Abstract :
了解马铃薯根际土壤中芽胞杆菌种类多样性,可为挖掘芽胞杆菌新资源提供基础。从新疆伊犁州9个地点采集了30份马铃薯根际土壤样品,采用可培养法从中分离芽胞杆菌,通过16S rRNA基因同源性鉴定了分离菌株的分类地位。共获得芽胞杆菌349株,基于菌落形态特征和16S rRNA基因序列确定了66个代表菌株,其中14株与近缘种模式菌株的16S rRNA基因序列相似性介于93.0–98.5%之间,为潜在芽胞杆菌新种。349株芽胞杆菌属于9属66种,分别为芽胞杆菌属(Bacillus)的41个种(41/66,62.1%),为最优势属;类芽胞杆菌属(Paenibacillus)有9种(9/66,13.6%),赖氨酸芽胞杆菌属(Lysinibacillus)共6种(6/66,9.1%),嗜冷芽胞杆菌属(Pscychrobacillus)共4种(4/66,6.1%),虚构芽胞杆菌属(Fictibacillus)有2种(2/66,3.0%),短芽胞杆菌属(Brevibacillus)、大洋芽胞杆菌属(Oceanobacillus)、鲁梅尔芽胞杆菌属(Rummelibacillus)和解硫胺素芽胞杆菌属(Aneurinibacillus)皆为1种。新疆9个地点马铃薯根际土壤中芽胞杆菌含量为2.20–8.86×104 cfu/g,其中特克斯蒙古乡马铃薯根际土壤中芽胞杆菌的菌落含量和种类最多,分别为8.86×104 cfu/g和37种;尼勒克县马场马铃薯根际土壤中的芽胞杆菌含量最少,特克斯去昭苏路上和阜康西沟村的芽胞杆菌种类最少,仅9种。但每个地点的优势种均相同,均为简单芽胞杆菌(Bacillus simplex)和阿氏芽胞杆菌(B.aryabhattai)。芽胞杆菌种类可划分为高频度分布型(简单芽胞杆菌等4种)、中频度分布型(植物内生芽胞杆菌等12种)和低频度分布型(其余51种芽胞杆菌,如蕈状芽胞杆菌)。新疆9个地点芽胞杆菌种类分布可分为高含量高丰度型(特克斯蒙古乡和农技推广园)和低含量低丰度型(如尼勒克县马场等地)。相关性分析发现,马铃薯根际芽胞杆菌种类分布与海拔高度无显著相关性。新疆马铃薯根际土壤蕴含种类丰富的芽胞杆菌资源,可为芽胞杆菌功能菌株开发提供丰富的菌种来源。

Abstract :
Ralstonia solanacearum causes bacterial wilt disease in a wide range of host plants, including tomato. To elucidate the invasion pattern of R. solanacearum, one avirulent strain, FJAT-1458, and one virulent strain, FJAT-91, were characterized according to their colonization and interaction with host in tomato roots. Both strains colonized in tomato roots. The highest colonization numbers were 1.66 x 10(8) cfu g(-1) by FJAT-91 at 5 days (d) after inoculation and 1.09 x 10(8) cfu g(-1) by FJAT-1458 at 6 d after inoculation. Infection with FJAT-91 caused tomato plant wilt with a disease index of 23.65% at 3 d after inoculation and 100% at 6 d after inoculation, and infection with FJAT-1458 did not cause plant wilt. Compared with FJAT-1458, infection with FJAT-91 reduced elongation of tomato roots, induced serious browning, and overflowed bacteria during the late stages of infection. Examination of cellular structure showed that infection with FJAT-1458 did not have obvious destructive effects on plant cells, while FJAT-91 induced a series of cytopathological changes, including swelling of mitochondria, degeneration of cytoplasm and nuclear heterochromatin, and collapse of host cells, which eventually resulted in the death of the host plant. The cytopathological changes appeared from the second to the fourth disease stages.

【目的】研究在茶园中合理间作绿肥对茶园节肢动物群落结构和多样性的影响。【方法】本文在茶园中分别设间作铺地木兰+罗顿豆、圆叶决明+白三叶、白三叶+平托花生3种不同绿肥组合处理,并设不间作绿肥、常规除草的茶园为对照。【结果】间作不同绿肥均能提高茶园节肢动物群落的物种丰富度和群落多样性,提高茶园捕食性天敌昆虫、蜘蛛和寄生性天敌在茶园冠层群落的比率;不同处理茶园冠层节肢动物Shannon-wiener多样性指数(H)、Simpson指数(D)和均匀度指数变化趋势基本一致;茶冠层的节肢动物群落多样性指数在4类不同处理茶园间差异显著;间作绿肥增加了茶园的生物多样性,4类不同处理茶园茶冠层群落相似性系数在0.741~0.892之间,表明4类不同处理茶园冠层群落在物种组成上有较高的相似性。4类不同处理茶园害虫与天敌两类群功能团的丰盛度之间成显著的正相关,表明天敌对害虫的空间数量跟随效应强,主要表现为二者数量间的相互作用。灰色关联度分析表明在4类不同处理茶园中的群落节肢动物物种数/个体数、天敌物种数/害虫物种数、害虫丰富度与多样性指数和均匀度指数的关联系数较高,反应了群落的多样性和稳定性。【结论】间作增强了茶园群落的生物多样性,提高害虫天敌的种类和个体数,有利有效发挥天敌对有害生物的生态控制。

