Abstract :
Hybrid Pennisetum, although a high-quality silage feedstock, faces utilisation challenges owing to its high fibre content. This study employed an innovative approach by inoculating white rot fungi prior to aerobic exposure of hybrid Pennisetum silage, aiming to enhance resource utilisation through efficient lignocellulose degradation. Instead of autoclave-based sterilisation before fungal inoculation, the experiment utilised ensiling treatments with either Lactiplantibacillus plantarum (LP) or non-inoculated control (CK) using fresh hybrid Pennisetum as the substrate. After 90 d of ensiling, Pleurotus ostreatus (CKPo, LPPo) and Irpex lacteus (CKIl, LPIl) were inoculated into the silage for solid-state fermentation. Among the treatments, CKIl achieved the highest degradation rates of neutral detergent fibre, acid detergent fibre, and acid detergent lignin with reductions of 5.42 %, 7.00 %, and 15.59 %, respectively. Furthermore, I. lacteus inoculation reduced eukaryotic richness, and the relative abundance of the dominant fungus Irpex in CKIl was higher than that in LPIl (85.32 % vs. 68.07 %). These findings suggest that I. lacteus fermentation not only enhances fibre degradation but also improves the eukaryotic community structure, thereby significantly increasing the nutritional value of hybrid Pennisetum as feed and offering a sustainable strategy for silage production.

Abstract :
本发明提供了一种快速检测鉴定巨菌草酵母双杂交筛库阳性克隆的PCR方法。所述方法需提前对酵母菌落进行破壁,使用无菌枪头挑取单克隆菌落于20μL NAOH溶液(0.2M)中吹打混匀,99℃均质破壁10min,以破壁后的上清液为模板,采用上游引物F:5'‑CTATTCGATGATGAAGATACCCC‑3'和下游引物R:5'‑GTGAACTTGCGGGGTTTTTCAG‑3',进行PCR反应;PCR反应循环设置为:95℃预变性3min;95℃变性30s,57℃退火30s,72℃延伸30s,35个循环;72℃延伸10min,反应结束。本发明具有快速、成功率高、价格低廉等特点,为巨菌草酵母双杂交筛库阳性克隆的检测和鉴定提供了方法依据,同时也为其他物种酵母克隆的鉴定提供了参考,对解决小众植物、多倍体植物的酵母克隆鉴定问题具有重要意义。

为探究植物乳杆菌、布氏乳杆菌对杂交狼尾草(Pennisetum glaucum×purpureum)青贮发酵品质及有氧稳定性的影响,本试验以杂交狼尾草为试验材料,设置对照组(CK组,无添加)、植物乳杆菌(Lactiplantibacillus plantarum)组(LP组,1×10~6 cfu·g~(-1) FW)、布氏乳杆菌(Lactobacillus buchneri)组(LB组,1×10~6 cfu·g~(-1)FW)、植物乳杆菌和布氏乳杆菌组(LPLB组,5×10~5 cfu·g~(-1) FW LP+5×10~5 cfu·g~(-1) FW LB),厌氧发酵60天(d)后,测定其营养成分、发酵品质和有氧稳定性。结果表明:与CK组相比,各处理组粗蛋白(CP)、乳酸(LA)和乙酸(AA)含量显著增加(P<0.05),pH值和丁酸(BA)含量显著下降(P<0.05);在有氧暴露阶段,各处理组乳酸(LA)含量显著增加(P<0.05),氨态氮含量显著降低(P<0.05),LB和LPLB组在有氧暴露9天时,酵母菌和好氧菌数量(P<0.05)显著降低,乳酸菌数量显著增加(P<0.05);用隶属函数法进行综合价值评定,LPLB组青贮效果最佳。

