Antifungal activity and action mechanism of Bacillus velezensis against Phyllosticta capitalensis isolated from guava fruits SCIE

Abstract :

The antifungal mechanism of Bacillus velezensis (B. velezensis) against Phyllosticta capitalensis (P. capitalensis), a pathogen isolated from guava fruits and its efficacy in controlling guava black spot disease were systematically investigated. The minimum inhibitory concentration (MIC) of B. velezensis against P. capitalensis was identified to be 10(4) CFU/mL. Microscopic observations revealed significant morphological alterations in P. capitalensis mycelium following B. velezensis treatment, suggesting compromised cell membrane integrity and a consequent increase in membrane permeability. Moreover, B. velezensis treatment induced elevated levels of malondialdehyde (MDA), hydrogen peroxide (H2O2) and superoxide anions (O-2 center dot(-)), accompanied by reduced catalase (CAT) and peroxidase (POD) activities in P. capitalensis, thereby effectively inhibiting its growth. Transcriptome analysis provided further evidence that B. velezensis might exert its antifungal effects through destabilizing the cell membrane and attenuating the antioxidant defense capacity of P. capitalensis. B. velezensis treatment significantly slowed the disease development caused by P. capitalensis in guavas, with diameters measuring only 0.097 cm at 4 days and 0.423 cm at 8 days, which substantially lower than those of the CK group (P < 0.01). These findings indicate that B. velezensis holds considerable potential as an effective biocontrol agent for managing guava black spot disease.

Abstract :

Phyllosticta capitalensis (P. capitalensis) is a major postharvest pathogen of guava, causing black spot disease, which results in serious rot and significant economic losses. This study demonstrated that the sterile fermentation supernatant of Bacillus velezensis (BT313-SFS) effectively inhibited P. capitalensis, with a minimum inhibitory concentration (MIC) of 300 mL/L. BT313-SFS maintained stable antifungal activity across a pH range of 4-8 and temperatures between 25 and 60 degrees C. However, its antifungal efficacy was significantly reduced by treatment with trypsin and alkaline protease. Cellular-level analysis revealed that BT313-SFS altered the morphology of P. capitalensis, destroyed the cell wall and membrane, suppressed intracellular antioxidant enzyme activities, raised reactive oxygen species (ROS) levels, and induced oxidative cellular damage. Furthermore, BT313-SFS impaired mitochondrial function and disrupted the tricarboxylic acid (TCA) cycle, interfering with cellular energy metabolism and inhibiting P. capitalensis proliferation. Thus, BT313-SFS exhibited excellent preservation effects by reducing guava respiration, retarding color browning, and maintaining cell membrane integrity, thereby extending the storage life of the fruit. These results established BT313-SFS as a promising natural antifungal agent for controlling postharvest black spot disease in guava.

Abstract :

Novel covalent organic frameworks (COFs) based PAN@TpBD(NH2)(2) electrospun composite nanofiber mem-branes (ECNMs) were fabricated as strong anion exchange sorbent by implementing electrospinning technology. The finished sorbent was characterized, and key parameters of pipette-tip solid phase extraction (PTSPE) pro-cedures were investigated. Inorganic arsenic (iAs) was successfully separated from rice under the optimal precondition conditions, and quantified by hydride generation-atomic fluorescence spectrometry (HG-AFS). This PTSPE-HG-AFS methodology achieved 0.015 mu g L-1 detection limit, 4.67 % relative standard deviation, and 86.48 similar to 99.11 % recoveries. In this work, preparation and characterization of this novel COFs-based anion exchange sorbent, PAN@TpBD(NH2)(2) ECNMs, is described and its suitability for PTSPE applications is demonstrated.

