Enhancing Monacolin K and GABA Biosynthesis in Monascus pilosus via GAD Overexpression: Multi-Omics Elucidation of Regulatory Mechanisms SCIE

Abstract :

Monascus produces various bioactive compounds, including monacolin K (MK), gamma-aminobutyric acid (GABA), and Monascus pigments (MPs). Studies have shown that overexpressing genes within the MK biosynthetic cluster significantly enhances MK production. Additionally, MK synthesis in Monascus is regulated by other genes. Based on previous transcriptomic analyses conducted in our laboratory, a significant positive correlation was identified between the expression level of the GAD gene and MK production in M. pilosus. In this study, the GAD gene from M. pilosus was selected for overexpression, and a series of engineered M. pilosus strains were constructed. Among the 20 PCR-positive transformants obtained, 13 strains exhibited MK production increases of 12.84-52.50% compared to the parental strain, while 17 strains showed GABA production increases of 17.47-134.14%. To elucidate the molecular mechanisms underlying the enhanced production of MK and GABA, multi-omics analyses were performed. The results indicated that GAD overexpression likely promotes MK and GABA synthesis in M. pilosus by regulating key genes (e.g., HPD, HGD, and FAH) and metabolites (e.g., alpha-D-ribose-1-phosphate, beta-alanine) involved in pathways such as tyrosine metabolism, phenylalanine metabolism, the pentose phosphate pathway, propanoate metabolism, and beta-alanine metabolism. These findings provide theoretical insights into the regulatory mechanisms of MK and GABA biosynthesis in Monascus and suggest potential strategies for enhancing their production.

Abstract :

为探究枳椇子和余甘子提取物共包埋微胶囊喷雾干燥工艺,改善它们的加工和贮藏特性,采用层次分析-指标相关性赋权系数法(AHP-CRITIC)确定指标权重系数(黄酮包埋率、多酚包埋率、黄酮载药量、多酚载药量),建立微胶囊综合评分体系,采用响应面法优化微胶囊的制备工艺,同时对制备得到的微胶囊颗粒理化特征、结构表征进行综合分析。结果表明,喷雾干燥法制备提取物微胶囊的最佳工艺条件为:芯壁质量比2∶3.5,进风温度162.5℃,壁材配比(麦芽糊精和阿拉伯胶质量比)为1∶1.39,在此条件下的微胶囊综合评分为67.498,颗粒大小均匀,结构完整,粒径为3.833μm。微胶囊性能研究表明,麦芽糊精+阿拉伯胶作为复合壁材能很好地包埋枳椇子和余甘子提取物,提高其热稳定性、防潮性以及贮藏稳定性。

Abstract :

In order to optimize the process of Hovenia dulcis—Phyllanthus emblica extract co—encapsulated microcapsules and improve the processing and storage characteristics of Hovenia dulcis-Phyllanthus emblica extract, the microcapsule was maded by spray drying method. Taking flavonoid embedding rate, polyphenol emhedding rate, flavonoid drug loading and polyphenol drug loading as investigation indexes, the analytic hierarchy process-criteria importance though intercrieria correlation (AHP-CRITIC) method was used to determine the weight coefficients of each index and establish a compre-hensive scoring System for microcapsules. Response surface methodology was used to optimize the preparation conditions of microcapsules. The physicochemical properties and structural characterization of microcapsules were analyzed. The re-sults showed that the Optimum conditions for the preparation of extract microcapsules by spray drying method of core-wall ratio, inlet-air temperature and wall material mass ratio (maltodextrin and gum arabic) were 2:3.5, 162.5 °C and 1:1.39, respectively. Under this condition, the microcapsules had uniform particle size and complete structure, with a particle size of about 3.833 p.m and a comprehensive score of 67.498. The study on microcapsule Performance showed that the wall material compositum of maltodextrin and gum arabic could be a good co—encapsulate on the Hovenia dul-cis-Phyllanthus emblica extractive, and improve the heat stability, moisture resistance and storage stability of the extractive. © 2025 Chinese Institute of Food Science and Technology. Characterization of Key Metabolic Markers in Hongqujiu Across Different Aging Years Using Metabolomics SCIE

Abstract :

