Structure, Function and Application of Tremella Fuciformis Polysaccharide: A Review SCIE

Abstract :

Tremella fuciformis (TF) is a valuable edible fungus rich in various nutrients, with TF polysaccharide (TFP) as its primary bioactive component. Extensive studies have demonstrated that TFP exhibits significant bioactivities, including antioxidant, antitumor, immunomodulatory, memory-enhancing, anti-inflammatory, hypoglycemic, and hypolipidemic effects, as well as skin-protective properties and gut microbiota modulation. Due to these multifunctional properties, TFP has garnered considerable attention in the food, pharmaceutical, and cosmetic industries. This review summarizes recent research on TFP, focusing on its preparation methods, structural characteristics, and diverse biological activities. Additionally, it provides a theoretical basis for the potential applications of TFP in the food, pharmaceutical, and cosmetic industries, while also discussing future research directions and development prospects.

Abstract :

High blood sugar is the main feature of diabetes. In order to effectively regulate blood glucose levels, in this study, we used Gracilaria as raw material and found that 70% ethanol extract of Gracilaria (GE) and complex protease hydrolysate of Gracilaria (GH) were beneficial for decreasing fasting blood glucose, improving insulin resistance, enhancing the function of islet beta-cells, and regulating the abnormal increase of TC, TG, LDL-c caused by hyperglycemia in diabetic mice. Moreover, 70% ethanol extract of Gracilaria (GE) and complex protease hydrolysate of Gracilaria (GH) affected the gut microbiota structure of diabetic mice, decreased the Firmicutes/ Bacteroidetes ratio, increased the abundance of Lactobacillus and Collinsella etc. and reduced the abundance of Helicobacter and Oscillibacter etc. In addition, the content of acetic acid and butyric acid in metabolites of shortchain fatty acids had increased, which had the effect of regulating blood glucose level. The results have showed that 70% ethanol extract of Gracilaria (GE) and complex protease hydrolysate of Gracilaria (GH) are potential marine foods for maintaining blood sugar health.

灰树花醇提物化学成分及其生物活性研究

Abstract :

为探明灰树花发挥活性作用的化学成分及其抗氧化活性和对肿瘤细胞的抑制作用,该文分析了灰树花醇提物不同极性溶剂萃取物的主要化学成分及其对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、2,2’-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)自由基和羟自由基的清除能力,以及对肝癌细胞(HepG2)和肺癌细胞(A549)的抑制作用。结果显示,乙酸乙酯萃取物和正丁醇萃取物的抗氧化活性最佳,总酚和黄酮的质量分数与抗氧化活性显著正相关(P<0.05);石油醚萃取物和乙酸乙酯萃取物对肿瘤细胞抑制效果最好,三萜和甾醇质量分数与肿瘤细胞抑制作用极显著正相关(P<0.01)。利用超高效液相色谱质谱联用技术(UPLC-MS)从灰树花醇提物中共鉴定出15个化合物,其中芹菜素和表儿茶素可能是灰树花发挥抗氧化作用的主要化学成分,麦角甾醇和过氧化麦角甾醇可能是发挥肿瘤细胞抑制作用的甾体类化合物。该研究结果可为灰树花作为功能食品进一步开发提供理论依据。

Abstract :

Grifola frondosa, an edible and medicinal resource, is widely used as functional foods worldwide. To explore bioactive compounds against alpha-glucosidase, human tumor cells and enterovirus 71 (EV71), eight compounds were isolated from G. frondosa by chromatographic column. Among the isolated compounds, heptadecanoic acid, uridine and adenosine exhibited potent inhibition activity against alpha-glucosidase, ergosterols and ergosterol-5,8-peroxide showed anti-proliferative activity on tumor cells, while ergosterol and methyl linoleate displayed inhibition against the replication of EV71. Also, to our knowledge, this is the first study to report that fatty acids isolated from G. frondosa show potent inhibition against alpha-glucosidase and EV71. Further molecular docking results revealed that the active compounds in G. frondosa form hydrogen bonding, hydrophobic interactive and pi-stacking with the active sites on the surface of alpha-glucosidase, CASP3 and VP1 proteins, thus promoting the active compounds to combine with the target protein to form a stable complex, thus playing an antagonistic role. Our results could provide a new active compound and mode of action for G. frondosa to treat diabetes, cancer and EV71-infected patients.

