍保持良好的稳定性,降解中间产物对水生环境微生物的毒害效应也较小.另外,两种废水中均未检测出由有机磷降解而生成的磷酸盐,说明Zn/TiO_2-OSP在光催化降解有机磷的同时也能吸附水体中的磷酸盐.

利用光催化与分子印迹技术,制备了一种对微囊藻毒素-LR(MC-LR)具有选择性降解功能的分子印迹型Zn/g-C_3N_4复合光催化材料(MIZn/g-C_3N_4).在此基础之上,对该复合光催化材料的形貌结构与光学性能进行了表征,并对其吸附-降解性能进行了探究.结果表明,分子印迹技术可显著提高光催化材料Zn/g-C_3N_4比表面积,减小其带隙并扩大对可见光的吸收范围,抑制光生载流子复合,进而提升光催化性能.MI-Zn/gC_3N_4对MC-LR的吸附过程可用Langmuir等温吸附模型进行表征,吸附行为属于单层吸附,最大吸附量(Qm)为8.18 mg·g~(-1).而吸附动力学研究则发现,一级和二级动力学模型拟合系数相差不大,表明该吸附体系同时伴随着物理吸附和化学吸附.避光条件下MI-Zn/C_3N_4在60 min内达到吸附平衡阶段,可吸附约75%的MC-LR.可见光照射下MI-Zn/g-C_3N_4降解速率最快,4 h内可将2 mg·L~(-1)的MC-LR全部降解.经过5次循环实验,去除效率仍保持在82.16%,表明MI-Zn/g-C_3N_4具有较好的光催化性能与重复利用能力.·O2-和h+是MI-Zn/g-C_3N_4光催化降解MC-LR的主要活性物质.

Abstract :
本发明涉及涉及微生物技术领域,具体涉及一种菠萝泛菌固态菌剂的保存方法。所述保存方法具体为:将菠萝泛菌固态菌剂与固体载体混合,搅拌均匀,室温条件下静置培养24~48h后,真空密封包装,置于20~25℃干燥处保存;其中,所述固体载体由40重量份麸皮、25重量份玉米芯、15重量份椰壳15和20重量份硅藻土组成。本发明的保存方法操作简便,容易实施,并且能够长期有效保存菠萝泛菌固态菌剂的活力,保本发存12个月后有效活菌数达2.8×1012 cfu/g。利用本发明的保存方法制备的菠萝泛菌菌剂,对雷公藤有明显的促生作用,能够明显提高雷公藤叶绿素含量、甲素含量及生物量,值得大力推广。