本试验旨在研究益生菌添加剂对青贮紫花苜蓿和杂交狼尾草以及二者混合青贮营养品质、发酵品质的影响。试验设置3种青贮:紫花苜蓿单独青贮(M青贮)、杂交狼尾草单独青贮(P青贮)和紫花苜蓿与杂交狼尾草重量比为7∶3的混合青贮(MP青贮);每种青贮处理再分别设4个组:对照组(无接种,CK组)、接种1.0×10~6CFU/g枯草芽孢杆菌组(BS组)、接种1.0×10~6CFU/g布氏乳杆菌组(LB组)和接种0.5×10~6CFU/g枯草芽孢杆菌+0.5×10~6CFU/g布氏乳杆菌组(BSLB组)。发酵60 d后,分析青贮营养品质、发酵品质,并使用V-score评分体系对发酵效果进行评价。结果表明:M青贮中粗蛋白质(CP)、乳酸(LA)含量显著高于MP和P青贮(P<0.05);P青贮中干物质(DM)含量显著高于M和MP青贮(P<0.05),且pH和丁酸(BA)含量显著低于M和MP青贮(P<0.05)。MP青贮中CP、乙酸(AA)和PA含量显著高于P青贮(P<0.05),中性洗涤纤维(NDF)和酸性洗涤纤维(ADF)含量显著低于P青贮(P<0.05)。3种青贮中,与CK组相比,BS、LB和BSLB组的AA含量显著提高(P<0.05),pH和氨态氮/总氮(NH_3-N/TN)显著降低(P<0.05)。接种益生菌添加剂可抑制青贮有氧暴露时的二次发酵,降低pH增速并抑制酵母菌和好氧细菌的繁殖。除MP青贮中CK、BS组,各组的V-score评分均在80分以上,属于优等级;其中,P青贮中BSLB组的V-score评分最高(98.27分)。综上所述,在杂交狼尾草青贮中同时接种布氏乳杆菌和枯草芽孢杆菌时青贮效果最佳,最具有推广意义。

Abstract :
Elephant grass is widely used in feed production and ecological restoration because of its huge biomass and low occurrence of diseases and insect pets. However, drought seriously affects growth and development of this grass. Strigolactone (SL), a small molecular phytohormone, reportedly participates in improving resilience to cope with arid environment. But the mechanism of SL regulating elephant grass to response to drought stress remains unknown and needs further investigation. We conducted RNA-seq experiments and identified 84,296 genes including 765 and 2325 upregulated differential expression genes (DEGs) and 622 and 1826 downregulated DEGs, compared drought rehydration with spraying SL in roots and leaves, respectively. Combined with targeted phytohormones metabolite analysis, five hormones including 6-BA, ABA, MeSA, NAA, and JA had significant changes under re-watering and spraying SL stages. Moreover, a total of 17 co-expression modules were identified, of which eight modules had the most significant correlation with all physiological indicators with weighted gene co-expression network analysis. The venn analysis revealed the common genes between Kyoto Encyclopedia of Genes and Genomes enriched functional DEGs and the top 30 hub genes of higher weights in eight modules, respectively. Finally, 44 DEGs had been identified as key genes which played a major role in SL response to drought stress. After verification of its expression level by qPCR, six key genes in elephant grass including PpPEPCK, PpRuBPC, PpPGK, PpGAPDH, PpFBA, and PpSBPase genes regulated photosynthetic capacity under the SL treatment to respond to drought stress. Meanwhile, PpACAT, PpMFP2, PpAGT2, PpIVD, PpMCCA, and PpMCCB regulated root development and phytohormone crosstalk to respond to water deficit conditions. Our research led to a more comprehensive understanding about exogenous SL that plays a role in elephant grass response to drought stress and revealed insights into the SL regulating molecular mechanism in plants to adapt to the arid environment.