灵芝复方对2型糖尿病小鼠的抗糖尿病潜力

本研究旨在探讨灵芝复方对2型糖尿病小鼠的抗糖尿病潜力。灵芝复方(Ganodermalucidumfunctional formulations, GFF)由灵芝提取物、蛋白核小球藻提取物、人参提取物和富铬酵母构成。通过使用50 mg/kg链脲佐菌素对小鼠进行2型糖尿病造模,使用GFF进行灌胃。处死小鼠后,测定小鼠的血清和肝脏生化指标,计算小鼠的胰岛功能系数和观察小鼠的脏器组织形态,对小鼠的盲肠内容物进行16SrRNA基因高通量测序,测定盲肠内容物中的短链脂肪酸含量,测定肝脏中与糖脂代谢相关基因的m RNA转录水平,并用Western Blot (WB)实验进行验证。在这项研究中,与2型糖尿病小鼠相比,GFF可能通过以下几种方式表达抗糖尿病潜力:调节血清和肝脏生化指标(包括总胆固醇,甘油三酯,低密度脂蛋白胆固醇和高密度脂蛋白胆固醇);促进炎症因子IL-2表达,抑制IL-6表达;降低胰岛素抵抗,恢复β细胞功能;减少肝细胞中脂肪堆积,减少肝细胞间距,减少肠道通透性,增加胰岛细胞大小;恢复肠道菌群结构,增加厚壁菌和拟杆菌的比例;促进肠道中短链脂肪酸合成,丁酸含量约增加10~12倍,乙酸、丙酸和戊酸含量约增加2~3倍;上调IRS、PI3K、GLUT4、AKT、m TOR、AMPK-α、IL-2R和FOXP3基因表达,下调GSK-3β、SREBP-1c、ACC、FAS、SCD1、NF-κB和STAT5基因表达。将各指标与富集的肠道菌群进行关联性分析后发现,治疗组小鼠肠道中富集的Acinetobacter、Vagococcus、Klebsiella、Proteus和Weissella等菌群与短链脂肪酸、IL-2、总胆固醇、高密度脂蛋白胆固醇、胰岛素抵抗水平和血糖值曲线下面积呈正相关,与IL-6呈负相关,说明肠道菌群与糖脂代谢紊乱的相关指标密切相关。总而言之,本研究首次证明了GFF可能通过改善各种生化指标,修复失衡的肠道菌群,促进肠道中短链脂肪酸的合成,调节PI3K/AKT等糖脂代谢相关通路上的基因表达等来维持2型糖尿病小鼠的血糖稳态,从而体现出一定的抗糖尿病水平。后续实验将对GFF进行纯化,寻求其中特有的降血糖物质。

麦角甾醇抗EV71型肠道病毒的研究

Abstract :

为探寻灰树花中治疗手足口病的潜在活性成分。利用硅胶柱层析、薄层色谱、半制备液相色谱从灰树花中分离得到8个化合物,使用CCK-8法研究各化合物对手足口病的主要病原体肠道病毒71型(enterovirus 71,EV71)的抑制作用。结果显示与阳性药物利巴韦林(TC_(50)=694.23±37.83μg/mL, IC_(50)=110.79±9.57μg/mL, TI=6.27)相比,麦角甾醇的半数毒性浓度(TC50=898.90±35.62μg/mL)更大(P<0.01),抗EV71活性(IC_(50)=28.24±2.58μg/mL)更强(P<0.01),抗病毒指数(TI=31.80)更高。实时荧光定量PCR表明50μg/mL的麦角甾醇就能够有效抑制EV71病毒衣壳蛋白VP1的mRNA表达量(P<0.01)。进一步的分子对接技术显示麦角甾醇与VP1有较好的结合活性(Score值为-6.21 kcal/mol),麦角甾醇主要通过谷氨酸、异亮氨酸、缬氨酸、丙氨酸、色氨酸和脯氨酸位点与VP1进行连接,从而阻止EV71进入宿主细胞。以上结果表明灰树花中的麦角甾醇具有比利巴韦林更高的安全性以及更强的抗EV71活性,低剂量的麦角甾醇能够有效抑制病毒的复制,同时可通过与病毒衣壳蛋白相结合的方式保护宿主细胞免受病毒侵染。本次研究提示麦角甾醇可作为治疗手足口病的潜在药物,同时为灰树花作为保健食品和功能性食品的高值化开发提供理论基础。