在福建隽永天香茶业公司茶洋有机茶基地研究了茶园间作不同绿肥对假眼小绿叶蝉及其天敌缨小蜂的影响。结果表明,不同间作处理茶园假眼小绿叶蝉虫口数量差异显著(F=3.15,P<0.05),缨小蜂虫口数量差异极显著(F=4.22,P<0.01)。间作铺地木蓝+罗顿豆茶园,假眼小绿叶蝉虫口数量最高1035头,间作铺地木蓝+猪屎豆茶园,假眼小绿叶蝉的虫口数量最低378头;间作白三叶草+圆叶决明茶园,叶蝉天敌缨小蜂的虫口数量最高263头,缨小蜂虫口数量比空白对照提高了61.98%,茶园间作绿肥显著提高了天敌缨小蜂的种群数量。不同间作处理茶园假眼小绿叶蝉与天敌缨小蜂均呈显著的正相关。间作白三叶草+圆叶决明,茶园平均益害比最高为0.56,对照茶园最低为0.18。

Ralstonia solanacearum is one of the most destructive plant bacterial pathogens worldwide. The population dynamics and genetic stability are important issues, especially when an avirulent strain is used for biocontrol. In this study, we developed a rapid method to differentiate the virulent and avirulent strains of R. solanacearum and to predict the biocontrol efficiency of an avirulent strain using high performance liquid chromatography (HPLC). Three chromatographic peaks P-1, P-2 and P-3 were observed on the HPLC spectra among 68 avirulent and 28 virulent R. solanacearum strains. Based on the HPLC peaks, 96 strains total were assigned to three categories. For avirulent strains, the intense peak is P1, while for virulent strains, P3 is the majority. Based on the HLPC spectra of R. solanacearum strains, a chromatography titer index (CTI) was established as CTi; = S-i;/(S-1+S-2+S-3) x 100% (i represents an individual HPLC peak; S-1, S-2 and S-3 represent peak areas of P-1, P-2 and P-3, respectively). The avirulent strains had high values of C-Tl-1 ranging from 63.6 to 100.0%, while the virulent strains displayed high values of CTI3 ranging from 90.2 to 100.0%. Biological inoculation studies of 68 avirulent strains revealed that the biocontrol efficacy was the best when CTI1 =100%. The purity and genetic stability of R. solanacearum strains were confirmed in the P-1 fraction of avirulent strain FJAT-1957 and P-3 fraction of virulent strain FJAT-1925 after 30 generations of consecutive subculture. These results confirmed that fractioning by HPLC and their deduced CTI can be used for rapid and efficient evaluation and prediction of an isolate of R. solanacearum. To the best of our knowledge, this is the first report that HPLC fractioning can be used for rapid differentiation of virulent and avirulent strains of R. solanacearum. (C) 2015 Elsevier Ltd. Abstract :
A novel Gram-stain-positive, endospore-forming bacterium, designated FJAT-22460(T), was isolated from a soil sample of a potato field in Xinjiang Autonomous Region, China. Cells were rods that were catalase-positive and motile by peritrichous flagella. The strain was found to grow at temperatures ranging from 10 to 40 degrees C (optimum 30 degrees C) and at pH 5.0-12.0 (optimum pH 7) with 0-5% (w/v) NaCl (optimum 0% NaCl). Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain FJAT-22460(T) belonged to the genus Paenibacillus and exhibited 16S rRNA gene sequence similarities of 97.3, 97.2, 97.2 and 97.0% with Paenibacillus glucanolyticus DSM 5162(T),Paenibacillus lautus DSM 3035(T), Paenibacillus lactis MB 1871(T) and Paenibacillus chibensis JCM 9905(T), respectively. DNA-DNA relatedness of strain FJAT-22460(T) with Paenibacillus glucanolyticus DSM 5162(T) and Paenibacillus lautus DSM 3035(T) was 62.6% and 33.3%, respectively, lower than the 70% accepted for species delineation. The menaquinone was identified as MK-7. The major fatty acids detected were anteiso-C-15:0 (51.4 %), iso-C-15 : 0 (5.3 %), C-16 : 0 (12.1 %), iso-C-16 : 0 (10.7 %) and anteiso-C-17 : 0 (6.9 %). The DNA G+C content was determined to be 50.9 mol%. Phenotypic, chemotaxonomic and genotypic properties clearly indicated that isolate FJAT-22460(T) represents a novel species within the genus Paenibacillus, for which the name Paenibacillus solani sp. nov. is proposed. The type strain is FJAT-22460T (=DSM 100999(T) =CCTCC AB 2015207(T)).

Seven species of Cosmocomoidea Howard (Hymenoptera: Mymaridae) from China are keyed and illustrated. The two new species are C. rugosa Aishan, Triapitsyn & Xu sp. n. (Guangdong and Yunnan) and C. tenuis Xu, Lin & Hu sp. n. (Fujian). Three species are newly recorded from China, C. atra (Foerster) s.l., C. kikimora (Triapitsyn), and C. oxypygus (Foerster).

对鸭脚树星室木虱的卵和各龄期若虫的形态特征进行了扫描电镜观察和描述,测量了各龄期虫体的体长和体宽.1龄若虫体长为0.22-0.35 mm;2龄若虫体长0.35-0.60 mm,身体长椭圆形,腹部分节明显;3龄若虫体长0.51-0.83 mm,身体扁平,腹部分节不明显,沿身体一周着生截状蜡腺毛;4龄若虫体长1.0-1.5 mm,身体扁平,腹部分节明显,翅芽明显;5龄若虫体长1.2-2.0 mm,身体肥胖柔软.