本试验旨在研究不同浓度丙酸对杂交狼尾草青贮营养品质、发酵品质及有氧暴露后发酵指标和微生物数量的影响,并筛选出适宜的丙酸浓度再进行微生物多样性分析。各组杂交狼尾草青贮中丙酸浓度分别为0(对照组,CK组)、0.1%(T_1组)、0.5%(T_2组)和1.0%(T_3组)。青贮60 d后测定杂交狼尾草青贮营养品质、发酵品质、有氧暴露后发酵指标及微生物数量,参考各指标和隶属函数法筛选出适宜浓度丙酸组(PA组),分析其微生物多样性。结果表明:1)青贮60 d后,与CK组相比,T_1、T_2、T_3组的干物质(DM)、粗蛋白质(CP)含量显著增加(P<0.05),T_2、T_3组的水溶性碳水化合物(WSC)含量显著增加(P <0.05), T_1、 T_2组的中性洗涤纤维(NDF)、酸性洗涤纤维(ADF)含量显著降低(P<0.05),T_2、T_3组的乳酸(LA)含量显著升高(P<0.05),T_2、T_3组的pH和乙酸(AA)、丁酸(BA)含量及氨态氮/总氮显著降低(P<0.05)。2)在有氧暴露期间,与CK组相比,T_2、T_3组有氧暴露0、3、6、9 d的pH显著降低(P<0.05),T_1、T_2、T_3组有氧暴露0、3、9 d的LA含量显著升高(P<0.05),T_1、T_2、T_3组有氧暴露0、3、6、9 d的氨态氮含量显著降低(P<0.05),T_2、T_3组有氧暴露6、9 d的乳酸菌(LAB)数量显著降低(P<0.05),T_2、T_3组有氧暴露0、3、6、9 d的酵母菌数量显著降低(P<0.05),T_2、T_3组有氧暴露6、9 d的好氧菌(AB)数量显著降低(P<0.05)。3) T_2组的综合价值评定值最高。4)在门水平上,杂交狼尾草青贮的优势菌门为厚壁菌门(Firmicutes)和变形菌门(Proteobacteria);在属水平上,杂交狼尾草青贮的优势菌属为乳杆菌属(Lactobacillus)和代尔夫特菌属(Delftia)。与CK组相比,PA组的厚壁菌门相对丰度降低,变形菌门相对丰度升高;PA组的乳杆菌属、魏斯氏菌属(Weissella)和肠杆菌属(Enterobacter)相对丰度降低,代尔夫特菌属相对丰度提高。综上所述,添加适宜浓度丙酸可以提高杂交狼尾草青贮的营养价值,改善发酵品质、有氧稳定性和微生物的群落组成,其中添加0.5%丙酸效果最好。

Abstract :
旨在研究布氏乳杆菌和不同糖类联用对紫花苜蓿青贮营养成分、发酵品质、瘤胃降解率以及有氧稳定性的影响。以紫花苜蓿为原料,采用了4种处理:LB(添加1×10~6 cfu·g~(-1)布氏乳杆菌);GLB(添加1×10~6 cfu·g~(-1)布氏乳杆菌和2%葡萄糖);FLB(添加1×10~6 cfu·g~(-1)布氏乳杆菌和2%岩藻多糖);CK(无添加)。青贮60 d后,测定以上4个处理紫花苜蓿的营养成分、发酵品质、瘤胃降解率和有氧稳定性。结果表明,在营养成分上,与CK相比各处理均显著提高了紫花苜蓿青贮料的干物质(DM)、粗蛋白(CP)和可溶性碳水化合物(WSC)含量(P<0.05),其中GLB处理DM和CP含量最高,FLB处理WSC含量最高。各处理均能显著降低酸性洗涤纤维(ADF)含量(P<0.05),GLB处理能显著降低青贮料中性洗涤纤维(NDF)含量(P<0.05)。发酵品质上,除FLB处理外其余处理均能显著降低青贮料pH(P<0.05),GLB处理显著增加了乳酸(LA)和丙酸(PA)含量(P<0.05),显著降低了氨态氮/总氮(AN/TN, P<0.05)。LB和FLB处理LA、乙酸(AA)含量和AN/TN显著降低(P<0.05),PA含量显著增加(P<0.05)。在瘤胃降解方面,除GLB处理的ADF有效降解率无显著提高外(P>0.05),各处理DM、CP、ADF和NDF有效降解率均显著提升(P<0.05)。与CK相比,各处理均能在有氧暴露6 d显著降低pH、酵母菌和好氧菌数(P<0.05),其中FLB处理表现最佳。根据隶属函数法综合评价各项指标,各处理按优劣排序为:GLB(0.76)>LB(0.53)>FLB(0.52)>CK(0.14)。综上所述,LB、GLB和FLB处理均对紫花苜蓿青贮营养成分、发酵品质、瘤胃降解率及有氧稳定性有积极影响,其中GLB处理效果最好。

