Abstract :
Selenium (Se) in paddy rice is one of the significant sources of human Se nutrition. However, the effect of arsenic (As) pollution in soil on the translocation of Se species in rice plants is unclear. In this research, a pot experiment was designed to examine the effect of the addition of 50 mg As/kg soil as arsenite or arsenate on the migration of Se species from soil to indica Minghui 63 and Luyoumingzhan. The results showed that the antagonism between inorganic As and Se was closely related to the rice cultivar and Se oxidation state in soil. Relative to the standalone selenate treatment, arsenite significantly ( p < 0.05) decreased the accumulation of selenocystine, selenomethionine and selenate in the roots, stems, sheaths, leaves, brans and kemels of both cultivars by 21.4%-100.0%, 40.0%-100.0%, 41.0%-100%, 5.4%96.3%, 11.3%-100.0% and 26.2%-39.7% respectively, except for selenocystine in the kemels of indica Minghui 63 and selenomethionine in the leaves of indica Minghui 63 and the stems of indica Luyoumingzhan. Arsenate also decreased ( p < 0.05) the accumulation of selenocystine, selenomethionine and selenate in the roots, stems, brans and kernels of both cultivars by 34.9%-100.0%, 30.2%-100.0%, 11.3%-100.0% and 5.6%-39.6% respectively, except for selenate in the stems of indica Minghui 63. However, relative to the standalone selenite treatment, arsenite and arsenate decreased ( p < 0.05) the accumulation of selenocystine, selenomethionine and selenite only in the roots of indica Minghui 63 by 45.5%-100.0%. Our results suggested that arsenite and arsenate had better antagonism toward Se species in selenate-added soil than that in selenite-added soil; moreover, arsenite had a higher inhibiting effect on the accumulation of Se species than arsenate. Nitrogen (N), phosphorus (P) and potassium (K) are three macroelements in agriculture production, but their combined effects on arsenic (As) toxicity and its translocation in rice plants are not clear. In this study, an orthogonal rotation combination based on different N, P and K (NPK) concentration was first designed to examine their combined effect on the As toxicity, its transformation and migration in rice plants based on the hydroponic culture and pot soil culture. The results showed that 2.0 mg/L arsenite (As(III)) had obvious toxicity on the growth of indica LuYouMingZhan (LYMZ) and the optimal NPK concentration was 28.41, 6 and 50 mg/L based on the quadratic regression of the recovery rate of chlorophyll SPAD value of indica LYMZ. The optimal NPK combination significantly alleviated the physiological toxicity of As(III) on indica LYMZ rice seedling and decreased the accumulation of inorganic As in their roots and shoots by 23.8+1.8 % and 33.4+2.4 % respectively; further pot culture from different As(III) polluted soil showed that the optimal NPK combination significantly increased the dry weight of roots, stems, sheaths and leaves of indica LYMZ rice plants as well as yield indicators by 6.4 %-61.7 % and 7.1 %-89.8 % respectively, decreased the accumulation of As(III) and arsenate by 6.25 %-100 % and 12.36 %-100 % respectively in their roots, stems, sheaths, leaves, brans and kernels except As (III) concentration in their sheaths, decreased the accumulation of dimethylarsenate in their sheaths, leaves, brans and kernels, and had the best repair effect on the translocation of As species in 50 mg/kg As(III)-added soil. Our study provided a desirable strategy for alleviating As toxicity in paddy soil and reducing As pollution in rice plants.

