Genome-wide identification, expression profiling, and protein interaction analysis of the CCoAOMT gene family in the tea plant (Camellia sinensis) SCIE

Abstract :

Background The caffeoyl-CoA-O methyltransferase (CCoAOMT) family plays a crucial role in the oxidative methylation of phenolic substances and is involved in various plant processes, including growth, development, and stress response. However, there is a limited understanding of the interactions among CCoAOMT protein members in tea plants. Results In this study, we identified 10 members of the CsCCoAOMT family in the genome of Camellia sinensis (cultivar 'HuangDan'), characterized by conserved gene structures and motifs. These CsCCoAOMT members were located on six different chromosomes (1, 2, 3, 4, 6, and 14). Based on phylogenetic analysis, CsCCoAOMT can be divided into two groups: I and II. Notably, the CsCCoAOMT members of group Ia are likely to be candidate genes involved in lignin biosynthesis. Moreover, through the yeast two-hybrid (Y2H) assay, we established protein interaction networks for the CsCCoAOMT family, revealing 9 pairs of members with interaction relationships. Conclusions We identified the CCoAOMT gene family in Camellia sinensis and conducted a comprehensive analysis of their classifications, phylogenetic and synteny relationships, gene structures, protein interactions, tissue-specific expression patterns, and responses to various stresses. Our findings shed light on the evolution and composition of CsCCoAOMT. Notably, the observed interaction among CCoAOMT proteins suggests the potential formation of the O-methyltransferase (OMT) complex during the methylation modification process, expanding our understanding of the functional roles of this gene family in diverse biological processes.

氟处理下茶树根系的组织细胞形态差异及蜡质合成相关基因WSD1的表达分析

茶树是一种氟含量较高的植物,具有聚氟性。基于扫描电镜技术,探究不同质量浓度(10 mg·L~(-1)和50 mg·L~(-1))及不同时间(1 d和16 d)氟处理下,氟高富集茶树品种黄棪和氟低富集茶树品种佛手根系吸收富集氟的特性差异,并对前期转录组数据筛选出氟处理下茶树蜡质合成的差异基因WSD1进行分析。结果显示,50 mg·L~(-1)氟处理后,黄棪根部表皮细胞表面覆盖的蜡质稍有增多,细胞排列较为疏松,而佛手根部表皮细胞界限模糊,覆盖蜡质明显增多,细胞壁出现扭曲断裂等氟不耐受症状。蜡质合成相关基因WSD1荧光定量结果显示,WSD1经外源氟处理后,其对茶树根部的蜡质有较为明显的上调作用;WSD1蛋白互作网络预测结果及相关性分析显示,WSD1受CSS0041298、CSS0012327及CSS0049082的负调控。本研究从茶树与蜡质合成互作角度探究茶树缓解氟胁迫的响应机制,为深入探究茶树氟吸收调控和耐氟茶树品种的选育提供科学依据。

茶梗基缓释尿素的养分释放特征及应用效果研究

Abstract :

为获得一种低成本、性能优良、生物可降解的缓释肥料,以铁观音茶梗为原材料,按照不同配比加入尿素混合,采用挤压膨化法制备缓释肥料。采用扫描电镜分析缓释肥料的三维显微结构,并通过静水培养法和土柱淋溶法结合凯氏定氮法测定各处理组样品在静水及土柱中的养分释放特征。最后,通过玉米苗盆栽试验研究缓释肥料的应用效果。试验结果表明,自制缓释肥料相较于普通尿素肥料,养分释放速率明显降低,在较长时间后才完全释放,且茶梗比例越低,养分释放速率越高,说明茶梗形成的三维网格结构具有良好的养分释控效果。应用试验结果表明,施用3号缓释肥料(茶梗40%、尿素60%)最有利于玉米苗的生长。研究结果可为茶梗资源的综合利用提供新途径,以期实现茶废弃资源的绿色应用,为“碳达峰、碳中和”目标作出贡献。

Abstract :