为研究添加剂甲酸对紫花苜蓿(Medicago sativa L.)青贮品质、有氧暴露后微生物数量以及瘤胃降解特性的影响,分别在新鲜紫花苜蓿中添加0(CK),4(FA1),6(FA2),8 mL·kg~(-1)(FA3)甲酸后,室温青贮60 d。通过隶属函数法比较青贮品质,筛选出青贮效果最佳的处理,测定其有氧暴露后微生物数量及瘤胃降解率。结果表明:与CK组相比,添加不同水平甲酸均能显著提高紫花苜蓿干物质(Dry matter, DM)、粗蛋白(Crude protein, CP)、可溶性碳水化合物及乳酸含量(P<0.05),显著降低中性洗涤纤维(Neutral detergent fiber, NDF)及酸性洗涤纤维(Acid detergent fiber, ADF)、pH值、氨态氮比总氮(P<0.05)。通过隶属函数法筛选出FA2处理总隶属度最高。FA2组在有氧暴露后第0~6天的pH值、酵母菌和好氧菌计数显著低于CK组(P<0.05)。瘤胃降解特性中,FA2组DM,CP,NDF及ADF有效降解率较CK组显著提高(P<0.05)。综上,添加6 mL·kg~(-1)甲酸青贮效果最佳。

The aim of this study was to compare the effect of different additives on nutritional quality, fermentation variables and microbial diversity of hybrid Pennisetum silages. A control (CK - no additives) and seven treatments were tested, namely, Lactiplantibacillus plantarum (LP), Lentilactobacillus buchneri (LB), propionic acid (PA), calcium propionate (CAP), LP + LB; LP + PA and LP + CAP. In comparison with CK, all treatments increased the contents of crude protein and lactic acid, decreased the content of butyric acid, and altered the bacterial communities of the silage. Except for the CAP and LP + CAP treatments, the additives decreased pH and the ammonia nitrogen:total nitrogen (NH3-N:TN) ratio. The results of principal component analysis revealed that the PA, LP + PA and LP + LB treatments ranked as the top three silages. The PA and LP + PA treatments exhibited higher water-soluble carbohydrate content, but lower pH, and NH3-N:TN ratio than the other treatments. With the PA and LP + PA treatments, the relative abundances of Lactobacillus and Enterobacter decreased, and of Proteobacteria and Delftia increased, while the carbohydrate metabolism of the microorganisms improved. The LP and LB treatments reduced the Shannon and Simpson diversities. In the beta diversity, PA and LP + PA separated from the other treatments, indicating that there were differences in the composition of bacterial species. The relative abundance of Lactobacillus increased in the LP and LB treatments and of Leucanostoc and Weissella increased in the CAP and LP + CAP treatments. In summary, the addition of L. plantarum, L. buchneri, propionic acid, calcium propionate, and their combinations improved fermentation quality, inhibited harmful bacteria and conserved the nutrients of hybrid Pennisetum.

Abstract :
采用生物信息学方法鉴定了巨菌草(Cenchrus fungigraminus Z. X. Lin&D.M. Lin&S. R. Lan sp. nov.)中CAMTA(calmodulin-binding transcription activator)转录因子的家族成员,并对其蛋白序列的理化性质、基因结构、保守基序、保守功能结构域、系统进化发育关系、启动子区域顺式作用元件进行了分析;探讨了巨菌草CAMTA((CfuCAMTA))家族成员在干旱胁迫下转录水平的变化,以及CfuCAMTA转录因子对响应干旱胁迫的影响.结果表明:巨菌草CAMTA家族共有15个成员,其中仅CfuCAMTA10和CfuCAMTA11较为稳定,不稳定系数小于40;多数成员为不稳定蛋白质;巨菌草和紫象草CAMTA基因的亲缘关系最近;4个CAMTA转录因子的表达水平随干旱时间的延长,在第14天表达量达到最大,说明这些CAMTA参与了干旱胁迫的响应.
