Improvement of Quality of Sour Camel Milk by Extract of Sparassis crispa: Physicochemical Properties, Sensory Quality and Metabolic Changes SCIE

Abstract :

Sour camel milk, as a nutritious fermented dairy product, faces challenges in terms of quality stability. Sparassis crispa, due to its antioxidant and antibacterial properties, shows potential in improving food quality. This study aimed to investigate the effects of different active components of Sparassis crispa on the quality of sour camel milk. The results indicated that Component I was the most effective Sparassis crispa component in enhancing the quality of sour camel milk. The components of Component I were identified as LysoPC(0_0_18_2(9Z,12Z)), LysoPC(18_1(11Z)_0_0), and N-(2-hydroxymethyl-3-chloro-4-hydroxyphenyl) anthranilic acid, among others. It increased the total viable count of lactic acid bacteria (LAB) and water-holding capacity (WHC) while improving the texture of sour camel milk. Metabolomics analysis revealed that the first component of sour camel milk (FCS) and Sparassis crispa sour camel milk (SS) have a high degree of similarity in the composition of flavor substances. The characteristic flavor metabolites included 2-amylfuran, isoamyl alcohol, 2-methylbutyraldehyde, and 2-ethyl-1-hexanol. Additionally, the supplementation of Component I increased the levels of metabolites such as amino acids, free fatty acids, organic acids, and carbohydrates, thereby contributing to the enhanced taste and nutritional quality of sour camel milk. This intervention also strengthened carbohydrate and amino acid metabolism in LAB. These findings provide a theoretical basis for utilizing Component I to improve the quality of sour camel milk.

菌草灵芝醇提物对免疫损伤小鼠的改善作用

Abstract :

菌草灵芝是用菌草代替木屑栽培出的灵芝,有多种生物活性,已被证实有较强的体外免疫作用。环磷酰胺作为烷化剂类抗肿瘤药物,也是典型的免疫抑制剂,会造成机体免疫损伤。本研究以环磷酰胺诱导小鼠免疫低下模型,灌胃100 mg/(kg·d)的菌草灵芝醇提物(JGEH),研究其对小鼠免疫损伤的改善情况。结果:JGEH可显著(P<0.05)提高小鼠体质量(6.32%)、脏器指数(胸腺38.50%、脾脏71.32%、肝脏1.27%和肾脏指数11.48%)、碳廓清能力(41.50%),迟发型变态反应程度(59.50%)。此外,JGEH还能显著(P<0.05)提高小鼠血清中IgG(27.73%)、IgA(26.51%)、IgM(23.41%)和IL-10(41.98%)等免疫球蛋白和抗炎因子含量,降低TNF-α(17.40%)、IFN-γ(17.49%)和IL-6(42.33%)等炎症细胞因子含量,显著提高(P<0.05)全血中白细胞(41.23%)、红细胞(18.91%)、血红蛋白(42.32%)、血小板含量(40.29%)及淋巴细胞百分比(159.94%),为菌草灵芝醇提物的发展提供研究思路。

Abstract :

Juncao Ganoderma lucidum, which is cultivated with Juncao instead of wood chips, has a wide ränge of bioactivities and has been shown to have a strong in vitro immune effect. Cyclophosphamide, as an alkylating antitumor agent, is also a typical immunosuppressant that causes immune damage to the body. In the present study, cyclophosphamide-induced immunocompromise model in mice was investigated by gavage of 100 mg/(kg-d) Juncao Ganoderma lucidum alcohol to study the improvement of immune injury in mice. Abstract :

灰树花多肽-锌螯合物(Grifola frondosa Polypeptide Zinc Chelate,GPs-Zn)是灰树花多肽(Grifola frondosa Polypeptide,GPs)和锌离子的螯合产物,利用电镜扫描、X-射线粉末衍射技术、红外光谱和紫外光谱等技术对GPs-Zn进行结构分析;通过构建ICR母鼠缺锌模型,进一步探究GPs-Zn对孕期缺锌鼠后代的影响。表征结果表明,GPs和锌离子螯合成一种新的物质;红外光谱对比图发现肽链上的氨基和羧基都参与了锌离子的配位反应;紫外光谱图发现,GPs-Zn中多肽的羰基原子和锌离子结合,诱导紫外光谱的红移,表明GPs与锌离子结合形成GPs-Zn。动物实验表明:GPs-Zn可将缺锌仔鼠胸腺指数提高78.69%(雌鼠)和87.55%(雄鼠);脾脏指数提高40.28%(雌鼠)和43.22%(雄鼠);体质量提高89.98%(雌鼠)和88.30%(雄鼠);且血清的超氧化物歧化酶(SOD)活力提高了108.07%(雌鼠)和26.16%(雄鼠);锌浓度提高14.74%(雌鼠)和29.33%(雄鼠);碱性磷酸酶(AKP)水平降低52.28%(雌鼠)和62.48%(雄鼠)。综上可知,GPs-Zn对孕期缺锌鼠后代仔鼠的胸腺指数、脾脏指数、超氧化物歧化酶活力、锌浓度和碱性磷酸酶水平有一定改善作用。