Lanthanum can affect the growth and development of the tea plant. Tieguanyin (TGY) and Shuixian (SX) cultivars of Camellia sinensis were selected to explore the mechanism underlying the accumulation of lanthanum (tea plants' most accumulated rare earth element) through proteomics. Roots and fresh leaves of TGY and SX with low- and high-accumulation potential for lanthanum, respectively, were studied; 845 differentially expressed proteins (DEPs) were identified. Gene ontology analysis showed that DEPs were involved in redox processes and related to molecular functions. Kyoto Encyclopedia of Genes and Genomes metabolic pathway analysis showed that DEPs were associated with glutathione (GSH) and alpha-linolenic acid metabolism, plant pathogen interaction, and oxidative phosphorylation. Thirty-seven proteins in the GSH metabolism pathway showed significant differences, wherein 18 GSH S-transferases showed differential expression patterns in the root system. Compared with the control, expression ratios of GST (TEA004130.1) and GST (TEA032216.1) in TGY leaves were 6.84 and 4.06, respectively, after lanthanum treatment; these were significantly higher than those in SX leaves. The LOX2.1 (TEA011765.1) and LOX2.1 (TEA011776.1) expression ratios in the alpha-linolenic acid metabolic pathway were 2.44 and 6.43, respectively, in TGY roots, which were significantly higher than those in SX roots. The synthesis of specific substances induces lanthanum-associated defense responses in TGY, which is of great significance for plant yield stability.

人工智能趋势下基于AHP-模糊数学的茶业机械课程教学改革效果综合评价研究

Abstract :

随着人工智能的不断发展,将其融入新农科课程已经是一种趋势。在此背景下,福建农林大学安溪茶学院开展了“人工智能+茶业机械”创新教育改革以培养高素质行业人才。为综合评价该教学改革的效果,对2018级和2019级茶学专业学生进行问卷调查,并采用AHP-模糊数学综合评价法进行定量评价。结果表明,教学改革后的教学质量评价为“很好”,学生接受和认可程度高,将人工智能与茶业机械相结合的教学改革效果较好,值得进一步深化与推广。

茶(Camellia sinensis (L.) O. Kuntze)是一种天然保健饮料,在世界饮料史上起着极其重要的作用。从种质资源的角度来看,不同茶树品种/种质间稀土元素(REE)、氟(F)和铝(Al)含量的差异达显著水平。本研究以前期鉴定出的REE、F和Al的低积累(T15)、中积累(本山)和高积累(水仙)的不同基因型茶树品种为材料,基于Illumina平台的转录组测序,分析差异基因表达情况。结果显示通过转录组测序共发现了83 357条基因,GO功能分类到44个类别中,主要涉及细胞、细胞器、细胞膜和大分子复合物;不同茶树种质间富集最显著的代谢通路为植物激素信号转导、谷胱甘肽代谢,随机选择的8个基因实时荧光定量(Q-PCR)结果进一步证实了RNA测序结果。同时ABC转运蛋白可能参与吸收和富集。这些新发现为低REE、F和Al积累茶树品种的筛选和培育提供了理论基础。

为优化茶园间作模式、丰富茶园间作模式的科学理论,本试验探讨了间作白三叶、金盏菊和金花菜三种间作模式下对茶园土壤营养成分及茶叶品质成分与茶叶产量的影响,并分析了茶叶品质成分和土壤营养成分的相关性。结果表明:三种间作模式条件下茶叶中茶多酚含量均显著降低,进而降低酚氨比。茶园间作金花菜,茶百芽重提高;而间作白三叶、金盏菊,茶百芽重与发芽密度均提高。茶园间作白三叶显著提高土壤有效磷、速效钾、碱解氮和全氮含量;间作金花菜则显著提高土壤速效钾、碱解氮和全氮含量,而全磷含量显著降低;间作金盏菊显著提高土壤有效磷、速效钾和有机质,而全磷与全钾含量显著降低。茶叶品质与土壤相关性分析发现:土壤pH值和土壤全磷、全钾及有机质含量变化与茶叶品质关系十分紧密。茶园间作白三叶、金盏菊和金花菜均不同程度上改良土壤性状,增强土壤肥力,进而改善茶叶品质。

