Abstract :
This study evaluated the effects of dietary synthetic astaxanthin (SA), Haematococcus pluvialis (HP) and Phaffia rhodozyma (PR) on the growth performance, antioxidant activity, innate immunity, morphology, and pigmentation of juvenile Litopenaeus vannamei. Shrimp (1.15 +/- 0.01 g) were fed with the control diet and astaxanthin diets containing 20 mg/kg of astaxanthin from three sources (SA, HP, and PR) for 56 days. The results indicated that, compared with the control group, growth performance was observably elevated in the HP and PR groups (p < 0.05). The astaxanthin (SA, HP, and PR) supplemented diets markedly elevated the activities of glutathione peroxidase (GPx) and GST in the intestine and hepatopancreas (p < 0.05), while observably reduced the MDA content (p < 0.05). The apoptosis rates in three astaxanthin groups were noticeably reduced in comparison with the control group (p < 0.05). Dietary astaxanthin (SA, HP, and PR) observably elevated the expression of the Toll, IMD, and CAT genes in the hepatopancreas (p < 0.05). Besides, dietary astaxanthin (SA, HP, and PR) noticeably improved the astaxanthin accumulation and pigmentation of shrimp (p < 0.05). The survival rates of shrimp challenged with Aeromonas hydrophila were markedly higher in the astaxanthin groups than in the control group (p < 0.05), and no significant difference was detected among three astaxanthin groups (p > 0.05). Moreover, our results suggested that natural astaxanthin (HP and PR) was more effective for enhancing growth and antioxidant capacity of shrimp. Nevertheless, no marked difference was detected between natural astaxanthin and SA in coloration performance and disease resistance.

以玉米DDGS替代饲料中不同比例[0(D0)、25%(D25)、50%(D50)和75%(D75)]的豆粕蛋白,配制成玉米DDGS含量分别为0、13.63%、27.26%和40.89%的4种等氮等能饲料,饲喂初始体重为(3.60±0.01) g的中华圆田螺(Cipangopaludina cathayensis)幼螺56d,探讨其对中华圆田螺幼螺生长性能、体成分、抗氧化能力和消化系统组织结构的影响。结果显示:(1) D25组终末体重(FBW)、特定生长率(SGR)和增重率(WGR)显著高于D0组(P<0.05), D25和D0组之间存活率(SR)、摄食率(FR)、饲料效率(FE)和蛋白质效率(PER)无显著差异(P>0.05),但D50和D75组的FBW、SGR和WGR显著低于D0组(P<0.05); D25组中华圆田螺幼螺螺体粗蛋白、粗脂肪和灰分含量均显著高于D0组(P<0.05); D50和D75组中华圆田螺幼螺肝胰腺和肠道的胰蛋白酶(TRY)及脂肪酶(LPS)的活性均显著低于D0组(P<0.05),而各组间淀粉酶(AMS)活性无显著差异(P>0.05)。(2) D25组中华圆田螺幼螺肝胰腺和肠道碱性磷酸酶(AKP)、酸性磷酸酶(ACP)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPx)和谷胱甘肽S转移酶(GST)活性与D0组相比均无显著差异(P>0.05), D50和D75组中华圆田螺幼螺肝胰腺和肠道CAT、SOD和GPx活性显著低于D0组(P<0.05), D75组肝胰腺和肠道丙二醛(MDA)含量显著高于D0组(P<0.05); D50和D75组肝胰腺GPx、GST和SOD和肠道GST基因表达量均显著低于D0组(P<0.05)。(3) D25组中华圆田螺幼螺肝胰腺和肠道组织结构与D0组无显著差异, D75组中华圆田螺幼螺肝胰腺部分肝小管结构紊乱,基膜破裂; D50和D75组中华圆田螺幼螺肠道上皮细胞受损,固有层萎缩。综上结果,玉米DDGS在中华圆田螺幼螺饲料中的添加比例以不超过13.63%为宜。

Abstract :
Acidifier has become one of the most important substitutes for antibiotics, and its beneficial effects on growth performance and digestive ability have been evidenced. Sodium benzoate is a commonly used acidifier. A 56-day feeding test was carried out to elucidate the effects of dietary sodium benzoate on growth performance, morphology, antioxidant activity and intestinal microflora of juvenile Litopenaeus vannamei with an average initial body weight of 0.55 +/- 0.01 g. Six experimental diets were prepared by adding 0, 750, 1000, 1250, 1500 and 1750 mg/kg sodium benzoate to the control diet. The results showed that: The final body weight (FBW) and weight gain (WG) of juvenile shrimp were significantly improved by diet supplemented with 1750 mg/kg sodium benzoate. Dietary 1750 mg/kg sodium benzoate significantly increased the trypsin activity. The activities of catalase (CAT), superoxide dismutase (SOD) and glutathione S-transferase (GST) in the intestine and hepatopancreas of the 1750 mg/kg sodium benzoate group were markedly higher than those of the control group. Furthermore, the expression levels of trypsin (TRY), amylase (AMS), lysozyme (LZM), interleukin-8 (IL8) and interleukin-10 (IL10) genes in the intestine significantly increased when the content of sodium benzoate in the diet was greater than or equal to 1000 mg/kg. The addition of 1500 and 1750 mg/kg sodium benzoate to the feed improved the intestinal and hepatopancreatic tissue structures of juvenile shrimp and enriched the diversity of intestinal microbiota. Sodium benzoate residues were not detected in each sodium benzoate supplemented group. At the end of the Aeromonas hydrophila challenge test, juvenile shrimp in the 1500 and 1750 mg/kg group exhibited significantly higher cumulative survival rates than that in the control group. Conclusively, dietary 1750 mg/kg sodium benzoate could improve the growth performance, antioxidant capacity, morphology, intestinal microflora and disease resistance of juvenile shrimp.

