Abstract :
Probiotics exert a favorable regulatory effect on human intestinal health. However, probiotics face challenges in overcoming the highly acidic environment of the stomach and effectively reaching the intestine, which limit their beneficial effects and bioavailability. We developed a pH-responsive probiotic embedding gel carrier, where the internal Tremella polysaccharide (TMP) and Ca2+ were cross-linked to create a porous and isolated water microenvironment for probiotics. The introduction of sodium alginate and TMP not only enhanced the mechanical properties of the gel but also contributed to the formation of an outer layer structure with pH response. The gel system had good gel properties, could be shaped freely, and protected Lactobacillus rhamnosus from stomach acid and bile salts. The in vitro simulation experiments demonstrated that the release rate of carrierprotected probiotics was found to be less than 1.99% in simulated gastric fluid, whereas nearly complete release occurred in simulated intestinal fluid, with a release rate over 95%. The survival rate of probiotics in simulated gastric and intestinal fluids could be enhanced by hydrogel carriers. Simultaneously, the carrier could enhance the resistance of probiotics to bile and antibiotic. The results showed that Alg-Ca2+-4 parts per thousand TMP gel carrier was an ideal material for probiotic delivery.

为改善鲜切芋艿贮藏期间的感官品质,以永定‘六月红’芋艿为试材,探究不同贮藏温度对L-抗坏血酸(L-ascorbic acid, AA)联合超声(ultrasound, US)处理鲜切芋艿品质及货架期的影响。采用质量分数2%的AA溶液浸泡试材15 min后,进行US处理,分析贮藏期间4个不同贮藏温度(4、9、15、25℃)对处理鲜切芋艿品质的影响,并据此建立鲜切芋艿的货架期预测模型。结果表明:经AA联合US处理的鲜切芋艿在4℃低温下能明显延缓品质劣变,主要体现在抑制褐变度和失重率增加,减缓硬度和pH值下降,保持细胞膜完整性和较高的感官评分,延长贮藏期等方面。通过动力学模型结合Arrhenius方程对芋艿货架期进行预测,其实测值与预测值之间的相对误差均在±10%以内,所建模型可有效预测鲜切芋艿在4~25℃温度范围内的货架期。综上,经AA联合US处理后的鲜切芋艿最佳贮藏温度为4℃,其在该条件下的货架期为20 d,研究结果为鲜切芋艿的贮藏保鲜和销售提供了参考。

Abstract :
Browning is the primary factor influencing the storage quality of Lyophyllum decastes fruiting bodies. Prior research has demonstrated that US treatment can enhance the storage quality of L. decastes by modulating amino acid consumption, redox balance, and energy metabolism post-harvest. This study identified differentially expressed genes across various storage periods of L. decastes using transcriptomics combined with WGCNA analysis. Sixteen core differentially expressed genes were identified, primarily enriched in pathways related to pyruvate metabolism and amino acid metabolism. qRT-PCR verification revealed an upward trend in the expression levels of LdPK genes associated with pyruvate metabolism, whereas genes such as LdPC, LdLYS21, LdDLAT, and LdlldD exhibited a downward trend. Similarly, genes involved in amino acid metabolism, including LdTRPD, LdOAT, LdIDO1/3, LdPRODH, LdMCC, and LdEGT2.2, also showed a downward trend. These findings indicate that US treatment influences key enzymes of pyruvate metabolism in postharvest L. decastes, sustains energy homeostasis, reduces amino acid consumption rates, and consequently delays postharvest quality deterioration of L. decastes.

本发明公开了一种杏鲍菇菌种Ple0400及其应用。本发明中,菌种根据基因组测序数据,Pl.e0400是针对袋式栽培选育。以‘日引1号’为亲本,自交选育的;采用Kmer差减组装技术对‘Pl.e0400’和亲本‘日引1号’全基因组测序数据进行两两互比,获得特异性差异片段序列(SV),获得差异片段(SV),随机选取4个SV序列设计特异性引物8对,对杏鲍基‘Pl.e0400’和亲本‘日引1号’进行扩增,Chr1A,Chr2A,Chr4A和Chr5A为日引1号与Pl.e0400共有,Chr1,Chr2,Chr4和Chr5为日引1号特有片段。从结果可以得出杏鲍菇‘Pl.e0400’亲本‘日引1号’的DNA指纹存在显著差异,确定是不同品种。Pl.e0400’菌盖为凹面型、浅灰色,菌柄短、米白色、硬度大,栽培周期较短,产量高,耐贮藏、耐运输,有很好的开发应用前景。