This study focused on the development of a gaseous ClO2 treatment system that contained dry ice that was used to form carbon dioxide (CO2) and an absorbent pad with sodium chlorite (NaClO2) solution for ClO2 gas gen-eration. Furthermore, the effects of ClO2 gas on the populations of Listeria monocytogenes and the quality properties of table grapes during cold storage were evaluated. The system continuously released the sufficient concentrations of ClO2 gas to treat berries during 30 d of cold storage. After 20 h of ClO2 gas treatment, the populations of Listeria monocytogenes decreased from 5.88 log CFU/g to a non-detectable (ND) level. There were no significant (p > 0.05) changes in the color parameters (L*, a*, and b*) of table grapes treated with ClO2 gas. The firmness, weight, total soluble solids (TSS), and titratable acid (TA) losses of table grapes were significantly (p < 0.05) reduced by ClO2 gas, while Vitamin C was unaffected. Additionally, the residual ClO2 of table grapes were <0.5 mg/kg, which is considered a safe level. Overall, the novel gaseous ClO2 treatment system may be used as an effective intervention technology to improve the safety and quality of table grapes during cold storage.

一株引起番石榴黑斑病病原菌的分离与鉴定

目的:旨在为明确番石榴黑斑病优势病原菌的种类及其特性提供科学依据。方法:从受黑斑病病害侵染的番石榴果实上分离纯化出一株优势病原菌,命名为S1,并对其进行形态学观察、致病性测定、生长特性研究及聚合酶链式反应(Polymerase Chain Reaction,PCR)测序比对分析,较为全面的对分离出的优势病原菌S1进行鉴定。结果:由形态学观察可知,S1菌落的气生菌丝蓬松且致密,颜色呈墨绿色或黑色,其孢子形态为单孢且透明,大小均匀,约为(6~10)μm×(6~7)μm,呈圆形或椭圆形,与《真菌鉴定手册》比对,初步判定其属叶点霉属菌,将其反接于健康番石榴果实上进行致病性测定。结果表明,S1为致使番石榴产生黑斑病的主导优势病原菌。通过生长特性研究表明,S1在偏碱性、黑暗、常温条件下较为适宜生长,结合PCR测序结果,确定出致使番石榴黑斑病的优势病原菌S1为首都叶点霉(Phyllosticta capitalensis)。结论:引起番石榴黑斑病的优势病原菌为首都叶点霉。

目的将聚乙烯亚胺(polyethyleneimine,PEI)修饰于氧化石墨烯(graphene oxide,GO)表面,制备PEI@GO合成材料以实现乌龙茶叶中无机砷的选择性吸附分离,并结合原子荧光光谱法对其进行检测。方法利用傅里叶红外光谱、电镜扫描、Zeta电位分析对PEI@GO进行表征,并建立基于PEI@GO的管尖固相萃取(pipette tip solid phase extraction,PTSPE)体系结合氢化物发生-原子荧光光谱法(hydride generation atomic fluorescence spectrometry,HG-AFS)实现乌龙茶中无机砷的选择性吸附分离和准确定量。结果傅里叶红外光谱图中出现N-H特征峰,表明PEI成功修饰于GO的表面,电镜下PEI@GO呈现多孔的山脊山谷结构,Zeta电位分析其等电点约为9.7,在p H 6下,5 mg PEI@GO对As (V)溶液吸附率达95.84%,所建立的PTSPE-HG-AFS法最低检出限为0.027μg/L,相对标准偏差为0.48%,不同品种乌龙茶样品中无机砷含量为13.00~122.00μg/kg,回收率在86.48%~108.98%之间。结论制备的PEI@GO材料可有效实现无机砷的选择性吸附分离,所建立的方法操作简便、灵敏度高,可准确用于实际茶叶样品中无机砷的分析检测。

Abstract :