Hongqujiu, one of the three principal varieties of yellow wine, is a traditional fermented beverage originating from China that employs Hongqu as the fermentation agent. In this study, an untargeted metabolomics approach based on gas chromatography-mass spectrometry (GC-MS) and liquid chromatography-mass spectrometry (LC-MS) was applied to systematically analyze the volatile compounds (VOCs) and non-volatile compounds (NVCs) in Hongqujiu across different aging years for the first time. The analysis identified a total of 262 VOCs and 2564 NVCs in samples of Hongqujiu aged for six distinct years. Based on metabolic differences, the samples were categorized into two groups: the low-year group (5-year, 6-year) and the high-year group (8-year, 10-year, 15-year, 20-year). Nineteen VOCs (e.g., 4-amino-butyric acid and diethanolamine) and thirty NVCs (e.g., palmitoylethanolamide and erinacine D) were identified as key differential metabolites distinguishing the low-year group from the high-year group. The higher-year group is enriched with a variety of substances with different flavors or biological activities, such as sugar derivatives, amino acids and their complexes, organic acids and their intermediate metabolites, steroids and terpenoid compounds, lipids and their derivatives, nitrogen-containing heterocycles, and aromatic compounds. The accumulation of these substances not only shapes the unique and rich flavor characteristics of aged red rice wine (such as the caramel aroma and umami peptide flavor), but also endows red rice wine with potential health benefits due to the physiological regulatory functions of some active ingredients. This study contributes to a deeper understanding of the composition and dynamic variations in metabolites in Hongqujiu, offering a scientific foundation for identifying aged Hongqujiu and conducting further research to enhance its quality.

不同酒龄红曲酒的抗氧化活性分析

Abstract :

以红曲酒为研究对象,研究不同酒龄对其活性成分和抗氧化能力的影响及其相关性。采用紫外分光光度法检测不同酒龄红曲酒的色价、总酚和总黄酮含量,以总抗氧化能力、羟自由基清除能力、DPPH自由基清除率、超氧化物歧化酶活性评价红曲酒的抗氧化能力。结果表明:20年红曲酒的色价最高;随着酒龄的增加,红曲酒的总酚和总黄酮含量变化趋势不同,其中15年红曲酒的总酚含量最高,而8年的总黄酮含量最高。在不同酒龄红曲酒中,20年的抗氧化能力总体效果最好。相关性分析结果表明,色价与总抗氧化能力和DPPH自由基清除率具有极显著相关性(P<0.01);总黄酮含量与DPPH自由基清除率和超氧化物歧化酶活性具有极显著相关性(P<0.01);总酚含量与超氧化物歧化酶活性无显著相关性(P>0.05),与其他3项抗氧化能力指标呈负相关。

Genome-wide identification and expression analysis of B-box genes during abiotic stress responses in the tea plant SCIE

Abstract :

BBX proteins are zinc-finger transcription factors that regulate light-dependent processes including photomorphogenesis, flowering, and circadian rhythms. While BBX families have been characterized in model plants, their evolutionary constraints and stress-response mechanisms in perennial woody crops remain poorly understood, limiting crop improvement strategies for climate resilience. Here, we show that the tea plant (Camellia sinensis) maintains precisely 32 CsBBX members despite its 20-fold larger genome compared to Arabidopsis, revealing remarkable evolutionary conservation on BBX family expansion. Through comprehensive genome-wide identification, phylogenetic analysis, and expression profiling across eight tissues and four abiotic stresses, we uncovered specialized regulatory modules within the CsBBX family. Tissue-specific analysis revealed distinct expression patterns, including root-specific CsBBX21 and reproductive tissue-enriched CsBBX6-1/2 and CsBBX19-3, indicating functional diversification beyond light responses. Cold stress emerged as the most potent inducer, with CsBBX7-1 showing 40-fold rapid induction, while CsBBX30-1 demonstrated unique droughtspecific accumulation (14-fold). Functional characterization revealed that CsBBX19-3 and CsBBX24 activate the promoter of the stress-responsive transcription factor CsDREB2A, establishing a novel regulatory hierarchy upstream of both DREB2A and DREB1/CBFs-dependent cold tolerance pathways. Our findings establish CsBBX proteins as master regulators integrating light and stress signals, expanding beyond canonical photoperiodic functions. The identification of stress-specialized CsBBX members provides immediate targets for developing climate-resilient tea cultivars through molecular breeding approaches.