本发明公开了硫量子点材料在缓解苗期水稻铅毒害中的应用,硫量子点材料的制备方法包括:将预设重量份的升华硫粉、氢氧化钠加入到水中混合,再加入预设量PEG‑400,然后在预设温度和搅拌速度下,反应预设时长,反应所得的产物在剧烈搅拌下,加入预设浓度的H2O2进行反应,制得硫量子点材料;本发明方案合成的硫量子点材料的成本低且性质稳定,原料只需要升华硫粉、氢氧化钠、蒸馏水、PEG‑400、过氧化氢;除此之外,本发明方案硫量子点具有纳米材料缓释性的特点,能够调节缓解水稻生长周期中的铅毒害效应。

Abstract :
Increased carbon emissions led to extreme weather, global warming, and other environmental problems. In order to control energy input and reduce carbon emissions, this study first combines a three-stage Slacks-Based Measure with Data Envelopment Analysis (SBM-DEA) and uses the Malmquist index to quantify energy consumption at the city scale and the related carbon emission efficiency in Fujian Province for the period 2015-2020. Second, we explore the carbon reduction potential on the city scale from the perspective of improving carbon emission efficiency. Our results demonstrate that (i) the carbon emission efficiency of the nine cities increases overall in the first stage, when technical efficiency approaches the efficiency frontier state and efficiency shortage is mainly caused by the lack of pure technical efficiency. (ii) Regression by stochastic frontier analysis in the second stage reveals that the secondary industry correlates positively at 1% significance with fossil energy consumption and power consumption, indicating that the carbon emission efficiency decreases as the secondary industry increases. (iii) Putian and Xiamen reduced their carbon emission efficiency in the third stage due to (a) the input redundancy of fossil energy and social power consumption and (b) excessive undesirable output carbon emissions. (iv) There were improvements in carbon emission efficiency peaks in 2015, with Longyan, Ningde, and Sanming improving by about 50%. This improvement then decreased up to the year 2020, when the improvement in the carbon emission efficiency of Ningde and Zhangzhou was 6.02% and 9.50%, respectively, and that of all other cities was less than 1%. Therefore, we suggest that carbon emission reduction in the future can be further improved by improving technology, optimizing industrial structure, and various other ways to further improve carbon emission efficiency.

Selenium (Se) can reduce cadmium (Cd) uptake/translocation via regulating pectins, hemicelluloses and lignins of plant root cell walls, but the detailed molecular mechanisms are not clear. In this study, six hydroponic ex-periments were set up to explore the relationships of uptake/translocation inhibition of Cd by selenite (Se(IV)) with cell wall component (CWC) synthesis and/or interactions. Cd and Se was supplied (alone or combinedly) at 1.0 mg L-1 and 0.5 mg L-1, respectively, with the treatment without Cd and Se as the control. When compared to the Cd1 treatment, the Se0.5Cd1 treatment 1) significantly increased total sugar concentrations in pectins, hemicelluloses and callose, suggesting an enhanced capacity of binding Cd or blocking Cd translocation; 2) stimulated the deposition of Casparian strips (CS) in root endodermis and exodermis to block Cd translocation; 3) stimulated the release of C-O-C (-OH- or -O-) and C--O (carboxyl, carbonyl, or amide) to combine Cd; 4) regulated differential expression genes (DEGs) and metabolites (DMs) correlated with synthesis and/or in-teractions of CWSs to affect cell wall net structure to affect root cell division, subsequent root morphology and finally elemental uptake; and 5) stimulated de-methylesterification of pectins via reducing expression abun-dances of many DMs and DEGs in the Yang Cycle to reduce supply of methyls to homogalacturonan, and regulated gene expressions of pectin methylesterase to release carboxyls to combine Cd; and 6) down-regulated gene expressions associated with Cd uptake/translocation.

Zearalenone (ZEN), a widespread mycotoxin, can cause great harm to people's health. In order to assay ZEN, an immobilization-free electrochemical sensor has been developed. A multifunctional hairpin DNA has been carefully designed, including three functions: the aptamer for zearalenone (ZEN), primer, and template sequence. This hairpin DNA can anchor on polydopamine nanospheres (PDANSs), which can protect DNA against the digestion of enzymes and prevent the occurrence of strand displacement amplification (SDA). In the presence of ZEN, the hairpin DNA is dissociated from PDANSs due to the interaction between ZEN and the aptamer, and the SDA reaction is initiated with the help of endonuclease and polymerase. During the SDA process, substantial amounts of negatively charged dsDNA are generated. The MB molecules are embedded into the dsDNA grooves to obtain the complex with a negative charge. The confined MB is repelled on the surface of the negatively charged ITO electrode, leading to the decline of the current. This immobilization-free method possesses high sensitivity (LOD of 0.18 pg mL(-1)) and good selectivity and can be applied to assay ZEN in corn flour.

