Abstract :
Low molecular weight polysaccharides had higher bio-activity and bioavailability compared to ultra-high molecular weight polysaccharides, this study aimed to obtain low molecular weight polysaccharides from Tremella fuciformis (TFLP) by using high-temperature and high-pressure assisted hydrochloric acid method to degrade Tremella fuciformis polysaccharides (TFP), and the structural characteristics, in vivo antioxidant and immune enhancing activities of TFP and TFLP was explored through Caenorhabditis elegans (C. elegans) and mice model. It was found that TFP and TFLP were acidic polysaccharides with molecular weights of 2238 kDa and 3 kDa, respectively. The glycosidic bonding of TFP and TFLP was mainly composed of different configurations of mannopyranose. TFP and TFLP had excellent in vivo antioxidant activity and stress resistance by regulating the mRNA transcription level and metabolites in C. elegans. Results also showed that TFP and TFLP could enhance the antioxidant capacity and immunity of serum, spleen and small intestine tissues in normal mice and cyclophosphamide-induced immunosuppressive mice through regulating the relative transcription and expression levels of anti-inflammatory related signaling factors, and it has found that TFLP showed better immune enhancement and antioxidant activity than TFP. In addition, Akkermansia, Bacteroides and Alloprevotella were characteristic bacteria at the genus level in immunosuppressed mice intervened with TFLP, with a significant increase in relative abundance. The content of SCFAs significantly increased in immunosuppressed mice by TFLP. These results indicated that TFP and TFLP had potential in vivo antioxidant and immune enhancing activities.

银耳富含营养素,银耳多糖是其中最主要的功能活性成分,具有多种生物活性。银耳多糖的制备方法多样,对于其结构的研究多集中在分子量、单糖组分和糖苷键类型等方面。本文综述了银耳多糖的制备、结构和抗氧化、抗肿瘤、调节免疫、改善记忆、抗炎、降血糖和降血脂等生物活性及其在食品、化妆品和医药方面的开发应用。未来需要优化银耳多糖的制备技术,结合现代分析技术解析银耳多糖的高级结构,对银耳多糖的功能及作用机理进行深入的研究,以期为银耳的精深加工与产品研发提供理论参考。

Abstract :
Different methods were used to degrade Tremella fuciformis polysaccharides (TFP) and prepare low molecular weight polysaccharides of Tremella fuciformis (TFLP) to improve their bioavailability. It was found that the TFLP prepared by ultrasonic-assisted H2O2-Vc method showed the highest level of antioxidant activity and stress resistance in C. elegans. The structural characteristics, in vivo antioxidant and stress resistance of TFLP-1 were evaluated after isolation and purification of TFLP, it was found that TFLP-1 was an acid polysaccharide with a molecular weight of 75770 Da, which mainly composed of mannose. Meanwhile, it could regulate the antioxi-dant activity and stress resistance in C. elegans by upregulating the transcription of fat-5, fat-7, acs-2, glp-1, hsf-1, hsp-1, mtl-1, nhr-49, skn-1 and sod-3 mRNA. The improvement effects were closely related to the significant regulation of galactose metabolism, alpha linolenic acid metabolism, and pantothenate and CoA biosynthesis metabolic pathways. These results provided insights into the high value application of Tremella fuciformis in the food industry and the development of antioxidant related functional foods.

Covalent organic frameworks (COFs) have attracted considerable attention in sample pretreatment because of their unique characteristics. However, the submicron or micron size of COFs has restricted their wider applications in solid-phase extraction (SPE). Herein, multiwalled nanotubes (MWNTs) were used as substrate materials to synthesize core-shell structured MWNTs@COFs composites (MWNTs@SNW-1) using a simple self-assembly method. The as-prepared MWNTs@SNW-1 composite exhibited a high BET surface area, good thermal stability, and good adsorption capacity. The MWNTs@SNW-1 composite was used as an adsorbent in cartridge-based SPE to extract four phytohormones before determining their levels by high-performance liquid chromatography. The experimental parameters affecting extraction efficiency, including the amount of adsorbents, solution pH, ionic strength, eluent type, and eluent volume, were investigated. The developed method showed a wide linear range (0.37-100 ng mL(-1)), low detection limits (0.11-0.32 ng mL(-1)), low limits of quantification (0.37-1.07 ng mL (-1)), high enrichment factors (45.9-49.3), and good reproducibility (< 4.8%) for phytohormones. The developed analytical method was used to analyze trace phytohormones in fruit juices with good recoveries, highlighting the potential of the MWNTs@SNW-1 composite as an adsorbent in sample preparation. 为探究石莼醇提物基于细胞膜损伤的抑菌作用,检测了其对金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌、沙门氏菌的抑菌性能,测定了不同质量浓度石莼醇提物对供试菌生长、核酸和蛋白质的泄露量、培养液电导率的影响,使用流式细胞仪、扫描电镜、透射电镜观察金黄色葡萄球菌菌体损伤率和超微结构的变化.结果表明:石莼醇提物可显著抑制金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌、沙门氏菌的正常生长,尤以对金黄色葡萄球菌的抑制作用最强,其最低抑制浓度(MIC)、最低杀菌浓度(MBC)分别为1.56、3.13 mg·mL~(-1);2×MIC的石莼醇提物可增大菌体细胞膜通透性,导致菌体内核酸、蛋白质的大量泄露,培养液的电导率极显著上升(P<0.01),菌体细胞膜严重萎缩甚至裂解,菌体损伤率可达96.42%.综上,石莼醇提物通过破坏细胞膜的完整性达到抑菌效果.