Monacolin K (MK),produced by Monascus fermentation, has significant hypolipidemic effects.In this study, under solid-state fermentation (SSF) quinoa was used as substrate to investigate the effect of the MK production byMonascusspecies quinoa. The results of the single factor experimentshowed that the amount of the substrate loaded, addition ofwater to the substrate, and fermentation temperature change time have a significant effect on the MK production of Monascus-fermented quinoa. It was found that neither M. pilosusfermented quinoa norM. ruber-fermented quinoa produced citrinin. Consequently, these three factors were selected for the response surface optimization to determine the optimal fermentation process. The results showed that the optimal fermentation process of M. pilosus-fermented quinoa wasas follows: quinoa loading amount of 30 g/300 ml, addition of 20 ml water to substrate, seed liquid inoculation volume of 10%, cultured at 30 degrees C for 2 days, followed by a temperature change to 25 degrees C and continued cultivation until 12 days. Under this fermentation process, the yield of MK reached 2.51 mg/g and without production of any citrinin. The optimal fermentation process of M. ruber-fermented quinoa was as follows: quinoa loading amount of 31 g/300 ml, water addition of substrate of 20 ml, seed liquid inoculation volume of 20%, cultured at 30 degrees C for 2 days, followed by a temperature change to 25 degrees C and continued cultivation until 12 days. Under this fermentation process, the yield of MK reached 3.22 mg/g and without any citrinin production.

Molecular Mechanism of Mok I Gene Overexpression in Enhancing Monacolin K Production in Monascus pilosus SCIE

Monascus species are capable of producing various active metabolites, including monacolin K (MK) and pigments. Studies have shown that the overexpression of the mok I gene from the MK synthesis gene cluster in Monascus species can significantly increase MK production; however, the molecular mechanism has not yet been fully elucidated. Therefore, this study focused on the mok I gene of Monascus pilosus to construct overexpression strains of the mok I gene, resulting in high-yield MK production. Sixteen positive transformants were obtained, seven of which produced 9.63% to 41.39% more MK than the original strain, with no citrinin detected in any of the transformants. The qRT-PCR results revealed that the expression levels of mok I in the transformed strains TI-13, TI-24, and TI-25 increased by more than 50% compared to the original strain at various fermentation times, with the highest increase being 10.9-fold. Furthermore, multi-omics techniques were used to analyze the molecular mechanisms underlying enhanced MK production in transformed strains. The results indicated that mok I overexpression may enhance MK synthesis in M. pilosus by regulating the expression of key genes (such as MAO, HPD, ACX, and PLC) and the synthesis levels of key metabolites (such as delta-tocopherol and alpha-linolenic acid) in pathways linked to the biosynthesis of cofactors, the biosynthesis of unsaturated fatty acids, tyrosine metabolism, ubiquinone and other terpenoid-quinone biosynthesis, alpha-linolenic acid metabolism, and glycerophospholipid metabolism. These findings provide a theoretical basis for further study of the metabolic regulation of MK in Monascus species and for effectively enhancing their MK production.

Analysis of Targeted Components in the Combination Extract of Hovenia dulcis and Phyllanthus emblica by UPLC-MS/MS SCIE

This study aimed to establish a UPLC-MS/MS method for rapid analysis and identification of target components in the combination extract of Hovenia dulcis and Phyllanthus emblica. The gradient elution was carried out with methanol (A) -0.2% formic acid solution (A) with low rate of 1.0 mL center dot min-1, column temperature of 30 degrees C, detection wavelength of 285 nm. The ESI ion source was used to collect data under the negative ion mode. Through analysis and identification, five target components were inferred, namely gallic acid, dihydromyricetin, corilagine, taxifolin, and ellagic acid in the combination extract. Quantitative analysis was conducted on the combination extract. By utilizing the theory of modern supramolecular technology in traditional Chinese medicine, the functional composite product was obtained by selecting the compound extraction of Hovenia dulcis and Phyllanthus emblica as raw materials, and quantitative analysis was conducted on on the combination extract. This indicates that the co-xtraction of Hovenia dulcis and Phyllanthus emblica not Delivered by Ingenta only simplified the extraction process but also enhances the extraction rate of the flavonoids in Hovenia dulcis. The developed UPLC-MS/MS method is rapid, sensitive, which provides a technical basis for the selection of raw material mass ratios and product development of Hovenia dulcis and Phyllanthus emblica combination.
