Abstract :
This study aimed to investigate the immune mechanisms of pedal mucus in Pacific abalone Haliotis discus hannai from different populations. Proteomic differences between the pedal mucus of the control group and the heat-resistance group of H. discus hannai were comparatively analyzed, and the proteins were annotated and analyzed to understand the related functions and roles of differentially expressed proteins (DEPs). The results showed that there were 4054 DEPs in total, among which the DEPs in the pedal mucus of the heat-resistance group and the control group were mainly immune-related heat shock proteins, calmodulin, Ig-like and fibronectin type III domain-containing proteins, histones and mucins, etc.; the DEPs related to growth metabolism included glutathione, growth factor receptor-bound protein, alanine aminotransferase, etc. Circadian entrainment signaling pathway and growth hormone synthesis, secretion and action signaling pathway was significantly enriched, cortisol synthesis and secretion signaling pathway was up-regulated in Haliotis discus hannai heat-resistance group. This study provided references for exploring the relationship between the mucus proteome difference in pedal mucus from different abalone populations sources. These results would contribute to further search for proteins related to immunity, growth and stress resistance, and provide theoretical basis for the development of high-quality germplasm resources of Pacific abalone.

本发明涉及贝类遗传育种领域,特别涉及一种人工诱导皱纹盘鲍三倍体的方法。该方法包括以下步骤:1)种鲍的促熟及选择:选取无病害种鲍置于种鲍培育池中同步促熟,从中选择性腺发育良好的二倍体鲍;2)人工催产:将挑选的种鲍进行催产,至雌雄种鲍产卵和排精为止;3)配子的选择和混合受精:分别滤除精卵中的杂质,将精子与卵子混合进行人工受精,得受精卵;4)热休克诱导:受精15min后,将受精卵置于预热温度为26~31℃的海水中处理10~15min,以抑制受精卵第二极体的排出;5)孵育:热休克结束后立即将受精卵收集至孵育水箱中进行孵化培育。此法选育出的皱纹盘鲍三倍体具有耐热性,可减少渔户因鲍度夏难而导致的经济损失。通过改良种鲍催产方式,优化诱导条件,可实现皱纹盘鲍三倍体的高效稳定获得。

一种皱纹盘鲍四倍体的诱导方法,涉及贝类四倍体的培育方法。包括以下步骤:1)种鲍选择及促熟:选取无外部损伤、健康有活力的雌雄种鲍分别置于种鲍培育池中同步促熟;2)人工催产:挑选性腺发育良好的二倍体鲍进行催产,至雌雄种鲍产卵和排精为止;3)精卵的选择和混合受精:分别滤除精卵中的杂质,将精子加入卵液中同步混合受精;4)冷休克诱导:当60%~80%受精卵准备第一次卵裂开始伸长时,立即将受精卵收集至预冷温度为2~8℃海水中处理10~15min,以抑制第一次卵裂;5)孵育:冷休克结束后立即将受精卵放入18~22℃孵育水箱中进行孵化。通过改良种鲍催产方式,优化诱导条件,从而实现皱纹盘鲍四倍体的高效稳定获得。

In recent years, the abalone aquaculture industry has been threatened by the bacterial pathogens. The immune responses mechanisms underlying the phagocytosis of haemocytes remain unclear in Haliotis discus hannai. It is necessary to investigate the immune mechanism in response to these bacterial pathogens challenges. In this study, the phagocytic activities of haemocytes in H. discus hannai were examined by flow cytometry combined with electron microscopy and transcriptomic analyses. The results of Vibrio parahaemolyticus, Vibrio alginolyticus and Staphylococcus aureu challenge using electron microscopy showed a process during phagosome formation in haemocytes. The phagocytic rate (PP) of S. aureus was higher than the other five foreign particles, which was about 63%. The PP of Vibrio harveyi was about 43%, the PP peak of V. alginolyticus in haemocyte was 63.7% at 1.5 h. After V. parahaemolyticus and V. alginolyticus challenge, acid phosphatase, alkaline phosphatase, total superoxide dismutase, lysozyme, total antioxidant capacity, catalase, nitric oxide synthase and glutathione peroxidase activities in haemocytes were measured at different times, differentially expressed genes (DEGs) were identified by quantitative transcriptomic analysis. The identified DEGs after V. parahaemolyticus challenge included haemagglutinin/amebocyte aggregation factor-like, supervillin-like isoform X4, calmodulin-like and kyphoscoliosis peptidase-like; the identified DEGs after V. alginolyticus challenge included interleukin-6 receptor subunit beta-like, protein turtle homolog B-like, rho GTPase-activating protein 6-like isoform X2, leukocyte surface antigen CD53-like, calponin-1-like, calmodulin-like, troponin C, troponin I-like isoform X4, troponin T-like isoform X18, tumor necrosis factor ligand superfamily member 10-like, rho-related protein racA-like and haemagglutinin/amebocyte aggregation factor-like. Some immune-related KEGG pathways were significantly up-regulated or down-regulated after challenge, including thyroid hormone synthesis, Th17 cell differentiation signalling pathway, focal adhesion, melanogenesis, leukocyte transendothelial migration, inflammatory mediator regulation of TRP channels, ras signalling pathway, rap1 signalling pathway. This study is the first step towards understanding the H. discus hannai immune system by adapting several tools to gastropods and providing a first detailed morpho-functional study of their haemocytes.