抗生素的耐药性如今已成为人类健康的巨大威胁,是全球关注的焦点问题。针对抗生素在水产养殖环境及水产中泛滥的现状,该文归纳了抗生素在水产养殖业中的应用现状、抗生素耐药性及其公共健康危害,分析了水产供应链中耐药细菌和耐药基因研究现状及耐药性传播风险,以期对水产业中抗生素耐药性危害的评估和削减提供新视角。

为提高丹桂干花的贮存稳定性,探索适宜丹桂干花贮存保质的安全、绿色的复合脱氧干燥剂,并确定丹桂干花产品的有效包装方式,本研究以丹桂干花的色度饱和值(Cab值)、菌落总数等为指标,结合肉眼观察,探讨复合脱氧干燥剂与聚乙烯塑料材质、不透明镀铝膜等不同包装方式对丹桂干花贮藏品质的影响。结果表明,采用单层不透明镀铝膜袋包装,并辅以理想的复合脱氧干燥剂(纤维干燥剂:葡萄糖氧化酶质量比=10:1),丹桂干花在干燥、室温下贮存12个月,可以保持色泽金黄,其Cab值为98.20±0.14,菌落总数3.16×103 CFU/g,其外观色泽与微生物指标仍维持在较理想水平。综上,采用纤维干燥剂与葡萄糖氧化酶以10:1为复合比组成的脱氧干燥剂与单层不透明镀铝膜袋包装能显著提升丹桂干花贮藏质量。

本研究基于TPA质构测定和感官评定,单因素探讨冷冻淮山紫薯球外表皮的生产工艺配方,并通过因子分析与响应面法优化其关键工艺配比,以提高冷冻淮山紫薯球的冻藏品质。结果显示,冷冻淮山紫薯球外表皮的主要工艺配比的优化组合为:紫薯添加量52%,小麦淀粉量5%,泡打粉添加量0.16%;结合单因素实验结果白糖添加量为10%,植物油使用量为4%。以上述工艺配方进行验证试验,加工出的冷冻淮山紫薯球产品品质优良,其规范化综合因子得分为0.778,感官评分49,平均硬度866.36 g、弹性0.66、咀嚼性144.83 g。

In the current study, a novel covalent organic frameworks COF-SCU1 incorporated electrospun nanofibers (PAN@COF-SCU1 nanofibers) was fabricated via a facile electrospinning method and utilized as adsorbent in pipette tip solid-phase extraction (PT-SPE) of tetracycline antibiotics (TCs) from foods. The prepared PAN@COF-SCU1 nanofibers possessed both of the unique characteristics of electrospun nanofibers and COFS-CU1, and thus improving the adsorption capacity of the electrospun nanofibers and preventing the problems of leakage and high pressure caused by directly using the nanosize COFs as adsorbent in PT-SPE. The experiments affected the adsorption and desorption efficiencies, such as the loading ratios of COFS-CU1 in nanofibers, the amount of nanofibers, the matrix pH and desorption solvent, were studied in detail. Eventually, a new pipette tip solid-phase extraction-high performance liquid chromatography (PT-SPE/HPLC) method was proposed for the analysis of three TCs from food. Satisfied linearity for TCs was obtained in the range of 4-70 ng mL(-1). The limits of detection and quantification were ranged from 0.6 to 3 ng mL(-1) and from 2 to 10 ng mL(-1), respectively. The interday and intraday precisions (RSD) were all lower than 9%. The proposed PT-SPE/HPLC method was used to determine TCs residues in grass carp and duck samples for the first time. The results could not only explore the availability of PT-SPE in the extraction of TCs in food samples, but also broadened the potential applications of COFs in sample preparation.