Positive effect of peptide-calcium chelates from Grifola frondosa on a mouse model of senile osteoporosis SCIE

Abstract :

Calcium supplementation has been shown to be efficacious in mitigating the progression of senile osteoporosis (SOP) and reducing the incidence of osteoporotic fractures resulting from prolonged calcium shortage. In this study, Grifola frondosa (GF) peptides-calcium chelate were synthesized through the interaction between peptide from GF and CaCl2. The chelation reaction was shown to involve the participation of the amino and carboxyl groups in the peptide, as revealed by scanning electron microscope, Fourier-transform infrared, and ultraviolet spectrophotometry. Furthermore, a mouse model of (SOP) induced by d-galactose was established (SCXK-2018-0004). Results demonstrated that low dosage of low-molecular weight GF peptides-calcium chelates (LLgps-Ca) could significantly improve serum index and pathological features of bone tissue and reduce bone injury. Further research suggested that LLgps-Ca could ameliorate SOP by modulating the disrupted metabolic pathway, which includes focal adhesion, extracellular matrix receptor interaction, and PI3K-Akt signaling pathway. Using Western blot, the differentially expressed proteins were further confirmed. Thus, calciumchelating peptides from GF could serve as functional calcium agents to alleviate SOP.

银耳多糖对益生菌发酵驼乳改善小鼠免疫和肠道黏膜屏障功能的影响

目的:研究银耳多糖对益生菌发酵驼乳改善环磷酰胺(cyclophosphamide,CTX)诱导的免疫抑制小鼠免疫和肠道黏膜屏障功能的影响。方法:测定益生菌发酵驼乳和银耳益生菌发酵驼乳的理化指标和抗氧化活性。BALB/C小鼠随机分为正常组(NC组)、模型组(MD组)、益生菌发酵驼乳组(FCM组)、银耳益生菌发酵驼乳组(FYCM组),每天分别灌胃0.2 mL生理盐水、生理盐水、益生菌发酵驼乳、银耳益生菌发酵驼乳,干预周期为30 d;最后4 d,除NC组之外,其余各组每天腹腔注射60 mg/kg m_b CTX构建免疫抑制模型。对小鼠脏器指数、脾淋巴细胞增殖活性、迟发型变态反应、细胞因子和免疫球蛋白含量、脾脏和空肠组织形态学进行分析。结果:低温贮藏21 d,FYCM的pH值从4.52缓慢下降到4.30,滴定酸度从95.7°T逐步上升到124.73°T,乳酸菌数量从9.40×10~9 CFU/mL下降到8.38×10~9 CFU/mL,持水力从39.23%下降到35.13%。与益生菌发酵驼乳相比,银耳益生菌发酵驼乳的抗氧化能力极显著提高(P<0.01)。小鼠实验表明,与MD组相比,FYCM可显著提高免疫抑制小鼠的脏器指数、脾淋巴细胞增殖率和迟发性超敏反应能力,上调白细胞介素-6、干扰素-γ、免疫球蛋白G、免疫球蛋白M、肠道分泌型免疫球蛋白A水平,修复脾脏和空肠黏膜结构,且其效果显著优于FCM。结论:银耳益生菌发酵驼乳具有调节免疫和肠道黏膜屏障功能的潜力,在功能性乳品市场具有广阔的应用前景。

灵芝是我国著名的药食两用真菌,含有多糖、三萜类化合物、蛋白质、多肽、甾醇等多种活性物质,具有抗肿瘤、抗菌、抗动脉粥样硬化、降血糖、降血脂、抗衰老、抗真菌、抗病毒和增强免疫等功效。灵芝一直作为预防、控制和治疗慢性疾病的药物或营养补充剂,其精深加工产品涉及医药、保健、食品、美妆等领域,但目前我国灵芝精深加工仍存在栽培人员技术水平低、技术研发与转化不足、产品质量参差不齐等问题。文中综述了灵芝精深加工的现状及存在问题,提出了今后的发展策略,对开发利用我国丰富的灵芝资源、促进灵芝产业发展、提高人民健康水平,具有重要意义。

Abstract :

The hypoglycemic effects of low-molecular-weight Laminaria japonica polysaccharide (LJO) were investigated in type 2 diabetes mellitus (T2DM) mice, focusing on its effect on the microbiome, metabolome, and transcriptome. The findings demonstrated that LJO significantly reduced fasting blood glucose levels, insulin levels, and inflammatory factors. Additionally, LJO induced changes in gut microbiota composition and increased the concentrations of cecal short-chain fatty acids. Analysis of transcriptomics and metabolomics data revealed that LJO primarily altered the endocrine and digestive systems, signal transduction, and lipid metabolism. It led to a decrease in palmitic acid levels and an increase in glutathione levels. Real-time quantitative polymerase chain reaction assay suggested that LJO upregulated Irs1 expression, consequently reducing insulin resistance. These findings strongly suggest that LJO holds promise in ameliorating T2DM and may serve as a potential dietary supplement for patients with T2DM.