Rab7 is a small GTPase that regulates vesicular traffic from early to late endosomal stages of the endocytic pathway. In this study, full-length cDNA sequences of Rab7 were cloned from Yellow River Carp Cyprinus carpio, which were designated as CcRab7, and its function during pathogen infection was also investigated. The results showed that the full-length cDNA of CcRab7 was composed of 1511 bp, and open reading frame is 618 bp encoding 205 amino acids with 23.10 kDa. The deduced amino acid sequence of CcRab7 shared high similarity to that of Pimephales promelas (99.02%). In this paper, real-time fluorescence quantitative PCR (qRT-PCR) was used to detect the expression levels in 11 tissues and organs of healthy Yellow River Carp, and the tissue distribution of CcRab7 as well as its defense mechanism against Spring viremia of carp virus (SVCV) and Aeromonas hydrophila in Yellow River Carp C. carpio was investigated. The results revealed that the mRNA transcripts of CcRab7 were widely detected in various tissues of C. carpio, with the highest expression levels in head kidney and blood, indicating that CcRab7 was involved in the innate immune process. Yellow River Carp was immunized with A. hydrophila and SVCV respectively, and the expression changes of CcRab7 were detected in gill, spleen, liver and skin of Yellow River Carp. The mRNA transcripts of CcRab7 in the main organs (gill, and liver, skin and spleen) were significantly affected by SVCV and A. hydrophila infection. The results showed that the CcRab7 expression levels was obviously up-regulated at different time points. The eukaryotic recombinant plasmids of CcRab7 and pEGFP-N3 transfected into Grass carp ovary cell lines (GCO) cells for subcellular localization. The results provide a basis for further investigation into the immunological role of CcRab7 in Yellow River Carp.

Rab5A play important roles in regulating trafficking organelles, especially in phagosome formation. In the present study, full-length cDNA sequences of Rab5A were cloned from Yellow River Carp Cyprinus carpio, which was designated as CcRab5A. The full-length cDNA of the CcRab5A cDNA sequence is 2434 bp and included an open reading frame (ORF) encoding 216 amino acids polypeptide with an estimated molecular weight of 23.47 kDa. Bioinformatics analysis showed that the CcRab5A protein was highly conserved during evolution. CcRab5A's deduced amino acid sequence showed high identity to Cyprinus carpio (99.54%) in comparison. The guanine-base binding motif (G), phosphate/magnesium-binding motif (PM), and Rab family motif (Rab F) of CcRab5A are highly conserved among various species, but the N- and C-terminal regions were hypervariable, according to the results of multiple sequence alignment and phylogenetic analysis. Additionally, 11 tissues of Yellow River Carp were examined using Real-time Fluorescence Quantitative PCR (qRT-PCR) to determine the expression levels, with the highest expression levels in head kidney and blood. Followed by heart, liver, muscle, brain, gill, skin, spleen and intestine; The expression level in body kidney was the lowest. Yellow River Carp was immunized with Aeromonas hydrophila and Spring viremia of carp virus (SVCV) respectively, and the expression changes of CcRab5A in gill, spleen, liver, intestine and skin of Yellow River Carp were detected. The results showed that the expression level of the gene was obviously up-regulated at different time points. The eukaryotic recombinant plasmids of CcRab5A, pEGFP-N3 were constructed and transfected into GCO cells for subcellular localization. The results showed that CcRab5A were mainly distributed in nuclear membrane and various endosome membranes. These results showed that CcRab5 were involved in viral and bacterial infection in the immune response of Yellow River Carp.

Hypoxia occurrence is potentiated by eutrophication caused by human activities and it may be the most dramatically increasing environmental factor as a result of the climate change-led increasing temperatures. The crucian carp Carassius auratus is an important aquaculture economical freshwater species and is also a potential model for the study of physiological adaptation. However, the transcriptomics and genomics data available for this species are still scarce. The gills of fish are the major organ of hypoxia and other stresses. In this study, we studied the effect of the exposure to hypoxia (1.0 mg O2/L for 7 d) and 2 mg/mL Poly I:C on the gene expression of the gills of Carassius auratus using the Illumina sequencing platform. Comparison of the gene expression levels between the experimental and control groups showed differentially expressed genes (DEGs). The DEGs were mainly annotated into glycolysis, oxidative phosphorylation and immune system categories based on the KEGG database. In glycolysis pathway, lactate dehydrogenase, malate dehydrogenase, glyceraldehyde 3-phosphate dehydrogenase, enolase and succinate dehydrogenase were identified. In addition, the DEGs were enriched in the immune-related pathway functions, including pattern recognition receptor (PRR), antigen processing and presentation, B cell and T cell receptor signaling pathways, natural killer (NK) cell mediated cytotoxicity, Fc gamma R-mediated phagocytosis, and the JAK-STAT, p53 and cytokines signaling pathways. This study also produced potential hypoxia biomarkers including FOS, JUN B, lysosomal acid lipase, and HIF-1 alpha. HIF-1 signaling pathway was significantly down-regulated after hypoxia and Poly I: C exposure. Hence, the data generated here may offer a valuable resource information for the further study of the immune response and also provide improved strategies against hypoxia and Poly I:C in teleost fish.