Abstract :
Ochratoxin A (OTA) is a terrible mycotoxin, which causes environmental problems and seriously endangers people's food safety. The development of the biosensor for OTA is be imperative. Here, taking advantage of CRISPR-Cas12a, a member of CRISPR-Cas family, as the amplification tool, a label-free ECL sensing platform for the detection of OTA is developed. A well-designed crRNA hybridized with its complementary strand (also as aptamer for OTA), and activated the activity of Cas12a (Activated stage). While in the existence of OTA, crRNA and its complementary strand dissociated due to the competitive effect of OTA, which inhibited the activity of Cas12a (Locked stage). Inert Cas12a could not digest hairpin DNA on the electrode, which declined ECL intensity because of steric hindrance. As a consequence, this ECL sensing platform can realize to assay OTA in the drinks. © 2022, The Authors. All rights reserved.

Roles of cell walls in restricting Cd uptake and translocation affected by selenium (Se) are not fully understood. A series of hydroponic experiments were conducted to explore the effects of selenite [Se(IV)] on pectins, hemicelluloses, lignins, callose, and Casparian strips (CS) in rice roots (hybrid Yongyou No.9) via physiological experiments, confocal laser scanning microscopy (CLSM) observations, and conjoint analysis of transcriptome and metabolome. Under 1 mg L–1 Cd exposure, 0.5 mg L–1 Se increased total sugar concentrations in pectins, hemicelluloses and callose, and stimulated the deposition of CS in the root endodermis and exodermis. However, Se did not increase lignin concentration possibly due to expression inhibition of genes correlated with cinnamyl–alcohol dehydrogenase and guaiacol peroxidase. Se stimulated the release of more functional groups of C–O–C (–OH– or –O–) and C=O (carboxyl, carbonyl, or amide) but decreased the C=O group (ester). Many differential expression genes (DEGs) and/or differential metabolites (DMs) correlated with callose, CS, lignin, pectins and hemicelluloses were found. In addition, the modification of side chains of different pectins and hemicelluloses, and the subsequent cross–links among pectins, hemicelluloses and lignin were discussed. © 2022, The Authors. Laver is one of the major arsenic contributors to human diets. The study on metabolic and residual characteristic of each arsenic species contained in laver is important to scientifically assess the intake risk of arsenic in the laver. The metabolic and residual characteristic of main arsenic species in laver, namely arsenate [As(V)], dimethylarsinic acid [DMA(V)] and two arsenosugars, was investigated by mouse experiments in this study. The results showed that the intake of higher-dose laver did not lead to a notable increase of As(V) concentration in mouse muscle/organs and feces. In contrast, DMA(V) excretion in feces and DMA(V) residue in muscle/organs showed a close correlation with laver-dose intake. Most DMAsSugarMethoxy was translated into other arsenic species and then was together excreted out via mouse feces; two dominant arsenic species, arsenosugar DMAsSugarMethoxy and DMAsSugarPhosphate, were not detected in mouse muscle/organs after 20-Day or 30-Day feeding whether in lower-dose laver groups containing 1/36 (mass ratio) of the laver in mouse feed or higher-dose laver groups contain-ing 1/6 (mass ratio) of the laver in mouse feed. About 65-77% of total arsenic digested by mouse was excreted out via feces; only 0.12-0.78% of it was accumulated in mouse organs/muscle. The results of this study provided valuable knowledge for comprehending the stability and metabolic characteristics of different arsenic species from Fujian laver in vivo, also for more scientifically assessing the intake risk of arsenic in laver.