This study investigated the protective effects of carboxymethyl chitosan-dialdehyde glucan/polydopamine carrier-delivered Bacillus subtilis (PBS) on Salmonella-induced intestinal inflammation in mice. The results indicated that PBS significantly lowered the organ index and inflammatory factor levels while increasing the levels of oxidase in mice. Interestingly, after PBS intervention, the mRNA expression levels of mucin, tight junction proteins, immune proteins, and transcription factors that constitute the intestinal barrier in mice exhibited a significant increase compared to those in the model group. The intestinal flora results showed that PBS improved the abundance of Ruminococcus, Bacillus, and Roseburia, simultaneously increasing the content of short-chain fatty acids. Furthermore, the downstream protein expression of pathway genes confirmed that PBS inhibited the inflammatory response by regulating the TLR4-NF-kappa B-NLRP3 inflammasome signaling pathway. These results indicate that carrier protection for Bacillus subtilis can enhance its relief effect on mice infected with Salmonella.

Fresh-cut taro, renowned for its high nutritional value and convenience, is prone to rapid browning post-cutting, which hinders its storage life. This study focused on the effects of L-ascorbic acid (AA) combined with ultrasound (US) treatment (AS) on the storage quality and transcriptome analysis of fresh-cut slices of Yongding June Red Taro. Compared to the control (CK) group, AS treatment effectively reduced the weight loss rate of taro slices, maintained higher hardness, delayed the increase of browning, and inhibited the accumulation of O-2(-) and H2O2. Furthermore, the AS group showed increased glutathione levels and maintained higher activities of ascorbate peroxidase and glutathione reductase, yet decreased the contents of flavonoids and reducing sugars. Simultaneously, in the AS group, the activities of tyrosinase and lipoxygenase were lowered, thereby preserving the high sensory quality of fresh-cut taro slices. Transcriptome analysis revealed that differentially expressed genes (DEGs) between the AS and CK groups were annotated and categorized into 50 and 20 functional groups based on the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) databases, respectively. Notably, both groups exhibited significant enrichment in processes related to photosynthesis, protein processing in the endoplasmic reticulum, and isoflavone biosynthesis. Therefore, we concluded that AS treatment could alleviate oxidative stress and maintain storage quality by regulating metabolic pathways. These findings provide insights into the physiological changes occurring in taro immediately after cutting and serve as an essential basis for developing effective storage and preservation techniques.

This study investigated the structure-activity relationships between SePEPs (selenium-enriched peptides) and PEPs (selenium-free peptides) and compared the antioxidant activities of SePEPs and PEPs. The results showed that SePEPs exhibited higher antioxidant activity than PEPs at the same molecular weight, with the molecular weights of 0-3500 Da exhibiting the highest in vitro antioxidant activity. Chelation between selenium and peptides led to a more compact structure and increased particle density in SePEPs. A spectroscopic analysis revealed new peaks and redshifts in SePEPs, along with a higher content of hydrophobic amino acids than PEPs. A molecular interaction modeling analysis indicated that hydrogen bonding and hydrophobic interactions primarily drove the binding between selenium-containing peptides and 1,1-diphenyl-2-picrylhydrazyl (DPPH). Moreover, the solid-phase synthesized MSePGP exhibited significantly greater antioxidant activity than glutathione at high concentrations. At 10 mg/mL, the DPPH radical scavenging rate of MSePGP was 68.5 +/- 2.2%. These findings provide a theoretical basis for the design and synthesis of selenium-enriched peptides with enhanced antioxidant properties.

This study investigated the hepatoprotective mechanisms of Volvariella volvacea fruiting body polypeptide (VVFP, 1-3 kDa) against acute alcohol-induced liver injury using multi-omics approaches. Male ICR mice pretreated with VVFP (100-400 mg/kg) showed significantly prolonged alcohol tolerance latency (p < 0.05) and accelerated sobriety recovery compared to controls. Integrated transcriptomics and metabolomics revealed VVFP's dual regulatory effects: (1) transcriptional regulation of 36 endoplasmic reticulum stress genes (e.g., ERP57, Derl) through protein processing pathways (KEGG:04141), and (2) metabolic modulation of 23 hepatic metabolites, particularly phosphatidylcholines and organic acids, via amino acid biosynthesis and glycerophospholipid metabolism. Cross-omics analysis identified eight coregulated genes (Got1, Arg2, Srm, etc.) interacting with key metabolites (4-guanidinobutyric acid, GABA) through linoleic acid metabolism. These findings demonstrate VVFP's therapeutic potential as a functional food ingredient by highlighting its ability to simultaneously target hepatic stress responses and metabolic homeostasis during alcohol detoxification.