以含不同浓度黄曲霉毒素B_1(AFB_1)(0、10、100和1000μg/kg饲料)的4种饲料饲喂初始均重为(0.382±0.005) g的克氏原螯虾(Procambarus clarkii)幼虾42d,探讨AFB_1对克氏原螯虾幼虾生长性能、饲料效率和肝胰腺组织结构的影响。结果显示, 100和1000μg/kg毒素组幼虾的存活率、摄食率、终末体重、特定生长率和饲料效率均显著低于对照组, 10μg/kg毒素组与对照组无显著差异。10μg/kg毒素组幼虾肝胰腺碱性磷酸酶(AKP)、谷丙转氨酶(ALT)、谷草转氨酶(AST)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)、谷胱甘肽转移酶(GST)活性和丙二醛(MDA)含量均与对照组无显著差异,超氧化物歧化酶(SOD)活性显著低于对照组。饲料AFB_1含量≥100μg/kg时显著影响了克氏原螯虾幼虾上述肝胰腺酶的活性。10μg/kg毒素组肝胰腺组织结构发生轻微变化, 100和1000μg/kg毒素组幼虾的肝胰腺表现出严重病变, R细胞数量减少而B细胞数量增多。超微结构结果显示,随着AFB_1浓度的增加,肝胰腺细胞内线粒体肿胀,粗面内质网扩张,囊泡增多,脂滴变大。饲料中AFB_1≤100μg/kg时克氏原螯虾幼虾体内均未检测出AFB_1。1000μg/kg毒素组幼虾虾体中AFB_1积累量为1.65μg/kg,未超过FDA食品安全限定标准。综上所述,饲料中AFB_1含量≥10μg/kg时即对克氏原螯虾幼虾产生不良影响,说明克氏原螯虾幼虾对AFB_1较为敏感。

以含辅酶Q10(CoQ10)分别为0、40、80和120 mg/kg的4种饲料饲喂平均初始体重为(19.97±0.13) g的吉富罗非鱼(Oreochromis niloticus, GIFT)幼鱼56d,探讨辅酶Q10对吉富罗非鱼幼鱼生长性能、体成分、抗氧化能力、组织结构和基因表达的影响。结果显示,各辅酶Q10组吉富罗非鱼幼鱼的终末体重、摄食率、特定生长率和饲料效率与对照组均无显著差异, 120 mg/kg辅酶Q10组终末体重、特定生长率和饲料效率均为最高;辅酶Q10含量为120 mg/kg时,吉富罗非鱼幼鱼的干物质消化率显著升高;各辅酶Q10组血清过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)活性均显著高于对照组;各辅酶Q10组肝脏CAT和GSH-Px活性均显著高于对照组, 80和120 mg/kg辅酶Q10组肝脏总超氧化物歧化酶(SOD)和谷胱甘肽S转移酶(GST)活性均显著升高,丙二醛(MDA)含量显著降低;各实验组去内脏全鱼的水分、粗蛋白和灰分含量均无显著性差异, 120 mg/kg辅酶Q10组去内脏全鱼粗脂肪显著低于对照组;各实验组内脏团水分、粗蛋白、粗脂肪和灰分含量均无显著性差异;各实验组肝脏组织学观察未发现明显的结构变化, 120 mg/kg辅酶Q10组肠道绒毛长度、绒毛密度和肌层厚度显著增加; 120 mg/kg辅酶Q10组的肝脏sod、cat、gsh-px、gst和igm基因表达量显著上调,各辅酶Q10组的il-1β和il-8基因表达量均显著低于对照组;在嗜水气单胞菌(Aeromonas hydrophila)腹腔注射攻毒实验中, 80与120 mg/kg辅酶Q10组吉富罗非鱼幼鱼累计存活率均显著高于对照组。结果表明,饲料中添加辅酶Q10≤120 mg/kg对吉富罗非鱼幼鱼无不良损伤,饲喂120 mg/kg辅酶Q10能提高吉富罗非鱼幼鱼的消化率、抗氧化能力、肝脏抗氧化相关基因的表达和对嗜水气单胞菌的抗病力。