Introduction: Luteolin is a plant-derived flavonoid that exhibits a broad range of pharmacological activ-ities. Studies on luteolin have mainly focused on its use for hyperlipidaemia prevention, whereas the capacity of the flavonoid to hinder hyperglycaemia development remains underexplored.Objectives: To probe the anti-hyperglycemic mechanism of 6,8-guanidyl luteolin quinone-chromium coordination (GLQ.Cr), and to assess its regulatory effect on intestinal microbiota in type 2 diabetes mel-litus (T2DM) mice.Methods: High-sucrose/high-fat diet-induced and intraperitoneal injection of streptozotocin was used to develop a T2DM model. Glycometabolism related indicators, histopathology, and gut microbiota compo-sition in caecum samples were evaluated, and RNA sequencing (RNA-seq) of liver samples was con-ducted. Faecal microbiota transplantation (FMT) was further used to verify the anti-hyperglycemic activity of intestinal microbiota.Results: The administration of GLQ.Cr alleviated hyperglycaemia symptoms by improving liver and pan-creatic functions and modulating gut microbe communities (Lactobacillus, Alistipes, Parabacteroides, Lachnoclostridium, and Desulfovibrio). RNA-seq analysis showed that GLQ.Cr mainly affected the peroxi-some proliferative activated receptor (PPAR) signalling pathway in order to regulate abnormal glucose metabolism. FMT significantly modulated the abundance of Lactobacillus, Alloprevotella, Alistipes, Bacteroides, Ruminiclostridium, Brevundimonas and Pseudomonas in the caecum to balance blood glucose levels and counteract T2DM mice inflammation.Conclusion: GLQ.Cr improved the abnormal glucose metabolism in T2DM mice by regulating the PPAR signalling pathway and modulating intestinal microbial composition. FMT can improve the intestinal microecology of the recipient and in turn ameliorate the symptoms of T2DM-induced hyperglycaemia. (c) 2023 The Authors. Abstract :

This study was aimed at investigating the hypoglycemic effect of Chlorella pyrenoidosa functional formulations (CFF), composed of extracts of C. pyrenoudosa, Ganoderma lucidum, and Panax ginseng with chromium-enriched yeast, on streptozotocin (STZ)-induced high-glucose and high-fat diet-fed mice. In this study, CFF significantly reduced the symptoms of weight loss, liver deformation, cecum injury, and jejunal inflammation in type 2 diabetic mice. In addition to enriching gut probiotics, CFF inhibited harmful microorganisms that could affect glycolipid metabolism disorders (GLD) and enhanced production of short-chain fatty acids. In conclusion, CFF may be able to alleviate GLD symptoms. The study suggests that CFF regulates GLD and the possible mechanisms for this regulation. This provides a theoretical basis for developing nutritious and healthy foods.

ScopeTo investigate anti-aging effects of probiotic-fermented kelp enzymatic hydrolysate culture (KMF), probiotic-fermented kelp enzymatic hydrolysate supernatant (KMFS), and probiotic-fermented kelp enzymatic hydrolysate bacteria suspension (KMFP) in D-galactose-induced aging mice. Methods and resultsThe study uses a probiotic-mixture of Lactobacillus reuteri, Pediococcus pentosaceus, and Lactobacillus acidophilus strains for kelp fermentation. KMF, KMFS, and KMFP prevent D-galactose-induced elevation of malondialdehyde levels in serum and brain tissue of aging mice, and they increase superoxide dismutase and catalase levels and total antioxidant capacity. Furthermore, they improve the cell structure of mouse brain, liver, and intestinal tissue. Compared with the model control group, the KMF, KMFS, and KMFP treatments regulate mRNA and protein levels of genes associated with aging, the concentrations of acetic acid, propionic acid, and butyric acid in the three treatment groups are more than 1.4-, 1.3-, and 1.2-fold increased, respectively. Furthermore, the treatments affect the gut microbiota community structures. ConclusionsThese results suggest that KMF, KMFS, and KMFP can modulate gut microbiota imbalances and positively affect aging-related genes to achieve anti-aging effects.

Abstract :

为了探明灰树花的降血糖与抗病毒作用,以及发挥活性作用的潜在活性物质,采用70%乙醇超声提取灰树花,依次用石油醚、乙酸乙酯、正丁醇3种不同极性溶剂对其提取物进行分步萃取,分别得到石油醚萃取物、乙酸乙酯萃取物、正丁醇萃取物和水溶物,测定其对α-葡萄糖苷酶和肠道病毒71型(EV71)的抑制作用.结果显示正丁醇萃取物对α-葡萄糖苷酶和EV71的抑制作用强于其他萃取物,其半抑制浓度分别为(174.12±10.67)、(84.41±5.12)μg·mL~(-1).利用超高效液相色谱质谱联用和分子对接技术发现,正丁醇萃取物中含量较高(11.58%)的亚油酸甲酯可通过与α-葡萄糖苷酶和EV71表面活性位点相结合而发挥活性作用.