应用气相-离子迁移谱技术对不同年份紧压白茶的挥发性物质进行分析,共鉴定出56种挥发性物质。采用主成分分析(principal component analysis,PCA)和线性判别分析(linear discriminant analysis,LDA)方法共筛选出20个特征挥发性物质,起主要贡献作用的物质是2-乙酰基呋喃、庚醛、E-2-辛烯醛、柠檬烯、2-苯基乙醇及异丁酸。其中,柠檬烯、2-苯基乙醇、异丁酸的含量和贮藏时间呈反比,而2-乙酰基呋喃、庚醛和E-2-辛烯醛的含量则和贮藏时间呈正比。进一步进行线性回归分析,建立贮藏时间(y)和主要特征挥发性物质峰强数据的预测模型方程,y=-0.03x_1+0.038x_2-0.09x_3-0.012x_4+0.002x_5+0.004x_6+0.022x_7+0.019x_8-0.018x_9-0.011x_(10)+0.005x_(11)-0.011x_(12)+0.000x_(13)-0.042x_(14)-0.015x_(15)+0.017x_(16)+0.033x_(17)+0.008x_(18)+0.008x_(19)-0.124x_(20)(其中x_1~x_(13)为未定性特征挥发性物质,x_(14)~x_(20)分别为2-乙酰基呋喃、柠檬烯、2-苯基乙醇二聚体、2-苯基乙醇单体、2-庚酮、E-2-辛烯醛、异丁酸),R~2=0.929,P<0.05。另外,在贮藏过程中紧压白茶的特征挥发性物质间会相互作用,共同形成紧压白茶花香、清香药草味和坚果味的独特风味特征。运用GCIMS技术、配合化学计量法能够科学辨别紧压白茶仓储年限,为规范紧压白茶市场提供理论依据。

Adsorption of Dye by Waste Black Tea Powder: Parameters, Kinetic, Equilibrium, and Thermodynamic Studies SCIE

Waste black tea powder was used as a potential adsorbent to remove methylene blue (MB) from aqueous solution. Several operating factors in adsorption of MB onto waste black tea powder were investigated, including contact time, initial MB concentration, solution pH, adsorption temperature, and dosage of waste black tea powder. Experimental results revealed that the adsorption efficiency increased with contact time and solution pH values and decreased with initial MB concentration and adsorption temperature. The equilibrium time was estimated to be around 60 min. The maximum adsorption capacity and the highest adsorption efficiency were 302.63 mg center dot g(-1) and 100%, respectively. In kinetic study, pseudo-first-order and pseudo-second-order kinetic models, intraparticle diffusion model, and Boyd and Elovich models were employed to analyze the adsorption behavior and the adsorption mechanism. It was found that the pseudo-second-order kinetic model was suitable to describe the adsorption process, and the calculated equilibrium adsorption capacity was well close to the experimental data for different initial MB concentrations. The internal diffusion was not the only rate-controlling step, and the existence of boundary effect was observed in this study. From isotherm analysis, the equilibrium data were well represented by the Langmuir model, rather than Freundlich, Dubinin-Redushckevich, or Temkin models. The nonlinear fitting for various isotherm models implied that the adsorption behavior between MB and waste black tea powder was complication. Thermodynamic parameters including changes in Gibb's free energy, enthalpy, and entropy suggested that adsorption of MB onto waste black tea powder was a spontaneous and exothermic process. The multiple regeneration/adsorption experiments indicated that the used black tea powder efficiently remained more than 75% after five cycles using NaOH as a regenerative reagent and thus be used for many times. Therefore, as a low-cost and easily available material, waste black tea powder could be applied in wastewater treatment.

茶作为我国重要经济作物,茶叶的质量安全问题与人的身体健康、数千万茶农的经济收益及我国茶产业的发展等息息相关。茶叶氟含量超标是茶叶质量安全的一大问题,研究茶树(Camellia sinensis(L.) O.Kuntze)富氟机制及控氟降氟技术已经成为一项重要课题。本文综述了茶树对氟的吸收富集与解毒耐受机制及控氟、降氟技术等方面的研究成果,以期为筛选低氟茶树品种、提升茶叶氟质量安全等方面提供参考。