以双低菜粕替代饲料中不同比例[0(R0)、25%(R25)、50%(R50)、75%(R75)和100%(R100)]的鱼粉蛋白,配制成双低菜粕含量分别为0、21.25%、42.50%、63.75%和85%的5种等氮等能饲料,饲喂平均初始体重为(2.49±0.01) g的克氏原螯虾(Procambarus clarkii)幼虾42d,探讨其对克氏原螯虾幼虾生长性能、饲料利用率、消化能力及抗氧化能力的影响。结果显示:(1)R0、R25和R50组之间终末体重(FBW)、特定生长率(SGR)、饲料效率(FE)和蛋白质效率(PER)无显著差异(P>0.05), R75和R100组的FBW、SGR、FE和PER显著低于R0组(P<0.05);与R0组相比, R50、R75和R100组克氏原螯虾幼虾粗蛋白和粗脂肪含量显著降低(P<0.05); R75和R100组克氏原螯虾幼虾肝胰腺蛋白酶活性、肝胰腺脂肪酶活性及肠道胰蛋白酶活性均显著低于R0组(P<0.05),各组间淀粉酶活性无显著差异(P>0.05); R75和R100组肝胰腺和肠道NPY和TRY基因表达量显著低于R0组(P<0.05), MSNP基因表达量显著高于R0组(P<0.05)。(2)与R0组相比, R100组肝胰腺碱性磷酸酶(AKP)、谷丙转氨酶(GPT)和谷草转氨酶(GOT)活性显著升高(P<0.05);随着菜粕水平的升高, R100组丙二醛(MDA)含量显著高于R0组(P<0.05),肝胰腺过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPX)和谷胱甘肽S转移酶(GST)活性呈下降趋势且R75和R100组的以上4种的酶活性显著低于R0组(P<0.05); R50、R75和R100组肝胰腺和肠道AKP基因表达量显著上升(P<0.05), CAT、SOD和GPX基因表达量显著下降(P<0.05)。(3)R25组克氏原螯虾幼虾肝胰腺和肠道组织结构与R0组无显著差异,随着替代比例的升高,克氏原螯虾幼虾肝胰腺上皮细胞空泡化逐渐加重, R细胞萎缩程度加深,肝小管管腔畸形并变大;肠道上皮细胞脱落,肠绒毛明显变短。综上结果,双低菜粕在克氏原螯虾幼虾饲料中的添加比例以不超过21.25%为宜。

The present study was conducted to investigate the effects of dietary Coenzyme Q10 (CoQ10) on the growth performance, body composition, digestive enzyme activity, antioxidant capacity, intestinal histology, immune -antioxidant gene expression and disease resistance of juvenile European eel (Anguilla anguilla). Fish were fed a diet supplemented with CoQ10 at concentrations of 0, 40, 80 and 120 mg/kg for 56 days. The results indicated that dietary CoQ10 supplementation did not significantly affect final body weight (FBW), survival rate (SR), weight gain (WG), feed rate (FR), viscerosomatic index (VSI) or hepatosomatic index (HSI) among all experi-mental groups. However, the highest FBW, WG and SR were found in the 120 mg/kg CoQ10 group. Dietary 120 mg/kg CoQ10 markedly improved feed efficiency (FE) and the protein efficiency ratio (PER). The crude lipid in the body and triglycerides (TG) and total cholesterol (TC) in serum were obviously lower in the 120 mg/kg CoQ10 group than in the control group. For digestive enzymes, protease activity in the intestine was markedly boosted in the 120 mg/kg CoQ10 group. The serum activities of SOD, CAT and GST in the 120 mg/kg CoQ10 group were significantly higher than those in the control group. Dietary 120 mg/kg CoQ10 efficiently enhanced superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione S-transferase (GST) activities in the liver, while the malondialdehyde (MDA) content was significantly decreased. No significant histological changes in the liver were identified in any group. Dietary supplementation with 120 mg/kg CoQ10 improved antioxidant capacity and immunity by upregulating the expression of cyp1a, sod, gst, lysC, igma1, igmb1 and irf3 in the liver. Furthermore, the cumulative survival rate of juvenile European eel against challenge with Aeromonas hydrophila was significantly elevated in the 80 and 120 mg/kg CoQ10 supplemented groups. Conclusively, our study suggested that supplementing the diet of juvenile European eel with CoQ10 at a con-centration of 120 mg/kg could promote their feed utilization, fat reduction, antioxidant capacity, digestibility, immune-antioxidant gene expression and resistance to Aeromonas hydrophila without negative effects on fish health status.

Abstract :
本发明公开了一种提高鲤鱼抗氧化能力和抗病力的配合饲料,其由以下重量份材料制成:鱼粉3‑8份、鸡肉粉7‑10份、豆粕25‑32份、菜粕9‑12份、棉粕6‑10份、面粉7‑12份、次粉7‑10份、麸皮7‑10份、豆油3‑5份、多维多矿预混合物0.5‑1份、胆碱0.1‑0.5份、羧甲基纤维素钠1‑3份、膨润土5‑9份、磷酸二氢钙1‑2.5份、蛋氨酸0.1‑0.3份、赖氨酸0.1‑0.5份、辅酶Q10 0.012‑0.02份。本发明所得配合饲料能增强鲤鱼的抗病能力,提高其生长性能和饲料的利用率,具有良好应用前景。