Background and objectives Rice contains an order of magnitude higher inorganic arsenic (iAs), a class 1 non-threshold carcinogen, than other terrestrial crops. To reduce iAs in cooked rice, several methods have been proposed such as washing until clear, cooking with high water-to-rice ratio (W/R) then discarding excess water, or continuously percolating near-boiling water. The traditional method practiced by 1.6 billion East Asians, washing and cooking with low W/R until dry, removes only a small percentage of iAs. A new approach was investigated to reduce iAs in cooked rice. Findings Brief (10 min) soaking (W/R 10) at above gelatinization temperature (80 degrees C) reduced iAs in rice by up to 48.4%. A soaking protocol was developed for public use that on average reduced 39.5 +/- 6.6% (n = 12) iAs in non-waxy white rice. Conclusions Brief above-gelatinization-temperature soaking effectively reduces iAs in cooked white rice. Significance and novelty Above-gelatinization-temperature soaking was applied for the first time to cut iAs in cooked rice; thereby chronic iAs exposure and cancer risk can be reduced for rice consumers.

山茶油微胶囊的复合凝聚制备工艺参数及其热氧稳定性研究 CSCD

以明胶和阿拉伯胶为壁材,茶多酚为固化剂,采用复合凝聚法制备山茶油微胶囊。通过单因素和正交试验考察壁材比、pH、壁材浓度、凝聚时间、芯壁比等复合凝聚条件对山茶油微胶囊包埋率和微观形态的影响,确定山茶油微胶囊复合凝聚的最佳制备工艺,并对微胶囊化前后的山茶油氧化稳定性进行分析比较。结果表明:山茶油微胶囊的最佳复合凝聚制备工艺参数为壁材比1∶1 (W/W)、pH 4.2、壁材浓度1.0%、凝聚时间30 min、芯壁比3∶2 (W/W),在此最佳条件下制备的山茶油微胶囊产品呈浅棕色粉末状,平均包埋率达82.36%,休止角39.2°,含水率3.67%。热稳定温度280℃,具有良好的流动性和热稳定性。同时,加速氧化贮藏试验表明微胶囊化显著提高了山茶油的氧化稳定性。

乌饭果花色苷复合酶法提取工艺优化及其抗氧化活性

以乌饭果花色苷含量为指标,采用二次回归正交旋转组合设计优化乌饭果花色苷的复合酶法提取工艺,并分析评价其体外抗氧化活性。结果表明:乌饭果花色苷复合酶法提取的最佳工艺条件为加酶量234 U·g~(-1)原料,纤维素酶:果胶酶配比2∶1,pH4.0,料液比1∶30 g·mL~(-1),酶解温度50℃,酶解时间180 min,在此条件下测得乌饭果花色苷的含量最高,为136.08 mg·100 g~(-1)。同时,体外抗氧化活性研究表明,乌饭果花色苷对·OH、O~-_2·清除能力和抗脂质过氧化能力的半抑制浓度IC_(50)分别为92.23、23.93和303.1μg·mL~(-1),与同质量浓度抗坏血酸相比,其·OH、O~-_2·清除能力弱于抗坏血酸,但抗脂质过氧化能力优于抗坏血酸。

目的筛选乌饭树果乙醇提取物中抗氧化活性最强的极性分部。方法采用石油醚、乙酸乙酯、氯仿、正丁醇和水等5种不同极性溶剂对乌饭树果乙醇提取物进行进一步梯度萃取,并分析比较其总抗氧化能力(total antioxidant capacity,T_(AC))、抗脂质过氧化能力及对羟基自由基OH·、超氧阴离子自由基O_2~-·的清除效果。结果乌饭树果乙醇提取物的5个不同极性分部均具有一定的抗氧化活性,但其活性大小差异极显著(P<0.01),T_(AC)大小依次为:乙酸乙酯部>氯仿部>正丁醇部>石油醚部>水部。同时,在OH·、O_2~-·自由基清除能力和抗脂质过氧化能力上,乙酸乙酯部均优于氯仿部和正丁醇部,且呈明显的量效关系。结论乌饭树果乙醇提取物的乙酸乙酯部具有最强的抗氧化活性,且对OH·、O_2~-·的清除能力及抗脂质过氧化能力均强于L-抗坏血酸及其它部位,可作为天然抗氧化剂的潜在有效来源。
